Effect Of Reduced Cardiomyocyte O-Glcnacylation On The Expression And Activity Of The Cardiac Voltage-Gated Sodium Channel, Chiagozie Dawn Bede Paul-Onyia

Browse all Theses and Dissertations

Abnormal glycosylation can impact cardiac proteins, affecting signaling, contraction, and metabolism, which leads to compromised cardiac function. O-GlcNAcylation, the only form of intracellular glycosylation, involves adding a single GlcNAc molecule to proteins via enzymes: O-GlcNAc Transferase (OGT) for addition and O-GlcNAcase (OGA) for removal. The Ednie/Bennett lab has previously demonstrated that in both chronic (OGT KO) and acute (OGT inhibition) models of reduced cardiomyocyte O-GlcNAcylation, a disruption of Ca2+ homeostasis occurs. This study delved into the cardiac voltage-gated sodium channel (NaV 1.5) in these models. NaV 1.5 expression was analyzed through western blotting and immunocytochemistry, while its functional activity was …

Novel Formulation Strategies With Hot-Melt Extrusion Technology To Address Physicochemical And Physiological Constraints For Improved Oral Drug Delivery, Sagar Narala

Electronic Theses and Dissertations

Most current active pharmaceutical ingredients (APIs) exhibit poor water solubility, necessitating higher doses or repeated administration. Numerous methods were explored to augment solubility, including but not limited to amorphous solid dispersions, cocrystals, salts, and nanoformulations. The hot-melt extrusion (HME) technique has gained immense popularity due to its solvent-free and continuous manufacturing capabilities, contributing to its increasing use to enhance solubility and bioavailability. In the first chapter, hydrochlorothiazide (BCS-IV class) cocrystals were produced using coformers such as nicotinamide (NIC), resorcinol (RSL), and catechol (CAT) through HME technology. The cocrystals were compared against liquid-assisted grinding (LAG) cocrystals and then analyzed by DSC, …

A Discrete Conley-Zehnder Index For Symplectic Polytopes, Yutian Yu Jan 2023

A Discrete Conley-Zehnder Index For Symplectic Polytopes, Yutian Yu

Electronic Theses and Dissertations

A Hamiltonian system is a dynamical system governed by Hamilton’s equations. Hamiltonian systems provide a framework for studying classical mechanics. One fundamental principle in Hamiltonian mechanics is the conservation of energy along the flow of the Hamiltonian vector field. The smooth hypersurface of a Hamiltonian function on a symplectic manifold has a contact structure, and the associated Reeb vector field is, up to a scale, the Hamiltonian vector field. We are interested in the non-smooth case and a symplectic polytope makes a good model to study.

In this dissertation, we study the Reeb dynamics on the boundary of a symplectic …

Textural And Mineral Analysis Of Pegmatites From The Pala And Mesa Grande Districts In San Diego, California, Gabrielle Potter Jan 2023

Textural And Mineral Analysis Of Pegmatites From The Pala And Mesa Grande Districts In San Diego, California, Gabrielle Potter

Williams Honors College, Honors Research Projects

San Diego pegmatites of the Pala and Mesa Grande mining districts are world renowned for their gem-quality minerals and their Li-bearing mineral phases. Pegmatite genesis, especially in San Diego mines, has been a debated topic since the 1900’s (Morgan and London, 2012). Pegmatite genesis is caused by partial melting processes and fractional crystallization that form granitic melts with high concentrations of rare earth elements (REE), such as lithium. Assessing the exact mineralogy and determining the overall textures can provide insight into the crystallization history of pegmatites. In the San Diego pegmatites, lithium-bearing phases, such as lepidolite and pink elbaite (a …

The diffusion in and out of these vesicles will be tracked by using singular lipid molecules. These singular lipid molecules will be tagged with fluorescence and viewed using an internal reflection fluorescence microscope. The data collected over the course of this project will then be analyzed using Matlab. The result of the project will be to find the specific lipid diffusion constant of these unilaminar vesicles. The relationship between the concentration of DHA and lipid diffusion and membrane fluidity will be found as …

Global Minimum Search For The Largest Ge Cage Stabilized By A Single Interstitial Metal Atom, Alexis Kiefer Jan 2023

Global Minimum Search For The Largest Ge Cage Stabilized By A Single Interstitial Metal Atom, Alexis Kiefer

Williams Honors College, Honors Research Projects

Recent joint experimental and theoretical studies showed that the largest Ge cage-like cluster features 14 Ge atoms that encapsulate a single transition metal atom in the [Nb@Ge14]3- cluster. The next larger experimentally confirmed Ge cluster exhibiting a cage-like geometry was found to be viable only when stabilized by two transition metal atoms in the [Co2@Ge16]4- cluster. So, as of now, it is not clear whether a cage-like cluster made out of 15 Ge atom stabilized by a single transition metal is viable. Hence, the aim of this project is to find the …

Intergranular And Pitting Corrosion Mechanisms Of Sensitized Aluminum Alloy Aa5083, Clayton Egleston Jan 2023

Intergranular And Pitting Corrosion Mechanisms Of Sensitized Aluminum Alloy Aa5083, Clayton Egleston

Williams Honors College, Honors Research Projects

The motivation and objectives of this project is to examine the mechanisms of intergranular corrosion (IGC) and pitting corrosion of sensitized AA5083. In this regard, different characterization techniques were used, including optical analysis of microstructure, cyclic potentiodynamic polarization with Tafel fitting, electrochemical impedance spectroscopy with electrical equivalent circuit (EEC) fitting, and potentiostatic current transient monitoring. The transition from IGC to pitting corrosion occurs when the grain boundaries become saturated with the β-phase (Mg2Al3). It was found that AA5083 becomes more vulnerable to pitting corrosion as the degree of sensitization increases.

Growing In Stem: Hands-On Education Via Tower Gardening, Jeffrey A. Nowak, Shelby Johnston-Werling

Indiana STEM Education Conference

Sole use of a textbook as the primary source of learning in the middle school setting has been linked to potentially limit students’ conceptual understanding of complex content (Driscoll et al., 1994), while the positive impact of hands-on learning has been well documented (Ekwueme et al., 2015; Satterthwait, 2010). Ideally, when a textbook is used in conjunction with content applied in a hands-on manner, student engagement and understanding of standards-based content are increased. In a grant funded collaboration between the Northeast Indiana STEM Education Resource Center housed at Purdue University Fort Wayne and an East Allen Community Schools middle school …

Improving Developers' Understanding Of Regex Denial Of Service Tools Through Anti-Patterns And Fix Strategies, Sk Adnan Hassan, Zainab Aamir, Dongyoon Lee, James C. Davis, Francisco Servant

Department of Electrical and Computer Engineering Faculty Publications

Regular expressions are used for diverse purposes, including input validation and firewalls. Unfortunately, they can also lead to a security vulnerability called ReDoS (Regular Expression Denial of Service), caused by a super-linear worst-case execution time during regex matching. Due to the severity and prevalence of ReDoS, past work proposed automatic tools to detect and fix regexes. Although these tools were evaluated in automatic experiments, their usability has not yet been studied; usability has not been a focus of prior work. Our insight is that the usability of existing tools to detect and fix regexes will improve if we complement them …

Department of Electrical and Computer Engineering Faculty Publications

Deep Neural Networks (DNNs) are being adopted as components in software systems. Creating and specializing DNNs from scratch has grown increasingly difficult as state-of-the-art architectures grow more complex. Following the path of traditional software engineering, machine learning engineers have begun to reuse large-scale pre-trained models (PTMs) and fine-tune these models for downstream tasks. Prior works have studied reuse practices for traditional software packages to guide software engineers towards better package maintenance and dependency management. We lack a similar foundation of knowledge to guide behaviors in pre-trained model ecosystems.

In this work, we present the first empirical investigation of PTM reuse. …

Teachers' Beliefs As A Tool To Extend The Cognitive Equity Concept, Ana-Maria Haiduc, Rose Mbewe, Signe E. Kastberg Jan 2023

Teachers' Beliefs As A Tool To Extend The Cognitive Equity Concept, Ana-Maria Haiduc, Rose Mbewe, Signe E. Kastberg

Indiana STEM Education Conference

This research aims to extend the framing of the cognitive equity concept in teaching mathematics during the enacted phase of the curriculum beyond race and culture. Cognitive equity characterizes actions of teachers and curricula makers toward valuing differences in cognitive processes. Since beliefs influence teachers' actions, we investigate their ideas about valuing students' unique mathematical cognitive processes. Framing cognitive equity in teaching mathematics contributes to discovering teachers' equitable practices, thus revealing teachers' ways of making more inclusive cognitive curricular decisions. Future research could focus on framing cognitive equity in learning mathematics within the intended and implemented phase of the curriculum.

Nowadays, multiphase transition metal-based AB2 metal hydride alloys and the rare-earth metal-based AB5 alloys (currently used in consumer NiMH batteries) are used as negative electrodes in NiMH batteries. It is of great interest to explore the possibility of significantly improving NiMH battery performance to compete with Li-ion batteries. In this area, the effect of dopant elements from Group-IA, -IIA, -IIIA non-transition metal family has not been thoroughly studied. In this study, lithium, magnesium, and boron (a metalloid considered metal in this study) were chosen as modifiers in the C14-phase predominant AB2 alloy by different dopant concentrations and synthesis methods, doping …

Improving Lidar Fidelity For Hd Mapping Using Pattern Transplanting And Temporal Infill, Micah Bojrab Jan 2023

Improving Lidar Fidelity For Hd Mapping Using Pattern Transplanting And Temporal Infill, Micah Bojrab

Wayne State University Dissertations

High Definition (HD) Mapping for Autonomous Driving creates an easily accessible wealth of information to confirm and enrich feedback from on-vehicle sensors. HD Maps built offline generally use dedicated ground collection vehicles equipped with high-cost, survey-grade LiDAR to accurately map a vehicle's areas of operation. We focus our work on controlled-access, divided highways characterized by wide roadways that require complex permutations to cover all directions of travel and access ramps. We study two common issues when collecting this road class that cause low-fidelity (LF) point clouds for HD Maps. First, aerial collection covers all directions and all ramps in a …

Sensemaking Of Michigan's Read By Grade Three Law, Amy Catherine Hawtin Jan 2023

Sensemaking Of Michigan's Read By Grade Three Law, Amy Catherine Hawtin

Wayne State University Dissertations

Research suggests that school leaders’ understanding and process of making sense of educational reform policies and/or laws is shaped by social, organizational, individual factors. This understanding can impact how a school leader understands and interprets the law/policy and ultimately may impact implementation actions. In 2016, Michigan’s Read by Grade Three law was enacted into law, which requires districts to identify struggling readers and provide tiered reading intervention for those students requiring additional support. Furthermore, the law also outlines mandatory eligible retention for students achieving below the pre-determined cut-score on the annually administered Michigan Student Test of Educational Progress (M STEP) …

Predictable Dnn Inference For Autonomous Driving, Liangkai Liu Jan 2023

Predictable Dnn Inference For Autonomous Driving, Liangkai Liu

Wayne State University Dissertations

Deep neural networks (DNNs) are widely used in autonomous driving due to their high accuracy for perception, decision, and control. Predictability of the perception module is essential for the AV's safety. Predictability generally consists of two aspects: temporal and functional. Temporal aspects mean the task should be finished before the deadline. Functional aspects mean the task should make correct decisions. However, non-negligible time and performance variations are observed in DNN inference. Current DNN inference studies either ignore the variation issue or rely on the scheduler or the algorithm itself to handle it. None of the current work explains the roots …

Observing the connection between complexity theory and the K-12 education field, I conducted an extensive literature review to discover much research had already been undertaken. However, given the pervasive, wicked problems that still exist, there may be gaps in the operationalization of the findings. The purpose of this research is to determine how school leaders understand the complexity of their school system and if examining school leadership from a complexity lens may uncover practices that could create more effective school operations in dynamic environments. Ten school principals and leaders were interviewed. Five major themes were derived from the data: a) …

Molecular Dynamics Simulations For The Mechanical Behavior Of Alpha Quartz With Void Defects Under Tension, Yu Jia Jan 2023

Molecular Dynamics Simulations For The Mechanical Behavior Of Alpha Quartz With Void Defects Under Tension, Yu Jia

Electronic Theses and Dissertations

The structural and mechanical properties of void-free and single-void α-quartz were investigated using MD simulations with three different interatomic potentials (BKS, Vashishta and Tersoff). Two distinct ensembles, NVT and NPT, were separately applied to investigate the tensile response of α-quartz under uniaxial strain and uniaxial stress states. A comprehensive comparison among different potentials, as well as comparison with currently available experiments, has been made. The BKS and Vashishta potentials accurately predicted the structural properties of void-free α-quartz, while the Tersoff potential was deviated significantly from the experimental data. Under tension, the BKS did a better description for the mechanical performance …
