Programmed reduction of synapses is a hallmark of the developing brain, with sensory systems emerging as useful models with which to study this pruning. The central projections (terminal field) of the gustatory glossopharyngeal nerve (GL) of the rat are a prime example of developmental pruning, undergoing an approximate 66% reduction in volume from postnatal day 15 (P15) to P25. Later in adulthood, developmental GL pruning can be experimentally reversed, expanding to preweaning volumes, suggesting mature volumes may be actively maintained throughout the life span. In simian-human immunodeficiency virus (SHIV)-infected non-human primates, broadly neutralizing antibodies (bNAbs) against the virus appear to stimulate T cell immunity. To determine whether this phenomenon also occurs in humans we measured HIV-1-specific cellular immunity longitudinally in individuals with HIV-1 starting antiviral therapy (ART) with or without adjunctive bNAb 3BNC117 treatment. Using the activation-induced marker (AIM) assay and interferon-γ release, we observe that frequencies of Pol- and Gag-specific CD8+ T cells, as well as Gag-induced interferon-γ responses, are significantly higher among individuals that received adjunctive 3BNC117 compared to ART-alone at 3 and 12 months after starting ART. Burkholderia glumae is a Gram-negative bacterium causing bacterial panicle blight (BPB) in rice. BPB can cause severe damage to rice and reduce rice yield up to 75%, while no effective treatment has been developed to control the disease. B. glumae produces and secretes an essential virulence factor, toxoflavin, which is mainly responsible for the symptoms of BPB. Synthesis and transport of toxoflavin are regulated in a quorum-sensing dependent manner. Members of the DedA superfamily are putative membrane transporters and found in nearly all species. Mutation of dedA in Burkholderia species results in sensitivity to colistin, an antibiotic belonging to the …

As many parts of the world continue to fight the innumerable waves of COVID-19 infection, SARS-CoV-2 continues to sculpt its antigenic determinants to enhance its virulence and evolvability. Several vaccines were developed and used around the world, and oral antiviral medications are being developed against SARS-CoV-2. However, studies showed that the virus is mutating in line with the antibody's neutralization escape; thus, new therapeutic alternatives are solicited. Establishing The Non-Canonical Nature Of A Leucyl-Trna Synthetase Paralog In Sulfolobus Islandicus, Jonathan Blair Brewster

Aminoacyl-tRNA synthetases (aaRSs) are essential proteins. Their canonical function is to catalyze the attachment of amino acids to cognate tRNA. The organism Sulfolobus islandicus contains two aaRSs for leucine. These leucyl-tRNA synthetase (LeuRS) paralogs were previously studied using a single tRNALeu isoacceptor, tRNALeu-UAG. One leucyl-tRNA synthetase, LeuRS-F, was shown to faithfully perform the necessary function of charging, the process of catalytically attaching leucine to tRNALeu. Furthermore, the essentiality of LeuRS-F was emphasized by an inability to remove its gene from S. islandicus. The paralog, LeuRS-I, was shown to have no aminoacylation activity, compared to LeuRS-F, even though it could activate …

Primary liver cancer is a heterogeneous disease in terms of its etiology, histology, and therapeutic response. Concurrent proteomic and genomic characterization of a large set of clinical liver cancer samples can help elucidate the molecular basis of heterogeneity and thus serve as a valuable resource for personalized liver cancer treatment. In this study, we perform proteomic profiling of ~300 proteins on 259 primary liver cancer tissues with reverse-phase protein arrays, mutational analysis using whole genome sequencing and transcriptional analysis with RNA-Seq. Patients are of Japanese ethnic background and mainly HBV or HCV positive, providing insight into this important liver cancer …

Type 1 diabetes is a complex disease characterized by the lack of endogenous insulin secreted from the pancreatic β-cells. Although β-cell targeted autoimmune processes and β-cell dysfunction are known to occur in type 1 diabetes, a complete understanding of the cell-to-cell interactions that support pancreatic function is still lacking. To characterize the pancreatic endocrine compartment, we studied pancreata from healthy adult donors and investigated a single cell surface adhesion molecule, desmoglein-2 (DSG2). Anthropogenic impacts on marine environments can impact metal fluxes and concentrations available to marine species. Monitoring these impacts is necessary to better understand the interactions between the biotic and abiotic components of these ecosystems and mitigate the risk posed by harmful toxins introduced by human activities. Biomoniters, like macroscopic algae, are useful indicators that illuminate the bioaccumulation of toxins commonly introduced from anthropogenic activity. Coastal environments of Humboldt County provide an optimal habitat for a diversity of macroinvertebrates. This study’s focus is to determine the differences in diversity of coastal macroinvertebrate (CM) species on intertidal boulders. Eleven CM species were visually counted within a one-square-meter quadrat along two height strata of five intertidal boulders. Based upon the Shannon-Wiener diversity index, diversity was measured between the CM species in the lower region (0-1 meter from base of boulder) and the upper region (1-2 meters from base of boulder). The Coral X-radiograph Densitometry System (CoralXDS) is a Windows-based program which provides a tool for measurement of linear extension, density, and calcification from coral X-radiographs. These quantities are determined for high-density, low-density, and annual bands. CoralXDS operates in two "modes": full mode and extension/luminance mode. Full mode measures linear extension, density, and calcification, while extension/luminance mode measures only linear extension. Extension/luminance mode requires only a coral image with scaling information. CoralXDS allows user specification of transect location and orientation on the coral image, and provides several options for automated and manual band selection. The output measurements …

The CPCe (Coral Point Count with Excel extensions) program is a Windows-based software (PC use only) that provides a tool for the determination of coral cover using transect photographs. A specified number of spatially random points are distributed on a transect image and the features underlying the points are user-identified. Coverage statistics are then calculated and the results sent to Excel spreadsheets automatically.
Additionally, CPCe can be used for image calibration and area analysis of benthic features. Excel sheets are automatically generated to summarize the area calculations for each image.

Over the past two decades, mounting evidence has demonstrated that a mechanism known as store-operated Ca2+ entry (SOCE) plays a crucial role in sustaining skeletal muscle contractility by facilitating Ca2+ influx from the extracellular space during sarcoplasmic reticulum (SR) Ca2+ depletion. We recently demonstrated that, in exercised fast-twitch muscle from mice, the incidence of Ca2+ entry units (CEUs), newly described intracellular junctions between dead-end longitudinal transverse tubular (T-tubule) extensions and stacks of sarcoplasmic reticulum (SR) flat cisternae, strictly correlate with both the capability of fibers to maintain contractions during fatigue and enhanced Ca2+ influx via SOCE. Purpose: This study examined the association of microalbuminuria (MAU), as determined by albumin-to-creatinine ratio (ACR), with hypertension (HTN) among Turkish immigrants with type 2 diabetes (T2D) living in deprived neighborhoods of The Hague, Netherlands. Methods: A total of 110 participants, physician-diagnosed with T2D, aged ≥ 30 years were recruited from multiple sources from The Hague, Netherlands in a cross-sectional design. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured using automated office blood pressure equipment. Urine albumin was measured by immunoturbidimetric assay. This thesis is dedicated to the development of new methodologies for efficient synthesis of carbohydrate building blocks and their application to chemical glycosylation. S-Indolyl (SIn) anomeric moiety was investigated as a new leaving group. Understanding of the reaction pathways for the SIn moiety activation was achieved via the extended mechanistic study. The activation profile of indolylthio glycosides required large excess of activators. This drawback was partially addressed by the development of N-alkylated SInR derivatives. The activation process was studied by NMR and the increased understanding of the mechanism led to a discovery of different activation pathways taking place with …

Host records and nesting behavior of the Nearctic trapdoor spider-hunting spider wasps (Hymenoptera: Pompilidae) Calopompilus Ashmead and Priocnemissus Haupt (Pepsinae: Pepsini) and Aporus Spinola and Psorthaspis Banks (Pompilinae: Aporini) are reviewed, investigated, compared, and discussed. First host record, prey transport, and burrow excavation are described for Hesperopompilus sp., an undescribed, rare spider wasp (Hymenoptera: Pompilidae) from Texas. Taxonomic, ecological, and behavioral examination of the genus subsequently led to an investigation of the previously related Perissopompilus Evans and Xerochares Evans. Taxonomic, host preference, nesting behavior, and phylogenomic relationships of the three taxa are discussed along with those of Xenopompilus Evans. The molecular connection of Perissopompilus and Allochares Banks is supported by their common use of host species of Filistatidae.
