Biomedical Sciences

Tumor-associated macrophages (TAMs), as a major and essential component of tumor microenvironment (TME), play a critical role in orchestrating pancreatic cancer (PaC) tumorigenesis from initiation to angiogenesis, growth, and systemic dissemination, as well as immunosuppression and resistance to chemotherapy and immunotherapy; however, the critical intrinsic factors responsible for TAMs reprograming and function remain to be identified. By performing single-cell RNA sequencing, transforming growth factor-beta-induced protein (TGFBI) was identified as TAM-producing factor in murine PaC tumors. TAMs express TGFBI in human PaC and TGFBI expression is positively related with human PaC growth. By inducing TGFBI loss-of-function in macrophage (MΦs) in vitro …

Oncolytic Tanapoxvirus Recombinants Expressing Mil-2, Flagellin-C, And Mccl2 Regress Human Cancer Xenografts In Immune Deficient Nude Mice And Immune Cell Reconstituted Balb/C Nude Mice., Michael Leonard Monaco

Dissertations

Tanapoxvirus (TPV) is a double stranded DNA virus from the genus Yatapoxvirus which has been engineered for the purpose of potentially treating a number of human cancers. Previous studies have shown TPV recombinants are capable of inducing tumor inhibition and even regression in athymic nude mice bearing human cancer xenografts of colorectal, melanoma and triple negative breast cancer (TNBC). However, TPV is host-restricted to humans and monkeys, including cancerous cells, which has provided a unique obstacle in its development as an oncolytic virus (OV). Namely, standard syngeneic mouse models cannot be used as test systems to evaluate TPV’s efficacy against …

Investigating The Therapeutic Potential Of Cannabigerol (Cbg) In Female Nonalcoholic Steatohepatitis (Nash) Mouse Model, Raznin A. Joly

Master's Theses

Nonalcoholic Steatohepatitis (NASH) is a progressive form of nonalcoholic fatty liver disease (NAFLD) characterized by inflammation, fibrosis, steatosis, and oxidative stress, in addition to excessive fat accumulation in the liver. Cannabigerol (CBG), a precursor molecule of Cannabidiol (CBD), has demonstrated anti-inflammatory and anti-oxidative effects in various studies. Our previous data showed that a low dosage of CBG reduced inflammation and fibrosis in male NASH mice. However, the impact of CBG on female mice remains unknown. Recent studies have identified changes in immune cell populations in NAFLD patients. The goal of this project was to test the impact of CBG treatment …

Beaches, Bumps, And Milkshakes: A Humanizing, Open, And Dialogic, Postformal Autoethnographic Study Of An Experiential Learning Academy, Eric Anthony Karahalis

Theses & Dissertations

The dominant educational paradigm is rooted in a Platonic, Cartesian, and Piagetian mind-supremacist epistemology that subordinates the body and experience to the mind, ideas, and objective concepts. This reduces students to cognitive receptors of information in a detached, decontextualized, and dehumanizing classroom, i.e., the banking educational model (Freire, 2018; hooks,1994). Students are forced to sit at desks for several hours a day and expected to open their minds and ignore the body’s needs. This educational model is dehumanizing because it only approaches students and learning through one epistemological lens and separates learning from the world. On the other hand, experiential …

Program Development And Expansion Of A Clinician-Driven Wound Care Program At An Outpatient Wound Care Center, Zhenhai Niu Dec 2023

Program Development And Expansion Of A Clinician-Driven Wound Care Program At An Outpatient Wound Care Center, Zhenhai Niu

Doctor of Nursing Practice

Problem. Patients with chronic wounds often present with additional physical and emotional problems because healing of chronic wounds may require months to resolve. Practitioners develop treatment plans based on their own training and clinical expertise, but unfortunately, there are no universal guidelines to inform the management of chronic wounds. Expanding multi-factorial assessments and interventions could improve patient outcomes. The purpose of this project was to implement a wound care protocol for outpatient wound care clients based on a nutritional and psychological self-assessment. Methods. A patient-completed checklist, done on arrival to the clinic, triggered clinician-driven protocol interventions addressing mental health and …

Decades Of Don Quixote: Tracking Social Progression Through Marcela And Grisóstomo, Allison Jackson Dec 2023

Decades Of Don Quixote: Tracking Social Progression Through Marcela And Grisóstomo, Allison Jackson

Honors Theses

In this essay, I will track the evolution of academic opinions in the late 20th and early 21st century of two main components of the episode: Marcela’s character and what it says about Cervantes’ perception of women’s autonomy, as well as Grisóstomo’s death and the rejection of the notion of suicide by past academics. Marcela’s dramatic rejection of societal demands and Gristósomo’s tragic and confusing death are radically exciting since there is so much left to be assumed about the two main characters that is not explicitly written. The story relies on irony and subversion to enunciate a certain message; …

Examining The Microbial Diversity Of A Generalist Ant Along An Urban-Rural Gradient, Abdol-Munim Abazid Dec 2023

Examining The Microbial Diversity Of A Generalist Ant Along An Urban-Rural Gradient, Abdol-Munim Abazid

Honors Theses

After sampling the exoskeleton microbiomes of 225 ants from 3 different locations along a rural-urban gradient in the Chattanooga area, a photo library was collected and microbial diversity and morphospecies evenness of each location was determined. It was determined that there was no significant difference in either microbial diversity or morphospecies evenness between sites along the rural-urban gradient, even after adjusting data to be between only rural and urban areas. Urbanization was quantified using % impervious surface; however, a future study could quantify urbanization by other means, such as soil nutrient composition. Additionally, it was determined that there was no …

On The Causation Of The Mexican-American War, Emery Benson

Honors Theses

In 1844, Whig, former President, and then-Representative John Quincy Adams reflected on President John Tyler’s bill to annex Texas, writing about his anxiety over “the degeneracy of my country… under the transcendent power of slavery and the slave-representation.” Adams celebrated the treaty’s failure later that year, praising the nation’s escape from “slave-tainted monarchy, and of extinguished freedom.” In 1847, in the midst of the Mexican-American War, Reverend John Dudley of Vermont gave a fiery sermon in which he excoriated “the two leading sins of this nation, SLAVERY AND WAR.” Reverend Dudley continued, claiming “that the present war has its origin …

Using A Toxic Aging Coin To Assess Hexavalent Chromium-Induced Neurotoxicity., Samuel Thomas Vielee Dec 2023

Using A Toxic Aging Coin To Assess Hexavalent Chromium-Induced Neurotoxicity., Samuel Thomas Vielee

Electronic Theses and Dissertations

We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. We address this crisis using a ‘toxic aging coin’ approach; heads examines how age impacts toxicity, tails examines how chemicals accelerate aging. This thesis applies the heads side for Cr(VI)-induced neurotoxicity across ages. We …

Breaking Virulent: The Coincidental Evolution Of Virulence Factors In Bacteria., Rhiannon Emmanuelle Cecil Dec 2023

Breaking Virulent: The Coincidental Evolution Of Virulence Factors In Bacteria., Rhiannon Emmanuelle Cecil

Electronic Theses and Dissertations

Understanding how innocuous organisms can evolve to be pathogenic to humans is of increasing global concern. Further, understanding how existing pathogens may evolved to be more virulent is also vital to our ability to provide healthcare to people afflicted with diseases that promote chronic bacterial infections, such as cystic fibrosis. With the rise of antibiotic resistance in both bacteria and fungi it is paramount that new therapeutics are identified. Understanding what mutations occur that result in increased virulence in microbes can potentially provide new targets for antimicrobial drugs to combat antibiotic resistance. The Coincidental Evolution Hypothesis is a fundamental hypothesis …

From "Smart Talk" To "Living Well": Commonplaces And Their Role In Narratives Of Rare Disease., Caitlin E. Ray Dec 2023

From "Smart Talk" To "Living Well": Commonplaces And Their Role In Narratives Of Rare Disease., Caitlin E. Ray

Electronic Theses and Dissertations

As healthcare becomes more complex, automated, and bureaucratic, patients often suffer from a lack of resources, agency, and visibility when seeking medical care. Rhetoric and Composition, specifically the subfield of Rhetoric of Health and Medicine (RHM), is interested in studying and intervening into such issues. One way to challenge our current understanding of healthcare is to consider how the rare disease patient experience reveals the gaps, limitations, and assumptions of illness and health. I argue here that through rare diseases, rhetoricians of health and medicine can better understand the representation, advocacy, and patient experience within healthcare, and potentially lead to …

Characterizing The Regulation Of Candida Albicans Hyphal Morphogenesis In The Context Of Host Macrophages, Hannah Wilson Dec 2023

Characterizing The Regulation Of Candida Albicans Hyphal Morphogenesis In The Context Of Host Macrophages, Hannah Wilson

Dissertations and Theses (Open Access)

Candida albicans is a commensal fungus that resides on the skin, mucosal surfaces, and in the gut of an estimated 80% of individuals. Though generally harmless, C. albicans is capable of causing uncomfortable mucosal infections as well as deadly disseminated infections depending on the immune status of the host. Although antifungal therapeutics exist, the mortality rate associated with disseminated disease still lingers around 50%. Further, C. albicans is the fourth most common cause of all bloodstream infections and continues to present as a major clinical issue with less than satisfactory treatment options. For this reason, it is imperative to understand …

Leptospirosis, the most widespread zoonotic disease in the world, is broadly understudied in multi-host wildlife systems. Knowledge gaps regarding Leptospira circulation in wildlife, particularly in densely populated areas, contribute to frequent misdiagnoses in humans and domestic animals. We assessed Leptospira prevalence levels and risk factors in five target wildlife species across the greater Los Angeles region: striped skunks (Mephitis mephitis), raccoons (Procyon lotor), coyotes (Canis latrans), Virginia opossums (Didelphis virginiana), and fox squirrels (Sciurus niger). We sampled more than 960 individual animals, including over 700 from target species in the greater Los Angeles region, and an additional 266 sampled opportunistically …

Computational Molecular Docking Studies Of Small Molecule Inhibitors With The Sars-Cov-2 Spike Protein Variants: In-Silico Drug Discovery Using Virtual Screening And Drug Repurposing Approaches, Grace Gupta

Computational and Data Sciences (MS) Theses

The pandemic caused by the emergence of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in 2019 has caused a global public health crisis of nearly unprecedented scale. In the years following the outbreak, the scientific community has mobilized to develop several vaccines and treatments. Drug repurposing as a strategy for drug development has produced many of the current therapeutic options. The greatest challenge to designing a therapeutic inhibitor of SARS-CoV-2 is the shifting mutational landscape of the virus as it evolves. In this study, we focus on the spike protein as a target for potential inhibitors. We explore two …

Biology

Mosquitoes have profoundly affected human history and continue to threaten human health through the transmission of a diverse array of pathogens. The phylogeny of mosquitoes has remained poorly characterized due to difficulty in taxonomic sampling and limited availability of genomic data beyond the most important vector species. Here, we used phylogenomic analysis of 709 single copy ortholog groups from 256 mosquito species to produce a strongly supported phylogeny that resolves the position of the major disease vector species and the major mosquito lineages. Our analyses support an origin of mosquitoes in the early Triassic (217 MYA [highest posterior density region: …
