The opioid crisis has led to an increased number of drug overdoses in recent years. Several approaches have been established to predict opioid prescription by health practitioners. However, due to the complex nature of the problem, the accuracy of such methods is not yet satisfactory. Dependable and reliable classification of opioid dependent patients from well-grounded data sources is essential. Majority of the previous studies do not focus on the users’ mental health association for opioid intake classification. The humoral antibody response against Kaposi sarcoma-associated herpesvirus (KSHV) in infected individuals has been characterized demonstrating the latency-associated nuclear antigen (LANA) as the most antigenic KSHV protein. Despite the antigenicity of the protein, specific LANA epitopes have not been systematically characterized. Here, we utilized a bacteriophage T7 library, which displays 56-amino acid KSHV LANA peptides with 28-amino acid overlap (VirScan), to define those epitopes in LANA targeted by antibodies from a cohort of 62 sub-Saharan African Kaposi Sarcoma (KS) patients and 22 KSHV-infected asymptomatic controls. Understanding the genetic diversity and mechanisms underlying genetic variation in pathogen populations is crucial to the development of effective control strategies. We investigated the genetic diversity and reproductive biology of Colletotrichum graminicola isolates which infect maize by sequencing the genomes of 108 isolates collected from 14 countries using restriction site-associated DNA sequencing (RAD-seq) and wholegenome sequencing (WGS). Clustering analyses based on single-nucleotide polymorphisms revealed three genetic groups delimited by continental origin, compatible with short-dispersal of the pathogen and geographic subdivision. The purpose of this phenomenological study was to describe foster parents’ trauma knowledge in Central Virginia. The theories guiding this study include childhood complex trauma theory and experiential learning theory (ELT). As discussed by Herman (1992) and Terr (1991), childhood complex trauma theory explains the behaviors and actions of foster children who have suffered complex trauma. Meaningful watershed educational experiences (MWEE) are student-centered, problem solving-oriented environmental educational experiences in which students investigate local environmental issues leading to civic engagement and informed action. The four components of an MWEE include issue definition, outdoor field experience, synthesis and conclusion, and action projects. Precision nutrition is an emerging concept that aims to develop nutrition recommendations tailored to different people's circumstances and biological characteristics. Responses to dietary change and the resulting health outcomes from consuming different diets may vary significantly between people based on interactions between their genetic backgrounds, physiology, microbiome, underlying health status, behaviors, social influences, and environmental exposures. Active continental rifts are ideal sites for understanding the break-up of continents, and long-lived rift lake environments are known as important reservoirs for endemic communities and biodiversity. The sedimentary fill of extensional continental rifts within the East Africa Rift System (EARS) records a long history of continental extension and variable tropical climate that is unparalleled in its duration and fidelity. Traditional data acquisition methods in Nuclear Magnetic Resonance (NMR) collect a subset of the total amount of generated data and discard the rest. In this work, a modified approach toward NMR data acquisition and analysis is formalized and applied in a series of different contexts, ultimately illustrating the utility of collecting and considering this traditionally discarded data. We set out to determine whether the C-terminus (amino acids 481–798) of peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1α, UniProt Q9UBK2), a regulatory metabolic protein involved in mitochondrial biogenesis, and respiration, is an arginine methyltransferase substrate. Arginine methylation by protein arginine methyltransferases (PRMTs) alters protein function and thus contributes to various cellular processes. In addition to confirming methylation of the C-terminus by PRMT1 as described in the literature, we have identified methylation by another member of the PRMT family, PRMT7. We performed in vitro methylation reactions using recombinant mammalian PRMT7 and PRMT1 at 37, 30, 21, 18, and 4 °C. …

Summary
Background
One challenge in the field of in-vitro fertilisation is the selection of the most viable embryos for transfer. Morphological quality assessment and morphokinetic analysis both have the disadvantage of intra-observer and inter-observer variability. A third method, preimplantation genetic testing for aneuploidy (PGT-A), has limitations too, including its invasiveness and cost. We hypothesised that differences in aneuploid and euploid embryos that allow for model-based classification are reflected in morphology, morphokinetics, and associated clinical information.

Objective: This study aims to investigate the effect of Anastatica Hierochuntica L. on the duration of labour in primiparous women.
Methods: A prospective, case-controlled study with an intervention (n=28) and control (n=29) group. Participants were not randomized; their preferences related to the use of the A. Hierochuntica L. determined the group in which they would be included. In the intervention group, Anastatica Hierochuntica L. was used by all women. Standard care continued for both the intervention and control group. The duration of the first and second stage of labour were evaluated and compared between groups.

Signed into law in 2016, the 21st Century Cures Act offers new hope to patients by empowering the FDA to expedite review of innovative, potentially lifesaving drugs. But these expedited approvals raise the risk that pivotal drug safety and efficacy data will not arise until after the drug is already on the market. The Cures Act failed to respond to two key aspects of shifting the discovery of safety and efficacy data to the postmarket phase. While early childhood circumstances influence the accumulation of skills and abilities, parental and policy responses can magnify or mitigate the effects of childhood circumstances. This work explores the two ideas mentioned above.
Chapter 1 briefs the historical and philosophical roots of the study of human capital and discusses the economic approach to the understanding of child wellbeing before the age of five. Because childhood circumstances influence the accumulation of skills, Chapter 2 estimates the impact of temperature shock during pregnancy on early childhood health. Melanoma is one of the most lethal tumors with highly aggressive and metastatic properties. Although immunotherapy and targeted therapy have certain therapeutic effects in melanoma, a significant proportion of patients still have drug resistance after treatment. Recent studies have shown that long noncoding RNAs (lncRNAs) are widely recognized as regulatory factors in cancer. Phosphatidylethanol (PEth) is an alcohol biomarker that provides a longer window of detection than direct alcohol measurement or other common alcohol metabolites, like ethyl glucuronide. PEth has become useful in assessing drinking history, such as for alcohol-withdrawal treatment, organ transplant settings or prenatal alcohol exposure determination. A method was developed and validated to quantify PEth 16:0/18:1 and 16:0/18:2 in whole blood and dried blood spots (DBS) by liquid chromatography tandem mass spectrometry (LC-MS/MS). The purpose of this qualitative phenomenological study was to explore the lived experiences of Black women in leadership positions in the healthcare field. The study used an intersectional lens to illustrate how gender, race, class, and spirituality contributed to the systemic structures of oppression, bias, and discrimination, limiting ascension to positions of power and authority. The theoretical framework guiding this inquiry was the great man leadership theory, which analyzed how socio-cultural identities influenced Black women’s perceptions and experiences of leadership within the organizational context. Many aquatic organisms are experiencing increasingly severe and frequent droughts and drying events. Simultaneously, drought effects are carrying over to non-drought years as ecosystems remain in incomplete states of recovery. Aquatic organisms are thus faced with fewer sequential years under degraded environmental conditions to prepare for increasingly severe droughts and potential drying events. We assessed the effect of droughts and sex on the growth, mass, and mass-dependent estivation potential of long-lived aquatic salamanders (Greater Sirens, Siren lacertina) that estivate during drying events brought on by severe droughts. The humoral antibody response against Kaposi sarcoma-associated herpesvirus (KSHV) in infected individuals has been characterized demonstrating the latency-associated nuclear antigen (LANA) as the most antigenic KSHV protein. Despite the antigenicity of the protein, specific LANA epitopes have not been systematically characterized. Here, we utilized a bacteriophage T7 library, which displays 56-amino acid KSHV LANA peptides with 28-amino acid overlap (VirScan), to define those epitopes in LANA targeted by antibodies from a cohort of 62 sub-Saharan African Kaposi sarcoma (KS) patients and 22 KSHV-infected asymptomatic controls. Wheat streak mosaic virus (WSMV; genus Tritimovirus, family Potyviridae) is the causal agent of the most economically important wheat streak mosaic disease of wheat (Triticum aestivum) in the Great Plains region of theUnited States.WSMVdeterminants responsible forwheat streak mosaic disease in wheat are unknown. Triticum mosaic virus (TriMV), a wheatinfecting virus,was used as an expression vector for the transient expression of each of the WSMV-encoded cistrons in wheat. The E3 ligase TNF receptor-associated factor 4 (TRAF4) is frequently overexpressed and closely related to poor prognosis in human malignancies. However, its effect on carcinogenesis and radiosensitivity in oral squamous cell carcinoma (OSCC) remains unclear. The present study found that TRAF4 was significantly upregulated in primary and relapsed OSCC tumor tissues. Depletion of TRAF4 markedly improved the sensitivity of OSCC cells to irradiation (IR) treatment, showing that tumor cell proliferation, colony formation and xenograft tumor growth were reduced. Mechanistically, IR promoted the interaction between TRAF4 and Akt to induce Akt K63-mediated ubiquitination and activation. TRAF4 knockout inhibited the phosphorylation of …
