A Comparison Of Metabolite Extraction And Data Manipulation Methods For ¹H Nmr Spectroscopy Of Skeletal Muscle Using Chicken Breast Tissue, Hannah Underhill
A Comparison Of Metabolite Extraction And Data Manipulation Methods For ¹H Nmr Spectroscopy Of Skeletal Muscle Using Chicken Breast Tissue, Hannah Underhill
Theses
1H NMR has become an incredibly useful tool in the field of metabolomics. Many tissue types may be examined via 1H NMR in order to obtain a quantitative metabolic profile for an infinite number of phenotypes. In this study, I have tested three methods of metabolite extraction in conjunction with three methods of data analysis, in an attempt to determine which combination of the six methods (if any) are appropriate for extracting aqueous metabolites from skeletal muscle tissue for metabolite description and quantification via 1H NMR. Metabolite extraction methods tested the use of methanol/ chloroform/ water, methanol/ …
The Need For International Ai Activities Monitoring, Parviz Partow-Navid, Ludwig Slusky
The Need For International Ai Activities Monitoring, Parviz Partow-Navid, Ludwig Slusky
Journal of International Technology and Information Management
This paper focuses primarily on the need to monitor the risks arising from the dual-use of Artificial Intelligence (AI). Dual-use AI technology capability makes it applicable for defense systems and consequently may pose significant security risks, both intentional and unintentional, with the national and international scope of effects. While domestic use of AI remains the prerogative of individual countries, the unregulated and nonmonitored use of AI with international implications presents a specific concern. An international organization tasked with monitoring potential threats of AI activities could help defuse AI-associated risks and promote global cooperation in developing and deploying AI technology. The …

Environmental Factors Influencing Selection Of Cave Entrances As Nesting Sites By The Eastern Phoebe (Sayornis Phoebe), J. Foster Sims
Environmental Factors Influencing Selection Of Cave Entrances As Nesting Sites By The Eastern Phoebe (Sayornis Phoebe), J. Foster Sims
Theses
The Eastern Phoebe (Saynoris phoebe) is a medium-sized flycatcher found throughout eastern North America that traditionally nested in cave entrance zones. However, with the onset of human civilization and the adoption of man-made structures (e.g., barns, porches, bridges), the Eastern Phoebe’s population has dramatically increased (over 90%) in recent years. There has been little recent research regarding the continued use of cave entrances as nesting sites by the Eastern Phoebe. In this study, I surveyed 50 cave entrances in northern Alabama and south-central Tennessee to determine the environmental factors that influence the use of cave entrances as nesting …
Testing Cold Atmospheric Pressure Plasma : A Model For Cleaning Single-Use Urinary Catheters To Prevent Infections, Elanie Francease Briggs
Testing Cold Atmospheric Pressure Plasma : A Model For Cleaning Single-Use Urinary Catheters To Prevent Infections, Elanie Francease Briggs
Theses
Urinary tract infections (UTIs) are one of the most abundant bacterial infections world-wide with over 10 million cases and 13,000 deaths each year in the United States alone. The use of urinary catheters in hospitals and in outpatients significantly increases the risk of catheter-associated urinary tract infections (CAUTI) that are promoted by uropathogenic biofilm formation. Uropathogenic Escherichia coli (UPEC) is the dominant cause for both UTI and CAUTI, which are becoming increasingly multidrug-resistant. Uropathogenic biofilm accumulation on urinary catheters is very resistant to treatment by traditional sterilization methods. Cold atmospheric pressure plasmas (CAPP) have been known for their strong microbicidal …

“My Ob/Gyn Has The Worst Bedside Manner…” A Qualitative Analysis Of Patient-Provider Communication And Mental Health Outcomes Of Women In Rural Georgia, Tobi F. Oloyede
College of Graduate Studies: Theses & Dissertations
Despite technological advances in medicine and public health innovations, maternal mortality in the United States remains significantly higher than in other developed countries. Specifically, Georgia ranks as the second worst place to give birth in the country. Maternal mental health conditions are common, manageable complications that can happen at any point during pregnancy, childbirth, or in the first year following delivery, with 1 in 5 women affected. Unfortunately, 75% of women who experience MMH symptoms are left untreated.
This study focuses on patient-provider communication through the birthing experiences of women who have been pregnant in Georgia to examine how their …
Assessment Of Atmospheric Correction Algorithms For The Remote Sensing Of Water Quality In Southeastern U.S. Estuaries, Jerome Reimers
Assessment Of Atmospheric Correction Algorithms For The Remote Sensing Of Water Quality In Southeastern U.S. Estuaries, Jerome Reimers
College of Graduate Studies: Theses & Dissertations
Water quality is a key indicator in understanding and representing an environment's overall health. Through developments in remote sensing, we can utilize satellite imagery to measure water parameters in each aquatic system. When accurate atmospheric correction is performed, remote sensing can account for atmospheric attenuation and scattering effects to better measure the reflectance and estimate optically active constituents (OAC) present in upper water columns. Atmospheric Correction for OLI lite (ACOLITE) is an atmospheric correction algorithm designed specifically for robust atmospheric correction of water surfaces, in comparison to algorithms designed more for land surfaces such as the European Space Agency’s (ESA) …

Gene Expression Under Combined Hypoxia And Acidosis In Chondrosarcoma, Michael Stacey, Kostika Vangjeli, Christopher Osgood
Bioelectrics Publications
Chondrosarcomas are the second most common cause of bone cancer and are removed surgically with wide margins. On recurrence, they are resistant to chemo and radiation therapy and new treatment options are critically required. This tumor type produces hyaline cartilage, a cartilage normally formed under hypoxic and acidic environment due to lack of vasculature in cartilage. Paradoxically, chondrosarcomas arise in the well vascularized, oxygen rich environment of the bone. Hypoxia and acidosis are two stressors where the cellular effects are typically reported separately even though cells experience combined effects of hypoxia and acidosis. Given the mechanistic links between hypoxia and …

Acute respiratory distress syndrome (ARDS) is a severe respiratory condition characterized by increased lung permeability, hyper-inflammatory state, and fluid leak into the alveolar spaces. ARDS is a heterogeneous disease, with multiple direct and indirect causes that result in a mortality of up to 40%. Due to the ongoing Covid-19 pandemic, its incidence has increased up to ten-fold. Extracellular vesicles (EVs) are small liposome-like particles that mediate intercellular communication and play a major role in ARDS pathophysiology. Indeed, they participate in endothelial barrier dysfunction and permeability, neutrophil, and macrophage activation, and also in the development of a hypercoagulable state. A more …

Nanodiamonds (ND) have attracted significant interest for their use in several biomedical applications. These applications can be very useful if the safety and compatibility of ND are proven. We assessed the effects of ND (100 nm, Carboxylated) on primary macrophages and a macrophage-like cell line and found that these particles are not toxic to these cells at lower concentrations but may interfere with cell functions and differentiation. Internalization of ND by these cells in a time- and dose-dependent manner was mostly via phagocytosis and clathrin-dependent endocytosis and localized to the cytoplasm but not into the nucleus. No significant induction of …

Cancer ablation therapies aim to be efficient while minimizing damage to healthy tissues. Nanosecond pulsed electric field (nsPEF) is a promising ablation modality because of its selectivity against certain cell types and reduced neuromuscular effects. We compared cell killing efficiency by PEF (100 pulses, 200 ns–10 µs duration, 10 Hz) in a panel of human esophageal cells (normal and pre-malignant epithelial and smooth muscle). Normal epithelial cells were less sensitive than the pre-malignant ones to unipolar PEF (15–20% higher LD50, p < 0.05). Smooth muscle cells (SMC) oriented randomly in the electric field were more sensitive, with 30–40% lower LD50 (p < 0.01). Trains of ten, 300-ns pulses at 10 kV/cm caused twofold weaker electroporative uptake of YO-PRO-1 dye in normal epithelial cells than in either pre-malignant cells or in SMC oriented perpendicularly to the field. Aligning SMC with the field reduced the dye uptake fourfold, along with a twofold reduction in Ca2+ transients. A 300-ns pulse induced a twofold smaller transmembrane potential in cells aligned with the field, making them …

Insights Into The Conservation Of The Rare Perennial Herb, Astragalus Applegatei, Sean Stewart
Cal Poly Humboldt theses and projects
As anthropogenic activities expand, conservation of rare and endangered species is critical to reduce extinction risks. Collecting and analyzing demographic data can assist in the development of conservation strategies and can help evaluate the success of existing strategies. In this study, I built integral projection models from five years of demographic data and conducted a population viability analysis on four populations of the rare perennial herb, Astragalus applegatei, to inform conservation decisions. I also assessed how planting A. applegatei seedlings might contribute to the growth of an extant population. I found that one population was already below the extinction …

Is The Area Postrema Resistant To The Anesthetic Effects Of Ms-222?, Austin T. Csiszar
Cal Poly Humboldt theses and projects
MS-222 is a common fish anesthetic that dampens neural activity by inhibiting neuronal voltage-gated sodium channels (Nav) required for action potentials. Previous experiments conducted by our lab’s primary investigator using fluorescence calcium imaging in zebrafish neurons resulted in the observation that neurons in the area postrema (AP), a circumventricular organ located in the dorsal hindbrain, displayed prominent fluorescence intensity during MS-222 induced sedation. I used the genetically encoded calcium indicator GCaMP6s to conduct fluorescence calcium imaging with the goal of recording neural activity in the larval zebrafish area postrema and optic tectum to compare the anesthetic effects of …
The Effects Of Physical Education On Social Outcomes In Children With Disabilities: A Meta-Analysis, Alyette J. Afont
The Effects Of Physical Education On Social Outcomes In Children With Disabilities: A Meta-Analysis, Alyette J. Afont
Cal Poly Humboldt theses and projects
Social outcomes are the foundation of human relationships and many children with disabilities may display deficits in the necessary behaviors to form those connections. Previous literature shows that physical activity can be an effective intervention for children with disabilities to promote social outcomes through the three domains of learning – cognitive, psychomotor, and affective. The purpose of this meta-analysis is to collect, index, and evaluate the available literature exploring how a physical education program can impact social outcomes in children with disabilities. By composing articles pertaining to social outcomes and physical education through multiple databases, researchers will be able to …
Indigenous Women In Active Drug Abuse Recovery: An Analysis Of Native And Non-Native Programs, Raquel J. Muñoz
Indigenous Women In Active Drug Abuse Recovery: An Analysis Of Native And Non-Native Programs, Raquel J. Muñoz
Cal Poly Humboldt theses and projects
In general, much has been written on the experiences of prototypical women in drug recovery programs, however there is only a scarcity of research on the experiences of rural women of color in drug recovery programs. Very few Northern American cultures had experience with alcohol before the first wave of European settlers. Responses to intergenerational trauma faced by Native women include substance abuse, depression, anxiety, low self-esteem, suicidal thinking, and more. Due to socioeconomic disadvantages drug and alcohol abuse tends to be a coping mechanism for many Native American women. Drawing on the narratives of ten Indigenous women who are …

Olfactory Organ Morphology And Ultrastructure Of The Larval Koh Tao Island Caecilian (Ichthyophis Kohtaoensis), Jaclyn M. Patmore
Cal Poly Humboldt theses and projects
The olfactory system of living amphibians (Lissamphibia) undergoes major changes as it transitions from an aquatic to a terrestrial system during metamorphosis. Patterns of change in the cellular morphology of the nose have been examined for frogs (Anura) and salamanders (Caudata). However, it remains unknown if caecilians (Gymnophiona) have similar patterns of change in their nasal ultrastructure. In particular, no data on larval caecilian olfactory cell types are available. Here, using light microscopy and transmission electron microscopy, I examined the olfactory organ of larvae of the caecilian Ichthyophis kohtaoensis, to establish the ultrastructure of the epithelium and compare it …

Techno-Economic Analysis Of A Utility-Scale Solar Power Plant In Bhutan., Minjur
Cal Poly Humboldt theses and projects
Solar Photovoltaic (PV) technology, which captures energy from the sun, is a rapidly growing technology. Bhutan’s heavy dependence on a single energy source, hydropower, has become a significant concern for energy security. To address this, the Government of Bhutan intends to diversify energy sources using alternative renewable sources.
This research determined that Bhutan has the total potential to develop 3 gigawatts (GW) of solar power based on the utilization of about 7,100 acres of suitable land. The study also identified Tenchhekha, near the capital city of Thimphu, as a potential solar power plant location, with a capacity of 4.85 MWp …

Negative Social Media And Its Influence On Athlete's Performance, Bernd R. Huber
Cal Poly Humboldt theses and projects
This study aimed to investigate the potential impact of negative social media content on athletes' cortisol levels and subsequent performance. The study focused on the change in cortisol levels and differences in free throw performance, based on previous research findings. We hypothesized that negative social media postings would increase the stress experienced by student-athletes, resulting in elevated cortisol levels and decreased performance. Additionally, participants (n = 8) completed a questionnaire to examine the interaction between preexisting fear and the biological stress response. Contrary to expectations, there was no significant change in stress response, and negative postings did not have …

Variable Selection And Estimation In Bayesian Causal Mediation Analysis, Shuang Wang
Variable Selection And Estimation In Bayesian Causal Mediation Analysis, Shuang Wang
Open Access Master's Theses
Causal mediation analysis has emerged as a valuable tool in various fields, allowing for decomposing total effects into direct and indirect effects. However, identifying the mediation effects relies on assumptions of no unmeasured confounding in the exposure-mediator, exposure-outcome, and mediator-outcome relationships. Moreover, there is often an interest in examining the effects of a mixture of exposures on the outcome, which poses challenges in determining the appropriate set of confounding variables. Therefore, we proposed a Bayesian mediation analysis that simultaneously selects common confounding variables associated with the two exposures, mediator, and outcome, by employing spike and slab group lasso using MCMC …

Department of Biochemistry: Faculty Publications
Transmembrane P-type ATPase pumps catalyze the extrusion of transition metal ions across cellular lipid membranes to maintain essential cellular metal homeostasis and detoxify toxic metals. Zn()-pumps of the P-type subclass, in addition to Zn , select diverse metals (Pb, Cdand Hg) at their transmembrane binding site and feature promiscuous metal-dependent ATP hydrolysis in the presence of these metals. Yet, a comprehensive understanding of the transport of these metals, their relative translocation rates, and transport mechanism remain elusive. We developed a platform for the characterization of primary-active Zn()-pumps in proteoliposomes to study metal selectivity, translocation events and transport mechanism in real-time, …

Examining The Regulation Of The Bs21 Homologs In Francisella Tularensis, Sierra Schmidt
Open Access Master's Theses
Francisella tularensis is a facultative intracellular pathogen, which relies on a network of regulation for its virulence. It contains three homologs of the ribosomal protein bS21. These three ribosomal protein homologs can independently associate with ribosomes, leading to ribosomal heterogeneity. The second homolog, bS21-2, is implicated in the positive regulation of a number of virulence proteins. Additionally, transcriptomic data indicates that bS21-2 is autoregulated. The mechanism by which this autoregulation occurs is unknown, as is the regulation of the other two homologs, bS21-1 and bS21-3. In the following chapters, I will show that bS21-1 and bS21-3 do not autoregulate their …

The Role Of Myocardin In The Progression Of Non-Small Cell Lung Cancer, Soromidayo Akinsiku
The Role Of Myocardin In The Progression Of Non-Small Cell Lung Cancer, Soromidayo Akinsiku
Biotechnology Theses
Lung cancer is the leading cause of cancer-related mortality in the world and NSCLC accounts for 85% of all lung cancer cases. The mainstay of treatment for patients with stage I, II and IIIA NSCLC is surgery, followed by post-operative cisplatin-based chemotherapy. Additional adjuvant therapy involving targeted tyrosine kinase inhibitors has been in use, however even for the targeted therapy, resistance eventually develops. Therefore, there is a need for identifying novel targets for this life-threatening disease. Given that preliminary studies in Ikebe lab revealed that myocardin knockdown significantly promoted caspase-3 degradation, in this study, using myocardin siRNA, we investigated the …
