Functional Characterization Of Pacific White Shrimp Litopenaeus Vannamei Heat Shock Protein 90 In Response To White Spot Syndrome Virus Infection, Warumporn Yingsunthonwattana
Chulalongkorn University Theses and Dissertations (Chula ETD)
Heat shock proteins (HSPs) are host chaperones that can be used by viruses to facilitate various steps of viral infections. In this study, we found that Litopenaeus vannamei heat shock protein 90 (LvHSP90) is a viral-responsive protein, due to a significant transcriptional and translational upregulation following white spot syndrome virus (WSSV) infection. Suppression of LvHSP90 significantly reduced WSSV infection which was corresponded with significantly decreased expressions of viral genes, including the immediate-early genes and viral DNA polymerase. Conversely, injecting shrimp with WSSV that had been co-incubated with a recombinant LvHSP90 (rLvHSP90) promoted WSSV infection as evidenced by an increased cumulative …

Crispr-Cas12a-Based Detection And Differentiation Of Mycobacterium Spp., Peeraphan Compiro
Chulalongkorn University Theses and Dissertations (Chula ETD)
Mycobacterium species cause several vital human diseases, including tuberculosis (TB) and non-tuberculous mycobacterial (NTM) infections. TB is a chronic infectious disease caused by the Mycobacterium tuberculosis complex (MTBC). Additionally, non-tuberculous mycobacteria infections are caused by major NTM species, including Mycobacterium abscessus, Mycobacterium avium, Mycobacterium fortuitum, Mycobacterium kansasii, Mycobacterium gordonae, and Mycobacterium intracelulare, are medically necessary. Both groups of infections can lead to outbreaks in immunocompromised individuals and AIDS patients. It is crucial to differentiate between these two groups of mycobacterial infections, as they require distinct treatment approaches. However, initial diagnosis poses limitations in terms of time and diagnostic tools. Therefore, …

Development Of Duplex Loop-Mediated Isothermal Amplification (Dlamp) For Detection Of Carbapenem Antibiotic-Resistant Genes Kpc And Ndm, Nindi Syahputri Lubis
Chulalongkorn University Theses and Dissertations (Chula ETD)
Carbapenems are regarded as a last-resort option for treating a wide range of Gram-positive and Gram-negative bacterial infections. Unfortunately, the prevalence of carbapenem-resistant genes has been on the rise among patients. Some factors contribute to this trend, including prolonged hospital stays, prior antibiotic usage, inappropriate or insufficient antibiotic treatment, and contamination through wounds or feces. The emergence of carbapenem-resistant (CR) genes gained significant attention, particularly after 2005. These CR genes are frequently carried on mobile genetic elements such as plasmids or transposons, enabling their transmission to other bacteria. Klebsiella pneumoniae carbapenemase (KPC) and New delhi metallo-β-lactamase (NDM) stand out as …

Background: Nanopore sequencing enables rapid DNA/RNA analysis without amplification. Implementing this for point-of-care testing reduces time delays in traditional AST methods, potentially enhancing patient outcomes. We aim to evaluate the performance of Oxford Nanopore Technologies sequencing from positive blood culture for bacterial identification and antimicrobial susceptibility prediction in S. aureus and S. epidermidis by detecting the mecA gene. Methods: We conducted a cross-sectional descriptive study on individuals with positive blood cultures identifying S. aureus and S. epidermidis over a 12-month period from April 2023 to March 2024. Bacterial DNA was extracted from the blood culture specimens, and then sequenced in …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Tuberculosis (TB) is one of the 13th leading causes of death from a single infectious agent in the world in 2019. Some of the factors that influence the tuberculosis incident in the world are caused by undernourishment, HIV infection, alcohol use disorders, smoking, and diabetes. The median prevalence of diabetes data from the Global Health Observatory in the 30 high TB burden countries approximated 8% (Interquartile range [IQR]: 6-9%) and more than >=10 % in other countries in Africa and Papua New Guinea. It is also predicted that between 2019 and 2045 the statistic of diabetes will increase approximately 50% …

Correlation Between Meteorological Conditions, Pm2.5, And Covid-19 Mortality In Thailand, Viranphat Singrakphon
Chulalongkorn University Theses and Dissertations (Chula ETD)
The appearance of a new strain of coronavirus led to a worldwide health crisis, affecting public health and economies globally, including Thailand since 2019. Previous research had suggested possible links between environmental factors and the surge in COVID-19 cases, but these connections remained uncertain. This study examined the ratios of indoor to outdoor (I/O) environments and investigated potential associations between fine particulate matter (PM2.5), meteorological conditions, and the Case Fatality Rate (CFR%) of COVID-19 in Bangkok and its surrounding metropolitan area from January to December 2021. Through Spearman’s Rank correlation analysis, the findings indicated a positive correlation between CFR% and …

Identification And Characterization Of Bacteriophages For Phage Cocktail Formulation Specific To Uropathogenic Escherichia Coli, Pattida Kongsomboonchoke
Chulalongkorn University Theses and Dissertations (Chula ETD)
Urinary tract infections (UTIs) are commonly found in elderlies, infants, hospitalized patients, and most women. UTIs are caused by a variety of pathogens with uropathogenic Escherichia coli (UPEC) is mostly prevalence. Even though UPEC can be treated with several antibiotics, the rapid emergence of multidrug resistant (MDR)-UPEC renders the antibiotic treatment no longer successful. Therapy using phages, which are bacterial viruses, has been considered but it is limited by bacterial resistance that develops during evolution. Fortunately, the suppression of phage resistance can be improved when used several phages as a cocktail. Here, I aim to formulate an efficient phage cocktail …

The Diagnostic Value Of Bispectral Index For Assessing Sleep Quality After Elective Surgery In Intensive Care Unit, Pongpol Sirilaksanamanon
Chulalongkorn University Theses and Dissertations (Chula ETD)
Sleep abnormality is common in the intensive care unit (ICU). Monitoring and improving sleep quality may help recovery and improve outcomes from a major illness. Although polysomnography is a gold standard for measuring sleep quality, routine use is impractical due to cost and complexity. This study aimed to investigate the diagnostic accuracy of an alternative monitor, the Bispectral Index (BIS), compared to polysomnography for evaluating the quality of postoperative sleep in the ICU. This was a prospective observational study. Enrolled patients were monitored with both BIS and polysomnography during the first night after surgery. Results: Thirty-three patients were enrolled. The …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Depression is a mood disorder that is a significant risk factor for suicidal behaviors (SB). This study aimed to explore the influence of the potential factors of depression, encompassing adverse childhood experiences (ACEs), negative life events (NLEs), rumination, neuroticism, and the immune-inflammatory and nitro-oxidative (IO&NS) biomarkers on the severity of depression and SB among young adults. The effects of IO&NS biomarkers in psychiatric patients with SB were identified through systematic reviews and meta-analyses. A case-control study involving 118 participants was conducted to determine the association between psychological factors, including ACEs, NLEs, rumination, and neuroticism, and the depression phenome using partial …
Immunogenicity And Safety Of The Third Booster Dose With Homologous Or Heterologous Covid-19 Vaccine After Completing Primary Series Of Inactivated Vaccine (Coronavac) Or Adenovirus Vector Vaccine (Azd1222) In Thai Adults, Suvichada Assawakosri
Chulalongkorn University Theses and Dissertations (Chula ETD)
COVID-19 has spread across 200 countries, causing 705 million confirmed cases and 7 million deaths. Several vaccines have been approved by the World Health Organization to prevent the spread of the virus. The Omicron variant has become the predominant strain globally. Although most people have been vaccinated, primary doses of CoronaVac or AZD1222 may not provide adequate protection against the Omicron variants. Studies show that vaccine-induced immunity has declined over time, leading to increase of breakthrough infection cases. A heterologous booster administered after primary vaccination, is hypothesized to enhance the immune response against the newly emerged SARS-CoV-2 variant. This study …

Bezlotoxumab Binding Specificity To Toxin B Of Clostridium Difficile, Kun Karnchanapandh
Bezlotoxumab Binding Specificity To Toxin B Of Clostridium Difficile, Kun Karnchanapandh
Chulalongkorn University Theses and Dissertations (Chula ETD)
Clostridioides difficile infection (CDI) is one of the significant global health issues. Toxin B is the main target protein preventing CDI recurrence. Clinical research has shown that bezlotoxumab (Bez), a monoclonal antibody designed to prevent CDI recurrences, is less effective against the B2 strain compared to the B1 strain, resulting in a high recurrence rate. Bez functions by binding to the epitope 1 and 2 regions in the combined repetitive oligopeptide (CROP) domain. Some binding residues are distinctively different between B1 and B2 strains. In this work, our goal was to understand and compare the interaction of toxins B1 and …
