The Case For Integrated Pest Management In Africa: Transition From A Pesticide-Based Approach, Ramasamy Srinivasan, Manuele Tamò, Sevgan Subramanian

All Peer-Reviewed Publications

Pest management in major cropping systems has long been dominated by chemical pesticides in Africa. Smallholder farmers have perceived pesticides as insurance to protect their crops. Consumers are less aware of pesticide-related food-safety issues, and markets providing economic incentives to producers who adopt alternatives to pesticides have been slow to emerge. Hence, overuse of pesticides has been constantly increasing. Although African countries have a number of pesticide-related policies, they are inadequately implemented. However, significant investment has been made on integrated pest management (IPM) innovations in recent decades. Resistant cultivars, agroecological approaches, biological control, and biopesticides have been developed against key …

Numerous molecular and physiological processes in the skeletal muscle undergo circadian time-dependent oscillations in accordance with daily activity/rest cycles. The circadian regulatory mechanisms underlying these cyclic processes, especially at the post-transcriptional level, are not well defined. Previously, we reported that the circadian E3 ligase FBXL21 mediates rhythmic degradation of the sarcomere protein TCAP in conjunction with GSK-3β, and Psttm mice harboring an Fbxl21 hypomorph allele show reduced muscle fiber diameter and impaired muscle function. To further elucidate the regulatory function of FBXL21 in skeletal muscle, we investigated another sarcomere protein, Myozenin1 (MYOZ1), that we identified as an FBXL21-binding protein from …

Failure of hypomethylation agent (HMA) treatments is an important issue in myelodysplastic syndromes (MDS) and chronic myelomonocytic leukemia (CMML). Recent studies indicated that function of wildtype TP53 positively impacts outcome of HMA treatments. We investigated the combination of the HMA azacitidine (AZA) with DS-3032b and DS-5272, novel antagonists of the TP53 negative regulator MDM2, in cellular and animal models of MDS and CMML. In TP53 wildtype myeloid cell line, combinational effects of DS-3032b or DS-5272 with AZA were observed. In Tet2-knockout mouse model of MDS and CMML, DS-5272 and AZA combination ameliorated disease-like phenotype. RNA-Seq analysis in mouse bone …

Molecular Characterization Of Nitrogenase Regulation In Methanosarcina Acetivorans, Melissa Chanderban Dec 2022

Molecular Characterization Of Nitrogenase Regulation In Methanosarcina Acetivorans, Melissa Chanderban

Graduate Theses and Dissertations

Nitrogenase is the metalloenzyme only found in bacteria and archaea that is essential for biological nitrogen fixation (diazotrophy), but it can also serve as a catalyst in biofuel production. All diazotrophs contain a molybdenum (Mo) nitrogenase, while some species contain additional alternative nitrogenases where either vanadium (V) or iron (Fe) replace Mo in the active site cofactor. Nitrogen fixation by bacteria has been extensively studied. The limited investigation of nitrogen fixation in methanogenic archaea (methanogens) indicates production of nitrogenase is simpler than in bacteria and methanogen nitrogenase has different biochemical properties. Thus, methanogen nitrogenases provide a promising alternative for genetic …

Hypothesis Testing And Parameter Estimation In Mixture Cure Models For Cancer Survival Data, Mohammod Mahmudur Rahman Dec 2022

Hypothesis Testing And Parameter Estimation In Mixture Cure Models For Cancer Survival Data, Mohammod Mahmudur Rahman

Graduate Theses and Dissertations

In oncology clinical trials, when a treatment is administered to the patient population, a certain subset of patients may respond to the treatment while the other does not. The positive responders with long-term survival are considered “statistically cured” and can be referred to as cured patients or long-term survivors. When a proportion of patients achieve long-term survival, the hazard functions of two arms (control vs. treatment) are no longer proportional. As a result, the traditional log-rank test, which is the most popular test to evaluate the effectiveness of a treatment in clinical trials, tends to lose its power. In this …

Comparative Analysis Of Conformational Transition Pathways In Homologous Proteins, Dylan Sebastien Ogden

Graduate Theses and Dissertations

Molecular dynamics (MD) simulations are routinely used to study the dynamics of proteins. However, conventional MD limited to the sampling of local conformational changes as the functionally important conformational transitions of proteins often extend beyond the timescales of the simulations employed, for example, membrane transport proteins. We have determined the combination of multiple MD based techniques that allows for a rigorous characterization of energetics and kinetics of large-scale conformational changes in membrane proteins. The methodology is based on biased, nonequilibrium collective variable based simulations including nonequilibrium pulling, string method with swarms of trajectories, bias-exchange umbrella sampling, and rate estimation techniques. …

Empirical and computational methods were combined to examine whether individual or dual-drug treatments can restore the deficit in long-term synaptic facilitation (LTF) of the Aplysia sensorimotor synapse observed in a cellular model of Coffin–Lowry syndrome (CLS). The model was produced by pharmacological inhibition of p90 ribosomal S6 kinase (RSK) activity. In this model, coapplication of an activator of the mitogen-activated protein kinase (MAPK) isoform ERK and an activator of protein kinase A (PKA) resulted in enhanced phosphorylation of RSK and enhanced LTF to a greater extent than either drug alone and also greater than their additive effects, which is termed …

Faculty, Staff and Student Publications

We propose a curve-free random-effects meta-analysis approach to combining data from multiple phase I clinical trials to identify an optimal dose. Our method accounts for between-study heterogeneity that may stem from different study designs, patient populations, or tumor types. We also develop a meta-analytic-predictive (MAP) method based on a power prior that incorporates data from multiple historical studies into the design and conduct of a new phase I trial. Performances of the proposed methods for data analysis and trial design are evaluated by extensive simulation studies. The proposed random-effects meta-analysis method provides more reliable dose selection than comparators that rely …

The Distinctive Bionomics Of Aedes Aegypti Populations In Africa, David P. Tchouassi, Sheila B. Agha, Jandouwe Villinger, Rosemary Sang, Baldwyn Torto

All Peer-Reviewed Publications

Aedes aegypti is the primary vector of dengue, chikungunya, and Zika viruses of medical importance. Behavioral and biological attributes contribute to its vectorial capacity. The mosquito domestic form, which resides outside Africa (Ae. aegypti aegypti (Aaa)), is considered to breed in artificial containers in and around homes and preferentially feeds on human blood but commonly indulges in a plant diet. Potential divergence in these attributes, in sub-Saharan Africa (SSA) where Aaa coexists with the forest ecotype (Ae. aegypti formosus), should impact the vectoring ability and hence disease epidemiology. A summary of current knowledge on Ae. aegypti blood feeding, oviposition, and …

Melanoidins can be diagnosed using the Fourier transform infrared (FTIR) technique. UV/Vis is an effective tool for both qualitative and quantitative analysis of chemical components in melanoidin polymers. The structural and vibrational features of melanoidin synthesized from D-glucose and D-fructose are identical, according to FTIR spectra, with the only difference being the intensity of bands. Using FTIR spectra, the skeleton of melanoidin is divided into seven major regions. The existence of the C=C, C=N, and C=O groups in all melanoidins formed from fructose and glucose with ammonia is confirmed by the areas ranging from 1600 to 1690 cm-1, and the …

Investigations Of Prokaryotic Defense Systems, Hannah Domgaard

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Bacteria are constantly threatened with infection by mobile genetic elements (MGE) such as bacteriophage and plasmids. Bacteriophage and plasmids require the bacteria's cellular infrastructure to replicate their genomes. Rampant replication can lead to cell death which is one reason why bacteria have developed a diverse array of immune systems to prevent or limit infection. This thesis studies three types of bacterial immune systems, type IV-A CRISPR-Cas (Clustered Regularly Interspaced Short Palindromic Repeat –CRISPR associated), type V-A2 CRISPR-Cas systems, and Wadjet systems.

The type IV-A system lies adjacent to a dinG-like helicase gene. Research has shown that this system can target …

Injury to quizalofop-resistant rice was reported in some fields following postemergence applications of quizalofop. Glyphosate-resistant (GR) corn, cotton, and soybean, and imidazolinone-resistant rice are grown near quizalofop-resistant rice. Herbicide drift from glyphosate and imazethapyr and the resulting crop injury and potential yield loss is a cause of concern for producers. Field experiments conducted near Colt, and Keiser, AR, in 2021 evaluated whether low rates of glyphosate or imazethapyr interact with sequential quizalofop applications to exacerbate injury to quizalofop-resistant rice compared to quizalofop applications alone. Herbicide treatments consisted of a low rate of glyphosate (90 g ae ha(-1)) or imazethapyr (10.7 …

Functional connectome organization is altered in schizophrenia (SZ) and bipolar disorder (BD). However, it remains unclear whether network reorganization during a task relative to rest is also altered in these disorders. This study examined connectome organization in patients with SZ (N = 43) and BD (N = 42) versus healthy controls (HC; N = 39) using fMRI data during a visual object-perception task and at rest. Graph analyses were conducted for the whole-brain network using indices selected a priori: three reflecting network segregation (clustering coefficient, local efficiency, modularity), two reflecting integration (characteristic path length, global efficiency). Group differences were limited …
