Pathogenic coronaviruses are a major threat to global public health. Here, using a recombinant reporter virus-based compound screening approach, we identified small-molecule inhibitors that potently block the replication of severe acute respiratory syndrome virus 2 (SARS-CoV-2). Among them, JIB-04 inhibited SARS-CoV-2 replication in Vero E6 cells with a 50% effective concentration of 695 nM, with a specificity index of greater than 1,000. JIB-04 showed

We examine the historical and statistical relationship between race and life insurance. Life insurance can play a central role in households’ financial security. Race has played an important and changing role in the provision of life insurance in the U.S. from slave insurance before the Civil War, to “Scientific Racism” continuing into the 20th century, to policies that do not explicitly mention race in recent decades. In empirical work using new data, we confirm earlier work showing that Black individuals have higher life insurance coverage rates than white individuals, controlling for observable characteristics. The unsaturated zone is a critical component of karstic groundwater systems and is shown to provide substantial storage capacities. Understanding the spatial patterns and controls on flow path activation is often a challenge. Previous research focused on remotely sensed data or inferential analyses to quantify these patterns. Here, we use two cave systems—one in Arizona, USA and a second in Kentucky, USA—to show the value of the cave survey and inventory data in the direct observation of speleogenesis and unsaturated zone processes. A global international initiative, such as the Earth BioGenome Project (EBP), requires both agreement and coordination on standards to ensure that the collective effort generates rapid progress toward its goals. To this end, the EBP initiated five technical standards committees comprising volunteer members from the global genomics scientific community: Sample Collection and Processing, Sequencing and Assembly, Annotation, Analysis, and IT and Informatics. The current versions of the resulting standards documents are available on the EBP website, with the recognition that opportunities, technologies, and challenges may improve or change in the future, requiring flexibility for the EBP to meet its goals. …

Life on Earth has evolved from initial simplicity to the astounding complexity we experience today. Bacteria and archaea have largely excelled in metabolic diversification, but eukaryotes additionally display abundant morphological innovation. How have these innovations come about and what constraints are there on the origins of novelty and the continuing maintenance of biodiversity on Earth? The history of life and the code for the working parts of cells and systems are written in the genome. The Earth BioGenome Project (EBP) is an audacious endeavor to obtain whole-genome sequences of representatives from all eukaryotic species on Earth. In addition to the project’s technical and organizational challenges, it also faces complicated ethical, legal, and social issues. This paper, from members of the EBP’s Ethical, Legal, and Social Issues (ELSI) Committee, catalogs these ELSI concerns arising from EBP. This study was carried out for the mycosynthesis of silver nanoparticles by Candida albicans supernatant. All the isolates used in this study were taken from the patients who existed at Al-Elweya children's teaching hospital in Baghdad, Iraq. Mycosynthesized silver nanoparticles were characterized by color visualization, ultraviolet-visible (UV) spectroscopy, and Fourier transform infrared spectroscopy (FTIR). The UV-Vis spectroscopy examination has shown the highest absorbance (λmax) at the wavelength of 429 nanometers, which indicated the creation of silver nanoparticles. Furthermore, the results of the antibacterial potential of AgNO3 and AgNPs against Klebsiella pneumoniae and Staphylococcus haemolyticus bacteria showed the highest effect of …

Background: Reactive balance is the last line of defense to prevent a fall when the body loses stability, and beneficial effects of various exercise-based interventions on reactive balance in older adults have been reported. The fidelity of a signaling pathway depends on its tight regulation in space and time. Extracellular signal-regulated kinase (ERK) controls wide-ranging cellular processes to promote organismal development and tissue homeostasis. In chronic lymphocytic leukemia (CLL), the B cell receptor (BCR) plays a critical role in disease development and progression, as indicated by the therapeutic efficacy of drugs blocking BCR signaling. However, the mechanism(s) underlying BCR responsiveness are not completely defined. Selective engagement of membrane IgM or IgD on CLL cells, each coexpressed by more than 90% of cases, leads to distinct signaling events. Since both IgM and IgD carry the same antigen-binding domains, the divergent actions of the receptors are attributed to differences in immunoglobulin (Ig) structure or the outcome of signal transduction. More than 10,000 years ago humans in multiple parts of Eurasia began to form increasingly more entangled and mutually dependent relationships with non-human animals. In some cases, the elaboration of these relationships would result in evolutionary change, resulting in biologically distinct or domesticated populations. The last several decades of research show that this process is ongoing and polymorphous, emerging out of sustained human-animal interactions that develop along historically contingent trajectories shaped by particular biological, environmental, and cultural contexts. Thus, understanding domestication as an evolutionary process necessitates investigation into species- and context-specific developmental trajectories of human-animal interaction. Worldwide, malnutrition (undernutrition) is the leading cause of death in children under 5. Studies of healthy members of birth cohorts from low- and middle-income countries have identified a program of gut microbial community development that is disrupted in children with severe acute malnutrition (SAM) and moderate acute malnutrition (MAM). This perturbed gut microbiome development has been linked to the pathogenesis of malnutrition. Abstract: Most phages of Gram-negative bacteria hosts encode spanins for disruption of the outer membrane, which is the last step in host lysis. However, bioinformatic analysis indicates that ∼15% of these phages lack a spanin gene, suggesting they have an alternate way of disrupting the outer membrane (OM). Here, we show that the T7-like coliphage phiKT causes an explosive cell lysis associated with spanin activity despite not encoding spanins. A putative lysis cassette cloned from the phiKT late gene region includes the hypothetical novel gene 28 located between the holin and endolysin genes and supports inducible lysis in Escherichia …

Therapeutics targeting osteoclasts are commonly used treatments for bone metastasis; however, whether and how osteoclasts regulate premetastatic niche and bone tropism are largely unknown. In this study, we report that osteoclast precursors (OPs) can function as a premetastatic niche component that facilitates breast cancer (BCa) bone metastasis at early stages. At the molecular level, unbiased GPCR ligand/agonist screening in BCa cells suggested that R-spondin 2 (RSPO2) and RANKL, through interaction with their receptor LGR4, promoted osteoclastic premetastatic niche formation and enhanced BCa bone metastasis. This was achieved by RSPO2/RANKL-LGR4 signal modulating the WNT inhibitor DKK1 through Gαq and β-catenin signaling. …

IMPORTANCE: Platelets represent a potential therapeutic target for improved clinical outcomes in patients with COVID-19.
OBJECTIVE: To evaluate the benefits and risks of adding a P2Y12 inhibitor to anticoagulant therapy among non-critically ill patients hospitalized for COVID-19.
DESIGN, SETTING, AND PARTICIPANTS: An open-label, bayesian, adaptive randomized clinical trial including 562 non-critically ill patients hospitalized for COVID-19 was conducted between February 2021 and June 2021 at 60 hospitals in Brazil, Italy, Spain, and the US. The date of final 90-day follow-up was September 15, 2021.

Oat, triticale, wheat and barley are small grain cereals used as forage in many temperate Mexican regions. The objective was to determine crude protein and van Soest fiber contents of these forages cut at 80, 96, 108, 121, 138 and 153 days after seeding. Cultivars used were: Chihuahua (OC) for oat; Arne (TA), Bicentenario (TB) and Siglo XXI (TS) for triticale; Saturno (WS) for wheat; and San Marcos (BSM) for barley. There is an urgent need for the prioritization and ex situ conservation of pasture grass species. Many pasture grasses are still in the process of domestication and grasslands in the tropical, subtropical, semi-arid and arid regions of the world are therefore a major source of genetic material for future forage improvement. Within South Africa, it was found that the Paniceae contains several species with high pasture value and that many populations of these species are found in water-limited environments. Several studies have evaluated separately forage production, botanical composition, leaf traits and animal performance. However, few of them have focused on defoliation patterns at the level of functional groups (FGs) under different and variable herbage allowance (HA), especially in natural, diverse grasslands. The objective was to evaluate the relationship between HA and leaf traits on defoliation patterns of mature beef cows in the autumn, winter and spring. Rio Grande Silvery Minnow (RGSM) are currently constrained to only 5% of their historic range, and their persistence is threatened by highly altered and impaired habitat conditions (Bestgen and Platania 1991). These habitat conditions have resulted from reduced spring and summer flows due to natural variability and anthropogenic water development and extraction, which have resulted in substantial geomorphic changes (Swanson et al. 2011). Successful conservation of this endangered species will require determination of how available flows can be managed to provide conditions supporting growth, reproduction, and survival of RGSM within and across a variety of water years. Bringing our collective experiences of past collaborations through a virtual connection, we created an international research team of 16 multidiscipline, multicultural, and multilingual academic women called “COVID GAP” (Gendered Academic Productivity) to explore the ongoing challenges and effects of COVID-19. Identifying as insider researchers, we engaged in a two-phase, primarily qualitative research project to better understand the lived experiences of academics during the pandemic. Ultrasound is a sound wave with frequencies ranging between 20 kHz and 20 MHz. Ultrasound is able to temporarily and repeatedly open the BBB safely and enhance chemotherapeutic delivery without adverse effects. This novel technique in drug delivery benefits from the powerful ability of ultrasound to produce cavitation activity. Cavitation is the generation and activity of gas-filled bubbles in a medium exposed to ultrasound. As the pressure wave passes through the media, gas bubbles expand at low pressure and contract at high pressure. The accumulation of various types of waste containing both organic and inorganic metal-containing compounds is extremely hazardous for living organisms. The possibility of polymer degradation, biohydrogen synthesis, and metal detoxification via the dark fermentation of model potato waste was investigated. For this purpose, the strict anaerobic strain was isolated and identified as Clostridium butyricum. The high efficiency of dark hydrogen fermentation of potatoes with yield of hydrogen in 85.8 ± 15.3 L kg−1 VSpotato was observed.
