กรดไขมันไม่อิ่มตัวที่มีพันธะคู่หลายตำแหน่งในกลุ่มของโอเมก้า-3 เช่น ดีเอชเอ และอีพีเอ เป็นกรดอะมิโนจำเป็นต่อร่างกาย (essential amino acid) เนื่องจากในร่างกายมนุษย์ไม่มีเอนไซม์ที่สามารถสร้างพันธะคู่ตำแหน่ง ω-3 ได้ มีรายงานว่าเส้นทางการสังเคราะห์กรดไขมันผ่านวิถีทางชีวภาพ (FAS pathway) พบในแบคทีเรียจากทะเลบางสายพันธุ์เท่านั้น ในงานวิจัยนี้ทำการคักแยกแบคทีเรียทางทะเลจากตัวอย่างกะปิซึ่งมีการตรวจสอบจากงานวิจัยก่อนหน้าว่ามีกลุ่มของกรดไขมันโอเมก้า-3, 6 และ 9 ผลการศึกษาสามารถคัดแยกแบคทีเรียสะสมไขมันสูง (oleaginous bacteria) ได้ 6 ไอโซเลต โดยเฉพาะ Bacillus amyloliquefaciens SR121 เมื่อทำการวิเคราะห์ whole genome พบยีน FabF (β-ketoacyl-ACP reductase), FabG (Ketoreductase), FabZ (Dehydratase) และ FabI (Enoyl reduction) ซึ่งเกี่ยวข้องกับ 4 กระบวนการสำคัญ ใน FAS pathway นอกจากนี้ B. amyloliquefaciens SR121 มีการผลิตกรดไขมันมากที่สุดร้อยละ 75.06 ± 2.05 ของน้ำหนักเซลล์แห้ง และเมื่อใช้อาหารสำหรับการเพาะเลี้ยงที่มี pH5 พบปริมาณการผลิตไขมันเพิ่มขึ้นถึงร้อยละ 89.46 ± 3.79 ของน้ำหนักเซลล์ สำหรับการปรับภาวะเพื่อผลิตกรดไขมันกลุ่มโอเมก้า-3 พบที่ 35 องศาเซลเซียส มีการผลิตมากสุดถึง 19.73% ของปริมาณกรดไขมันทั้งหมด หรือคิดเป็นกรดไขมันไม่อิ่มตัว 92.97 มิลลิกรัม และมีการผลิตกรดไขมันกลุ่มโอเมก้า-3 มากที่สุดโดยปริมาณ 72 มิลลิกรัม ต่อน้ำหนักเซลล์ 1 กรัม

Silver nanoparticles (AgNPs) and microplastics are emerging water contaminants of the decade. They share a similar fate and transport in wastewater treatment plants as they tend to accumulate in sludge of aeration tanks. Since both contaminants have negative effects on microbial growth, the ammonia-oxidizing microorganisms in the aeration tanks are at risk for inhibition and consequently nitrification process fails. This study investigated the effects of AgNPs and microplastics on ammonia-oxidizing activity and community. No inhibition of ammonia oxidation rate was observed at 0.1 mg/L AgNPs. การนับจำนวนไวรัสด้วยจำนวน Plaque Forming Unit (PFU) เพื่อทำการทดลองในด้านต่างๆ เช่นการผลิตวัคซีน สามารถทำได้ด้วยการใช้ตาเปล่าซึ่งส่งผลให้เกิดความเหนื่อยล้า ใช้เวลานานและเกิดข้อผิดพลาดได้ง่าย การนับด้วยวิธีที่รวดเร็วและแม่นยำต้องคำนึงหลายปัจจัย เนื่องจาก PFU มีรูปร่างไม่แน่นอน ขอบไม่เด่นชัด ขนาดไม่สม่ำเสมอ รวมถึงความแตกต่างกันของสีระหว่าง PFU และพื้นหลังภายในจานหลุมไม่คงที่ วิทยานิพนธ์ฉบับนี้เสนอสร้างกรรมวิธีการนับ PFU แบบกึ่งอัตโนมัติจากภาพจานเพาะเลี้ยงที่ได้จากกล้องถ่ายรูปทั่วไปที่ไม่มีการควบคุมแสง เริ่มจากการจำแนก PFU กับพื้นหลังด้วยค่าขีดเริ่มเปลี่ยนแบบปรับตัวได้ โดยถ้าเป็น PFU ขนาดเล็กจะใช้ค่าขีดเริ่มเปลี่ยนบริเวณขอบและกลางหลุมแตกต่างกัน ขณะที่ถ้าเป็น PFU ขนาดใหญ่จะใช้ค่าขีดเริ่มเปลี่ยนเดียวกันทั้งหลุม ปัญหาเรื่องสีย้อมที่ต่างกันถูกแก้โดยใช้ผลต่างของสองช่องสัญญาณในปริภูมิสี CIE-XYZ ที่มีการถ่วงน้ำหนักให้ความสว่างของพื้นหลังเท่ากัน การแบ่งพื้นที่ของ PFU ที่ติดกันประกอบด้วยสองส่วนคือ กระบวนการวิธีสันปันน้ำร่วมกับการแปลงระยะทางและการใช้เกณฑ์ขนาดร่วมกับการขยายของจุดศูนย์กลางที่ได้จากภาพการแปลงระยะทางเป็นวงกลม จากการทดลองพบว่าเมื่อปรับค่าถ่วงน้ำหนักให้เหมาะสมแล้ว กรรมวิธีที่นำเสนอสามารถนับ PFU ผิดพลาดน้อยกว่าร้อยละ 10 สำหรับ PFU ที่เป็นสีขาว มีขอบราบเรียบและไม่มีรูตรงกลาง จึงสามารถนำมาใช้กับ PFU ของไวรัสไข้เลือดออก ไวรัสไข้ปวดข้อยุงลายและไวรัสไข้สมองอักเสบ การนับจะได้ผลผิดพลาดมากขึ้นสำหรับ PFU ที่มีสีชมพูปะปน เช่น ไวรัสไข้ซิก้าและ PFU ที่มีขอบฟุ้ง เช่น PFU ของไวรัสโคโรนา

มะเร็งท่อน้ำดีเป็นมะเร็งที่เกิดขึ้นบริเวณท่อทางเดินน้ำดี โดยมีความหลากหลายของเซลล์มะเร็งบริเวณเนื้อเยื่อมะเร็งและมีความรุนแรง มะเร็งท่อน้ำดีจัดเป็นสาเหตุหลักในการเสียชีวิตของผู้ป่วยในประเทศไทย เนื่องจากผู้ป่วยได้รับการวินิจฉัยเมื่อมะเร็งเจริญพัฒนาไปยังระยะท้ายและยังขาดวิธีการรักษาที่มีประสิทธิภาพ การรักษามะเร็งท่อน้ำดีด้วยวิธีมาตรฐานในปัจจุบันในผู้ป่วยที่ไม่สามารถผ่าตัดได้ หรือในผู้ป่วยหลังการผ่าตัด คือ การรักษาด้วยยาเคมีบำบัด ชนิดเจมไซทาบีนหรือยาเจมไซทาบีนร่วมกับยาซิสพลาติน แต่อย่างไรก็ตามผู้ป่วยส่วนใหญ่พบปัญหาการดื้อต่อยาเคมีบำบัดนำไปสู่การพยากรณ์โรคที่ไม่ดี ดังนั้นการพัฒนาการรักษาที่มีประสิทธิภาพมากยิ่งขึ้นถือว่ามีความจำเป็นเร่งด่วน โดยได้มีการศึกษาการตายของเซลล์มะเร็งแบบใหม่ที่ทำให้เซลล์มะเร็งตาย และกระตุ้นการทำงานของระบบภูมิคุ้มกันแบบจำเพาะ เรียกว่า การตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการตายของเซลล์แบบเนคโครปโทซิส จัดว่าเป็นการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยได้มีการศึกษาอย่างแพร่หลายในมะเร็งหลายชนิด โดยพบว่าการกระตุ้นให้เซลล์มะเร็งตายแบบเนคโครปโทซิสสามารถกระตุ้นระบบภูมิคุ้มแบบจำเพาะและยับยั้งการเจริญพัฒนาของก้อนมะเร็งในหนูทดลอง การศึกษาภายในห้องปฏิบัติการก่อนหน้าของคณะผู้วิจัยพบว่าการเหนี่ยวนำร่วมกันระหว่างยาเคมีบำบัดเจมไซทาบีนและสารสะแมคไมเมติกสามารถเหนี่ยวนำให้เซลล์มะเร็งท่อน้ำดีตายแบบเนคโครปโทซิสในสภาวะที่มีการยับยั้งการทำงานของเอนไซม์แคสเปส ทั้งนี้จากข้อจำกัดที่ยาเคมีบำบัดมีความเป็นพิษที่สูง การศึกษานี้จึงมีจุดมุ่งหมายเพื่อค้นหาสารที่สามารถนำมาทำการเหนี่ยวนำร่วมกับสารสะแมคไมเมติก โดยมีสมมติฐานของงานวิจัย คือ สารพฤกษเคมีที่มีฤทธิ์ทำให้เกิดการทำลายของดีเอ็นเอและหยุดวัฏจักรของเซลล์จะสามารถเสริมฤทธิ์กับสารสะแมคไมเมติก โดยกระตุ้นการตายของเซลล์แบบในคโครปโทซิสในเซลล์มะเร็งท่อน้ำดี โดยงานวิจัยนี้ได้ทำการศึกษาฤทธิ์ยับยั้งการแบ่งตัวและการตายเซลล์มะเร็งท่อน้ำดีของสารพฤกษเคมี ได้แก่ ชิโคนิน พลัมบาจิน เควอซิติน โดยพบว่าสารเควอซิตินมีค่าความเข้มข้นของสารที่ยับยั้งการรอดชีวิตของเซลล์ได้ 50% (IC50) ที่ต่ำที่สุดเมื่อเปรียบเทียบกับสารพฤกษเคมีชนิดอื่นๆ ในการวิเคราะห์ด้วยวิธีการทางชีวสารสนเทศน์พบว่าสารเควอซิตินมีเป้าหมายไปยังยีนที่มีบทบาทในการควบคุมวัฏจักรเซลล์ระยะ G2/M และยีนเป้าหมายของสารเควอซิตินมีการแสดงออกที่เพิ่มมากขึ้นอย่างมีนัยสำคัญในเนื้อเยื่อมะเร็งท่อน้ำดีเมื่อเปรียบเทียบกับเซลล์ปกติ การทดลองในเซลล์มะเร็งท่อน้ำดี (in vitro) พบว่าสารเควอซิตินสามารถหยุดวัฏจักรเซลล์ที่ระยะ G2/M นอกจากนี้สารเควอซิตินช่วงความเข้มข้นค่าต่ำที่มีความเป็นพิษน้อยในรูปแบบสารเดี่ยว เมื่อนำไปเหนี่ยวนำร่วมกับสารสะแมคไมเมติกชนิด LCL-161 แล้วพบว่าสามารถเสริมฤทธิ์กันกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดี โดยเมื่อทำการวิเคราะห์ชนิดการตายของเซลล์เพิ่มเติมพบว่ามีการแสดงออกที่เพิ่มขึ้นของโปรตีนเอ็มแอลเคแอลที่ถูกเติมหมู่ฟอสเฟต (phosphorylated MLKL; pMLKL) ซึ่งเป็นตัวบ่งชี้ที่จำเพาะต่อการตายแบบเนคโครปโทซิส นอกจากนั้นยังพบว่าการตายของเซลล์ถูกยับยั้งเมื่อมีการเติมตัวยับยั้งที่จำเพาะกับโปรตีนที่สำคัญต่อการเกิดเนคโครปโทซิส แสดงให้เห็นว่าการเหนี่ยวนำเซลล์ด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นให้เซลล์มะเร็งท่อน้ำดีตายผ่านเนคโครปโทซิส การศึกษาเพิ่มเติมเพื่อศึกษาการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการวิเคราะห์การเคลื่อนย้ายของโปรตีนเอชเอ็มจีบีวัน (HMGB-1) จากภายในนิวเคลียสมายังบริเวณไซโตพลาสซึมด้วยวิธีการย้อมแบบอิมมูโนฟลูออเรสเซนส์ พบว่าการเหนี่ยวนำเซลล์มะเร็งท่อน้ำดีด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นการตายของเซลล์ที่กระตุ้นภูมิคุ้ม กล่าวโดยสรุป การเหนี่ยวนำร่วมกันระหว่างสารเควอซิตินและสารสะแมคไมเมติกในสภาวะที่มีการยับยั้งเอนไซม์แคสเปสสามารถเสริมฤทธิ์กันโดยกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดีแบบเนคโครปโทซิสและการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน ดังนั้นงานวิจัยนี้อาจนำไปสู่การพัฒนากลยุทธ์การรักษามะเร็งท่อน้ำดีแบบใหม่ที่เพิ่มประสิทธิภาพในการรรักษาในอนาคต อย่างไรก็ตาม งานวิจัยนี้ยังต้องมีการศึกษาเพิ่มเติม เช่น การศึกษากลไกการเสริมฤทธิ์และการกระตุ้นการตายของเซลล์ การศึกษาฤทธิ์ในเซลล์มะเร็งท่อน้ำดีชนิดอื่นๆ และการเมินความเป็นพิษในเซลล์ปกติ เป็นต้น

Eighteen compounds were obtained from hexane and dichloromethane extracts from the rhizomes of Stemona curtisii Hook f. Their chemical structures were determined by extensive spectroscopic studies including 1D and 2D NMR, MS, UV, IR, and comparison with data from the literature. Dengue infection is one significant global health issue, as climate change exacerbated the spread of mosquito vectors. The Chikungunya virus is also a re-emerging arbovirus infection, which is transmitted by some similar vectors. Currently, no commercial antiviral drug is available to treat them. Previous reports suggested that several flavonoid derivatives inhibited mosquito-borne flaviviruses even at submicromolar levels, and one of them is a halogenated flavone derivative, 8-bromobaicalein, was a potential dengue and Zika replication inhibitor targeting flaviviral polymerase and also 3CLpro of SARS-CoV-2. However, forensic entomological studies on corpse relocation in Thailand is limited, therefore, this research aimed to study insect succession on pig carcasses relocated from indoor to outdoor conditions and pig carcasses refrigerated for 3 days before relocated to the outdoor condition. Glioblastoma (GBM) is the most common and highly aggressive primary cancer within the central nervous system (CNS). The mTORC2 is a primary regulator of glycolytic reprogramming in highly malignant cells through both Akt-dependent and AKT-independent pathways. However, the mechanism that cancer glycolysis through AKT-independent pathways remains largely unexplored. This study aimed to investigate the mechanism of mTORC2/ PKCδ signaling-regulated glycolysis of GBM cells by AKT-independent mechanisms. We examined the impact of various pharmacological inhibitors on the suppression of mTORC1, mTORC2, AKT, and PKCδ signaling. Isolation Of Ace2 Inhibitors From Rice Bran Oil By-Product, Muhammad Waqar Nasir

This study aimed to isolate γ-oryzanol from the crude oil acid fraction (a by-product of rice bran oil refinery) and to evaluate their Angiotensin-Converting Enzyme 2 (ACE2) inhibitory activity. Chromatographic fractionation of the crude oil acid fraction gave 3 isolated fractions of γ-oryzanol containing cycloartenyl ferulate, campesteryl ferulate, sitosteryl ferulate, stigmasteryl ferulate, and cyclobranyl ferulate. The γ-oryzanol containing cycloartenyl ferulate and campesteryl ferulate as major component showed good ACE2 inhibitory activity with the IC50 value 10.57 μg/mL. Oxidative stress, resulting from an imbalance between reactive oxygen species (ROS) and antioxidants, contributes to diseases and induces endoplasmic reticulum (ER) stress. Antioxidants can mitigate ER stress. We investigated synthetic peptides from riceberry rice hydrolysates against ER stress. Ten peptides were evaluated for toxicity and antioxidant activity in SH-SY5Y and L929 cell lines. Peptides ID 3 to ID 5 ,and ID 10 showed significant antioxidant activity protecting cells from oxidative stress induced by IAA and H2O2. Cutibacterium acnes is one of the major factors involved in the pathogenesis of acne vulgaris which sometimes develops antibiotic resistance particularly clindamycin- and doxycycline-resistant C. acnes. In this study, human cathelicidin (LL-37) which is a type of human antimicrobial peptide against a wide range of microorganisms was used. We investigated the antimicrobial activity of LL-37 on C. acnes isolated from patients with acne at the King Chulalongkorn Hospital using broth microdilution. Moreover, the synergistic effects of LL-37 with clindamycin and doxycycline were investigated using checkerboard microdilution, and time-killing assays. In this study, a plant from the Orchidaceae family, Aerides falcata, was investigated for its chemical constituents and anti-neuroinflammatory activity. A total of ten compounds were isolated and characterized. The isolated compounds included a new compound which was named aerifalcatin and nine known compounds: n-eicosyl-trans-ferulate, denthyrsinin, 2,4-dimethoxy-3,7-dihydroxyphenanthrene, 2,7-dihydroxy-3,4,6-trimethoxyphenanthrene, 3,7-dihydroxy-2,4,6-trimethoxyphenanthrene, agrostonin, syringaresinol, trans-n-feruloyltyramine, and trans-n-coumaroyltyramine. All the isolated compounds were evaluated for their anti-neuroinflammatory activity, except for trans-n-coumaroyltyramine, which was excluded due to its insufficient amount. Quantifying historical patterns of fire regimes in peatlands can help contextualise current fire behaviour and aid in planning on ecosystem and landscape scales. However, current methods for detecting the evidence of past fires in peat soils are laborious or expensive. Our goal was to develop an effective and inexpensive method for detecting pyrogenic carbon (PyC) concentration in peat which could be used to estimate the occurrence of fires by analysis of discrete soil samples. As human impacts from wildfires mount, there is a pressing need to understand why structures are lost in destructive fires. Despite growing research on factors contributing to structure loss, fewer studies have focused on why some fires are destructive and others are not. We characterized overall differences between fires that resulted in structure loss (“destructive fires”) and those that did not (“non-destructive wildfires”) across three California regions. Then, we performed statistical analyses on large fires only (≥100 ha) to distinguish the primary differences between large destructive large fires and large non-destructive fires. Legal theory and doctrine depend on underlying assumptions about human nature and sociality. Perhaps the most common and basic assumption is that we are separate persons who communicate imperfectly with one another. While this separation thesis has been questioned, it still dominates legal theory. However, I show that understanding separation and connection as alternative perspectives, rather than as ontologically true or false, reveals that legal conflict often arises when these perspectives give rise to clashing intuitions concerning the meaning of community and what constitutes goals and harms. Fraud, misrepresentation, and other unfair trade practices plague the market for human reproductive tissue. The sale of sperm, eggs, and embryos is virtually unregulated in almost all states, and courts have been inhospitable to victims. As a result, children are born with genetic disorders that impose extreme financial and personal hardship. Proposals for direct government oversight have, for the most part, failed to gain traction, and litigation has yielded inadequate remedies.

The purpose of this pilot study was to examine the attitudes of college-age students on the use of essential oils, alternative medicines, and modern medicine. Essential oils are natural oils that contain the essence/fragrance of a plant and are often used for cleaning purposes (e.g., in soaps, detergents, disinfectant sprays) and therapeutic purposes (e.g., relieving aches and pains, calming a person, healing illnesses and injuries, improving mood). Although essential oils are primarily used in alternative medicine, some oils have also been used on an experimental basis in modern medical settings (Corner et al., 1995).

Sleep is related to cognitive functioning, learning, and brain development in the adolescent population. Recent research indicates a rise in the presence of chronic sleep disorders such as insomnia in adolescents, particularly following the COVID-19 pandemic. Immigrant mothers face numerous challenges and unique stressors related not only to their role as asylum-seekers but also to their roles as women and caretakers. Some studies suggest that immigrant mothers may exhibit high internalizing symptoms related to pre-migration trauma exposure, while others claim that such symptoms may be due to the internalization of their children’s mental health. The replacement of natural lands with urban structures has multiple environmental consequences, yet little is known about the magnitude and extent of albedo-induced warming contributions from urbanization at the global scale in the past and future. Here, we apply an empirical approach to quantify the climate effects of past urbanization and future urbanization projected under different shared socioeconomic pathways (SSPs). We find an albedo-induced warming effect of urbanization for both the past and the projected futures under three illustrative scenarios. The albedo decease from urbanization in 2018 relative to 2001 has yielded a 100-year average annual global warming of 0.00014 …
