Context: The effects of leptin, an adipocyte-derived hormone that signals overall energy sufficiency, can only be studied in leptin-deficient conditions. In patients with lipodystrophy, a rare disease and unique model of leptin deficiency, treatment with recombinant leptin (metreleptin) improves glycemia and decreases energy expenditure. We hypothesized that these improvements might be mediated by reduced gluconeogenesis (GNG), an energy-requiring process.
Objective: To determine the effects of metreleptin on GNG and GNG substrates.
Methods: This was a single-arm prospective study of metreleptin administration in 15 patients with lipodystrophy, 9 of whom had data on GNG (NIH, 2013-2018). We analyzed total GNG, insulin-mediated …

The metazoan-specific Integrator complex catalyzes 3' end processing of small nuclear RNAs (snRNAs) and premature termination that attenuates the transcription of many protein-coding genes. Integrator has RNA endonuclease and protein phosphatase activities, but it remains unclear if both are required for complex function. Here, we show IntS6 (Integrator subunit 6) over-expression blocks Integrator function at a subset of Drosophila protein-coding genes, although having no effect on snRNAs or attenuation of other loci. Over-expressed IntS6 titrates protein phosphatase 2A (PP2A) subunits, thereby only affecting gene loci where phosphatase activity is necessary for Integrator function. Mental health can be interpreted as a social taboo in Asia, ensuring that students with mental health stigmas (SWMHS) face complex educational journeys that impact their wellbeing. This article provides a conceptual interdisciplinary commentary that illustrates how in Asian higher education (HE) settings, the psychosocial phenomena of face culture, a sociolinguistic blend of high-context power relations, sense-making, and cultural capital, defines human-to-human (HTH) dialogue. It suggests that human-to-computer-interaction (HCI) through artificial intelligence (AI) technologies, such as ChatGPT, could improve university wellbeing strategies in Asia. Immunosequencing has emerged as a newer clinical test for assessment of T-cell clonality in the blood and skin of cutaneous T-cell lymphoma (CTCL) patients. Utilization of immunosequencing, also known as high-throughput sequencing of the T-cell receptor (HTS-TCR), enables identification and quantification of the precise genetic signature of dominant T-cell clones. Although immunosequencing is more sensitive than commonly used methods such as polymerase chain reaction (PCR) paired with capillary electrophoresis or flow cytometry, it remains underutilized for CTCL management. The hedonic value of salt fundamentally changes depending on the internal state. High concentrations of salt induce innate aversion under sated states, whereas such aversive stimuli transform into appetitive ones under sodium depletion. Neural mechanisms underlying this state-dependent salt valence switch are poorly understood. Using transcriptomics state-to-cell-type mapping and neural manipulations, we show that positive and negative valences of salt are controlled by anatomically distinct neural circuits in the mammalian brain. The hindbrain interoceptive circuit regulates sodium-specific appetitive drive , whereas behavioral tolerance of aversive salts is encoded by a dedicated class of neurons in the forebrain lamina terminalis (LT) …

Results obtained from rat studies indicate that, even at low concentrations, mercurial species cause harmful effects on the kidneys, by inducing the nephrotic oxidative stress response. In the present work, Hg-associated proteins were identified as possible mercury-exposure biomarkers in rat kidneys exposed to low mercury chloride concentrations for 30 days (Hg-30) and 60 days (Hg-60), using metalloproteomic strategies. The renal proteomic profile was fractioned by two-dimensional electrophoresis and the mercury determinations in kidney samples, protein pellets and protein spots were performed using graphite furnace atomic absorption spectrometry. Artificial Intelligence For Mental Health Support During Covid-19: Experiences Of Graduate Counseling Students, Russell Fulmer, Eric T. Beeson, Carl Sheperis, Daniel Rosello, Rebecca Edelman

The purpose of this study was to examine how an AI chatbot could provide mental health support to counselors-in-training during the COVID-19 pandemic. The chatbot “Tess” was available to participants for two weeks. Participants responded to questions about their experience and the content of this qualitative data was analyzed. Themes emerged that focused on mental health during the pandemic, utility of the AI chatbot during the pandemic, and potential therapeutic use in general. Findings were mixed and suggest some skepticism among counseling students towards the use of an AI chatbot.

As artificial intelligence and robotic technologies accelerate economic transformation, outdated property and tax laws will increasingly fail American workers with ordinary skills that perform routine job functions. Because technology may render millions of workers redundant, U.S. policymakers must make significant social, economic, and legal structural changes to (1) improve the lives of average workers, (2) support the economy, and (3) maintain political stability.

Lapatinib, an oral tyrosine kinase inhibitor used as a first-line treatment for HER2-positive breast cancer, has been reported to be associated with hepatotoxicity; however, the underlying mechanisms remain unclear. In this study, we report that lapatinib causes cytotoxicity in multiple types of hepatic cells, including primary human hepatocytes, HepaRG cells, and HepG2 cells. A 24-h treatment with lapatinib induced cell cycle disturbances, apoptosis, and DNA damage, and decreased the protein levels of topoisomerase in HepG2 cells. The molecular basis of prostate cancer (PCa) progression from the primary disease to metastatic castration-resistant prostate cancer (CRPC) followed by therapy-induced neuroendocrine prostate cancer is not fully understood. In this study, we elucidate the role of miR-410, a little-studied microRNA located on chromosome 14q32.31 within the DLK1-DIO3 cluster, in PCa. miR-410 expression analyses in primary and metastatic PCa tissues and cell lines show that its levels are decreased in initial stages and increased in advanced PCa. Olanzapine is one of the most widely used antipsychotics since its initial approval by the US Food and Drug Administration in 1996 and has undergone extensive pharmacokinetic study. Despite being utilized in clinical psychiatry for decades, there remain questions regarding the variety of available formulations, the utility of therapeutic drug monitoring, altered kinetic properties in special populations/medical illnesses, the use of high-dose olanzapine, and drug interactions, among many others. Tuberculosis remains an international health threat partly because of limited protection from pulmonary tuberculosis provided by standard intradermal vaccination with Bacillus of Calmette and Guérin (BCG); this may reflect the inability of intradermal vaccination to optimally induce pulmonary immunity. In contrast, respiratory Mycobacterium tuberculosis infection usually results in the immune-mediated bacillary containment of latent tuberculosis infection (LTBI). Here we present RNA-Seq-based assessments of systemic and pulmonary immune cells from LTBI participants and recipients of intradermal and oral BCG. This study aims to confirm the effect of industrial education 4.0 on data, technology, and human literacy among postgraduate students in Indonesia. The research adopted covariance-based structural equation modeling by employing AMOS 25 to examine the relationship between variables. The model estimation was performed using confirmatory factor analysis as a standard model for measuring industrial education 4.0 and involved structural model analysis to confirm the hypotheses. Public libraries serve as repositories for a movement described as cultivation of the Library of Things. In the wake of COVID-19, the West Lafayette Public Library enhanced its existing Library of Things collection through the creation of science, technology, engineering, art, and mathematics (STEAM) kits. Since 2017, the West Lafayette Public Library has held regular free STEAM programs for the community; those programs were put on hold during the height of the COVID-19 pandemic, which concurred with a library renovation. The Wabash River is the most extensive river system in Indiana. The river drains two-thirds of Indiana’s 92 counties, which can have issues associated with stormwater runoff (Weist, 1981). As precipitation runs off impervious surfaces, it can carry high concentrations of pollutants, such as excess nitrogen and phosphorus, pet waste, and automobile fuel. Prior to impervious runoff entering the river, preexisting contaminants include industrial waste (acid mine drainage and oil field brines), municipal waste, and sediment (Weist, 1981). The Physics is Fun Project is an initiative funded by Purdue’s Office of Engagement and aims to promote an interest in physics among young learners aged 5 to 12. The project’s unique approach blends instructional design and motivational design to create an engaging and relevant learning experience. Indiana’s Wabash River is being polluted with contaminated water runoff from precipitation events. A lack of pervious land cover has led to an accumulation of fertilizer, sediment, and waste in the river. Green infrastructure, a stormwater management practice that mimics the natural ecosystem, is one of the most effective ways to prevent pollution from stormwater runoff and benefit the community. A new sub-type of folia named “reverse pseudo-gour” has been observed and described in the Nerja Cave, southern Spain. It consists of fairly vertical, thin barriers (about 5 mm high and 2 mm thick) that develop on the underside of a sub-horizontal surface (shelfstone) and grow in the opposite direction to normal gours (rimstone dams), generating sinuous shapes. Their mineral composition is essentially calcium carbonate, although globular aggregates composed of clay and phosphate minerals have also been identified.
