Methamphetamine use is increasing in the U.S. and in Missouri, as are the number of deaths associated with its use. Many systemic and mental health issues are associated with methamphetamine use or methamphetamine use disorder (MUD). Given the range of health issues associated with methamphetamine use or MUD, a collaborative approach to the care of patients can improve outcomes. This article provides an overview of a collaborative approach to caring for patients using or have used methamphetamine, from the perspective of the primary care, behavioral and dental clinician.

Aging is a medical problem affecting about 800 million people worldwide. However, the molecular mechanism of aging and aging-related disease has not yet been explored. Skin aging is a complex biological process primarily due to cellular senescence. Cellular senescence is an irreversible growth arrest that caused tissue to lose their cell-specific functionality, secreting pro-tumorigenic and pro-inflammatory cytokines, which eventually causes cancer. Citrus aurantium-derived secondary metabolites, including hesperidin, hesperetin, tangeretin, naringenin, nobiletin, and quercetin, are of vital importance to inhibiting aging due to their active properties. However, the effect of C. aurantium-derived secondary metabolites on the cells senescence and its molecular …

Open Access has evolved into the most complex challenge of the scholarly communication landscape and something libraries grapple with on a regular basis. But although librarians hold increasingly positive perceptions about OA, including its richness of unique content and immediacy of access, many lack the understanding, training, documentation, and knowledge of best practices that would allow them to engage with it confidently. This book helps to fill that gap, using a holistic approach that walks readers through the steps of integrating OA resources into library collections and supporting OA initiatives irrespective of budget, institution type, collection size, and staffing. Explaining …

Evidence of social learning is growing across the animal kingdom. Researchers have long hypothesized that social interactions play a key role in many animal migrations, but strong empirical support is scarce except in a few unique systems and species. In this review, we aim to catalyze advances in the study of social migrations by synthesizing research across disciplines and providing a framework for understanding when, how, and why social influences shape the decisions animals make during migration. Integrating research across the fields of social learning and migration ecology will advance our understanding of the complex behavioral phenomena of animal migration …

Several quantitative diagnostic techniques are available to estimate gastrointestinal parasite counts in the feces of ruminants. Comparing egg and oocyst magnitudes in naturally infected samples has been a recommended approach to rank fecal techniques. In this study, we compared the Mini-FLOTAC (sensitivity of 5 eggs per gram (EPG)/oocysts per gram (OPG)) and different averaged replicates of the modified McMaster techniques (sensitivity of 33.33 EPG/OPG) in 387 fecal samples from 10 herds of naturally infected North American bison in the Central Great Plains region of the USA. Both techniques were performed with fecal slurries homogenized in a fill-FLOTAC device. In the …

Urban rivers often contain elevated concentrations of contaminants such as organic pollutants and heavy metals which can be amplified in lotic ecosystems receiving effluents from wastewater treatment plants (WWTP). However, the impact of WWTPs on the microbial parameters of the urban river sediments has not been well documented compared to urban surface water. Collecting sediment samples at five different locations over a 9,000 m transect during four sampling periods, we investigated spatiotemporal variations of microbial parameters in sediments of lower Des Plaines River; the largest effluent-dominated stream in the United States and the effects of sediment physicochemical properties on the …
Towards The Detection And Analysis Of Performance Regression Introducing Code Changes, Deema Adeeb Alshoaibi
Towards The Detection And Analysis Of Performance Regression Introducing Code Changes, Deema Adeeb Alshoaibi
Theses
In contemporary software development, developers commonly conduct regression testing to ensure that code changes do not affect software quality. Performance regression testing is an emerging research area from the regression testing domain in software engineering. Performance regression testing aims to maintain the system's performance. Conducting performance regression testing is known to be expensive. It is also complex, considering the increase of committed code and developing team members working simultaneously. Many automated regression testing techniques have been proposed in prior research. However, challenges in the practice of locating and resolving performance regression still exist. Directing regression testing to the commit level …

The increasing demand for complex devices that utilize unique, three-dimensional nanostructures has spurred the development of controllable and versatile semiconductor fabrication techniques. However, there exists a need to refine such methodologies to overcome existing processing constraints that compromise device performance and evolution. Conventional wet etching techniques (e.g., crystallographic KOH etching of Si) successfully generate textured Si structures with smooth sidewalls but lack the capabilities of controllably producing high aspectratio structures. Alternatively, dry etching techniques (e.g., reactive-ion etching), while highly controllable and capable of generating vertically aligned, high aspect-ratio structures for IC technologies, introduce considerable sidewall and lattice damage as a …

Previous genome-wide association studies (GWASs) of stroke - the second leading cause of death worldwide - were conducted predominantly in populations of European ancestry1,2. Here, in cross-ancestry GWAS meta-analyses of 110,182 patients who have had a stroke (five ancestries, 33% non-European) and 1,503,898 control individuals, we identify association signals for stroke and its subtypes at 89 (61 new) independent loci: 60 in primary inverse-variance-weighted analyses and 29 in secondary meta-regression and multitrait analyses. On the basis of internal cross-ancestry validation and an independent follow-up in 89,084 additional cases of stroke (30% non-European) and 1,013,843 control individuals, 87% of the primary …

Female carriers of pathogenic/likely pathogenic (P/LP) BRCA1/2 variants are at increased risk of developing breast and ovarian cancer. Currently, the only effective strategy for ovarian cancer risk reduction is risk-reducing bilateral salpingo-oophorectomy (RR-BSO), which carries adverse effects related to early menopause. There is ongoing investigation of inhibition of the RANK ligand (RANKL) with denosumab as a means of chemoprevention for breast cancer in carriers of BRCA1 P/LP variants. Through the NCI Division of Cancer Prevention (DCP) Early Phase Clinical Trials Prevention Consortia, a presurgical pilot study of denosumab was developed in premenopausal carriers of P/LP BRCA1/2 variants scheduled for RR-BSO …

Alterations in the intestinal physiology caused by pathogen colonization result in immune activation. To provide insights into the mechanisms underlying the control of immune activation by changes in intestinal homeostasis, we conducted a forward genetic screen for suppressors of immune activation by intestinal distension in Caenorhabditis elegans. Our results indicate that C. elegans ACC-4, a member of a family of acetylcholine receptors, is required in immune activation by defects in the defecation motor program or by pathogen infection. ACC-4 acts postsynaptically in non-cholinergic RIM neurons to regulate several immune genes and a Wnt-mediated host immune response. These findings uncover a …

Brain metastases are the most common brain malignancy. This review discusses the studies presented at the third annual meeting of the Melanoma Research Foundation in the context of other recent reports on the biology and treatment of melanoma brain metastases (MBM). Although symptomatic MBM patients were historically excluded from immunotherapy trials, efforts from clinicians and patient advocates have resulted in more inclusive and even dedicated clinical trials for MBM patients. The results of checkpoint inhibitor trials were discussed in conversation with current standards of care for MBM patients, including steroids, radiotherapy, and targeted therapy. Advances in the basic scientific understanding …

OBJECTIVE: Risk factors predisposing infants to community-acquired bacterial infections during the first 2 months of life are poorly understood in South Asia. Identifying risk factors for infection could lead to improved preventive measures and antibiotic stewardship.
METHODS: Five sites in Bangladesh, India and Pakistan enrolled mother-child pairs via population-based pregnancy surveillance by community health workers. Medical, sociodemographic and epidemiological risk factor data were collected. Young infants aged 0-59 days with signs of possible serious bacterial infection (pSBI) and age-matched controls provided blood and respiratory specimens that were analysed by blood culture and real-time PCR. These tests were used to build …

The role of DNA methylation and its interplay with gene expression in the susceptibility to pancreatic cancer (PanC) remains largely unexplored. To fill in this gap, we conducted an integrative two-phase epigenome-wide association study (EWAS) of PanC using genomic DNA from 44 cases and 556 controls (20 local controls and 536 public controls in the Framingham Heart Study) in phase 1 and 23 cases and 22 controls in phase 2. We validated the findings using pre-diagnostic blood samples from 13 cases and 26 controls in the Women's Health Initiative (WHI) Study. We further examined gene expression in peripheral leukocytes of …

BACKGROUND: Nephroblastoma, also known as Wilms' tumor (WT), remains one of the major causes of tumor-related deaths worldwide in children. Cancer stem cells (CSCs) are considered to be the main culprits in cancer resistance and disease recurrence, which are reported in multiple types of tumors. However, the research on CSCs in WT is limited. Therefore, our study aimed to identify the key genes related to CSCs in WT to provide new ideas for treating WT.
METHODS: The RNA-seq and clinical data of WT samples were obtained from the University of California Santa Cruz (UCSC) Xena database, which included 120 WT …

Bile acids, originally known to emulsify dietary lipids, are now established signalling molecules that regulate physiological processes. Signalling targets several proteins that include the ion channels involved in regulating intestinal motility and bile viscosity. Studies show that bile acids regulate the epithelial sodium channel (ENaC) in cultured cell models and heterologous expression systems. ENaC plays both local and systemic roles in regulating extracellular fluids. Here we investigated whether bile acids regulate ENaC expressed in native tissues. We found that taurocholic acid and taurohyodeoxycholic acid regulated ENaC in both the distal nephron and distal colon. We also tested the hypothesis that …

Neoantigens are the key targets of antitumor immune responses from cytotoxic T cells and play a critical role in affecting tumor progressions and immunotherapy treatment responses. However, little is known about how the interaction between neoantigens and T cells ultimately affects the evolution of cancerous masses. Here, we develop a hierarchical Bayesian model, named neoantigen-T cell interaction estimation (netie) to infer the history of neoantigen-CD8

Background
Immune exhaustion and senescence are dominant dysfunctional states of effector T cells and major hurdles for the success of cancer immunotherapy. In the current study, we characterized how acute myeloid leukemia (AML) promotes the generation of senescent-like CD8+ T cells and whether they have prognostic relevance.
METHODS
We analyzed NanoString, bulk RNA-Seq and single-cell RNA-Seq data from independent clinical cohorts comprising 1,896 patients treated with chemotherapy and/or immune checkpoint blockade (ICB).
Results
We show that senescent-like bone marrow CD8+ T cells were impaired in killing autologous AML blasts and that their proportion negatively correlated with overall survival …

Rubisco activase (Rca) facilitates the catalytic repair of Rubisco, the CO2-fixing enzyme of photosynthesis, following periods of darkness, low to high light transitions or stress. Removal of the redox-regulated isoform of Rubisco activase, Rca-α, enhances photosynthetic induction in Arabidopsis and has been suggested as a strategy for the improvement of crops, which may experience frequent light transitions in the field; however, this has never been tested in a crop species. Therefore, we used RNAi to reduce the Rca-α content of soybean (Glycine max cv. Williams 82) below detectable levels and then characterized the growth, photosynthesis, and Rubisco activity of the …

The rapid loss of topsoil, biodiversity, and water quality across agricultural land in the United States and the industrialized world poses some of the most important risks to the future of global and domestic food security. Not only is arable land being lost at an unsustainable pace, but the average age of farmers has also steadily been rising due to a myriad of barriers young, BIPOC (Black, Indigenous, and People of Color), and beginning farmers face. In an attempt to address these issues, worm castings, bokashi tea, fish hydrolysate, and biochar were applied alone and in combination to an extremely …

The rapid increase in population, urbanization, and industrialization, along with inadequate water quality monitoring and wastewater management, contribute heavily to the pollution of water resources. Among the water contaminants of major concern, heavy metals are particularly considered as the most worrisome. Heavy metals tend to accumulate in the tissues of aquatic organisms (Ahmed et al., 2019; Rajeshkumar & Li, 2018; Tabrez et al., 2021) and thus increase in concentration upon moving higher up the food chain. This indicates that once humans ingest these organisms, they have a high risk of experiencing health problems as heavy metals are absorbed into their …

Dignity: A Journal of Analysis of Exploitation and Violence
This study aimed to establish how socio-cultural and economic factors support the endemicity of child labour trafficking in Nigeria. The research was conducted among rural and urban households and stakeholders in southern Nigeria. A field survey was conducted in Ekiti, Edo, Kwara, Lagos, and Osun States. The study utilised cultural relativism and the margin of appreciation theories. The qualitative research approach used in-depth interviews, focus groups, and personal observation methods to collect data. Researchers interviewed 70 participants, including parents/guardians, stakeholders (government officials and private agency representatives), traffickers, trafficked children, and their employers. Societal context, especially the perception of child rights, …

Dignity: A Journal of Analysis of Exploitation and Violence
This article reviews the goals, history, and impact of the new gender identity politics. Based on the Yogyakarta Principles, these new ideas and policies will profoundly affect the rights of women and girls worldwide. The Principles are a document from an international meeting about sexual orientation and gender identity in 2006. In 2017, the document was updated to the Yogyakarta Principles Plus 10. The Principles recommend legal changes by states worldwide, resulting in the erasure of sex as a legal and cultural category. These principles have been widely used to lobby for legal changes resulting in profound structural …

Chronic inflammation is associated with lung tumorigenesis, in which NF-κB–mediated epigenetic regulation plays a critical role. Lung tumor suppressor G protein–coupled receptor, family C, member 5A (GPRC5A), is repressed in most non–small cell lung cancer (NSCLC); however, the mechanisms remain unclear. Here, we show that NF-κB acts as a transcriptional repressor in suppression of GPRC5A. NF-κB induced GPRC5A repression both in vitro and in vivo. Intriguingly, transactivation of NF-κB downstream targets was not required, but the transactivation domain of RelA/p65 was required for GPRC5A repression. NF-κB did not bind to any potential cis-element in the GPRC5A promoter. Instead, p65 was …
