The development of whole exome and whole genome sequencing technologies has allowed geneticists to gradually uncover a wide spectrum of rare, deleterious mutations with large effect sizes, which are responsible for Mendelian diseases. Roughly 70% of human genes are single copy and have orthologs across the vertebrate lineage, suggesting that they are under purifying selection. This suggests that at least 10,000 genes will have Mendelian phenotypes when mutated but have yet to be discovered1. Exhaustive research efforts have been directed at finely categorizing the coding, and more recently noncoding, genome. The actual number of large effect noncoding mutations, and of …

Tuberculosis (TB) is the leading cause of death from any single infectious disease, killing over 1.5 million people each year. Treatment of TB is extraordinarily difficult, requiring at least 6 months of combination antibiotic therapy to cure patients. Even with this prolonged treatment, 5 to 10% of patients are not fully cured and experience disease relapse. Mycobacterium tuberculosis (Mtb), the causative agent of TB, is intrinsically resistant to most antibiotics and can acquire an additional level of antibiotic resistance through specific chromosomal mutations. A better understanding of intrinsic and acquired drug resistance in Mtb is essential to develop faster treatments …

Receptor activity-modifying proteins (RAMPs) have emerged as modulators of many aspects of G protein-coupled receptor (GPCR) biology and pharmacology. The RAMP family was discovered more than two decades ago and GPCR-RAMP interactions and their functional consequences on receptor trafficking and ligand selectivity have been documented for several GPCRs, mostly belonging to the secretin family. However, the pervasiveness of GPCR-RAMP interactions and the mechanisms of their effects are not well understood. Recent bioinformatics and experimental studies suggest that GPCR-RAMP interactions might be much more widespread than previously anticipated. The diversity of behaviors exhibited by animals is astounding. Even within a species, members constantly select between a myriad of ways to interact with their environment, ranging from foraging to fighting. Yet, why an individual behaves as it does in each moment of time typically remains nebulous to us as observers. In this thesis, I explore the possibility that these seemingly idiosyncratic behavioral decisions can be understood by accessing the hidden internal motivations that spur an animal to do one thing in lieu of another. Phenotypic sex differences are essential byproducts and drivers of evolution in sexually reproducing species. Knowing how sex differences arise and how they influence evolution is central to our understanding of evolution. Although many facets of sex differences have been explored, many remain unknown. Here I focus on sex differences and the evolution of gene expression in three parts. First, I examine the evolutionary properties of genes with sex differences in mean expression levels in the Drosophila brain – a surprisingly understudied organ in this context. Neuronal signals relevant for spatial navigation have been described in many species, however, a circuit-level understanding of how such signals interact to guide behaviour is lacking. In this thesis, I characterize a neuronal circuit in the Drosophila central complex that compares internally generated estimates of the fly's heading and goal angles––both encoded in world-centred, or allocentric, coordinates––to generate a body-centred, or egocentric, steering signal. Past work has argued that the activity of EPG cells, or "compass neurons", represents the fly's moment-to-moment angular orientation, or heading angle, during navigation. An animal's heading angle, however, is not always aligned with its goal …

Artificial drainage is a common agricultural management technique in the United States used to remove excess water from poorly drained soils. Approximately 22.48 million hectares of crop land are drain-tiled in the United States, providing long-term economic benefits to farmers. However, artificial drainage increases sediment transport in agricultural watersheds, which can degrade aquatic systems, destroy habitats, and limit biological diversity. Biotic indices based on benthic macroinvertebrates are commonly used to assess surface water quality, but recent studies show potential in developing sediment biotic indices using benthic macroinvertebrates to estimate fine sediment in streams. The theory of gerotranscendence was developed as a means of addressing limitations in current developmental theories. It is defined as the natural developmental process in which people gradually change their fundamental views of themselves, the world, their relationships, and reality in general. Research has shown that gerotranscendent behaviors are viewed as pathological by younger adults. The purpose of this study was to further explore differences in perception of gerotranscendent behaviors between college students and older adults. The current issue of locating, diagnosing, and treating cancer and other diseases linked to specific target genes necessitates the creation of a reliable system for precisely identifying target genes that are initially extracted from a human chromosome. Current methodologies often suffer from overlapping gene regions in the target gene that occurs during the analysis process, which can have a substantial impact on the accuracy of the results. Our recommended approach, which was the appropriate model to apply for this particular problem, is set to enhance the analytical process by utilizing neural networks' U-Net with an attention mechanism. Pediatric brain cancer is a rare and potentially deadly diagnosis that affects children in Kentucky at a higher incidence than those nationally. Limited exposures in children early in life to warrant the development and progression of brain cancer point to the role parental exposures experienced in the period prior to conception and throughout pregnancy may play in its development. Many of these exposures may be experienced through occupation. Data on a subset of pediatric brain cancer cases in Kentucky was compared to all cases diagnosed in the state from 1995 to 2019. Starry stonewort (Nitellopsis obtusa) (Desvaux in Loiseleur) J. Groves is an invasive macro alga that can take over entire water columns and outcompete native species. Previous research has quantified seasonal life history and phenology but there is no research quantifying carbohydrate allocation which was the aim for Chapter II. Management efforts to control Nitellopsis obtusa have been limited to stressing the thalli and have not been able to directly target the reproductive bulbils on larger scales. Smaller scale efforts such as the use of hand pulling can be used but hand pulling is not realistic for larger infestations. Chapter III …

Zebra mussel Dreissena polymorpha veligers were first found in the Red Lake system in Beltrami County in 2017, however no adult zebra mussels have been documented. When zebra mussels first invade a system, fish often switch their prey use to this new abundant resource. Freshwater drum Aplodinotus grunniens is one of these fish species known to consume physidae and bivalves, including zebra mussels, using pharyngeal teeth, the molar-like grinders in the back of their throats. This research aims to identify any presence of adult zebra mussels in the system and determine freshwater drum diet composition and feeding patterns by analyzing …

Algae are of scientific and commercial interest due to their ease of culture and high fatty acid content. The extracted fatty acids from these phytoplankton may potentially be used as an additional test for phylogenetic classification of new algal strains (Tonon et al. 2002), as well as in a supplement for human consumption and producing next-generation biofuels. Of interest is the fatty acid content contained within various algal isolates within the class Eustigmatophyceae. Cellular slime molds, Dictyostelium, are bactivorous, soil-dwelling amoebae. When food is available, cells reproduce asexually by binary fission. Under certain environmental conditions, such as darkness and humidity, Dictyostelium reproduces sexually. The sexual cycle is uniquely social and has unusual features. First, sexual reproduction in most eukaryotes usually involves fusion of two gametes. But, in Dictyostelium, triparental inheritance is observed (Bloomfield et al. 2019), in which gamete fusion involves more than two gametes. Second, most eukaryotic species have two sexes or mating types, but Dictyostelium species are known to have more than two mating types. Even though Dictyostelium species show variations …

Noteworthy Parasites (Trematoda, Cestoda, Phthiraptera) Of Three Birds (Aves: Passeriformes; Pelecaniformes; Piciformes) From Arkansas, C.T. Mcallister, H.W. Robison

Between November 2020 and March 2021, three species of birds, one each of a tufted titmouse (Baeolophus bicolor), pileated woodpecker (Dryocopus pileatus), and great blue heron (Ardea herodias) were salvaged from Lawrence and Polk counties, Arkansas, and examined for parasites. Found were four taxa of parasites, including a dicrocoeliid trematode, a cyclophyllidean cestode, and species of philopterid and menoponid lice. We document new host and distributional records for these parasites from select birds of the state.

Spermiogenesis In The Western Siren, Siren Nettingi (Caudata: Sirenidae), S.E. Trauth

I investigated spermiogenesis in the Western Siren, Siren nettingi (formerly Western Lesser Siren, Siren intermedia nettingi) from salamanders collected periodically over a span of 21 yr (1994–2015) in northeastern Arkansas. My primary objective in this study is to present histologically and ultrastructurally, for the first time, the novel sequence of spermatological events of the spermiogenic process in this species. I describe the transformational stages from secondary spermatocytes to mature biflagellated spermatozoa, all of which occur within the lumen of testicular lobules. These new-to-science processes begin with nuclear displacement and expansion within spermatids, which occur after their detachment from Sertoli cells …

We examined the microanatomy of the distal urogenital ducts in the male Northern Crawfish Frog (Rana areolata circulosa) from a small sample (n = 5) collected in February and March from central Arkansas over a 5-year period (2017–2022). Specifically, we sought information on the following structures: (1) the paired Wolffian (urogenital) ducts caudally from the kidneys to their merging with the urodeum of the cloaca, (2) the paired seminal vesicles (sperm storage structures), and (3) the paired Müllerian ducts and glands. This study provides new information on these urogenital ducts and promotes a clearer understanding of this anatomical region for …

No Sperm Morphometric Differences Between Two Snapping Turtle Species, J.D. Konvalina, S.E. Trauth

We examined sperm morphometrics between Chelydra serpentina and Macrochelys temminckii. When both species are combined, we found significant correlations between total sperm length and sperm head length as well as sperm tail length and sperm head length. In addition, we observed significant correlations between total sperm length and sperm tail length. T-tests revealed no significant differences in any of the sperm morphometrics between the two species. Both snapping turtle species had greater values for every sperm morphometric when compared to another freshwater turtle. The strong correlation between total sperm length and sperm tail length is concurrent with what has previously …
