Effects Of Hepatocyte Growth Factor On Porcine Mammary Cell Growth And Senescence, Chi-Ping Huang, Liang-Chih Liu, Hsin-Ling Lu, Chih-Rong Shyr

BioMedicine

Background: The porcine mammary glands share morphological and physiological similarities with human ones, making primary porcine mammary cells (PMC) suitable for biomedical research and a potential cellular therapeutic for breast cancer xenogeneic cell immunotherapy. Primary cells isolated from tissues remain the physiological functions of origin tissues but their self-renewal ability is restricted and cells acquire senescence during in vitro expansion. To overcome these drawbacks, here we sought to establish an approach to efficiently increase PMC’s in vitro growth. We studied the effects of the hepatocyte growth factor (HGF) to maintain the expansion capacity of porcine mammary cells and identify the …

Selenium Nanoparticles Attenuate Bleomycin-Induced Pulmonary Oxidative Stress And Fibrosis In Rats, Azza M A. Abouelella, Bahig A. El Deeb, Eman Mohammed Ali

Bulletin of Faculty of Pharmacy Cairo University

Pulmonary fibrosis is a fatal progressive lung disorder. Bleomycin (BLM) has been used for induction of experimental pulmonary fibrosis. Selenium nanoparticles (SeNPs) have been synthesized and investigated for their ability to ameliorate BLM-induced pulmonary fibrosis. SeNPs (0.5 mg⁄kg. ip) were administered for four weeks; two weeks pre and two weeks post BLM (2.5 U⁄kg. ip). BLM induced biochemical and histological deterioration of lungs. SeNPs induced significant histopathological and biochemical improvement of oxidative stress (MDA, SOD and GSH), inflammatory (IL6 and IL10) and fibrotic markers (hydroxyproline). In conclusion; SeNPs has potential therapeutic rather than protective effect on management of pulmonary damage.

Phenotypic Variability For Root Traits In Andean Common Beans Grown With And Without Aluminum Stress Conditions, Daniel Ambachew, Abay Asfaw, Tennessee State University

Agricultural and Environmental Sciences Faculty Research

Genetic variation in wild relatives, GenBank accessions, landraces, and cultivars can unlock key alleles for the traits of interest for breeding programs. Breeding programs often utilize different strategies to quantify the source of heritable variation for target traits. One neglected area of study is the root traits of diverse genotypes, and this is especially the case for aluminum toxicity effects on legumes such as the common bean, which is the most used pulse for direct human consumption. This study evaluated 267 genotypes of common bean that were part of the global Andean Diversity Panel (ADP), consisting mainly of genotypes assembled …

Sperm Dna Damage Shown To Correlate With Dna Alterations, Hailey E. Johnson, Chad Pollard, Timothy Jenkins Feb 2023

Sperm Dna Damage Shown To Correlate With Dna Alterations, Hailey E. Johnson, Chad Pollard, Timothy Jenkins

Library/Life Sciences Undergraduate Poster Competition 2023

DNA methylation is a biological process where methyl groups are added to the DNA molecule, changing the physical structure

Sperm DNA methylation has shown to predict pregnancy outcomes for those with infertility

DNA damage can happen through single and double stranded breaks. The Comet and TUNEL assay are two different and common ways to quantify levels of sperm DNA damage.

It is suggested DNA damage might relate with global DNA methylation patterns in sperm genome, however regional differences have been overlooked.

We wanted to assess the relationship between sperm DNA damage and regional DNA methylation, and compare the outcomes of …

The Effect Of Student Perception Of Parent Involvement And Socioeconomic Status On Academic Intrinsic Motivation, Pallavi Aggarwal, Taylor Lawson Smith Feb 2023

The Effect Of Student Perception Of Parent Involvement And Socioeconomic Status On Academic Intrinsic Motivation, Pallavi Aggarwal, Taylor Lawson Smith

Dissertations

Many teachers struggle to help students become intrinsically motivated in their school work. Research has shown that students who are intrinsically motivated tend to have higher academic achievement than students who are not (Mendoza, 2012). There are many factors that contribute to students’ intrinsic motivation. Building on existing research, the relationship between perceived parental involvement and students’ intrinsic motivation, as well as the relationship of perceived socioeconomic status and intrinsic motivation was investigated. A mixed methods research approach was used to determine a) the relationship between perceived parent involvement and students’ intrinsic motivation, b) the relationship between perceived socioeconomic status …

Dpc29 Promotes Post-Initiation Mitochondrial Translation In Saccharomyces Cerevisiae, Kyle A. Hubble, Michael F. Henry Feb 2023

Dpc29 Promotes Post-Initiation Mitochondrial Translation In Saccharomyces Cerevisiae, Kyle A. Hubble, Michael F. Henry

Rowan-Virtua School of Osteopathic Medicine Departmental Research

Mitochondrial ribosomes synthesize essential components of the oxidative phosphorylation (OXPHOS) system in a tightly regulated process. In the yeast Saccharomyces cerevisiae, mitochondrial mRNAs require specific translational activators, which orchestrate protein synthesis by recognition of their target gene's 5'-untranslated region (UTR). Most of these yeast genes lack orthologues in mammals, and only one such gene-specific translational activator has been proposed in humans-TACO1. The mechanism by which TACO1 acts is unclear because mammalian mitochondrial mRNAs do not have significant 5'-UTRs, and therefore must promote translation by alternative mechanisms. In this study, we examined the role of the TACO1 orthologue in yeast. We …

Apicomplexan parasite growth and replication relies on nutrient acquisition from host cells, in which intracellular multiplication occurs, yet the mechanisms that underlie the nutrient salvage remain elusive. Numerous ultrastructural studies have documented a plasma membrane invagination with a dense neck, termed the micropore, on the surface of intracellular parasites. However, the function of this structure remains unknown. Here we validate the micropore as an essential organelle for endocytosis of nutrients from the host cell cytosol and Golgi in the model apicomplexan Toxoplasma gondii. Detailed analyses demonstrated that Kelch13 is localized at the dense neck of the organelle and functions as …

Hemodynamic Investigations In Intracranial Aneurysms: A Commentary, Hang Yi, Mark Johnson, Luke Bramlage, Zifeng Yang, Bryan Ludwig

Mechanical and Materials Engineering Faculty Publications

Intracranial aneurysms (IAs) are abnormal bulges in a blood vessel in the brain that have a potential to rupture and even causing a stroke, which can lead to lasting brain damage, long-term disability, or even loss of life. It has been widely acknowledged that hemodynamic factors, e.g., instantaneous wall shear stress, time-averaged wall shear stress, wall shear stress gradient, gradient oscillatory number, oscillatory shear index, pulsatile blood flow waveform (flow rate magnitude and shape, physical flow period), relative residence time/turnover time, blood pressure, and vortex (i.e., size, location, and numbers), have a close relationship with the pathobiology (i.e., initiation, growth, …

Soybean Seed Protein Concentration Is Limited By Nitrogen Supply In Tropical And Subtropical Environments In Brazil, E. H. Figueiredo Moura Da Silva, N. Cafaro La Menza, G. G. Munareto, A. J. Zanon, Santos Carvalho, F. R. Marin

Department of Agronomy and Horticulture: Faculty Publications

Soybean production contributes to ca. 60% of global plant-based protein used for food and feed. Brazil is the largest soybean producer and exporter, with 60% from tropical and 40% from subtropical environments. Nitrogen (N) can play an essential role in the storage of proteins in seeds; thus, it could be a key factor in increasing the quantity and quality of seeds in high-yielding soybean crops. Unlike in temperate environments, there is a gap of knowledge on whether soybean grown under tropical and subtropical climates are limited by N-fertilization to sustain the seed yield increase without detriments in seed protein concentration. …

Incarcerated Bodies – Embodied Autoethnography In Prison, Shulamit Kitzis‬‎ Feb 2023

Incarcerated Bodies – Embodied Autoethnography In Prison, Shulamit Kitzis‬‎

The Qualitative Report

Prison is a study field in which everyone – inmates, guards, and prison researchers – experiences powerful sensory stimuli comprised of sounds, sights, and smells in a crowded, closed space. Yet traditional academic research has socialized researchers to “wash away” their physical and emotional feelings for fear they would jeopardize the scientific nature and validity of their studies. Nevertheless, at times in a prison setting, the researchers’ bodies are the only tool that enables them to document what goes on; so much so that ignoring their bodies and emotions leads to a loss of valuable information. Using embodied autoethnography (EA), …

Boron Nitride Nanotubes: Force Field Parameterization, Epoxy Interactions, And Comparison With Carbon Nanotubes For High-Performance Composite Materials, Swapnil S. Bamane, Michael B. Jakubinek, Krishan Kanhaiya, Behnam Ashrafi, Hendrik Heinz, Gregory Odegard

Michigan Tech Publications, Part 1

Boron nitride nanotubes (BNNTs) are a very promising reinforcement for future high-performance composites because of their excellent thermo-mechanical properties. To take full advantage of BNNTs in composite materials, it is necessary to have a comprehensive understanding of the wetting characteristics of various high-performance resins. Molecular dynamics (MD) simulations provide an accurate and efficient approach to establish the contact angle values of engineering polymers on reinforcement surfaces, which offers a measure for the interaction between the polymer and reinforcement. In this research, MD simulations and experiments are used to determine the wettability of various epoxy systems on BNNT surfaces. The reactive …

For a continuous healthcare or environmental monitoring system, it is essential to reliably sense the analyte concentration reported by electrochemical sensors. However, environmental perturbation, sensor drift, and power-constraint make reliable sensing with wearable and implantable sensors difficult. While most studies focus on improving sensor stability and precision by increasing the system’s complexity and cost, we aim to address this challenge using low-cost sensors. To obtain the desired accuracy from low-cost sensors, we borrow two fundamental concepts from communication theory and computer science. First, inspired by reliable data transmission over a noisy communication channel by incorporating redundancy, we propose to measure …

Real-Time Visualization Of Cytosolic And Mitochondrial Atp Dynamics In Response To Metabolic Stress In Cultured Cells, Donnell White Iii, Lothar Lauterboeck, Parnia Mobasheran, Tetsuya Kitaguchi, Antoine H. Chaanine, Qinglin Yang

School of Medicine Faculty Publications

Adenosine 50 triphosphate (ATP) is the energy currency of life, which is produced in mitochondria (~90%) and cytosol (less than 10%). Real-time effects of metabolic changes on cellular ATP dynamics remain indeterminate. Here we report the design and validation of a genetically encoded fluorescent ATP indicator that allows for real-time, simultaneous visualization of cytosolic and mitochondrial ATP in cultured cells. This dual-ATP indicator, called smacATPi (simultaneous mitochondrial and cytosolic ATP indicator), combines previously described individual cytosolic and mitochondrial ATP indicators. The use of smacATPi can help answer biological questions regarding ATP contents and dynamics in living cells. As expected, 2-deoxyglucose …

Asprosin Promotes Feeding Through Sk Channel-Dependent Activation Of Agrp Neurons, Bing Feng, Hesong Liu, Ila Mishra, Clemens Duerrschmid, Peiyu Gao, Pingwen Xu, Chunmei Wang, Yanlin He Feb 2023

Asprosin Promotes Feeding Through Sk Channel-Dependent Activation Of Agrp Neurons, Bing Feng, Hesong Liu, Ila Mishra, Clemens Duerrschmid, Peiyu Gao, Pingwen Xu, Chunmei Wang, Yanlin He

Faculty, Staff and Students Publications

Asprosin, a recently identified adipokine, activates agouti-related peptide (AgRP) neurons in the arcuate nucleus of the hypothalamus (ARH) via binding to protein tyrosine phosphatase receptor δ (Ptprd) to increase food intake. However, the intracellular mechanisms responsible for asprosin/Ptprd-mediated activation of AgRPARH neurons remain unknown. Here, we demonstrate that the small-conductance calcium-activated potassium (SK) channel is required for the stimulatory effects of asprosin/Ptprd on AgRPARH neurons. Specifically, we found that deficiency or elevation of circulating asprosin increased or decreased the SK current in AgRPARH neurons, respectively. AgRPARH-specific deletion of SK3 (an SK channel subtype highly expressed in AgRPARH neurons) blocked asprosin-induced …

Alternative Splicing Mediates The Compensatory Upregulation Of Mbnl2 Upon Mbnl1 Loss-Of-Function, Larissa Nitschke, Rong-Chi Hu, Andrew N Miller, Lathan Lucas, Thomas A Cooper

Faculty, Staff and Students Publications

Loss of gene function can be compensated by paralogs with redundant functions. An example of such compensation are the paralogs of the Muscleblind-Like (MBNL) family of RNA-binding proteins that are sequestered and lose their function in Myotonic Dystrophy Type 1 (DM1). Loss of MBNL1 increases the levels of its paralog MBNL2 in tissues where Mbnl2 expression is low, allowing MBNL2 to functionally compensate for MBNL1 loss. Here, we show that loss of MBNL1 increases the inclusion of Mbnl2 exon 6 and exon 9. We find that inclusion of Mbnl2 exon 6 increases the translocation of MBNL2 to the nucleus, while …

The periodic emergence of SARS-CoV-2 variants of concern (VOCs) with unpredictable clinical severity and ability to escape preexisting immunity emphasizes the continued need for antiviral interventions. Two small molecule inhibitors, molnupiravir (MK-4482), a nucleoside analog, and nirmatrelvir (PF-07321332), a 3C-like protease inhibitor, have recently been approved as monotherapy for use in high-risk patients with COVID-19. As preclinical data are only available for rodent and ferret models, here we assessed the efficacy of MK-4482 and PF-07321332 alone and in combination against infection with the SARS-CoV-2 Delta VOC in the rhesus macaque COVID-19 model. Macaques were infected with the SARS-CoV-2 Delta variant …

Abstract Global antimicrobial resistance is a health crisis that can change the face of modern medicine. Exploring diverse natural habitats for bacterially-derived novel antimicrobial compounds has historically been a successful strategy. The deep-sea presents an exciting opportunity for the cultivation of taxonomically novel organisms and exploring potentially chemically novel spaces. In this study, the draft genomes of 12 bacteria previously isolated from the deep-sea sponges Phenomena carpenteri and Hertwigia sp. are investigated for the diversity of specialized secondary metabolites. In addition, early data support the production of antibacterial inhibitory substances produced from a number of these strains, including activity against …

Mutations in the TRPV4 ion channel can lead to a range of skeletal dysplasias. However, the mechanisms by which TRPV4 mutations lead to distinct disease severity remain unknown. Here, we use CRISPR-Cas9-edited human-induced pluripotent stem cells (hiPSCs) harboring either the mild V620I or lethal T89I mutations to elucidate the differential effects on channel function and chondrogenic differentiation. We found that hiPSC-derived chondrocytes with the V620I mutation exhibited increased basal currents through TRPV4. However, both mutations showed more rapid calcium signaling with a reduced overall magnitude in response to TRPV4 agonist GSK1016790A compared to wildtype (WT). There were no differences in …
