Structural Biology Of Mcr-1-Mediated Resistance To Polymyxin Antibiotics Oct 2023

Structural Biology Of Mcr-1-Mediated Resistance To Polymyxin Antibiotics

Faculty, Staff and Students Publications

Polymyxins, a last resort antibiotic, target the outer membrane of pathogens and are used to address the increasing prevalence of multidrug-resistant Gram-negative bacteria. The plasmid-encoded enzyme MCR-1 confers polymyxin resistance to bacteria by modifying the outer membrane. Transferable resistance to polymyxins is a major concern; therefore, MCR-1 is an important drug target. In this review, we discuss recent structural and mechanistic aspects of MCR-1 function, its variants and homologs, and how they are relevant to polymyxin resistance. Specifically, we discuss work on polymyxin-mediated disruption of the outer and inner membranes, computational studies on the catalytic mechanism of MCR-1, mutagenesis and …

Bioresorbable perivascular scaffolds loaded with antiproliferative agents have been shown to enhance arteriovenous fistula (AVF) maturation by inhibiting neointimal hyperplasia (NIH). These scaffolds, which can mimic the three-dimensional architecture of the vascular extracellular matrix, also have an untapped potential for the local delivery of cell therapies against NIH. Hence, an electrospun perivascular scaffold from polycaprolactone (PCL) to support mesenchymal stem cell (MSC) attachment and gradual elution at the AVF's outflow vein is fabricated. Chronic kidney disease (CKD) in Sprague-Dawley rats is induced by performing 5/6th nephrectomy, then AVFs for scaffold application are created. The following groups of CKD rats are …

(1) Background: We previously reported the development of a recombinant protein SARS-CoV-2 vaccine, consisting of the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein, adjuvanted with aluminum hydroxide (alum) and CpG oligonucleotides. In mice and non-human primates, our wild-type (WT) RBD vaccine induced high neutralizing antibody titers against the WT isolate of the virus, and, with partners in India and Indonesia, it was later developed into two closely resembling human vaccines, Corbevax and Indovac. Here, we describe the development and characterization of a next-generation vaccine adapted to the recently emerging XBB variants of SARS-CoV-2. (2) Methods: We conducted preclinical studies …

Idiopathic pulmonary fibrosis (IPF) is a progressive fatal interstitial lung disease without an effective cure. Herein, we explore the role of 3,5,3′-triiodothyronine (T3) administration on lung alveolar regeneration and fibrosis at the single-cell level. T3 supplementation significantly altered the gene expression in fibrotic lung tissues. Immune cells were rapidly recruited into the lung after the injury; there were much more M2 macrophages than M1 macrophages in the lungs of bleomycin-treated mice; and M1 macrophages increased slightly, whereas M2 macrophages were significantly reduced after T3 treatment. T3 enhanced the resolution of pulmonary fibrosis by promoting the differentiation of Krt8+ transitional …

Hmg-Coa Reductase Inhibitors For Traumatic Brain Injury, Claudia Robertson

Faculty, Staff and Students Publications

Traumatic brain injuries (TBIs) are associated with high morbidity and mortality due to both the original insult as well as the destructive biological response that follows. Medical management aims to slow or even halt secondary neurological injury while simultaneously laying the groundwork for recovery. Statins are one class of medications that is showing increased promise in the management of TBI. Used extensively in cardiovascular disease, these drugs were originally developed as competitive inhibitors within the cholesterol production pipeline. They are now used in diverse disease states due to their pleiotropic effects on other biological processes such as inflammation and angiogenesis. …

Neuromodulation For Adjunctive Treatment In Postmastectomy Pain Syndrome

Faculty, Staff and Students Publications

Postmastectomy pain syndrome (PMPS) affects nearly half of patients who undergo mastectomy to treat breast cancer. As the survival rate of breast cancer increases with advancements in treatment, the incidence of PMPS is also increasing. Patients with PMPS can experience unrelenting, chronic pain refractory to traditional management with oral pharmacotherapy in conjunction with nonpharmacologic treatment (physical therapy, transcutaneous electrical nerve stimulation (TENS)). Neuromodulation is an emerging treatment modality for numerous chronic pain conditions. This case report highlights the tremendous success of spinal cord stimulator placement for a patient with PMPS.

The molecular mechanisms and evolutionary changes accompanying synapse development are still poorly understood1,2. Here we generate a cross-species proteomic map of synapse development in the human, macaque and mouse neocortex. By tracking the changes of more than 1,000 postsynaptic density (PSD) proteins from midgestation to young adulthood, we find that PSD maturation in humans separates into three major phases that are dominated by distinct pathways. Cross-species comparisons reveal that human PSDs mature about two to three times slower than those of other species and contain higher levels of Rho guanine nucleotide exchange factors (RhoGEFs) in the …

Absolution: Divine And Human, Cathal Doherty Oct 2023

Absolution: Divine And Human, Cathal Doherty

Theology Faculty Research and Publications

This article is a contribution to an ecumenical theology of the Word, examining how divine absolution is realized in concrete human experience. It addresses both the scriptural and sacramental mediation of divine absolution, emphasizing the commonality of both resting on the power of God's word spoken in human concrete actuality. The philosophical literature concludes that human beings, in order for forgiveness to be real to them, need to hear and know that they are forgiven. In other words, for forgiveness to be complete, the wrongdoer requires an act of “absolution” (even in a secular sense), which may be verbal or …

Coronary artery calcification (CAC), a measure of subclinical atherosclerosis, predicts future symptomatic coronary artery disease (CAD). Identifying genetic risk factors for CAC may point to new therapeutic avenues for prevention. Currently, there are only four known risk loci for CAC identified from genome-wide association studies (GWAS) in the general population. Here we conducted the largest multi-ancestry GWAS meta-analysis of CAC to date, which comprised 26,909 individuals of European ancestry and 8,867 individuals of African ancestry. We identified 11 independent risk loci, of which eight were new for CAC and five had not been reported for CAD. These new CAC loci …

Broodmother: What The Lack Of Bathroom Breaks Reveals About Dehumanizing Teachers, Jonathan Michael Coker

Educational Leadership

This chapter asks, “What is it like to be a public school teacher in a school that doesn't allow for adequate bathroom breaks?” Using van Manen's interpretive phenomenological methodology, the authors analyzed interviews from nine participants to create an empathetic portrayal of teachers who have inadequate bathroom breaks. These experiences were then distilled into a series of insights through the concept of “broodmothering.” These broodmothers deny their own physical and mental needs in the interest of supervising children, and show a profession burdened by increasing parentification and dehumanization of teachers. After connecting this phenomenon to larger issues in the profession, …

A Mindful Search For Basic Writing Identities: Constellating Contemplative Classroom Pedagogies, Classroom Community Circles, And Critical Discourse Analysis In A Cultural Rhetorics Ecology, Elizabeth Edwards Weems

English Theses & Dissertations

Many Basic Writing students have stories which are informed by traumas, othering, and marginalization. Finding pedagogies which address students’ stories, in addition to meeting academic goals, is crucial. Sometimes, students question how improving their reading and writing skills can help them navigate other areas of their education and experiences, especially their lives outside of academia. I have worked to create a kairotic pedagogy which invites students to read and write ‘the word and the world’ (Freire, 1970; 1988). Using Contemplative Classroom Pedagogies, I attempted to follow Freire’s (1992) call to make “teaching a creative act, a critical act, and not …

Dramatic Storytelling Develops Emergent Literacy In Efl: Evidence From The Uae Kindergartens, Ghada Yahya Alkilani

Dissertations

There is increasing importance of the English language for educational excellence and career success in the UAE. The early years is a sensitive period for language acquisition, and preschool age is the best time to learn English as a foreign language (EFL). Stories are a main vehicle used in early childhood education (ECE) to engage children in language learning. Although previous studies have found dramatic storytelling to be particularly effective in developing children’s emergent literacy in English as a native language, little is known about the effect of dramatic storytelling on developing young children’s emergent literacy in EFL. Also, there …

Identification Of A Potential Pr50gag Binding Site Critical For Selective Encapsidation Of Feline Immunodeficiency Virus (Fiv) Genomic Rna, Anjana Krishnan Oct 2023

Identification Of A Potential Pr50gag Binding Site Critical For Selective Encapsidation Of Feline Immunodeficiency Virus (Fiv) Genomic Rna, Anjana Krishnan

Dissertations

Feline immunodeficiency virus (FIV) is an important lentivirus that causes acquired immunodeficiency syndrome (AIDS) in cats, making FIV a potential small animal model for studying the pathogenesis of human AIDS. Furthermore, due to its phylogenetic distance from primate retroviruses like human immunodeficiency virus (HIV) and its ability to infect non-dividing cells, FIV is also being exploited for the development of vectors for human gene therapy. Therefore, it is of paramount importance that pertinent steps in the life cycle of FIV, such as genomic RNA (gRNA) packaging, are delineated at the molecular level. The process of retroviral gRNA packaging relies on …

Nerds V. Nintendo: Video Game Decompilations Versus Rights-Holder Interests, Kirk A. Sigmon

Cardozo Law Review

Video game “decompilations”—a potentially technically inaccurate term referring to fan efforts to entirely reprogram video games based on reverse engineering those games—present an interesting case study for evaluating the scope of video game copyrightability, fair use, and public expectations about content availability. Decompilations usually comprise entirely new code and do not comprise any assets of the original video game, suggesting that the decompilations, if viewed as mere code, do not apparently infringe any video game copyrights. That said, decompilations illustrate why copyright protects more than the discrete assets (e.g., art or music) of a video game, as decompilations are generally …

Beneficial Conflicts Of Interest, Anita K. Krug Oct 2023

Beneficial Conflicts Of Interest, Anita K. Krug

Cardozo Law Review

Conflicts of interest exist in both professional and private settings, and everyone experiences them from time to time. If a person harboring a conflict acts on it—meaning the person acts against interests she ought to uphold—innocent parties may be harmed. Accordingly, the key to addressing a conflict in most settings is to eliminate it, such as by prohibiting conflicted behavior or recusing oneself from a deliberative process. However, conflicts of interest have a special character in the securities realm, both because they are ubiquitous given financial firms’ myriad competing interests and because the goal of the agency charged with addressing …

Reforming The Mississippi Criminal Code Part Iv: Offenses Against Property; Theft And Related Crimes, Judith J. Johnson Oct 2023

Reforming The Mississippi Criminal Code Part Iv: Offenses Against Property; Theft And Related Crimes, Judith J. Johnson

MC Law Review

Clear and fair criminal laws are foundational to criminal justice, and any meaningful reform effort should begin with the criminal laws. The Mississippi Code has been justifiably criticized as often being neither clear nor fair. This article about reforming the theft crimes is the fourth in a series of articles advocating for change to the Mississippi criminal laws. The first article explained why change is needed. Briefly, Mississippi criminal laws have been justifiably criticized because of gross sentencing disparities, vague definitions of the conduct prohibited, as well as confusing or absent definitions of states of mind required to commit the …
