Entomological and microbiological evidence can be used in the estimation of postmortem intervals (PMI). This study observes the horizontal and vertical transmission of the microbial community associated with blow flies of forensic importance. Variable region four (V4) of the 16S ribosomal DNA (16S rDNA) was amplified and sequenced from various life stages of blow flies (n=72) using dual-index high throughput sequencing strategy on the MiSeq FGx platform. No significant difference in bacterial community structure was observed between blowfly adults with carrion access and blowfly adults with no carrion access. Introduction: The growing opioid epidemic in North America presents a call to action to develop new techniques for the identification and quantitation of novel synthetic opioids in uncommon matrices. In instances of extreme decomposition and/or significant scavenging, bone may be the only matrix available. In order to develop an extraction method for synthetic opioids in bone, bone samples were obtained from animal models. The animal model of choice is the rabbit due to their exhibition of spontaneous cortical bone remodeling, a process that is exhibited in humans but not in rats and mice.

Due to advancements in DNA analysis technology and an increased ability to detect trace quantities of low template DNA, ‘touch’ or trace DNA has become a prevalent form of evidence for forensic investigations. Trace DNA is biological material left on a surface from skin cells after being touched or handled. The success rate of casework could be improved with new methods to non-destructively screen trace DNA to predict DNA yield and whether multiple cell populations are present in the evidence prior to STR analysis. Evidence containing biological mixtures can make DNA interpretation more difficult due to ambiguous allele calls and artifacts that may be present within electropherograms. Separating out these cell mixtures can be advantageous to the analysts’ analysis methods for identifying STR profiles with higher accuracy. A potential cell separation method that could be implemented in the beginning of the forensic DNA workflow is optical trapping.

The implementation of rapid DNA analysis systems is of interest to forensic DNA casework, as this technology exhibits a sample-to-profile time of approximately two hours with limited handling of samples. This decreases the likelihood of contamination, and can potentially reduce DNA backlogs; however, current rapid DNA analysis systems do not execute one crucial step of the forensic DNA analysis workflow: human DNA quantitation. This step is necessary for DNA profiles to be compliant with the FBI’s Quality Assurance Standard 9.4, allowing DNA profiles to be uploaded and searched within the CODIS database. While efforts have been made to reduce the pervasive backlog of sexual assault evidence collection kits, the actual laboratory process remains very time-consuming due to the requirement of a differential lysis step before DNA purification, as well as intricate mixture analysis towards the end of the DNA workflow. Electrical power outages are of increasing interest to US urban scholars, government officials and stakeholders, as they have increased in number and duration with significant health and economic, among other, impacts. This analysis examines reports of power outages in New York City in relation to socioeconomic and health characteristics of neighborhoods. Using the city’s 311-call database we examine complaint calls for power outages from 2014 to 2022. While 311-calls for power outages occur all year long, volume trended higher during the warmer months (June, July and August), and as minimum daily temperatures exceeded 20 °C (68°F), the number of calls …

High concentrations of phosphate are known to adversely affect the environment. Excess phosphate can lead to eutrophication that eventually fosters uncontrollable growth of aquatic plants and algae. This can result in depletion of oxygen content which adversely impacts underwater organism’s survival rates. Successful crop production relies on soils with balanced physical, chemical and biological properties. Demand for greater crop yields has led to the breakdown of soil properties through detrimental agricultural practices. To combat soil degradation, farmers employ field management practices including cover crop application, crop rotation strategies and organic soil amendment addition. In this work, I explore the development of computational methods for automatically creating after-action reports of JTAC radio conversations. Prior research has investigated related issues of sentence compression, text summarization, and conversation summarization (Banerjee, Mitra, & Sugiyama, 2015; Clarke & Lapata, 2008; L. Wang & Cardie, 2012; Raffel et al., 2020). However, this work makes limiting assumptions about what features are relevant to a summary and what sources of information should be included. Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease which targets motoneurons (MNs), yet underlying disease mechanisms are not well understood. Evaluating the intrinsic excitability of MNs in ALS could lead to a better understanding of mechanisms causing neurodegeneration. The SOD1-G93A (SOD) model is the most commonly studied animal model of ALS. However, past studies have shown highly conflicting results on SOD MN excitability. Interestingly, I show that depending on the level of membrane depolarization, SOD MNs show opposite results of both hyper- and hypo-excitability. This reveals that differences in the methodology of measuring excitability can heavily impact the study results. …

Duchenne muscular dystrophy (DMD) is an X-linked genetic disorder that affects 1 in 3500 male births worldwide. It is characterized by mutations in the dystrophin gene that results in the loss of functional dystrophin. Dystrophin deficiency leads to instability of the sarcolemma alongside increased inflammation, necrosis and fibrosis resulting in membrane rupture and eventual muscle fiber death. There are currently no effective treatments for DMD. Here we show that Lipin1 expression is significantly downregulated at the mRNA and protein levels in gastrocnemius muscle of mdx mice, the DMD mouse model. The study was conducted to investigate the prevalence of four enteric viruses in mouse research facilities that may not be captured by routine pathogen screening. Fecal samples were collected from contact-sentinel mouse cages between April 2022 and January 2023, and screened for the presence of viral genome by RT-qPCR. Out of a total of 50 samples, murine norovirus and murine astrovirus were detected in two (4%) and one (2%) sample, respectively, while murine adenovirus and murine rotavirus were not detected. Although α-synuclein (α-syn) has been linked genetically and pathologically to Parkinson’s disease, the mechanism by which it drives disease pathogenesis is not fully understood. When α-syn is overexpressed in yeast, it forms abnormal cytoplasmic accumulations and leads to cell death, resembling the phenotypes observed in neurons. Such yeast models have been widely used to uncover molecular mechanisms of α-syn toxicity, but the induction of α-syn relies on growing yeast in galactose, a fermentable carbon source. Yeast does not require cellular respiration when grown in galactose, thus limiting the understanding of α-syn toxicity when cellular respiration is essential for neurons. We …

Duchenne muscular dystrophy (DMD) is a severe and progressive muscular dystrophy that develops in the skeletal muscles because of mutations in the dystrophin gene. Dystrophin stabilizes sarcolemma and assembles neuronal nitric oxide synthase (nNOS) into the dystrophin-associated protein complex on the sarcolemma. The absence of dystrophin triggers the delocalization of nNOS and contributes to the misregulation of muscle development, blood flow, muscle fatigue, and inflammation. Lipin1 was reduced in the skeletal muscles of patients with DMD and the mdx mouse model of DMD. In this study, we explored the role of lipin1 in the regulation of nNOS expression and function. …

The various environmental factors like toxic chemicals, chemotherapeutic agents and UV radiation impact human health by altering physiological functions of the normal living cells. One of the main consequences of exposure to these factors is DNA damage. Although, living system have inherent DNA repair mechanisms such as Nucleotide Excision Repair (NER), which removes DNA lesions from the genome in the form of small, approximately 30-nt-long DNA oligonucleotides. Environmental exposure to organophosphate (OP) pesticides, such as malathion, is a risk factor for neuropathy and neurodegeneration. Toxic levels of OPs irreversibly inhibit acetylcholinesterase (AchE) activity, leading to acute paralysis and even death as the neurotransmitter acetylcholine accumulates at cholinergic synapses. In addition, there is compelling evidence that repeated low-level “occupational-like” exposure to OPs is associated with somatosensory defects but the cellular mechanisms for this effect are unclear. We show sensory neuron cell size in the dorsal root ganglia is significantly reduced in rats exposed to occupational level malathion. However, co-administration of a reversible AchE inhibitor, galantamine, prevented this effect.

Short statutes of limitations for sex crimes ask the impossible of many vic- tims: report the crime before they have recovered from the trauma. Perpetra- tors go free as a direct result of the injury they caused. Nearly a third of victims of rape and sexual assaulthave PTSD during their lifetimes. PTSD is associated with three symptoms pertinent to reporting a crime: avoidance cop- ing (avoidingdistressing thoughts, feelings, or reminders of the attack), disso- ciative amnesia (forgetting important or all aspects of the attack), and depression. Most Americans do not realize that, notwithstanding the First Amendment's free speech guarantee, for most of our nation's history, judges sent men and women to prison for expressing ideas considered too "dangerous." It was not until the late 1960s that the Supreme Court rejected the clear and present danger doctrine, insisting that statutes banning speech must draw a distinction between advocacy of ideas and advocacy of imminent lawless action. The Court held that under that constitutional norm, the government could not send a Klansman to prison for expressing racist, anti-Semitic, or otherwise dangerous or offensive ideas. This paper analyzes the barriers and supports for Dominican University of California students in meeting their occupational needs and what services might be beneficial to them. To be able to answer this question, we conducted a survey using convenience sampling with twenty-seven Dominican undergraduate students, interviewed staff, and conducted a focus group with the students who participated in the survey. Our results indicated four main themes which include: occupational needs, barriers to meeting occupational needs, supports to meeting occupational needs, and services recommended. Environmental law is pragmatic, inevitable, and intentional. In the aggregate, the numerous federal environmental statutes are not simply a patchwork of ad hoc responses or momentary political breakthroughs to isolated public health problems and resource concerns. Together, they are a group of repeated, legislatively-backed commitments to embrace self-restraint for self-preservation.

Sugar maple canopy dieback has been reported in the upper Great Lakes since the mid-2000s. This thesis builds on research conducted to determine some of the underlying causes of canopy dieback, which include invasive earthworm impacts, with a view toward adaptations in management of northern hardwood forests in our region. The first chapter reports a study of understory communities in a subsection of forest health monitoring plots located in four national forests near Lake Superior. Based on plot assessments from 2010 and 2021, responses of plant community composition and traits to earthworm impacts and sugar maple canopy dieback were explored. …

Cellular communication is important in embryonic development. It facilitates timely migration and maturation of cells to produce a fully functional organism using cell signaling pathways. These pathways involve receptors, ligands, and intracellular enzymes to produce different effects in the cell. One such pathway, the EphA4 signaling pathway, is involved in altering the cytoskeleton of eukaryotic cells. LIM kinases (LIMKs) are enzymes that regulate important components of the cytoskeleton. Puppetry is a special effect- media magic that enhances the flavor of a fictional world. My dream is to harness that magic in order to teach preschoolers about their physical world. Puppets have a unique ability to provide an analogue that children can relate to, which makes them an incredibly effective and engaging vehicle for educational entertainment. Under the influence of climate change, the vulnerability of grassland ecosystems in the Mongolian Plateau has increased. Correspondingly, herdsmen need to adopt more livelihood adaptation strategies to cope with changes in climate and grassland environment. The purpose of this study is to analyze the impact of breeding structure on grassland quality in Mongolian plateau. We select 20 border provinces/banners of Mongolia and Inner Mongolia Autonomous Region for research, and combined statistical data with remote sensing data of 2007-2016. The Normalized Difference Vegetation Index (NDVI) is used to quantify the grassland vegetation quality. Results show that there are differences in livestock …
