Endothelial cells (ECs) are the primary cellular constituent of blood vessels that are in direct contact with hemodynamic forces over their lifetime. Throughout the body, vessels experience different blood flow patterns and rates that alter vascular architecture and cellular behavior. Because of the complexities of studying blood flow in an intact organism, particularly during development, the field has increasingly relied on in vitro modeling of blood flow as a powerful technique for studying hemodynamic-dependent signaling mechanisms in ECs. While commercial flow systems that recirculate fluids exist, many commercially available pumps are peristaltic and best model pulsatile flow conditions. However, there …

Sexual Health Education And Life Satisfaction For People With Congenital Neurological Disabilities, Alie Kriofske Mainella, Susan Miller Smedema
College of Education Faculty Research and Publications
People with disabilities are sexual beings, yet there is little research on sexuality in this population. The present study explored the impact of sex education for people with congenital neurological disabilities, largely, spina bifida and cerebral palsy, on sexual self-concept and life satisfaction. This study included 104 adults with spina bifida, cerebral palsy, and other congenital neurological disabilities. Hierarchical regression analysis was used to examine the relationships between demographic variables, sexual health education variables, and outcome variables (sexual self-concept and life satisfaction). Serial mediation analysis was conducted to examine the mediating relationship of sexual self-concept variables (sexual anxiety and sexual …

Health Literacy And Its Association With Human Papilloma Virus Vaccination Among Adults: Findings From The Behavioral Risk Factor Surveillance System, Vinodh Bhoopathi, Bhagavatula Pradeep, Maharaj Singh
College of Nursing Faculty Research and Publications
Health literacy is associated with the utilization of preventive health services. We examined the association between health literacy (HL) levels and receipt of at least one dose of the human papilloma virus (HPV) vaccination. We analyzed the data from the 2016 Behavioral Risk Factor Surveillance System (BRFSS) among adults aged 18 to 32. The primary outcome variable was the ‘yes/no’ response to the question that assessed whether the participant received at least the first dose of HPV vaccination. The primary independent variable was a summative HL score (range 3 through 12) we created for each respondent by adding the scores …

The Association Of Preoperative Frailty And Neighborhood-Level Disadvantage With Outcome In Patients With Newly Diagnosed High Grade Glioma, Pranav Mirpuri, Maharaj Singh, Richard Rovin
College of Nursing Faculty Research and Publications
Objective
Individual patient and socioeconomic factors are underexplored prognostic factors for glioblastoma (GBM). Frailty, a measure of physiological vulnerability, and area deprivation, a measure of socioeconomic status, are easily obtained during the preoperative evaluation. These metrics are predictors of outcome and access to treatments for other cancers. Therefore, we sought to determine the association of frailty and neighborhood disadvantage with outcomes of patients with newly diagnosed GBM.
Methods
This was a retrospective review of newly diagnosed patients with GBM undergoing surgery from 2015 through 2020. The 5-factor modified frailty index and national area deprivation index were determined for each patient. …

Defining Foundational Competence For Prelicensure And Graduate Nursing Students: A Concept Analysis And Conceptual Model, Katie L. Mcdermott, Kristina Dreifuerst
College of Nursing Faculty Research and Publications
Aim
To define the concept of foundational competence as it pertains to prelicensure and graduate nursing students during their educational program, thereby informing pedagogical and assessment practices.
Background
Competence has many definitions and uses, however a lack of consensus of a clear and useful definition in nursing persists. Without shared language, competence remains open for interpretation across and among groups and impedes the ability to teach and assess it in a competency-based education framework.
Method
Using the Walker & Avant method of concept analysis, literature was reviewed and synthesized to define and explain the concept of foundational competence in nursing …

Quantitative Single-Cell Analysis Of Leishmania Major Amastigote Differentiation Demonstrates Variably Extended Expression Of The Lipophosphoglycan (Lpg) Virulence Factor In Different Host Cell Types, Michael A. Mandell, Wandy L. Beatty, Stephen M. Beverley
2020-Current year OA Pubs
Immediately following their deposition into the mammalian host by an infected sand fly vector, Leishmania parasites encounter and are engulfed by a variety of cell types. From there, parasites may transit to other cell types, primarily macrophages or dendritic cells, where they replicate and induce pathology. During this time, Leishmania cells undergo a dramatic transformation from the motile non-replicating metacyclic stage to the non-motile replicative amastigote stage, a differentiative process that can be termed amastigogenesis. To follow this at the single cell level, we identified a suite of experimental 'landmarks' delineating different stages of amastigogenesis qualitatively or quantitatively, including new …

Nuclear envelope membrane proteins (NEMPs) are a conserved family of nuclear envelope (NE) proteins that reside within the inner nuclear membrane (INM). Even though Nemp1 knockout (KO) mice are overtly normal, they display a pronounced splenomegaly. This phenotype and recent reports describing a requirement for NE openings during erythroblasts terminal maturation led us to examine a potential role for Nemp1 in erythropoiesis. Here, we report that Nemp1 KO mice show peripheral blood defects, anemia in neonates, ineffective erythropoiesis, splenomegaly, and stress erythropoiesis. The erythroid lineage of Nemp1 KO mice is overrepresented until the pronounced apoptosis of polychromatophilic erythroblasts. We show …

UNLABELLED: As part of the Multiple Myeloma Research Foundation (MMRF) immune atlas pilot project, we compared immune cells of multiple myeloma bone marrow samples from 18 patients assessed by single-cell RNA sequencing (scRNA-seq), mass cytometry (CyTOF), and cellular indexing of transcriptomes and epitopes by sequencing (CITE-seq) to understand the concordance of measurements among single-cell techniques. Cell type abundances are relatively consistent across the three approaches, while variations are observed in T cells, macrophages, and monocytes. Concordance and correlation analysis of cell type marker gene expression across different modalities highlighted the importance of choosing cell type marker genes best suited to …

A Mixed Methodological Examination Of Older Adults’ Psychological Reactance Toward Caregiving Messages From Their Adult Children, Hannah Ball, Keith Weber, Alan K. Goodboy, Christine E. Kunkle, Christa L. Lilly, Scott A. Myers
Communication Faculty Articles and Research
This study extends psychological reactance theory (PRT) to family caregiving by exploring autonomy-threatening messages adult child caregivers use to gain compliance from older adult parents. Results of focus groups and interviews with older adult care recipients (Study One) and caregivers (Study Two) corroborated three types of autonomy-threatening messages, which were used to test PRT (Study Three). Older adults (N = 281) were randomly assigned a caregiving message and answered reactance-related survey questions. Results supported serial mediation: relative to an autonomy-supporting message, two types of autonomy-threatening messages (i.e., offering directives, expressing doubt) triggered greater freedom threat, which amplified reactance. In …

Pyrus calleryana is one of the most problematic invasive species in the eastern United States. The mechanisms that enable Py. calleryana to establish and outcompete native plants are not fully understood but likely include a profile of advantageous traits. Extended leaf phenology is a characteristic noted in many woody invasive plants. Leaf phenology of Py. calleryana and two native woody species, Populus deltoides and Platanus occidentalis, was observed in natural areas near Dayton, OH from December 2019 to November 2020. A frost event in May also gave us the serendipitous opportunity to assess frost tolerance of these species. We …

Research outputs 2022 to 2026
Despite improvements made with checkpoint inhibitor (CPI) therapy, a need for new approaches to improve outcomes for patients with unresectable or metastatic melanoma remains. EVX-01, a personalized neoepitope vaccine, combined with pembrolizumab treatment, holds the potential to fulfill this need. Here we present the rationale and novel design behind the KEYNOTE - D36 trial: an open label, single arm, phase II trial aiming to establish the clinical proof of concept and evaluate the safety of EVX-01 in combination with pembrolizumab in CPI naive patients with unresectable or metastatic melanoma. The primary objective is to evaluate if EVX-01 improves best overall …

Sediment Accumulation And Carbon Burial In Four Hadal Trench Systems, Kazumasa Oguri, Pere Masqué, Matthias Zabel, Heather A. Stewart, Gillian Mackinnon, Ashley A. Rowden, Peter Berg, Frank Wenzhöfer, Ronnie N. Glud
Research outputs 2022 to 2026
Hadal trenches are considered to act as depocenters for organic material, although pathways for the material transport and deposition rates are poorly constrained. Here we assess focusing, deposition and accumulation of material and organic carbon in four hadal trench systems underlying different surface ocean productivities; the eutrophic Atacama and Kuril-Kamchatka trenches, the mesotrophic Kermadec trench and the oligotrophic Mariana Trench. The study is based on the distributions of naturally occurring 210Pbex, 137Cs and total organic carbon from recovered sediment cores and by applying previously quantified benthic mineralization rates. Periods of steady deposition and discreet mass-wasting deposits were …

State Trends Of Cannabis Liberalization As A Causal Driver Of Increasing Testicular Cancer Rates Across The Usa, Albert Stuart Reece, Gary Kenneth Hulse
Research outputs 2022 to 2026
Background:
The cause of the worldwide doubling-tripling of testicular cancer rates (TCRs) in recent decades is unknown. Previous cohort studies associated cannabis use with TCR including dose–response relationships but the contribution of cannabis to TCRs at the population level is unknown. This relationship was tested by analyzing annual trends across US states and formally assessed causality. Four US datasets were linked at state level: age-adjusted TCRs from Centers for Disease Control Surveillance Epidemiology and End Results database; drug use data from annual National Survey of Drug Use and Health including 74.1 % response rate; ethnicity and median household income data …

Proviral integration of Moloney virus 2 (PIM2) is a pro‑survival factor of cancer cells and a possible therapeutic target in hematological malignancies. However, the attempts at inhibiting PIM2 have yielded underwhelming results in early clinical trials on hematological malignancies. Recently, a novel pan‑PIM inhibitor, JP11646, was developed. The present study examined the utility of targeting PIM2 in multiple solid cancers and investigated the antitumor efficacy and the mechanisms of action of JP11646. When PIM2 expression was compared between normal and cancer tissues in publicly available datasets, PIM2 was found to be overexpressed in several types of solid cancers. PIM2 ectopic …

Increasing evidence shows that the host gut microbiota might be involved in the immunological cascade that culminates with the formation of tissue granulomas underlying the pathophysiology of hepato-intestinal schistosomiasis. In this study, we investigated the impact of Schistosoma mansoni infection on the gut microbial composition and functional potential of both wild type and microbiome-humanized mice. In spite of substantial differences in microbiome composition at baseline, selected pathways were consistently affected by parasite infection. The gut microbiomes of infected mice of both lines displayed, amongst other features, enhanced capacity for tryptophan and butyrate production, which might be linked to the activation …

Teratogenic Effects Of Cannabinoid And Serotonin Receptor Disruption On Neural Crest Development, Brian K. Wells
Teratogenic Effects Of Cannabinoid And Serotonin Receptor Disruption On Neural Crest Development, Brian K. Wells
Biology Theses
Neurotransmitters are well-studied in adults and have often been the focus of many treatments for both physical and psychological ailments. This thesis explores the teratogenicity of altering the activity at the serotonin 2B and 2C receptors (5-HT2B; 5-HT2C) and the cannabinoid 1 receptor (CB1R) in the serotonergic and endocannabinoid systems, respectively. This investigation is focused on neural crest derivative structures. Chapter 1 reviews the background information behind cardiac and cranial neural crest cells and overviews the serotonergic and endocannabinoid systems. In Chapter 2, using a pharmacological agent, 1-methylpsilocin (1-MP), we disrupt the activity of 5-HT2B and 5-HT2C in the early …
