DNA double-strand breaks (DSB) are one of the most serious DNA lesions because improper repair of a DSB can lead to loss of heterozygosity, aneuploidy, and cancer. One of the primary pathways to repair DSBs is homologous recombination (HR). HR resects the DNA around the DSB and then uses homologous DNA as a template to restore the broken sequence. RAD51 has a vital function in this pathway by forming a nucleoprotein filament on a resected end of the DSB. The nucleoprotein filament searches for homology within the homologous DNA. Once homology is located, strand invasion followed by strand exchange occurs. …
Exploring Visitor Perceptions Of Popular Culture Tourism In Sleepy Hollow, New York, Danielle Baker Kreh
Exploring Visitor Perceptions Of Popular Culture Tourism In Sleepy Hollow, New York, Danielle Baker Kreh
All Theses
Popular culture tourism can present itself in many forms including travel based on popular literature, film, and television series. Popular culture tourism destinations exist from the assumption that tourists wish to visit destinations based on their popular culture connections. In fact, destinations may choose to emphasize their popular culture connections for the purpose of attracting more visitors. Such is the case with Sleepy Hollow, New York, which has many popular culture connections including the original short story “The Legend of Sleepy Hollow” by Washington Irving. Using the village of Sleepy Hollow as an example, the purpose of this study is …

Cardiovascular diseases (CVDs) are the leading cause of death worldwide in males (XY) and females (XX). Prior to menopause, females have a relative protection against serious cardiac pathologies compared to age-matched males. This phenomenon is widely attributed to the ovarian hormone estrogen. Unfortunately, hormone replacement therapy to maintain estrogen levels in postmenopausal females has overall adverse effects, and it is not recommended for long-term use or as a preventative measure for eCVDs. A major driver of CVDs, specifically heart failure, is cardiac fibrosis: the continued buildup of scar tissue that reduces the heart’s ability to pump. There are currently no …
Isolation And Characterization Of Extracellular Vesicles From Various Biological Matrices Using Capillary-Channeled Polymer (C-Cp) Fiber Solid-Phase Extraction Spin-Down Tips, Kaylan Jackson
All Dissertations
A number of recent works have emphasized the need to isolate nanometer-scale analytes, like extracellular vesicles (EVs), from various biologically-relevant fluids. Exosomes are a subset of small EVs that range from 30-200 nm in diameter that serve as biomolecular snapshots of their cell of origin containing mother cell-specific DNA, miRNA, mRNA, and proteins. As critical components of intercellular communication, exosomes and other EVs play significant roles in many physiological and pathological processes. Diverse populations of these vesicles can be collected from biofluids, including blood, saliva, and urine, from cell culture conditioned media and primary cells, and even from plant …

Modeling Electrostatics In Molecular Biology And Its Relevance With Molecular Mechanisms Of Diseases, Mahesh Koirala
All Dissertations
Electrostatics plays an essential role in molecular biology. Modeling electrostatics in molecular biology is complicated due to the water phase, mobile ions, and irregularly shaped inhomogeneous biological macromolecules. This dissertation presents the popular DelPhi package that solves PBE and delivers the electrostatic potential distribution of biomolecules. We used the newly developed DelPhiForce steered Molecular Dynamics (DFMD) approach to model the binding of barstar to barnase and demonstrated that the first-principles method could also model the binding. This dissertation also reflects the use of existing computational approaches to model the effects of Single Amino Acid Variations (SAVs) to reveal molecular mechanisms …

The overall theme is to use nanoparticles with interesting optical properties compared to their atomic levels and bulk …
Reading Toward Breath: A Poetic Ecology Of Creative Reading, Jessica Schad Manuel
Reading Toward Breath: A Poetic Ecology Of Creative Reading, Jessica Schad Manuel
All Dissertations
Reading is an act of perception that begins in wonder and leads to wisdom. It is not a response to writing but a response-ability we have to interact with the world around us and the phenomena before us. Reading with the body leads to wisdom, and when we participate in our existence by being in the world, we are reading. So what is reading? Reading is making. We make connections, and we form relationships. The act of reading is more than a cognitive process and even surpasses the phenomenological encounter.
My research describes the reader’s relationship to the text as …

Heat Stress Response And Excystation In Entamoeba Histolytica, Irem Bastuzel
All Dissertations
Entamoeba histolytica is a water- and food-borne intestinal protozoan parasite that causes amoebiasis and liver abscess and is responsible for symptomatic disease in approximately 100 million people each year leading to ~ 100,000 deaths. The most common disease transmission follows the oral-fecal route, but it can also be transmitted by mechanical vectors such as animals carrying the amoeba from contaminated sources to water systems. In rare cases, disease transmission has been recorded in some patients in which men-to-men sexual practices were preferred.
The life cycle of E. histolytica starts through ingestion of infectious cysts, which are non-dividing, quadri-nucleated structures surrounded …
Integration Of Techno-Economic Analysis (Tea) And Life Cycle Assessment (Lca) For Sustainable Process Design, Roksana Mahmud
Integration Of Techno-Economic Analysis (Tea) And Life Cycle Assessment (Lca) For Sustainable Process Design, Roksana Mahmud
All Dissertations
For sustainable design, technology developers need to consider not only technical and economic aspects but also potential environmental impacts while developing new technologies. Techno-economic analysis (TEA) evaluates the technical performance and economic feasibility of a technology. Life cycle assessment (LCA) evaluates the potential environmental impacts associated with a product system throughout its life cycle from raw material extraction to disposal. Generally, TEA and LCA are performed separately for technology assessment. Understanding of the trade-off between economic and environmental performances is crucial for sustainable process design, which is not fully available if TEA and LCA is performed separately. In contrast, integration …

Gender Neutral Parenting: Raising A Generation Outside The Gender Binary, Toni Noelle Martinez
Graduate Theses and Dissertations
In the United States, the social and cultural reality remains organized around the gender binary. The binary legitimizes itself on the widely held belief that gender is determined by biology and, therefore, is “natural.” By exploring and firmly placing gender as a cultural construct, this thesis looks at the possibilities of fracturing the binary. Borrowing from Stephan Hirschauer (1994) and Judith Butler’s (2004), this thesis theorizes what a gender neutral world could look like and examines how Gender Neutral Parents contribute toward a gender revolution. Gender Neutral Parents, a community that is mostly found online, represent a small group that …

Operationalizing The Gift Of Love (Gol) In Interpersonal Reconstructive Therapy (Irt): An Examination Of The Role Of Meaning Reconstruction In Therapeutic Change., Eliza Stucker-Rozovsky
Dissertations, 2020-current
Interpersonal Reconstructive Therapy (IRT) (Benjamin, 2003/2006) is an integrative, principles-based treatment approach and theory of psychopathology that conceptualizes the motivating factor that underlies personality psychopathology as the gift of love (GOL). That is, copy processes are learned and maintained to achieve psychic proximity to the important people and their internalized representations (IPIRs) that were the original teachers. Relinquishing this gift of love allows a person to re-orient their life towards uncovering and living by their own individually held meanings that will then define how they relate to themselves and their world. Spirituality and religion have historically helped individuals make meaning …

Fibrodysplasia ossificans progressiva (FOP) is a rare and devastating genetic disease, in which soft connective tissue is converted into heterotopic bone through an endochondral ossification process. Patients succumb early as they gradually become trapped in a second skeleton of heterotopic bone. Although the underlying genetic defect is long known, the inherent complexity of the disease has hindered the discovery of effective preventions and treatments. New developments in the gene therapy field have motivated its consideration as an attractive therapeutic option for FOP. However, the immune system's role in FOP activation and the as-yet unknown primary causative cell, are crucial issues …

Department of Medical Oncology Faculty Papers
In luminal gastrointestinal tumors, immune checkpoint inhibitors (ICIs) targeting PD-1, PD-L1 and CTLA-4 have been investigated in multiple settings. The indications for these drugs are primarily dependent on specific biomarkers that imply immunogenicity: overexpression of PD-L1, tumor mutational burden, loss of mismatch repair proteins (dMMR) and/or high microsatellite instability status. Although these markers can be both predictive and prognostic, there is variability in how they are measured and used to guide therapies. Moreover, the use of ICIs can be further refined with a better understanding of the tumor microenvironment and interactions with other available therapies. The purpose of this review …
