A Study Of The Snd1/Prmt5 Axis In Liver Cancer By Genetic Mouse Models, Tanner Wright, Tanner Wright Dec 2023

A Study Of The Snd1/Prmt5 Axis In Liver Cancer By Genetic Mouse Models, Tanner Wright, Tanner Wright

Dissertations and Theses (Open Access)

Arginine methylation is an essential post-translational modification (PTM) in cells. Protein arginine methyltransferase 5 (PRMT5) is the primary enzyme that catalyzes symmetric dimethyl arginine (SDMA) and requires methylosome protein 50 (MEP50) for stability and enzymatic activity which are necessary for life and development. Effector proteins bind different types of PTM’s to facilitate signaling. Staphylococcal nuclease Tudor domain containing 1 (SND1) is an effector that specifically binds SDMA via its single C-terminal Tudor domain. Both SND1 and PRMT5 have been implicated in hepatocellular carcinoma (HCC). SND1 has been confirmed as a driver of HCC using genetically engineered mouse models (GEMMs), though, …

Ideological Parameters For Deradicalization Programs To Measure Changes In Terrorist Ideology In Indonesia, Yudi Zulfahri, Aldi Nur Fadil Auliya Dec 2023

Ideological Parameters For Deradicalization Programs To Measure Changes In Terrorist Ideology In Indonesia, Yudi Zulfahri, Aldi Nur Fadil Auliya

Journal of Terrorism Studies

The paper analyzes the inefficiency of the Deradicalization strategy implemented by The National Counter Terrorism Agency (BNPT), causing the issue of recidivist terrorism in Indonesia. Although the occurrence of recidivism is common in various crimes, it poses unique challenges in terrorism cases, given that the government has allocated a specific budget and established a special agency, BNPT, to tackle terrorism-related issues. The high recidivism rate in terrorism cases has prompted public concern and criticism regarding the effectiveness of Deradicalization programs. Despite the ongoing debate, this paper argues that the success or failure of Deradicalization programs cannot be measured simply by …

Butyrate Suppresses Experimental Necrotizing Enterocolitis-Induced Brain Injury In Mice., Maribel Martinez, Wei Yu, Heather Menden, Tianhua Lei, Paula Monaghan-Nichols, Venkatesh Sampath

Manuscripts, Articles, Book Chapters and Other Papers

BACKGROUND: Necrotizing enterocolitis (NEC) is a devastating disease in premature infants, and 50% of infants with surgical NEC develop neurodevelopmental defects. The mechanisms by which NEC-induced cytokine release and activation of inflammatory cells in the brain mediate neuronal injury, and whether enteral immunotherapy attenuates NEC-associated brain injury remain understudied. Based on our prior work, which demonstrated that experimental NEC-like intestinal injury is attenuated by the short-chain fatty acid, butyrate, in this study, we hypothesize that NEC-induced brain injury would be suppressed by enteral butyrate supplementation.

METHODS: A standardized NEC mouse model [enteral formula feeding, lipopolysaccharide (LPS), and hypoxia] was used. …

Children’s Nutrition Research Center Staff Publications

It is increasingly assumed that there is no one-size-fits-all approach to dietary recommendations for the management and treatment of chronic diseases such as obesity. This phenomenon that not all individuals respond uniformly to a given treatment has become an area of research interest given the rise of personalized and precision medicine. To conduct, interpret, and disseminate this research rigorously and with scientific accuracy, however, requires an understanding of treatment response heterogeneity. Here, we define treatment response heterogeneity as it relates to clinical trials, provide statistical guidance for measuring treatment response heterogeneity, and highlight study designs that can quantify treatment response …

Mineta Transportation Institute

Nearly 5,000 people are killed and more than 418,000 are injured in weather-related traffic incidents each year. Assessments of the effectiveness of statistical models applied to crash severity prediction compared to machine learning (ML) and deep learning techniques (DL) help researchers and practitioners know what models are most effective under specific conditions. Given the class imbalance in crash data, the synthetic minority over-sampling technique for nominal (SMOTE-N) data was employed to generate synthetic samples for the minority class. The ordered logit model (OLM) and the ordered probit model (OPM) were evaluated as statistical models, while random forest (RF) and XGBoost …

Assessing The Thermodynamic Potential Of Deep Eutectic Solvents In The Absorption Of Greenhouse Gases Through Computational Methods, Thomas Michael Quaid

Theses and Dissertations

Greenhouse gas (GHG) capture is a fundamental technology in the fight against climate change. Some species of devastating GHGs like fluorinated compounds are released directly to the air from industrial processes. Some GHGs are co-produced in sustainably derived biofuels which require absorptive upgrading. The development of more efficient, cost effective, and environmentally friendly methods of capturing GHG’s from pollutant sources and industrial streams is of utmost importance in the fight against climate change. Deep eutectic solvents have entered the separations stage as potential disruptors to conventional solvent choices for a variety of applications. Deep eutectic solvents (DES) are compounds of …

Low Reynolds Number Locomotion Near Interfaces In Two-Fluid Media, Avriel Rowena Mae Cartwright Dec 2023

Low Reynolds Number Locomotion Near Interfaces In Two-Fluid Media, Avriel Rowena Mae Cartwright

Theses and Dissertations

Microorganisms often swim within complex fluid environments composed of multiple materials with very different properties. Biological locomotion, including swimming speed, is significantly impacted by the physical composition and rheology of the surrounding fluid environment, as well as the presence of phase boundaries and free interfaces, across which physical properties of the fluid media may vary greatly. Through computational simulations, we first investigate the classical Taylor’s swimming sheet problem near interfaces within multi-fluid environments using a two-fluid immersed boundary method. The accuracy of the methodology is illustrated through comparisons with analytical solutions. Our simulation results indicate that the interface dynamics and …

Through The Lens Of A Naturalist: How Learning About Nature Promotes Nature Connectedness Via Awe, Shu Tian Ng, Angela K. Y. Leung, Sarah Hian May Chan

Research Collection School of Social Sciences

Environmental educators stress the importance of engaging with the wonders of the Earth in promoting nature connectedness. However, it remains unclear if learning about nature has an incremental effect beyond mere exposure to nature and what psychological mechanism can explain such a learning effect if it exists. To fill this gap, we propose a mediation model in which learning about nature promotes a sense of awe—a self-transcendent emotion associated with the recognition of vastness in nature. A sense of awe, in turn, promotes nature connectedness. Study 1 employed a cross-sectional survey and offered preliminary support for the proposed model, with …

Kape: Knn-Based Performance Testing For Deep Code Search, Yuejun Guo, Qiang Hu, Xiaofei Xie, Cordy Maxime, Mike Papadakis, Yves Le Traon

Research Collection School Of Computing and Information Systems

Code search is a common yet important activity of software developers. An efficient code search model can largely facilitate the development process and improve the programming quality. Given the superb performance of learning the contextual representations, deep learning models, especially pre-trained language models, have been widely explored for the code search task. However, studies mainly focus on proposing new architectures for ever-better performance on designed test sets but ignore the performance on unseen test data where only natural language queries are available. The same problem in other domains, e.g., CV and NLP, is usually solved by test input selection that …

Geography Faculty Publications and Presentations

With climate change and urbanization, city planners and developers have increasing interest and practice in constructing, restoring, or incorporating wetlands as forms of green infrastructure to maintain water-related ecosystem services (WES). We reviewed studies that valued in functional or monetary units the water regulation and purification services of urban wetlands around the globe. We used the adaptive management cycle (AMC) as a heuristic to determine the step that a study would represent in the AMC, the connections between the cycle steps that were used or considered, and the stakeholders involved. Additionally, we identified the social, ecological, and/or technological dimension(s) of …

Shaping Supervised Agricultural Experiences In Rural American Schools: Support, Supervision, And Culture, Eric D. Rubenstein, Andrew Thoron, Taylor D. Bird Dec 2023

Shaping Supervised Agricultural Experiences In Rural American Schools: Support, Supervision, And Culture, Eric D. Rubenstein, Andrew Thoron, Taylor D. Bird

Journal of Research in Technical Careers

Over the years, examination of barriers to the development and implementation of Supervised Agricultural Experience (SAE) programs has gained traction. This has led the profession to investigate the relevance of SAE. In the conducted studies, the profession continually notes that SAE remains a foundational component and perhaps the only distinguishing difference between school-based agriscience education (SBAE) and other Career and Technical Education programs or specialty courses that tie in a student leadership organization. While collecting evidence of perceived barriers of SAE implementation is important, at some point the question must be asked: What is right with student SAE programs in …

Development Of Novel Methods To Study Host-Microbe Interactions In The Larval Zebrafish Gastrointestinal Tract, Anh K. Trinh Nguyen

Dissertations and Theses (Open Access)

The dynamic nature and inaccessible location of the intestine pose significant challenges to the study of intestinal physiology and pathology. Zebrafish larvae, possessing optical transparency and genetic tractability, offer an accessible and clinically relevant model for investigating dynamic events in the intestine via time-lapse imaging. In the first part of this work, I discuss our efforts to optimize the parameters of a foodborne infection assay using paramecia as a vehicle. This method provides an effective, high-throughput alternative to infection via immersion or oral gavage, and replicates the most common route of transmission of gastrointestinal (GI) infection in humans. The foodborne …

Telomeric Regulation Of Chromosomal Instability In Colorectal Cancer, Kyle Labella, Kyle A Labella Dec 2023

Telomeric Regulation Of Chromosomal Instability In Colorectal Cancer, Kyle Labella, Kyle A Labella

Dissertations and Theses (Open Access)

Telomere dysfunction drives chromosomal instability (CIN) during the transition from benign adenoma to malignant adenocarcinoma. While CIN provides a mutator mechanism for cancer-relevant genomic events, its role in shaping tumor biology during carcinogenesis is not well understood. Here, we explored the molecular and biological impact of telomere dysfunction and associated CIN in vivo in a faithful model of CRC. In vivo lineage tracing revealed that CIN increased neoplastic cell competition and clonal expansion through accelerated differentiation of neighboring stem cells, resulting in increased number of adenomas and decreased survival in CIN-high Apcmin mice. Mechanistically, CIN represses EZH2 leading to …

Cortical Representation Of Learning Social Interactions In Freely Moving Non-Human Primates, Melissa Franch Dec 2023

Cortical Representation Of Learning Social Interactions In Freely Moving Non-Human Primates, Melissa Franch

Dissertations and Theses (Open Access)

The motivation and capacity to be social is necessary for human survival. Successful learning of complex, prosocial behavior stems from the ability to perceive and respond to visual cues, such as the body language and facial expressions, from others in our environment. This dependence on visual information to guide social interaction is especially true for humans and non-human primates. Although recent studies in primate neurophysiology discovered neurons that can encode socially relevant variables, like reward and social actions, the underlying neural mechanisms of learning advanced social concepts, such as cooperation, are not well understood. Further, previous work has identified …

Downregulation Of Mir-10a-5p As A Potential Therapeutic Target To Improve The Detrimental Effects Of Post-Stroke Social Isolation, Patrick Mooz

Dissertations and Theses (Open Access)

Introduction

Social isolation is detrimental to health. Isolation and loneliness are linked to all-cause mortality and accelerated disease progression in patients with cancer, dementia, and vascular disease. The number of individuals that live alone is increasing in our society, especially in the elderly, and loneliness and isolation represent a profound growing public health challenge. Previous studies have found that the detrimental effects of social isolation can be recapitulated in animal models. Mice that are socially isolated (SI) prior to an experimental stroke have larger infarcts and significantly worse outcomes. Importantly, animals that remain pair housed (PH) when an experimental stroke …

A Signal To Divide: Apoptotic Extracellular Vesicles As Carriers Of Mitogenic And Immunogenic Signals, Safia Essien, Safia Essien

Dissertations and Theses (Open Access)

Efficient replacement of dead cells in epithelial tissue is crucial for maintaining barrier function and tissue homeostasis. Apoptotic cells can signal to neighboring cells to stimulate proliferation and compensate for cell loss and maintain overall cell numbers in normal physiology and cancer. While dying cells can transmit instructive cues to neighboring cells, the molecular mechanisms that induce cell division are not well understood. Recent evidence suggests that apoptotic bodies (ABs) or apoptotic extracellular vesicles (AEVs) mediate cell-to-cell communication and carry diverse biologically active cellular cargo which can influence cell proliferation. This dissertation visualizes and characterizes AEVs in larval zebrafish and …

In terms of metabolism, eating disorders manifest in two extreme directions: overnutrition, which can lead to obesity, and malnutrition, which can result in underweight or even starvation. Both extremes compromise the quality of life. According to the Diagnostic and Statistical Manual of Mental Disorders-5 (DSM-5) standard, eating disorders affect up to 17.9% of young women and 2.4% of young men. Although eating disorders are primarily defined as mental disorders rather than metabolic disorders, they are intertwined with complex emotions and sensory perceptions. However, in contemporary animal research related to eating disorders and feeding behaviors, the majority of neuroscientists still examine …

Oncogenic Kras And Telomere Biology In Crc Progression, Ronald Depinho Dec 2023

Oncogenic Kras And Telomere Biology In Crc Progression, Ronald Depinho

Dissertations and Theses (Open Access)

While colorectal cancer (CRC) patients diagnosed with localized stage disease (as defined by SEER) have a 5-year survival rate of 90%, this rate plunges to 14% for patients diagnosed with metastatic CRC. Consequently, there is an immediate imperative to elucidate the mechanisms that drive the transition to advanced CRC.

Human CRCs carrying oncogenic mutations in the KRAS oncogene, henceforth referred to as KRAS*, exhibit a 25% higher propensity for developing liver metastases. Similarly, in our CRC mouse model, engineered with an inducible Kras* transgene and conditional null alleles of Apc and Tp53 (referred to as iKAP), KRAS* has been …
