INTRODUCTION AND OBJECTIVE: Despite the improvements in management and treatment of chordomas over time, the risk of disease recurrence remains high. Consequently, there is a push to develop effective systemic therapeutics for newly diagnosed and recurrent disease. In order to tailor treatment for individual chordoma patients and develop effective surveillance strategies, suitable clinical biomarkers need to be identified. The objective of this study was to systematically review all prognostic biomarkers for chordomas reported to date in order to classify them according to localization, study design and statistical analysis.

Background
Microbes are associated with many human diseases and influence drug efficacy. Small-molecule drugs may revolutionize biomedicine by fine-tuning the microbiota on the basis of individual patient microbiome signatures. However, emerging endeavors in small-molecule microbiome drug discovery continue to follow a conventional “one-drug-one-target-one-disease” process. A systematic pharmacology approach that would suppress multiple interacting pathogenic species in the microbiome, could offer an attractive alternative solution.
Results
We construct a disease-centric signed microbe–microbe interaction network using curated microbe metabolite information and their effects on host. We develop a Signed Random Walk with Restart algorithm for the accurate prediction of effect of microbes …

The intestinal microbiota is an important modulator of graft-versus-host disease (GVHD), which often complicates allogeneic hematopoietic stem cell transplantation (allo-HSCT). Broad-spectrum antibiotics such as carbapenems increase the risk for intestinal GVHD, but mechanisms are not well understood. In this study, we found that treatment with meropenem, a commonly used carbapenem, aggravates colonic GVHD in mice via the expansion of Bacteroides thetaiotaomicron (BT). BT has a broad ability to degrade dietary polysaccharides and host mucin glycans. BT in meropenem-treated allogeneic mice demonstrated upregulated expression of enzymes involved in the degradation of mucin glycans. These mice also had thinning of the colonic …

This research makes use of a hypothesis generation technique known as Literature-Based Discovery (LBD) to study certain under-researched topics related to online game addiction problems in our society. This research work tries to primarily address two problems related to game addiction which have been under-studied according to the literature available on this topic. In the first problem, we try to identify certain moderating factors of massively multiplayer games which weakens the relationship between the psychological flow state and game addiction. In IDH-wildtype glioblastomas which meet the histopathological or molecular diagnosis criteria, it remains unclear whether the presence of TERT promotor mutations provides additional prognostic information. Based on a multicenter cohort of 466 IDH-wildtype glioblastomas (including 396 with and 70 patients without TERT promotor mutations), we found that TERT promotor mutations were neither associated with progression-free survival nor overall survival. This held true in various treatment-based or molecular subgroups. This argues against standardized analysis for TERT promotor mutation status for the purpose of prognostic or therapeutic relevance in newly diagnosed IDH-wildtype glioblastoma that otherwise meets the histopathological and …

Turbidity currents commonly bypass sediment in submarine channels on the continental slope, and deposit sediment lobes farther down-dip on the flat and unconfined abyssal plain. Seafloor and outcrop data have shown that the transition from bypass to deposition usually occurs over complex zones referred to as channel–lobe transition zones (CLTZs). Recognition of these zones in cores and outcrop remains challenging due to a lack of characteristic sedimentary facies and structures. Background: The endoplasmic reticulum (ER)-membrane protein complex (EMC) is a multi-protein transmembrane complex composed of 10 subunits that functions as a membrane-protein chaperone. Variants in EMC1 lead to neurodevelopmental delay and cerebellar degeneration. Multiple families with biallelic variants have been published, yet to date, only a single report of a monoallelic variant has been described, and functional evidence is sparse.
Methods: Exome sequencing was used to investigate the genetic cause underlying severe developmental delay in three unrelated children. The ability to monitor mental effort during a task using a wearable sensor may improve productivity for both work and study. The use of the electrodermal activity (EDA) signal for tracking mental effort is an emerging area of research. Through analysis of over 92 h of data collected with the Empatica E4 on a single participant across 91 different activities, we report on the efficacy of using EDA features getting at signal intensity, signal dispersion, and peak intensity for prediction of the participant's self-reported mental effort. In this study, the response surface methodology (RSM) and artificial neural network (ANN) were employed to study the adsorption process of 2,4-dichlorophenoxyacetic acid (2,4-D) by using modified hydrogel, i.e., activated carbon poly(dimethylaminoethyl methacrylate) (AC/PDMAEMA hydrogel). The effect of pH, the initial concentration of 2,4-D and the activated carbon content on the removal of 2,4-D and adsorption capacity were investigated through the face-centered composite design (FCCD), optimal design and two-level factorial design. The response surface plot suggested that higher removal of 2,4-D and adsorption capacity could be achieved at the higher initial concentration of 2,4-D and lower pH and activated carbon …

Intrusion detection systems examine the computer or network for potential security vulnerabilities. Time series data is real-valued. The nature of the data influences the type of anomaly detection. As a result, network anomalies are operations that deviate from the norm. These anomalies can cause a wide range of device malfunctions, overloads, and network intrusions. Developing high-performance and low-cost electrocatalysts for oxygen evolution reaction (OER) is the key to implementing polymer electrolyte membrane water electrolyzer (PEMWE) for hydrogen production. To date, iridium (Ir) is the state-of-the-art OER catalyst, but still suffers from the insufficient activity and scarce earth abundance, which results in high cost both in stack and electricity. Environments where geothermal waters and glacier meltwater mix are common on Earth yet little is known about the biogeochemical processes that occur when hot, reduced geothermal water mixes with cold, oxidized glacial meltwater in natural systems. Mount St. Helens provides an ideal location to study the interaction between geothermal and glacier waters since the water sources, and their mixing environment in Step Creek, are exposed in the volcanic crater. We find that the two water sources contain distinct major ion, trace element, dissolved organic matter (DOM), and biological signatures. Human pegivirus (HPgV) is best known for persistent, presumably non-pathogenic, infection and a propensity to co-infect with human immunodeficiency virus or hepatitis C virus. However, unique attributes, such as the increased risk of malignancy or immune modulation, have been recently recognized for HPgV. We have identified a unique case of a woman with high levels HPgV infection in two pregnancies, which occurred 4 years apart and without evidence of human immunodeficiency virus or hepatitis C virus infection. Early-life experience often shapes behaviors like innovation and exploration. These behaviors are important to animals encountering novel food resources in diverse habitats, such as mesocarnivores in urban areas. To understand if early-life experiences impact later-life behavior, we examined how coyotes (Canis latrans) responded to a multi-access puzzle box at two life stages: pup (~ 7 weeks) and dispersal (~ 10 months). We first exposed pups, still living with their parents and littermates, to a baited puzzle box. The data in this article are associated with the research paper “GigaAssay – an adaptable high-throughput saturation mutagenesis assay". The raw data are sequence reads of HIV-1 Tat cDNA amplified from cellular genomic DNA in a new single-pot saturation mutagenesis assay designated the “GigaAssay”. A bioinformatic pipeline and parameters used to analyze the data. Raw, processed, analyzed, and filtered data are reported. The data is processed to calculate the Tat-driven transcription activity for cells with each possible single amino acid substitution in Tat. Objectives: In chronic diseases such as rheumatoid arthritis (RA), carbamylation may cause the production of autoantibodies such as the anti-carbamylated protein antibody (anti-CarP) and protein malfunctioning. However, its relation to periodontitis (PD) remains unclear. Therefore, this study aimed to assess the serum levels of anti-carbamylated protein antibodies (anti-CarP) in four groups of participants, the RAPD, RA, PD, and healthy groups and correlate the findings with rheumatological parameters.
