The current mortality statistics for cardiovascular disease (CVD) in the United States (US) and on a global scale are staggering. CVD is responsible for approximately 31% of global deaths,1 and approximately 25% of adults in the United States (US) have CVD.2 The American Heart Association has identified eight primary measures for improving and maintaining cardiovascular health: Life’s Essential 8.3 However, a lesser recognized measure that has a profound effect on cardiovascular health has recently emerged,4-5 that of the gut microbiome. The link between cardiovascular health and diet is not merely due to essential nutrients found in …

Long noncoding RNAs (lncRNAs) are involved in glioma initiation and progression. Glioma stem cells (GSCs) are essential for tumor initiation, maintenance, and therapeutic resistance. However, the biological functions and underlying mechanisms of lncRNAs in GSCs remain poorly understood. Here, we identified that LINC00839 was overexpressed in GSCs. A high level of LINC00839 was associated with GBM progression and radiation resistance. METTL3-mediated m6A modification on LINC00839 enhanced its expression in a YTHDF2-dependent manner. Mechanistically, LINC00839 functioned as a scaffold promoting c-Src-mediated phosphorylation of β-catenin, thereby inducing Wnt/β-catenin activation. Combinational use of celecoxib, an inhibitor of Wnt/β-catenin signaling, greatly sensitized GSCs to …

Evaluating Vaccine Management Strategies For Edwardsiella Ictaluri Infections In Zebrafish (Danio Rerio), Brandy Malbrough
Evaluating Vaccine Management Strategies For Edwardsiella Ictaluri Infections In Zebrafish (Danio Rerio), Brandy Malbrough
LSU Master's Theses
Aquaculture is a globally important industry that faces significant challenges due to infectious diseases, which can result in substantial financial losses. In 2011, a new strain of Edwardsiella ictaluri emerged as a major pathogen affecting zebrafish in the ornamental fish industry, leading to high mortality rates and posing a severe threat to the sector. Vaccines provide potential benefits for disease prevention in aquaculture, particularly live-attenuated vaccines (LAVs), which induce strong host immune responses. Bacterial attenuation for vaccine development has evolved from serial passage to more targeted genetic modification techniques. However, regulatory constraints limit the availability of licensed LAVs for aquaculture. …

In 2020, educators and students were faced with a global pandemic that created unprecedented challenges to classrooms across the nation. For many students, the shift to online learning in a necessary effort to maintain educational continuity lasted for an entire academic year. Students attended online synchronous and asynchronous class sessions, interacted with their peers in exclusively online settings, and were isolated to the social and economic constraints of their own households. This study examines the dramatic impact that these virtual learning experiences had on middle school students’ learning, engagement, and development during the COVID-19 pandemic. By interviewing middle school students, …

Pharmacy Faculty Articles and Research
Proteoglycan 4 (PRG4) is a mucinous glycoprotein secreted by synovial fibroblasts and superficial zone chondrocytes, released into synovial fluid, and adsorbed on cartilage and synovial surfaces. PRG4′s roles include cartilage boundary lubrication, synovial homeostasis, immunomodulation, and suppression of inflammation. Gouty arthritis is mediated by monosodium urate (MSU) crystal phagocytosis by synovial macrophages, with NLRP3 inflammasome activation and IL-1β release. The phagocytic receptor CD44 mediates MSU crystal uptake by macrophages. By binding CD44, PRG4 limits MSU crystal uptake and downstream inflammation. PRG4/CD44 signaling is transduced by protein phosphatase 2A, which inhibits NF-κB, decreases xanthine oxidoreductase (XOR), urate production, and ROS-mediated IL-1β …

Background: Nearly 80% of patients with pancreatic cancer suffer from glucose intolerance or diabetes. Pancreatic cancer complicated by diabetes has a more immunosuppressive tumor microenvironment (TME) and is associated with a worse prognosis. The relationship between glucose metabolism and programmed cell death-Ligand 1 (PD-L1) is close and complex. It is important to explore the regulation of high glucose on PD-L1 expression in pancreatic cancer and its effect on infiltrating immune effectors in the tumor microenvironment. Methods: Diabetic murine models (C57BL/6) were used to reveal different immune landscape in euglycemic and hyperglycemic pancreatic tumor microenvironment. An epidemic of obesity has affected large portions of the world, increasing the risk of developing many different age-associated diseases, including cancer, cardiovascular disease, and diabetes. In contrast with the prevailing notion that "a calorie is just a calorie," there are clear differences, within and between individuals, in the metabolic response to different macronutrient sources. Recent findings challenge this oversimplification; calories from different macronutrient sources or consumed at different times of day have metabolic effects beyond their value as fuel. Here, we summarize discussions conducted at a recent NIH workshop that brought together experts in calorie restriction, macronutrient composition, and …

The seasonal availability of light and micronutrients strongly regulates productivity in the Southern Ocean, restricting biological utilization of macronutrients and CO2 drawdown. Mineral dust flux is a key conduit for micronutrients to the Southern Ocean and a critical mediator of multimillennial-scale atmospheric CO2 oscillations. While the role of dust-borne iron (Fe) in Southern Ocean biogeochemistry has been examined in detail, manganese (Mn) availability is also emerging as a potential driver of past, present, and future Southern Ocean biogeochemistry. Here, we present results from fifteen bioassay experiments along a north–south transect in the undersampled eastern Pacific sub-Antarctic zone. In addition to …

Morphological And Molecular Characterization Of The Olfactory System In The Formosan Subterranean Termite, Paula Castillo
LSU Doctoral Dissertations
The Formosan subterranean termite, Coptotermes formosanus, is a eusocial insect species. Its members live in colonies consisting of reproductive (queen and king) and non-reproductive individuals (workers and soldiers). Olfaction is an essential sense for the social life of many insects. However, in termites, little is known about how olfactory cues are perceived in the peripheral sensory system (antennae) and processed in the central nervous system (brain) to elicit different behavioral responses across castes. In this study, we investigated the olfactory system in all castes (soldiers, workers, and reproductive alates), from the antennal and brain morphology to the antennal gene …

Most microbes have developed responses that protect them against stresses relevant to their niches. Some that inhabit reasonably predictable environments have evolved anticipatory responses that protect against impending stresses that are likely to be encountered in their niches–termed “adaptive prediction”. Unlike yeasts such as Saccharomyces cerevisiae, Kluyveromyces lactis and Yarrowia lipolytica and other pathogenic Candida species we examined, the major fungal pathogen of humans, Candida albicans, activates an oxidative stress response following exposure to physiological glucose levels before an oxidative stress is even encountered. Why? Using competition assays with isogenic barcoded strains, we show that “glucose-enhanced oxidative stress resistance” phenotype …

Purpose: Cancer survivors develop cancer and treatment-related morbidities at younger than normal ages and are at risk for early mortality, suggestive of an aging phenotype. The Cumulative Illness Rating Scale for Geriatrics (CIRS-G) is specifically designed to describe the accumulation of comorbidities over time with estimates of severity such as total score (TS) which is a sum of possible conditions weighted by severity. These severity scores can then be used to predict future mortality.
Methods: CIRS-G scores were calculated in cancer survivors and their siblings from Childhood Cancer Survivor Study cohort members from two time points 19 years apart and …
