An effective vaccine against Chlamydia trachomatis is urgently needed as infection rates continue to rise and C. trachomatis causes reproductive morbidity. An obligate intracellular pathogen, C. trachomatis employs a type 3 secretion system (T3SS) for host cell entry. The tip of the injectosome is composed of the protein CT584, which represents a potential target for neutralization with vaccine-induced antibody. Here, we investigate the immunogenicity and efficacy of a vaccine made of CT584 epitopes coupled to a bacteriophage virus-like particle (VLP), a novel platform for Chlamydia vaccines modeled on the success of HPV vaccines. Volunteer-contributed geographic data (VGI) is an important source of geospatial big data that support research and applications. A major concern on VGI data quality is that the underlying observation processes are inherently biased. Detecting observation hot-spots thus helps better understand the bias. Enabled by the parallel kernel density estimation (KDE) computational tool that can run on multiple GPUs (graphics processing units), this study conducted point pattern analyses on tens of millions of iNaturalist observations to detect and visualize volunteers’ observation hot-spots across spatial scales. In coastal wetlands and tropical reefs, snails can regulate foundation species by feeding on marsh grasses and hard corals. In many cases, their impacts are amplified because they facilitate microbial infection in grazer-induced wounds. Whether snails commonly graze live plants and facilitate microbial growth on plants in tropical seagrass systems is less explored. On a Belizean Caye, we examined patterns in snail-generated grazer scars on the abundant turtlegrass (Thalassia testudinum). Our initial survey showed the occurrence of snail-induced scarring on live turtlegrass blades was common, with 57% of live leaves scarred. Feeding trials demonstrated that two of five common snails …

Bovine respiratory disease (BRD), as one of the most common and costly diseases in the beef cattle industry, has significant adverse impacts on global food security and the economic stability of the industry. The bovine respiratory microbiome is strongly associated with health and disease and may provide insights for alternative therapy when treating BRD. The niche-specific microbiome communities that colonize the inter-surface of the upper and the lower respiratory tract consist of a dynamic and complex ecological system. Genetic studies on telomere length are important for understanding age-related diseases. Prior GWAS for leukocyte TL have been limited to European and Asian populations. Here, we report the first sequencing-based association study for TL across ancestrally-diverse individuals (European, African, Asian and Hispanic/Latino) from the NHLBI Trans-Omics for Precision Medicine (TOPMed) program. We used whole genome sequencing (WGS) of whole blood for variant genotype calling and the bioinformatic estimation of telomere length in n=109,122 individuals. Evidence-based studies on the benefits of integrating STEM into the arts are limited; however, some suggest that it can lead to improved scientific literacy and new approaches for artistic scholarship. Unfortunately, undergraduate education often creates disciplinary silos where the two are not integrated. Here, we discuss a unique collaboration between professors in the art and biology departments. Our goal was to integrate science into art courses using an agar art activity. We hypothesized that art students could effectively learn microbiology laboratory techniques and use them as novel tools for artistic practice. The activity was integrated into two to four sessions …

Background
The Glanville fritillary (Melitaea cinxia) butterfly is a model system for metapopulation dynamics research in fragmented landscapes. Here, we provide a chromosome-level assembly of the butterfly's genome produced from Pacific Biosciences sequencing of a pool of males, combined with a linkage map from population crosses.
Results
The final assembly size of 484 Mb is an increase of 94 Mb on the previously published genome. Estimation of the completeness of the genome with BUSCO indicates that the genome contains 92–94% of the BUSCO genes in complete and single copies. We predicted 14,810 genes using the MAKER pipeline and …

Nurses' roles facilitating bonding between adoptive parents and their infants have been premised on the idea that the experience is the same as for biological parents. Developmental theorists Bowlby (1969) and Erikson (1959) maintained that bonding between an adoptive parent and their adopted child is essential for normal growth and development. The purpose of this study was to study the experience of bonding between adoptive parents and children.

Severe under-representation of Black women academic nurse leaders persists in United States higher education, and a major research gap still exists regarding experiences of these leaders, and facilitators of and barriers to their success. Our objective was to examine how race and gender influence how Black women academic nurse leaders’ function in their leadership positions, how they are perceived by their peers, and how their perception of race, gender, class, and power influences diversity, equity, and inclusion initiatives in the workplace. Genetic studies on telomere length are important for understanding age-related diseases. Prior GWAS for leukocyte TL have been limited to European and Asian populations. Here, we report the first sequencing-based association study for TL across ancestrally-diverse individuals (European, African, Asian and Hispanic/Latino) from the NHLBI Trans-Omics for Precision Medicine (TOPMed) program. We used whole genome sequencing (WGS) of whole blood for variant genotype calling and the bioinformatic estimation of telomere length in n=109,122 individuals. We identified 59 sentinel variants (p-value <5×10-9) in 36 loci associated with telomere length, including 20 newly associated loci (13 were replicated in external datasets). There was little evidence of effect size heterogeneity across populations. Fine-mapping at OBFC1 indicated the independent signals colocalized with cell-type specific eQTLs for OBFC1 (STN1). Heart valve development is governed by both genetic and biomechanical inputs. Prior work has demonstrated that oscillating shear stress associated with blood flow is required for normal atrioventricular (AV) valve development. Cardiac afterload is defined as the pressure the ventricle must overcome in order to pump blood throughout the circulatory system. In human patients, conditions of high afterload can cause valve pathology. Whether high afterload adversely affects embryonic valve development remains poorly understood. Here we describe a zebrafish model exhibiting increased myocardial afterload, caused by vasopressin, a vasoconstrictive drug. The endogenous lysosomal cysteine protease inhibitor SERPINB3 (squamous cell carcinoma antigen 1, SCCA1) is elevated in patients with cervical cancer and other malignancies. High serum SERPINB3 is prognostic for recurrence and death following chemoradiation therapy. Cervical cancer cells genetically lacking SERPINB3 are more sensitive to ionizing radiation (IR), suggesting this protease inhibitor plays a role in therapeutic response. Here we demonstrate that SERPINB3-deficient cells have enhanced sensitivity to IR-induced cell death. Knock out of SERPINB3 sensitizes cells to a greater extent than cisplatin, the current standard of care. IR in SERPINB3 deficient cervical carcinoma cells induces predominantly necrotic cell death, …

Lysosomal membrane permeabilization (LMP) and cathepsin release typifies lysosome-dependent cell death (LDCD). However, LMP occurs in most regulated cell death programs suggesting LDCD is not an independent cell death pathway, but is conscripted to facilitate the final cellular demise by other cell death routines. Previously, we demonstrated that Caenorhabditis elegans (C. elegans) null for a cysteine protease inhibitor, srp-6, undergo a specific LDCD pathway characterized by LMP and cathepsin-dependent cytoplasmic proteolysis. A graphene oxide (GO)-based cortisol biosensor was developed to accurately detect corti-sol concentrations from sweat samples at point-of-care (POC) sites. A reference electrode, counter electrode, and working electrode make up the biosensor, and the working electrode was functional-ized using multiple layers consisting of GO and antibodies, including Protein A, IgG, and anti-Cab. Sweat samples contact the anti-Cab antibodies to transport electrons to the electrode, resulting in an electrochemical current response. The sensor was tested at each additional functionalization layer and at cortisol concentrations between 0.1 and 150 ng/mL to determine how the current response differed. Objectives: To assess the prevalence of antibiotic-resistant gram-negative bacteria (R-GNB) among patients without recent hospitalization and to examine the influence of outpatient antibiotic exposure on the risk of acquiring R-GNB in this population. Design: 2-year retrospective cohort study. Setting: Regional Veterans Affairs healthcare system. Patients: Outpatients at 13 community-based clinics. Methods: We examined the rate of acquisition of R-GNB within 90 days following an outpatient visit from 2018 to 2019. We used clinical and administrative databases to determine and summarize prescriptions for systemic antibiotics, associated infectious diagnoses, and subsequent R-GNB acquisition among patients without recent hospitalizations. Background: Irreversible electroporation (IRE) has emerged as a viable consolidative therapy after induction chemotherapy, in which this combination has improved overall survival of locally advanced pancreatic cancer (LAPC). Optimal timing and patient selection for irreversible electroporation remains a clinically unmet need. The aim of this study was to investigate preoperative factors that may assist in predicting progression-free and overall survival following IRE.

Extracellular signal-regulated kinase 3 (ERK3) is an atypical member of the mitogen-activated protein kinase (MAPK) family, members of which play essential roles in diverse cellular processes during carcinogenesis, including cell proliferation, differentiation, migration, and invasion. Unlike other MAPKs, ERK3 is an unstable protein with a short half-life. Although deubiquitination of ERK3 has been suggested to regulate the activity, its ubiquitination has not been described in the literature. Here, we report that FBXW7 (F-box and WD repeat domain-containing 7) acts as a ubiquitination E3 ligase for ERK3. The dramatic increase in greenhouse gas (GHG) emissions by humans over the past century and a half has created an urgency for monitoring, reporting, and verifying GHG emissions as a first step toward mitigating the effects of climate change. Fifteen percent of global GHG emissions come from agriculture, and companies in the food and beverage industry are starting to set climate goals. Given the rapid development of genome mining in this decade, the substrate channel of paclitaxel might be identified in the near future. A robust microbial cell factory with gene dbat, encoding a key rate-limiting enzyme 10-deacetylbaccatin III-10-O-transferase (DBAT) in paclitaxel biosynthesis to synthesize the precursor baccatin III, will lay out a promising foundation for paclitaxel de novo synthesis. Here, we integrated gene dbat into the wild-type Escherichia coli BW25113 to construct strain BWD01. Yet, it was relatively unstable in baccatin III synthesis. Integrated Orderly Classification System of Grassland (IOCSG) was initiated by Professor Ji-Zhou Ren in the 1960s and it has satisfactorily served to direct theory and practice of grassland classification for the last several decades in China. In this study IOCSG is adopted to classify the grasslands in Yobe state Nigeria, using 30 years (1984-2013) data on precipitation and temperature from 12 satellite stations across the state. Humidity (moisture index) was estimated as the ratio of precipitation per annum to annual temperature. An analysis of forbs from 9 years of Plant Census protocol (NRI 2019) is compared to data from the Line Point Intercept (LPI) protocol. Both protocols were conducted on the same geospatially separate macroplots (N=1230). The macroplots locations were stratified by NRI based on a land hierarchy which has had long-term use in the United States (Salley etal 2015). This ties vegetation and land together in the data. The analysis showed that the 15-minute plant census method increased forb species data by 29 percent over the LPI method which takes 40-60 minutes. Grasslands are known to host crop pests and may act as the reservoir of pests affecting maize production. This research aimed at determining the effect of grassland composition on abundance of stemborers on grasses sorrouding maize farms in western Kenya. The experimental set up followed a complete random block design (CRBD); with four elevations at Lambwe, Homabay, Luanda and Mt. Elgon assigned as blocks based on different altitudes. Sampling was done in two seasons, during the short rain season of 2019 and the long rain season of 2020. Grasslands surrounding four pairs of push-pull and non-push-pull maize farms were assessed. …

This study used global positioning systems (GPS) to understand grazing distribution of three cattle in the communal rangeland of Eastern Cape, South Africa. Free ranging cattle were randomly selected from participating households and were fitted with neck GPS collars, which recorded geographic position every five-minute interval during wet and dry season.
