As Americans live longer, the prevalence of Alzheimer’s disease and related dementias will increase. Caregivers are critical to the care and life quality of people with dementia. Yet, dementia caregivers are at increased risk for health issues, social isolation, and financial challenges. To help educators with the University of Kentucky Cooperative Extension Service work with their local Alzheimer’s Association and the University of Louisville to better support dementia families, we explored caregiver needs and service utilization using an online survey. This article describes the process our team followed to create a series of career podcasts for middle school students using Anchor. Connecting Diverse Communities Through A 4-H Urban-Rural Experience, Laurie Murrah-Hanson, Brittani Lee, Pamela Bloch

Misconceptions regarding agricultural careers are exacerbated by increasing urbanization and are one of many differences in perspective between urban and rural communities. The One Georgia 4-H Urban-Rural Experience addresses this divide through a statewide youth exchange focusing on the agricultural sector. The program includes a multifaceted look at agricultural careers through visits to urban and rural communities. Focus group evaluations revealed the impact on youth through exposure to new careers, increased understanding of the diversity of agriculture, awareness of the interconnected nature of urban and rural communities, and a sense of belonging to a diverse group of youth.

New Jersey 4-H Junior Explorers Virtual Short-Term Exploratory Program (Step), Matthew Newman, Alayne Torretta

During the worldwide pandemic, youth were isolated from each other. In response, New Jersey 4-H, a part of Rutgers Cooperative Extension, created the Junior Explorers, a virtual exchange program bringing together 4-H members with youth from other countries. This program provides youth ages 10-13 with access to international exchange opportunities that would otherwise be sparse for this age group. This program has demonstrated that Extension professionals can successfully adapt exchanges to a virtual platform. With youth the world over learning virtually, Extension professionals have a unique opportunity to learn from and collaborate with international partners to enhance existing programs.

Community, local, and regional food systems (CLRFS) programming reflects important issues and priorities that intersect with Extension and the sustainability of our food system. CLRFS programming in Extension, however, is still developing slowly while food movements grow nationally. This article describes a CLRFS model and complementary process for conducting listening sessions with Extension professionals and community leaders to develop and enhance CLRFS programming to address critical food system needs. A recommendation for Cooperative Extension is that such a tool may aid CLRFS program potential as an integrated “food, farm, and health” approach for community-level application.

We used an online survey to document challenges experienced by aquaponic hobbyists (n = 81), producers (n = 117), and educators (n = 75). Responses were distilled into the following categories: 1) operations and management; 2) facilities, location, and system design; 3) knowledge and educational resources; 4) funding; 5) economic viability; 6) plant culture; 7) marketing and distribution; 8) fish culture; 9) human factors; 10) regulations and certifications. Training and research in these areas are needed to advance the aquaponics industry.

Leveraging The Covid-19 Fermentation Trend To Enhance Nutrition And Food Safety Extension Efforts, Thais M. Ramos, Hanna Louvau, Heesun Kim, Maria L. Marco, Erin Dicaprio

Our program aimed to increase knowledge related to fermented foods. Over 400 stakeholders registered for a webinar series that focused on defining fermented foods, health benefits of fermenting foods, and the safety of fermented foods. Participants indicated increases in knowledge and overall satisfaction with the content of the fermentation curriculum. The impact of the COVID-19 pandemic on the program outcomes are discussed.

Is There An Economic Advantage To Planting Diverse Summer Annual Forage Mixtures?, Kelly Mercier, Chris Teutsch, Ray Smith, Kenny Burdine, Edwin Ritchey, Eric Vanzant

This study examined economic implications of planting summer annual mixtures of grasses, legumes, and forbs at varying nitrogen rates. No differences in yield occurred between the three mixtures, indicating that mixtures with lowest seed cost will be most economical. Applying N resulted in yield increases of 12.26 lb DM per lb N applied. Although yield responses to N were positive, sensitivity analyses showed that applying N resulted in positive net returns only when hay prices were high and N prices were low. When utilization rates are accounted for, enterprise budgets determined grazing to be 18% cheaper to implement than haying.

Based on results collected through a comprehensive needs assessment of farmers market (FM) vendors in Pennsylvania, the purpose of this study was to develop and pilot-test a customized, food safety training program for FM vendors. We set out to determine whether the C-terminus (amino acids 481–798) of peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1α, UniProt Q9UBK2), a regulatory metabolic protein involved in mitochondrial biogenesis, and respiration, is an arginine methyltransferase substrate. Arginine methylation by protein arginine methyltransferases (PRMTs) alters protein function and thus contributes to various cellular processes. In addition to confirming methylation of the C-terminus by PRMT1 as described in the literature, we have identified methylation by another member of the PRMT family, PRMT7. The humoral antibody response against Kaposi sarcoma-associated herpesvirus (KSHV) in infected individuals has been characterized demonstrating the latency-associated nuclear antigen (LANA) as the most antigenic KSHV protein. Despite the antigenicity of the protein, specific LANA epitopes have not been systematically characterized. Here, we utilized a bacteriophage T7 library, which displays 56-amino acid KSHV LANA peptides with 28-amino acid overlap (VirScan), to define those epitopes in LANA targeted by antibodies from a cohort of 62 sub-Saharan African Kaposi Sarcoma (KS) patients and 22 KSHV-infected asymptomatic controls. The E3 ligase TNF receptor-associated factor 4 (TRAF4) is frequently overexpressed and closely related to poor prognosis in human malignancies. However, its effect on carcinogenesis and radiosensitivity in oral squamous cell carcinoma (OSCC) remains unclear. The present study found that TRAF4 was significantly upregulated in primary and relapsed OSCC tumor tissues. Depletion of TRAF4 markedly improved the sensitivity of OSCC cells to irradiation (IR) treatment, showing that tumor cell proliferation, colony formation and xenograft tumor growth were reduced. Mechanistically, IR promoted the interaction between TRAF4 and Akt to induce Akt K63-mediated ubiquitination and activation. TRAF4 knockout inhibited the phosphorylation of …

International current best practice recommends the discontinuation of antipsychotics for challenging behaviours in people with intellectual disabilities (ID), due to lack of evidence of efficacy and risks of harmful side-effects. In clinical practice, discontinuation may be difficult. The aim of this study was to gain insight into prescribers’ practice by investigating their experiences with the discontinuation of long-term antipsychotics for challenging behaviour. From professionals’ associations thirty-four registered ID physicians, psychiatrists and specialist mental healthcare nurses were recruited who completed an online questionnaire in this survey–study. Natural products, specifically complex, carbocyclic natural products, have served as the inspiration for novel methods development for decades. Over that time, metallacycle-mediated cross-coupling has arisen as a powerful means for the synthesis of fused carbocycles. This work details three methods for the synthesis and functionalization of carbocyclic motifs that have been enabled by Ti-mediated cross-coupling reactions. The first of these studies include a complementary method to the Pauson–Khand reaction for the synthesis of highly substituted cyclopentenones via Ti-mediated alkyne–β-ketoester coupling. Infection with the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is associated with acute and postacute cognitive and neuropsychiatric symptoms including impaired memory, concentration, attention, sleep and affect. Mechanisms underlying these brain symptoms remain understudied. Here we report that SARS-CoV-2-infected hamsters exhibit a lack of viral neuroinvasion despite aberrant blood-brain barrier permeability. Hamsters and patients deceased from coronavirus disease 2019 (COVID-19) also exhibit microglial activation and expression of interleukin (IL)-1β and IL-6, especially within the hippocampus and the medulla oblongata, when compared with non-COVID control hamsters and humans who died from other infections, cardiovascular disease, uraemia or trauma. Signed into law in 2016, the 21st Century Cures Act offers new hope to patients by empowering the FDA to expedite review of innovative, potentially lifesaving drugs. But these expedited approvals raise the risk that pivotal drug safety and efficacy data will not arise until after the drug is already on the market. The Cures Act failed to respond to two key aspects of shifting the discovery of safety and efficacy data to the postmarket phase. Every year, millions of Americans struggle with serious mental illness. Of them, thousands experience civil, or involuntary, commitment—that is, hospitals invoke the coercive power of the state to force these individuals into psychiatric hospitals against their will. Whether someone requires hospitalization is a complex question of psychology, medicine, and substantive law.

But the process of civil commitment itself is troubling. Across the board, states fail to afford those facing civil commitment meaningful procedural protections. Scholars have established cross-sectional connections between how married couples navigate their finances and their sexual relationship. For example, financial management behaviors have been shown to predict sexual satisfaction among newlywed couples. However, we know very little about the direction of the association between financial management behaviors and sexual satisfaction. Introduction

Pretreatment with the antiemetic trimethobenzamide has been recommended practice in the United States (US) to address the risk of nausea and vomiting during initiation of apomorphine treatment. However, trimethobenzamide is no longer being manufactured in the US, and despite the recent update to the US prescribing information, there may be uncertainty regarding how to initiate apomorphine.

Methods

To better understand why antiemetic pretreatment was recommended and if it is necessary when initiating apomorphine therapy, we performed a literature review of subcutaneous apomorphine therapy initiation with and without antiemetic pretreatment in patients with PD.
