The Winchcombe Meteorite, A Unique And Pristine Witness From The Outer Solar System, Aj King, L Daly, J Rowe, Et Al

School of Geography, Earth and Environmental Sciences

Direct links between carbonaceous chondrites and their parent bodies in the solar system are rare. The Winchcombe meteorite is the most accurately recorded carbonaceous chondrite fall. Its pre-atmospheric orbit and cosmic-ray exposure age confirm that it arrived on Earth shortly after ejection from a primitive asteroid. Recovered only hours after falling, the composition of the Winchcombe meteorite is largely unmodified by the terrestrial environment. It contains abundant hydrated silicates formed during fluid-rock reactions, and carbon- and nitrogen-bearing organic matter including soluble protein amino acids. The near-pristine hydrogen isotopic composition of the Winchcombe meteorite is comparable to the terrestrial hydrosphere, providing …

Chronic exposure to airborne carbon black ultrafine (nCB) particles generated from incomplete combustion of organic matter drives IL-17A-dependent emphysema. However, whether and how they alter the immune responses to lung cancer remains unknown. Here, we show that exposure to nCB particles increased PD-L1+ PD-L2+ CD206+ antigen-presenting cells (APCs), exhausted T cells, and Treg cells. Lung macrophages that harbored nCB particles showed selective mitochondrial structure damage and decreased oxidative respiration. Lung macrophages sustained the HIF1α axis that increased glycolysis and lactate production, culminating in an immunosuppressive microenvironment in multiple mouse models of non-small cell lung cancers. Adoptive transfer of lung APCs …

We report the quaternary structure of core transcriptional complex for the full-length human progesterone receptor-B (PR-B) homodimer with primary coactivator steroid receptor coactivator-2 (SRC-2) and the secondary coactivator p300/CREB-binding protein (CBP). The PR-B homodimer engages one SRC-2 mainly through its activation function 1 (AF1) in N-terminus. SRC-2 is positioned between PR-B and p300 leaving space for direct interaction between PR-B and p300 through PR-B's C-terminal AF2 and its unique AF3. Direct AF3/p300 interaction provides long-desired structural insights into the known functional differences between PR-B and the PR-A isoform lacking AF3. We reveal the contributions of each AF and demonstrate their …

Currently, there are no treatments that ameliorate cardiac cell death, the underlying basis of cardiovascular disease. An unexplored cell type in cardiac regeneration is cardiac Purkinje cells; specialized cells from the cardiac conduction system (CCS) responsible for propagating electrical signals. Purkinje cells have tremendous potential as a regenerative treatment because they may intrinsically integrate with the CCS of a recipient myocardium, resulting in more efficient electrical conduction in diseased hearts. This study is the first to demonstrate an effective protocol for the direct reprogramming of human cardiomyocytes into cardiac Purkinje-like cells using small molecules. The cells generated were genetically and …

The Perceived Impact Of Project-Based Learning (Pbl) On Middle School Students’ Science, Technology, Engineering, And Mathematics (Stem) Identity And Engagement, Jonathan Olivera Nov 2022

The Perceived Impact Of Project-Based Learning (Pbl) On Middle School Students’ Science, Technology, Engineering, And Mathematics (Stem) Identity And Engagement, Jonathan Olivera

Theses and Dissertations

Despite multiple calls to action, the United States educational system is not producing enough viable contributors in the areas of science, technology, engineering, and mathematics (STEM). These fields continue to grow, and the STEM workforce continues to expand. However, the pool of citizens prepared to enter these professions is not keeping up with the demand. Part of this issue can be attributed to a diminishing interest in STEM by students, particularly during their identity forming adolescent years. Active learning strategies have proven successful in preventing this decline with project-based learning (PBL) being one of the most successful active learning strategies. …

Applying Retrieval Practice In Climate Change Education: How Retrieval-Based Learning May Enhance High School Students’ Conceptual Understanding On Climate Change Topics, Mengyu Wang Nov 2022

Applying Retrieval Practice In Climate Change Education: How Retrieval-Based Learning May Enhance High School Students’ Conceptual Understanding On Climate Change Topics, Mengyu Wang

Dissertations

This dissertation explored ways to apply retrieval practice strategy on climate change learning at the high school level. Three studies presented that retrieval practice could be used broadly on different topics of climate change and in different learning scenarios. The first study focused on the effect of question placement on learning when students studied sea level rise from an educational video lecture. The results suggested that applying retrieval practice in a style of embedded short-answer questions in videos has clear advantage over restudying the video lecture in both immediate and delayed tests, but no difference in students' performance was found …

Molecular Basis For Integrin Adhesion Receptor Binding To P21-Activated Kinase 4 (Pak4), Byung Hak Ha, Sezin Yigit, Nailini Natarajan, Elizabeth M. Luoma, David A. Calderwood, Titus J. Boggon

SHU Faculty Publications

Integrin adhesion receptors provide links between extracellular ligands and cytoplasmic signaling. Multiple kinases have been found to directly engage with integrin β tails, but the molecular basis for these interactions remain unknown. Here, we assess the interaction between the kinase domain of p21-activated kinase 4 (PAK4) and the cytoplasmic tail of integrin β5. We determine three crystal structures of PAK4-β5 integrin complexes and identify the PAK-binding site. This is a region in the membrane-proximal half of the β5 tail and confirmed by site-directed mutagenesis. The β5 tail engages the kinase substrate-binding groove and positions the non-phosphorylatable integrin residue Glu767 at …

Histone Deacetylase 4 And 5 Translocation Elicited By Microsecond Pulsed Electric Field Exposure Is Mediated By Kinase Activity, Zahra Safaei, Gary L. Thompson Iii

Henry M. Rowan College of Engineering Departmental Research

Electroporation-based technologies using microsecond pulsed electric field (µsPEF) exposures are established as laboratory and clinical tools that permeabilize cell membranes. We demonstrate a µsPEF bioeffect on nucleocytoplasmic import and export of enzymes that regulate genetic expression, histone deacetylases (HDAC) -4 and -5. Their μsPEF-induced nucleocytoplasmic transport depends on presence and absence of extracellular calcium ions (Ca2+) for both MCF7 and CHO-K1 cells. Exposure to 1, 10, 30 and 50 consecutive square wave pulses at 1 Hz and of 100 µs duration with 1.45 kV/cm magnitude leads to translocation of endogenous HDAC4 and HDAC5. We posit that by eliciting a rise …

Where Are The Black Teachers? There Should Be More To Make It Even, Harolyn Harris Nov 2022

Where Are The Black Teachers? There Should Be More To Make It Even, Harolyn Harris

Dissertations

For years, Black teachers lived and worked in the communities that they served. I was one of those teachers who lived and worked in my community, but I noticed that there were not many of us. As a Black woman, mother and teacher, I was fully aware of the numbers of Black teachers declining. I wanted to know why.

In this study, I utilized my experience as a Black student, a Black teacher, and a Black administrator to reflect my feelings. I used Critical Race Theory as the theoretical lens on my stories using autoethnography as the research method. I …

Regulation Of Parkinson’S Disease-Associated Genes By Pumilio Proteins And Micrornas In The Human Neuroblastoma Cell Line Sh-Sy5y, Lisa J. Snoderly-Foster Nov 2022

Regulation Of Parkinson’S Disease-Associated Genes By Pumilio Proteins And Micrornas In The Human Neuroblastoma Cell Line Sh-Sy5y, Lisa J. Snoderly-Foster

Dissertations

Parkinson’s disease (PD) is the second most common age-related, neurodegenerative disease. A small collection of genes has been linked to PD, including SNCA encoding the protein alpha-synuclein that aggregates in Lewy bodies, a hallmark of PD. Overexpression of these genes can lead to pathogenesis, yet the regulatory mechanisms that control protein production of the genes are not fully understood. The SH-SY5Y neuronal cell line is a common model for the study of neurodegenerative diseases, including PD. However, SH-SY5Y cells are difficult to transfect, a process necessary for genetic manipulations and downstream analysis of RNA and protein. Herein, I describe the …

Life History And Ecology Of The San Emigdio Blue Butterfly (Lepidoptera: Lycaenidae), Gregory R. Ballmer Nov 2022

Life History And Ecology Of The San Emigdio Blue Butterfly (Lepidoptera: Lycaenidae), Gregory R. Ballmer

The Taxonomic Report of the International Lepidoptera Survey

The San Emigdio blue butterfly, Plebulina emigdionis (Grinnell, 1905), occurs in small, scattered colonies in and near the southwestern Mojave Desert of California. Colonies depend on a symbiotic relationship with the ant Formica francoueri (Bolton), and occur only where the ant’s range (primarily in more mesic cis-montane habitats) narrowly overlaps that of the butterfly’s more widely distributed Atriplex larval hosts in more xeric habitats. Colonies of P. emigdionis are often localized around a few host plants and, therefore, sensitive to habitat changes due to anthropocentric causes and environmental stochasticity. The biology, ecology, and status of known colonies of P emigdionis …

Bison Movements Change With Weather: Implications For Their Continued Conservation In The Anthropocene, Nicholas A. Mcmillan, Samuel D. Fuhlendorf, Barney Luttbeg, Laura E. Goodman, Craig A. Davis, Brady W. Allred, Robert G. Hamilton Nov 2022

Bison Movements Change With Weather: Implications For Their Continued Conservation In The Anthropocene, Nicholas A. Mcmillan, Samuel D. Fuhlendorf, Barney Luttbeg, Laura E. Goodman, Craig A. Davis, Brady W. Allred, Robert G. Hamilton

Department of Agronomy and Horticulture: Faculty Publications

Animal movement patterns are affected by complex interactions between biotic and abiotic landscape conditions, and these patterns are being altered by weather variability associated with a changing climate. Some animals, like the American plains bison (Bison bison L.; hereafter, plains bison), are considered keystone species, thus their response to weather variability may alter ecosystem structure and biodiversity patterns. Many movement studies of plains bison and other ungulates have focused on point-pattern analyses (e.g., resource-selection) that have provided information about where these animals move, but information about when or why these animals move is limited. For example, information surrounding the …

Protein Phosphatase 2a Regulates Xanthine Oxidase-Derived Ros Production In Macrophages And Influx Of Inflammatory Monocytes In A Murine Gout Model, Sandy Elsayed, Khaled A. Elsaid Nov 2022

Protein Phosphatase 2a Regulates Xanthine Oxidase-Derived Ros Production In Macrophages And Influx Of Inflammatory Monocytes In A Murine Gout Model, Sandy Elsayed, Khaled A. Elsaid

Pharmacy Faculty Articles and Research

Background: Gout is a common arthritis, due to deposition of monosodium urate (MSU) crystals which results in IL-1β secretion by tissue-resident macrophages. Xanthine oxidase (XO) catalyzes uric acid (UA) production and in the process, reactive oxygen species (ROS) are generated which contributes to NLRP3 inflammasome activation. Protein phosphatase 2A (PP2A) may be involved in regulating inflammatory pathways in macrophages. The objective of this study was to investigate whether PP2A regulates gout inflammation, mediated by XO activity modulation. We studied UA and ROS generations in MSU stimulated murine bone marrow derived macrophages (BMDMs) in response to fingolimod phosphate, a PP2A activator, …

Epigenetic And Genetic Associations And Gene Expression For The Risk Of Eczema In Two Consecutive Generations, Rumana Siddique Nov 2022

Epigenetic And Genetic Associations And Gene Expression For The Risk Of Eczema In Two Consecutive Generations, Rumana Siddique

Electronic Theses and Dissertations Archive

Studies demonstrated genetic and epigenetic associations with eczema. However, not many studies investigated the interaction of epigenetic and genetic associations with eczema. Additionally, few studies examined gene expression (GE) levels along with epigenetic and genetic associations with eczema. Using the multigenerational epigenetic, genetic, and GE data of Isle of Wight (IOW) birth cohort, I investigated the interplay between DNA methylation (DNAm) and a genetic variant (SNP) with eczema and GE. First, I evaluated epigenome-wide associations of eczema using the DNAm data of two generations of the IOW (namely, the F1 and F2 generations) and examined the overlap of genes of …

To Brag Or Not To Brag: Two Essays On The Effective Promotion Of Prosocial Behaviors, Eric Narcum

Electronic Theses and Dissertations Archive

Whether it is an individual or a corporation, it is natural to desire credit for good deeds. However, promoting one’s own prosocial behaviors has the potential to backfire by raising suspicions about one’s motives. In two essays, this dissertation examines how self-promotion impacts others’ perceptions of prosocial actors. Essay 1 focuses on how corporate bragging in CRM advertisements affects consumers’ attitudes towards and preference for a prosocial company. Results from four studies demonstrate a number of important findings. First, participants have less of a preference for companies that brag in their CRM advertisements. Secondly, perceived motives mediate the relationship between …

Deep Learning -Powered Computational Intelligence For Cyber-Attacks Detection And Mitigation In 5g-Enabled Electric Vehicle Charging Station, Manoj Basnet Nov 2022

Deep Learning -Powered Computational Intelligence For Cyber-Attacks Detection And Mitigation In 5g-Enabled Electric Vehicle Charging Station, Manoj Basnet

Electronic Theses and Dissertations Archive

An electric vehicle charging station (EVCS) infrastructure is the backbone of transportation electrification. However, the EVCS has various cyber-attack vulnerabilities in software, hardware, supply chain, and incumbent legacy technologies such as network, communication, and control. Therefore, proactively monitoring, detecting, and defending against these attacks is very important. The state-of-the-art approaches are not agile and intelligent enough to detect, mitigate, and defend against various cyber-physical attacks in the EVCS system. To overcome these limitations, this dissertation primarily designs, develops, implements, and tests the data-driven deep learning-powered computational intelligence to detect and mitigate cyber-physical attacks at the network and physical layers of …
