Intracellular Negative Feedback Mechanisms In Blubber And Muscle Moderate Acute Stress Responses In Fasting Seals, Jessica G. Avalos, Elizabeth R. Piotrowski, Allison D. Northey, Daniel E. Crocker, Jane I. Khudyakov

Pacific Faculty Work

Animals may limit the cost of stress responses during key life history stages such as breeding and molting by reducing tissue sensitivity to energy-mobilizing stress hormones (e.g. cortisol). We measured expression of genes encoding glucocorticoid receptor (GR, NR3C1), GR inhibitor (FKBP5) and cortisol-inactivating enzyme (HSD11B2) in blubber and muscle of northern elephant seals before and after stress axis stimulation by adrenocorticotropic hormone (ACTH) early and late in a fasting period associated with molting. ACTH elevated cortisol levels for >24 h and increased FKBP5 and HSD11B2 expression while downregulating NR3C1 expression in blubber and muscle, suggesting robust intracellular negative feedback in …

Faculty Research 2023

Three-dimensional (3D) organoid cultures are flexible systems to interrogate cellular growth, morphology, multicellular spatial architecture, and cellular interactions in response to treatment. However, computational methods for analysis of 3D organoids with sufficiently high-throughput and cellular resolution are needed. Here we report Cellos, an accurate, high-throughput pipeline for 3D organoid segmentation using classical algorithms and nuclear segmentation using a trained Stardist-3D convolutional neural network. To evaluate Cellos, we analyze ~100,000 organoids with ~2.35 million cells from multiple treatment experiments. Cellos segments dye-stained or fluorescently-labeled nuclei and accurately distinguishes distinct labeled cell populations within organoids. Cellos can recapitulate traditional luminescence-based drug response …

A New Cobell: The Need For A Continued Buy-Back Program, Liam C. Conrad Dec 2023

A New Cobell: The Need For A Continued Buy-Back Program, Liam C. Conrad

American Indian Law Journal

The General Allotment Act of 1887 divided Indian reservations into smaller plots for the supposed benefit of individual Indians. Today, these allotments are severely fractionated, with some 160-acre plots having as many as a thousand owners. Since allotment, Congress has repeatedly attempted to solve this problem. However, only the Cobell Land Buy-Back Program has made any sizeable impact on fractionation levels. This paper examines the fractionation problem and the Cobell Program. Now that the Cobell Program has ended in November 2022, this paper argues that Congress must quickly reauthorize a similar program or fractionation will soon exceed pre-Cobell levels.

Mechanical Characterization Of Vertically Aligned Carbon Nanotube Forest Microelectrodes For Neural Interfacing, Spencer Mcleod Roberts Dec 2023

Mechanical Characterization Of Vertically Aligned Carbon Nanotube Forest Microelectrodes For Neural Interfacing, Spencer Mcleod Roberts

Theses and Dissertations

Silicon intracortical microelectrodes arrays (MEA) provide some of the best performance for brain computer interfaces but suffer from lifetime limitations that prevent clinical adoption. A primary contributor of lifetime limitations is the chronic foreign body response (FBR) which remains active in response to the brain tissue strain caused by implanted probes during brain micromotion. Designing probes with softer materials can reduce the chronic FBR and improve device lifetime. A high-aspect ratio CNT MEA could provide several advantages as an intracortical MEA but has yet to be mechanically characterized. CNT MEAs are infiltrated with 3 degrees of carbon infiltration and mechanically …

Evaluating Principal Effectiveness: A Review Of The Literature, Jerry Burkett

Journal of Educational Leadership in Action

The evaluation of a campus principal can be a challenging process due largely to the complicated factors that exist to capture an accurate assessment of a principal’s leadership effectiveness. Generally, principal evaluations are conducted by district-level officials who often do not have the time or the resources to observe campus principals on a regular basis. Further, principal evaluation systems (PES) are designed to improve the practice of principals (Clifford & Ross, 2012; Davis, Kearney, Sanders, Thomas, & Leon, 2011; Fuller & Hollingworth, 2014a; Fuller et al., 2015), and there has historically been an acknowledgment that these evaluations do not always …

The Contested Exempla Of Lucius Cornelius Sulla, Victoria Theresa White Dec 2023

The Contested Exempla Of Lucius Cornelius Sulla, Victoria Theresa White

Dissertations and Theses

In 82 BCE, Lucius Cornelius Sulla emerged the victor of his civil war against the supporters of Marius and Cinna. The brutal aftermath of this victory, characterized by the creation of the proscriptions, would brand Sulla as a cruel and immoderate victor. Still, his successes in Rome’s previous wars against foreign enemies had enshrined Sulla as a brilliant military commander within Roman memory. Previous scholarship about Sulla has focused on the dichotomy of Sulla’s actions, often in order to evaluate his clemency or cruelty and his aims of his political reforms. This thesis analyzes how ancient Roman and Greek authors …

Theses and Dissertations

This work contains two key contributions to the development of 3D displays. First a successful exploratory human trial research study. By comparing multiple real displays in human trials, the factors of ease of use, time, and accuracy were able to be examined in a general setting, instead of isolated to only a specific type of 3D display. These human tests showed that 3D displays in general offer better ease of use and accuracy compared to 2D displays when showing 3D media. This differs from previous studies where only a single display was examined or theoretical papers discussing factors on paper. …
