Bacterial cell division and the decision to divide is a process that requires a wide variety of factors. In bacterial cells, such as the widely studied Escherichia coli, a large protein complex, termed the Z-ring, assembles at midcell to enable division. The Z-ring is subject to many regulators which govern the spatiotemporal positioning of the ring. The Z-ring is made up of the eukaryotic tubulin homolog FtsZ, which polymerizes to recruit division proteins to the midcell. FtsZ polymers are tethered to the cytoplasmic membrane by the actin homolog FtsA. Together, FtsZ and FtsA act as the first step in …

In the spring of 2020, the COVID-19 pandemic rapidly brought college baseball to a halt. Most college baseball players saw disruption of their training and development due to interruptions of their seasons. With these interruptions, it is necessary to examine the effects of the COVID-19 pandemic on collegiate baseball injury epidemiology. Data from a small-school Midwest collegiate conference was tallied for the number of collegiate conference injuries per year, and for the injuries per anatomic region. What’s more, immigrants, territorial residents, and tribal members encounter deteriorated corners of the American welfare state. The present study examines the suitability of the DIBELS 8th edition assessment for use within the scope and context of the Energy Express summer literacy program. The 6-week summer program helps children in grades K-5 from rural and low-income West Virginia communities improve or maintain their reading levels over the summer months. Standard, grade-appropriate DIBELS 8th subtests were administered to 1,065 participants at the beginning and end of Summer 2022. The resulting pre- and post-scores were cross-referenced with demographic data and attendance rates from 20 counties and 35 sites in West Virginia. They were then analyzed for correlations …

This study aimed to evaluate and model the survival of Listeria monocytogenes on organic HoneyCrisp (HC) and Fuji (FJ) apples during storage at various temperatures. Fresh organic HC and FJ apples (without wax coating) obtained from a local wholesale market were inoculated with a 2-strain mix of L. monocytogenes followed by storage at 5.0 (22.9% RH), 12.0 (37.0% RH), and 22.5oC (50.4% RH) for 60, 35, 7 days, respectively, and periodically (day-0 to 60) analyzing microbial populations. A significant proportion of adults in the United States (US) suffer from various inflammatory chronic conditions (ICCs). They include a wide range of diseases such as diabetes, rheumatoid arthritis, asthma, and chronic obstructive pulmonary diseases. ICCs are not curable, and their management typically involves medications such as NSAIDs and corticosteroids. Recently, a whole health approach that places patients’ well-being at the center is increasingly being used for better management of ICCs. As protein aggregation is the defining hallmark of all amyloid diseases, a common therapeutic strategy is to develop molecules that inhibit aggregation. However, this approach has yielded limited success. Many amyloid proteins directly interact with lipid membranes. These interactions promote distinct aggregation pathways and often result in membrane damage leading to toxicity. As a result, directly targeting the ability of amyloids to bind lipid membranes represents a novel therapeutic strategy. As a proof of principle, the interaction between lipid membranes and mutant huntingtin protein (htt) aggregates was used to test this strategy. For women, two of the greatest risk factors for affective disorders are adversity experienced during puberty and later becoming pregnant. We have created a translationally relevant mouse model where we address these complex risk factors. Previously, we discovered that pregnant mice (dams) that experienced chronic variable stress (CVS) during puberty display a blunted hypothalamic-pituitary-adrenal (HPA) response when exposed to an acute stressor. Interestingly, this alteration only first becomes apparent during pregnancy, which is a sensitive period for these effects due to normative neuroendocrine changes. Ammonia is critical to supporting human life on earth because of its use as fertilizer. The Haber-Bosch process to produce ammonia has been practiced for over 100 years. This process operates at high pressure and temperature to overcome the thermodynamic and kinetic limitations of the ammonia synthesis reaction thus researchers have tried to overcome it for decades. At present this process represents 1% of global energy usage and 2.5% of global CO2 emissions. Efficient feed utilization is critical for both economic sustainability and environmental responsibility in modern livestock production. While there has been extensive research, the multifaceted nature of feed efficiency remains complex, with many biological factors still unexplored. This dissertation examines the physiological foundations of feed efficiency by elucidating some of the complex biological mechanisms associated with residual feed intake (RFI) phenotype in beef cattle, using a range of Omics approaches. Coenzyme A (CoA) is a crucial cofactor required to support many essential metabolic processes, including fatty acid oxidation and synthesis, the TCA cycle, and bile acid synthesis. In order to support the proper functioning of these pathways, CoA levels within the cell must be properly regulated. The regulation of CoA levels is achieved through a balance between CoA synthesis and CoA degradation. While the regulation of CoA synthesis has been extensively characterized, the degradation of CoA has received less attention. A rigorous research process is the foundation for the development and ultimate recommendation for use of dietary supplements and functional foods. This dissertation uses three seemingly disparate studies to explore the diverse research methodologies used in dietary supplement and functional food development and use. The first study focuses on primary research on the development of microgreen lemon balm herbal tea, employing experimental methodologies to assess its nutritional and bioactive properties compared to adult lemon balm herbal tea. Birdsfoot trefoil (Lotus corniculatus L.) (BFT) is a high quality, tanniniferous forage legume that has been shown to mitigate gastrointestinal nematode (GIN) infection in sheep. Reliable establishment of birdsfoot trefoil in existing pastures is difficult due to BFT’s poor seedling vigor, susceptibility to crown and root rot, and, perhaps chiefly, competition from other pasture plants. Most existing pastures in the Northeastern United States are primarily composed of cool-season grasses, many of which can be highly competitive. In Chapter 2, an experiment was conducted to evaluate the effect of different mixer types (MTY) and mix times (MTI) on mix uniformity (MU). A 2 (mix time) x 3 (mixer type) factorial was utilized in this experiment to create six treatments, each mixed in three randomized complete blocks. Mix times were designated adequate or inadequate, were based on the manufacturers’ recommendation, and varied based on the mixer utilized. Fresh produce and poultry have been associated with several foodborne illness outbreaks. Strategies to mitigate these risks are a much-needed research priority; therefore, five experiments were conducted to analyze pathogen cross-contamination capabilities on produce and poultry. This dissertation explores the identity and role of immunoglobulin-like (Ig-like) receptors in chickens, with focus on their implications in disease and disease progression. These receptors, wisely expressed across immune cells, interact with the major histocompatibility complex (MHC) class I molecules to modulate immune responses in mammals. The purpose of this study was to identify how integrating learning theories into the design of a formal university course helps facilitate students’ resources, goals, and orientations (Schoenfeld, 2011) of learning theories for their future career work in coaching, sport psychology consulting, or other sport-related professions. Sixteen students signed up for a fifteen-week fall semester course at a Mid-Atlantic university in America and were asked to annotate, create, and reflect upon examples of future work in their desired fields for their three major written assignments in the course. Resistance training is related to disease prevention, improved health, and positive psychosocial well-being in adults and adolescents. Further, resistance training is positively associated with body image and self-esteem. However, most adolescents do not meet activity guidelines, particularly for resistance training. Schools play an essential role for increasing physical activity. Little is known about the effects of different classroom formats for resistance training, i.e., single-gendered versus coeducational, on body image and self-esteem in female adolescents.

Background: Intestinal polyps are noticeable mucosal outgrowths that extend into lumen of gastrointestinal tract. In adults, the most frequently detected polyps are adenomatous polyps which are thought to be precursors of large bowel cancer. Colonoscopy is the most widely accepted method for colon cancer screening. Congenital heart defects (CHD) are the most common developmental abnormalities seen at birth. Therefore, it is critical to understand the molecular mechanisms that ensure cardiogenesis proceeds correctly. These mechanisms include those facilitated by the essential evolutionarily conserved Trithorax Group (TrxG) and Polycomb Group (PcG) protein complexes that govern proper gene expression during development and cardiogenesis. In this paper, a new pharmaceutical cocrystal was synthesized using apigenin (AP) and pharmaceutically acceptable conformer nicotinamide (Nico), and the drug delivery between AP-Nico pharmaceutical cocrystal and human serum albumin (HSA) in vivo was studied at atomic scale. The pharmaceutical cocrystal was characterized using Fourier-transform infrared (FTIR) spectroscopy, 1H NMR spectroscopy, differential scanning calorimetry (DSC), and powder X-ray diffraction (PXRD), and the self-assembling mechanism was explored. The dissolution and cumulative release in vitro were investigated. This paper focuses on the effect of okra bast fillers on biodegradation of poly(vinyl alcohol) composites. Fibers were obtained from okra plant stems via biological degumming and subjected to different chemical treatments such as scouring, alkalizing, maleic anhydride treatment, and vinyl acetate grafting. The fibers were ground and PVA-okra bast filler composites were produced at 20 wt% fiber load via the solution casting method. The obtained composites were tested in terms of mechanical properties and exposed to biodegradation in soil. Dietary patterns are inherently related to greenhouse (GHG) emissions via agricultural practices and food production systems. As the global population is predicted to increase from 8 billion (current) to 9.6 billion by 2050 added pressure will be placed on existing agricultural systems, resulting in increased GHG emissions thus exacerbating climate change.
