In phenylketonuria (PKU), an important component of the UK dietary management system is a 50 mg phenylalanine (Phe)/1 g protein exchange system used to allocate the Phe/natural protein intakes according to individual patient tolerance. Any foods containing protein ≤ 0.5 g/100 g or fruits/vegetables containing Phe ≤ 75 mg/100 g are allowed without measurement or limit. In children with PKU, we aimed to assess the difference between the prescribed natural protein intake and their actual consumed intake, and to calculate the natural protein/Phe intake from foods given without measurement or restriction. Objectives Health and care resources are under increasing pressure, partly due to the ageing population. Physical activity supports healthy ageing, but motivating exercise is challenging. We aimed to explore staff perceptions towards a virtual reality (VR) omnidirectional treadmill (MOTUS), aimed at increasing physical activity for older adult care home residents. Design Interactive workshops and qualitative evaluation. Settings Eight interactive workshops were held at six care homes and two university sites across Cornwall, England, from September to November 2021. This research underscores the critical role of Tennessee's transportation network, particularly its heavy reliance on trucking along rural roads, in facilitating the movement of goods and significantly impacting the state's economy. It emphasizes the need to comprehend the implications of freight transportation on both urban and rural areas, shedding light on the latter's frequent neglect despite substantial impacts. To bridge this disparity, the research focuses on evaluating and mitigating freight-related impacts in suburban and rural communities. This study interrogates the ways wilderness spaces become understood as sacred places through rhetoric, using Grandfather Mountain in Western North Carolina as a case study. Using close reading, rhetorical field methods, and social media content analysis, this dissertation examines the rhetorical interpretation, ritualization, contestation, and commodification of wilderness as it comes to be understood and interacted with as sacred. This study offers three significant and interrelated interventions. First, it interrogates how rhetoric works to designate a sacred place. Locusts are a historically important model organism, and have been at the core of many discoveries related to olfaction. In recent years they have become less popular compared to organisms with genetic toolkits such as drosophila, but they have many advantages for studying principles of olfactory chemosensation and related behaviors. In this thesis there are two primary focuses; the development of a surface-bound odorant treadmill (odor-trail treadmill) for studying trail-following behaviors, and the second is the development of tools and procedures to enable simultaneous neural and behavioral recording in locusts. Securities law has long regulated securities-related speech—and until recently, it did so with little, if any, First Amendment controversy. Yet the antiregulatory turn in the Supreme Court’s twenty-first-century Free Speech Clause doctrine has inspired corporate speakers’ increasingly successful efforts to resist regulation in a variety of settings, settings that now include securities law. This doctrinal turn empowers courts, if they so choose, to dismantle the securities regulation framework in place since the Great Depression. Everyday our body’s immune and detoxification systems are working to protect us from pathogens, cancerous cells, foreign substances, and chemical exposures. Declining trends in per capita longevity in the US suggest that these systems are being stressed beyond their natural resiliency. Several of these trends are discussed in light of funding for preventive vs. illness care. The authors then use a systems and interdisciplinary approach to introduce the immunobiological and biotransformation systems, and the concept of the exposome for those unfamiliar. Parentheses in statutes have been at issue in an increasing number of court cases, even at the Supreme Court. Parentheses have a slightly different story from other punctuation marks and they have been used consistently throughout legal history. The Federal Constitution, early statutes, and a large part of our modern state and federal law separate words from their sentences using parentheses. Neuroblastomas have shed light on the differentiation disorder that is associated with spontaneous regression or differentiation in the same tumor at the same time. Long noncoding RNAs (lncRNAs) actively participate in a broad spectrum of biological processes. However, the detailed molecular mechanisms underlying lncRNA regulation of differentiation in neuroblastomas remain largely unknown. Here, we sequenced clinical samples of ganglioneuroma, ganglioneuroblastoma, and neuroblastoma. We compared transcription profiles of neuroblastoma cells, ganglion cells, and intermediate state cells; verified the profiles in a retinoic acid-induced cell differentiation model and clinical samples; and screened out the lncRNA ADAMTS9 antisense RNA 2 (ADAMTS9-AS2), which contributed …

The stemless carline thistle (Carlina acaulis subsp. caulescens) is a plant species commonly found at lower altitudes in the Central European mountains. Although it is still relatively frequent in the area, there has been a decrease in its numbers in the last decades. This research aims to determine the impact of the management of mountain mesic meadows and habitat factors on the morphometric variability of C. acaulis and species composition patterns of accompanying vegetation. The current study targeted to perform qualitative and quantitative analysis of medicinal plant taxa used in folk medicine in Trabzon. Due to its historical significance, diversity of plant species, and hosting a wide range of cultures, the Turkish city of Trabzon has significant ethnobotanical potential. Ethnobotanical data including family name, scientific name, local name, used parts, therapeutic uses, preparation methods, administration routes, and cited locations were gathered by face-to-face interviews with 645 participants mostly over 50 years old. his study examined the effect of biochar application to the soil at different times on the phytochemical composition (phenolic compounds, carotenoids, vitamin C, sugars, proteins, MDA, and antioxidant activity) and expression of HCT, F3’H and CHS genes (which are involved in the accumulation of phenolic compounds) in cabbage kale (Brassica oleracea L. var. acephala). Clove (Syzygium aromaticum) is a cross-pollinated crop with significant genetic variation in planting materials derived from seeds. Vegetative propagation through grafting is a valuable strategy to achieve genetically identical characteristics in progeny. This study investigates the influence of rootstock age and hormone (gibberellin) treatments on the grafting union and survival rate of top clove grafting under greenhouse conditions. In this study, it was aimed to determine the relationship between β carbonic anhydrase (βCA) gene expression levels and drought stress severity. For this purpose, the expression levels of six βCA genes in the roots and leaves of Arabidopsis thaliana were investigated under mild, moderate, and severe drought conditions. In addition, changes in the biomass, leaf water content, photosynthetic efficiency, CA enzyme activity, free proline content, and lipid peroxidation were also determined. The malignant brain cancer glioblastoma (GBM) contains groups of highly invasive cells that drive tumor progression as well as recurrence after surgery and chemotherapy. The molecular mechanisms that enable these GBM cells to exit the primary mass and disperse throughout the brain remain largely unknown. Bacterial infections in the United States are becoming increasingly resistant to existing antibiotic treatments. Due to projected increases in resistance and the recent decrease in novel antibacterials, experts have determined that the United States is in the “post-antibiotic era.” The scientific community has failed to resolve resistance despite the continual discovery of new antibiotic compounds. Methylphenidate (MPD), known as Ritalin, is a psychostimulant used to treat children, adults, and the elderly. MPD exerts its effects through increasing concentrations of dopamine (DA), norepinephrine (NE), and serotonin (5-HT) in the synaptic cleft. Concomitant behavioral and neuronal recording from the ventral tegmental area (VTA), locus coeruleus (LC), and from the dorsal raphe (DR) nucleus, which are the sources of DA, NE, and 5-HT to the mesocorticolimbic circuit, were investigated following acute and repetitive (chronic) saline, 0.6, 2.5, or 10.0 mg/kg MPD. Spondyloepimetaphyseal dysplasia with severe short stature, RPL13-related (SEMD-RPL13), MIM#618728), is a rare autosomal dominant disorder characterized by short stature and skeletal changes such as mild spondylar and epimetaphyseal dysplasia affecting primarily the lower limbs. The genetic cause was first reported in 2019 by Le Caignec et al., and six disease-causing variants in the gene coding for a ribosomal protein, RPL13 (NM_000977.3) have been identified to date. This study presents clinical and radiographic data from 12 affected individuals aged 2-64 years from seven unrelated families, showing highly variable manifestations. The affected individuals showed a range from mild to severe short stature, …

Millions of RNA sequencing samples have been deposited into public databases, providing a rich resource for biological research. These datasets encompass tens of thousands of experiments and offer comprehensive insights into human cellular regulation. However, a major challenge is how to integrate these experiments that acquired at different conditions. We propose a new statistical tool based on beta-binomial distributions that can construct robust gene co-regulation network (CoRegNet) across tens of thousands of experiments. Our analysis of over 12 000 experiments involving human tissues and cells shows that CoRegNet significantly outperforms existing gene co-expression-based methods. Although the majority of the genes …

Despite their high degree of effectiveness in the management of psychiatric conditions, exposure to antipsychotic drugs, including olanzapine and risperidone, is frequently associated with substantial weight gain and the development of diabetes. Even before weight gain, a rapid rise in circulating leptin concentrations can be observed in most patients taking antipsychotic drugs. To date, the contribution of this hyperleptinemia to weight gain and metabolic deterioration has not been defined. Here, with an established mouse model that recapitulates antipsychotic drug-induced obesity and insulin resistance, we not only confirm that hyperleptinemia occurs before weight gain but also demonstrate that hyperleptinemia contributes directly …

Ventricular arrhythmias (VAs) in heart failure are enhanced by sympathoexcitation. However, radiotracer studies of catecholamine uptake in failing human hearts demonstrate a proclivity for VAs in patients with reduced cardiac sympathetic innervation. We hypothesized that this counterintuitive finding is explained by heterogeneous loss of sympathetic nerves in the failing heart. In a murine model of dilated cardiomyopathy (DCM), delayed PET imaging of sympathetic nerve density using the catecholamine analog [11C]meta-Hydroxyephedrine demonstrated global hypoinnervation in ventricular myocardium. Although reduced, sympathetic innervation in 2 distinct DCM models invariably exhibited transmural (epicardial to endocardial) gradients, with the endocardium being devoid of sympathetic nerve …
