Animal studies implicate one-carbon metabolism and DNA methylation genes in hepatocellular carcinoma (HCC) development in the setting of metabolic perturbations. Using human samples, we investigated the associations between common and rare variants in these closely related biochemical pathways and risk for metabolic HCC development in a multicenter international study. We performed targeted exome sequencing of 64 genes among 556 metabolic HCC cases and 643 cancer-free controls with metabolic conditions. Multivariable logistic regression was used to calculate odds ratios (ORs) and 95% confidence intervals (CIs), adjusting for multiple comparisons. The prevalence of obesity is rapidly increasing worldwide, while the development of effective obesity therapies lags behind. Although new therapeutic targets to alleviate obesity are identified every day, and drug efficacy is improving, adverse side effects and increased health risks remain serious issues facing the weight-loss industry. Serotonin, also known as 5-HT, has been extensively studied in relation to appetite reduction and weight loss. Women account for a sizeable proportion of the unhoused population in the U.S. Over one-third (38.7%) of unhoused individuals are women, which is a 17% increase from 2016 (United States Department of Housing and Urban Development, 2019). The increased prevalence of women experiencing homelessness calls for a renewed examination of their service needs. This dissertation presents three studies examining factors impacting the service needs of unhoused women. Harmful cyanobacterial blooms (cyanoHABs) are a complex and widespread disturbance in freshwater water bodies, impacting water quality for wildlife and human populations. While cyanobacteria often bloom in warm lakes impacted by human development like agriculture, blooms are increasingly reported in cooler waters with limited development in the surrounding watershed. As much of cyanoHAB research has focused on lakes in highly developed watersheds, the understanding of factors leading to cyanobacteria dominance and blooms in the absence of major development remains limited. The study conducted biosynthesis of copper oxide nanoparticles using an aqueous extract of Aspergillus niger and in-vestigated their potential biomedical applications. The nanoparticles were characterized using various techniques in-cluding UV-Vis, SEM, TEM, and EDX, revealing their spherical to crystalline shape with sizes ranging from 21 nm to 48 nm. The nanoparticles demonstrated notable antimicrobial activity against Staphylococcus aureus with a zone of 8 mm and Escherichia coli recorded high synergistic activity with 20.93%, as well as antioxidant properties. Furthermore, selective cytotoxicity towards breast cancer cells was observed with an IC50 of 107.81 ug/ml and an IC50 of 250.93 ug/ml on …

Brain transcriptional variation is a heritable trait that mediates complex behaviors, including addiction. Expression quantitative trait locus (eQTL) mapping reveals genomic regions harboring genetic variants that influence transcript abundance. In this study, we profiled transcript abundance in the striatum of 386 Diversity Outbred (J:DO) mice of both sexes using RNA-Seq. All mice were characterized using a behavioral battery of widely-used exploratory and risk-taking assays prior to transcriptional profiling. We performed eQTL mapping, incorporated the results into a browser-based eQTL viewer, and deposited co-expression network members in GeneWeaver. Background: Child sexual abuse (CSA) in a preventable national public health problem that results in adverse health consequences. Research suggests many victims are currently attending school. With approximately 80% of readmissions experiencing post-discharge complications (Agri et al., 2020), quality improvement (QI) initiatives to increase ostomy patient’s self-efficacy and quality of living (QOL) are needed. The metabolism of phosphatidylcholine (PC), a major eukaryotic membrane lipid, has been shown to be an important aspect of pathogenesis in Candida albicans. Of note, PC turnover product, glycerophosphocholine (GPC), is abundant renal metabolite available within the host. GPC is readily imported by C. albicans, and deletion of the major GPC transporter, Git3, leads to decreased virulence in a bloodstream infection model. According to a 2022 report, organs destined for transplantation are discarded 15% of the time for various reasons (Israni et al. 2022). This highlights a critical need in transplant practices. Transplant donor tissues are in need of essential nutrients, including molecular oxygen, to maintain integrity during preservation. A current trend in preservation is to perfuse the ex vivo tissue graft with nutrient-rich aqueous perfusate on a closed circuit. There are about 6.2 million individuals in the U.S. total with heart failure (Kilgore et al., 2017). About one-quarter of patients with heart failure are readmitted within thirty days of discharge, while one-quarter of hospital readmissions are preventable. Background The sugarcane aphid (SCA; Melanaphis sacchari) has emerged as a key pest on sorghum in the United States that feeds from the phloem tissue, drains nutrients, and inflicts physical damage to plants. Previously, it has been shown that SCA reproduction was low and high on sorghum SC265 and SC1345 plants, respectively, compared to RTx430, an elite sorghum male parental line (reference line). In this study, we focused on identifying the defense-related genes that confer resistance to SCA at early and late time points in sorghum plants with varied levels of SCA resistance.

AMD contaminants pose hazards to ecosystems and to human health by lowering the pH and by suspending toxic heavy metals in water. Over 50% of pregnant women are anemic and the majority of these are iron deficient. Micronutrient deficiency, the symptom of heavy menstrual bleeding in nonpregnant individuals, and loss of blood associated with pregnancy and obstetric delivery contribute to iron deficiency (ID). Poor outcomes with low maternal iron can affect not only the pregnancy but can also have major bearings on the offspring. True collaboration is a crucial component of implementing change and promoting a healthy work environment in the healthcare setting. This project highlights the significance of collaboration in implementing a standardized prenatal breastfeeding education curriculum. The outcome demonstrated successful improvement in breastfeeding initiation rates and achievement of Baby Friendly Hospital Designation.
