How Capuchin Monkeys Use Their Semi-Prehensile Tails, Madhur Mangalam, Callum F. Ross, Patricia Izar, Elisabetta Visalberghi, Dorothy M. Fragaszy

Among primates, prehensile/semi-prehensile tails have evolved independently in the families Atelidae and Cebidae of the infraorder Platyrrhini (Neotropical monkeys). They facilitate maintaining stability during locomotion on thin, flexible branches and while reaching for food on challenging substrates. How a prehensile/semi-prehensile tail is coordinated with the hind limbs to facilitate controlled, flexible adoption of postures remains unknown. In an experimental set-up in the wild, we induced capuchin monkeys to adopt a tail-assisted, head-down tripodal posture to reach for food, documenting from slow-motion video recordings (120 fps) both qualitative changes in the monkeys’ positional behaviour – the relative orientation of their limbs …

Chimeric antigen receptor (CAR) T cells have transformed the therapeutic options for relapsed/refractory (R/R) B-cell acute lymphoblastic leukemia. Data for CAR therapy in extramedullary (EM) involvement are limited. Retrospective data were abstracted from the Pediatric Real World CAR Consortium (PRWCC) of 184 infused patients from 15 US institutions. Response (complete response) rate, overall survival (OS), relapse-free survival (RFS), and duration of B-cell aplasia (BCA) in patients referred for tisagenlecleucel with EM disease (both central nervous system (CNS)3 and non-CNS EM) were compared with bone marrow (BM) only. In 2021, Eastern Michigan University Archives lecturer Matt Jones began documenting the story of Ypsilanti’s Human Rights Ordinance #1279 in an effort to explore the ways in which local queer activism has evolved multi-generationally in Ypsilanti. What began as a refusal of service by a local print shop to a small EMU student group quickly turned into a years-long battle over who was deserving of basic human rights. To the LGBTQ activists and community members documented here, they had always been present in the community: working, paying taxes, painting their houses, mowing their lawns, attending council meetings, and even serving …

BACKGROUND: An increase in opioid use has led to an opioid crisis during the last decade, leading to declarations of a public health emergency. In response to this call, the Houston Emergency Opioid Engagement System (HEROES) was established and created an emergency access pathway into long-term recovery for individuals with an opioid use disorder. A major contributor to the success of the program is retention of the enrolled individuals in the program.

Watercress is an enriched source of phenethyl isothiocyanate (PEITC), among other phytochemicals, with an antioxidant capacity. The aim of this study was to (i) chemically characterize and (ii) biologically evaluate the profile of the main health-promoting compounds contained in edible (i.e., mixture of leaves and lateral buds) and non-edible (i.e., stems) parts of watercress in an in vitro model of malignant melanoma consisting of human malignant melanoma (A375), non-melanoma (A431) and keratinocyte (HaCaT) cells. Background: Ocular infection with Toxoplasma gondii is a major preventable cause of blindness, especially in young people. The aim of the present study was to assess detection rate of T. gondii DNA in blood samples of clinically diagnosed of ocular toxoplasmosis using uracil DNA glycosylase-supplemented loop-mediated isothermal amplification (UDG-LAMP) and real-time quantitative PCR (qPCR) based on REP-529 and B1.
Methods: One hundred and seventeen patients with clinically diagnosed ocular toxoplasmosis (OT) were participated in the study as well as 200 control patients. Peripheral blood samples were assessed using UDG-LAMP and qPCR techniques targeting REP-529 and B1.

Sodium and sulfate ions are among the suggested abundant ions on Europa, a moon of Jupiter. In order to investigate the potential habitability of Europa, we study the effects of sodium sulfate (Na2SO4) on a non-halophilic bacterium by subjecting Escherichia coli (E. coli) to a wide range of Na2SO4 concentrations (0–1.0 m). Nischarin (Nisch) is a cytosolic scaffolding protein that harbors tumor‐suppressor‐like characteristics. Previous studies have shown that Nisch functions as a scaffolding protein and regulates multiple biological activities. In the current study, we prepared a complete Nisch knockout model, for the first time, by deletion of exons 5 and 6. This knockout model was confirmed by Qrt– PCR and Western blotting with products from mouse embryonic fibroblast (MEF) cells. Embryos and adult mice of knockouts are significantly smaller than their wild‐type counterparts. The developmental origins of psychopathology begin before birth and perhaps even prior to conception. Understanding the intergenerational transmission of psychopathological risk is critical to identify sensitive windows for prevention and early intervention. Prior research demonstrates that maternal trauma history, typically assessed retrospectively, has adverse consequences for child socioemotional development. However, very few prospective studies of preconception trauma exist, and the role of preconception symptoms of posttraumatic stress disorder (PTSD) remains unknown. The current study prospectively evaluates whether maternal preconception PTSD symptoms predict early childhood negative affectivity, a key dimension of temperament and predictor of later psychopathology. Adolescents with low von Willebrand factor (VWF) levels and heavy menstrual bleeding (HMB) experience significant morbidity. There is a need to better characterize these patients genetically and improve our understanding of the pathophysiology of bleeding. We performed whole-exome sequencing on 86 postmenarchal patients diagnosed with low VWF levels (30-50 IU/dL) and HMB and compared them with 660 in-house controls. We compared the number of rare stop-gain/stop-loss and rare ClinVar "pathogenic" variants between cases and controls, as well as performed gene burden and gene-set burden analyses. The specificity of CRISPR/Cas9 genome editing is largely determined by the sequences of guide RNA (gRNA) and the targeted DNA, yet the sequence-dependent rules underlying off-target effects are not fully understood. To systematically explore the sequence determinants governing CRISPR/Cas9 specificity, here we describe a dual-target system to measure the relative cleavage rate between off- and on-target sequences (off-on ratios) of 1902 gRNAs on 13,314 synthetic target sequences, and reveal a set of sequence rules involving 2 factors in off-targeting: 1) a guide-intrinsic mismatch tolerance (GMT) independent of the mismatch context; 2) an "epistasis-like" combinatorial effect of multiple mismatches, which are …

Reports of COVID-19-associated mucormycosis have been increasing in frequency since early 2021, particularly among patients with uncontrolled diabetes. Patients with diabetes and hyperglycaemia often have an inflammatory state that could be potentiated by the activation of antiviral immunity to SARS-CoV2, which might favour secondary infections. In this Review, we analysed 80 published and unpublished cases of COVID-19-associated mucormycosis. Uncontrolled diabetes, as well as systemic corticosteroid treatment, were present in most patients with COVID-19-associated mucormycosis, and rhino-orbital cerebral mucormycosis was the most frequent disease. Preclinically, enasidenib and azacitidine (ENA + AZA) synergistically enhance cell differentiation, and venetoclax (VEN), a small molecule Bcl2 inhibitor (i) is particularly effective in IDH2 mutated acute myeloid leukemia (IDH2mutAML). This open label phase II trial enrolled patients (pts) with documented IDH2mutAML. All patients received AZA 75 mg/m2/d x 7 d/cycle and ENA 100 mg QD continuously. Concomitant Bcl2i and FLT3i were allowed (NCT03683433).Twenty-six pts received ENA + AZA (median 68 years, range, 24-88); 7 newly diagnosed (ND) and 19 relapsed/refractory (R/R). Biopolymers and alkali-activated materials have attracted a great deal of attention as adsorbents for the removal of heavy metal contaminants from aqueous solutions. Both materials are sustainable and feature unique properties, but biopolymers are relatively more expensive or difficult to prepare and exhibit low mechanical and surface properties, a narrow pH range, and thermal stability. In this study, hybrid adsorbents were prepared from both types of material, by alkali activation of low-cost fly ash precursors accompanied by incorporation of 0-2%mass chitosan biopolymer. MeCP2 is associated with Rett syndrome (RTT), MECP2 duplication syndrome, and a number of conditions with isolated features of these diseases, including autism, intellectual disability, and motor dysfunction. MeCP2 is known to broadly bind methylated DNA, but the precise molecular mechanism driving disease pathogenesis remains to be determined. Using proximity-dependent biotinylation (BioID), we identified a transcription factor 20 (TCF20) complex that interacts with MeCP2 at the chromatin interface. Importantly, RTT-causing mutations in MECP2 disrupt this interaction. TCF20 and MeCP2 are highly coexpressed in neurons and coregulate the expression of key neuronal genes. Chronic obstructive pulmonary disease (COPD) is a leading cause of death worldwide, however our understanding of cell specific mechanisms underlying COPD pathobiology remains incomplete. Here, we analyze single-cell RNA sequencing profiles of explanted lung tissue from subjects with advanced COPD or control lungs, and we validate findings using single-cell RNA sequencing of lungs from mice exposed to 10 months of cigarette smoke, RNA sequencing of isolated human alveolar epithelial cells, functional in vitro models, and in situ hybridization and immunostaining of human lung tissue samples. Allogeneic transplant (alloHCT) and chimeric antigen receptor modified (CAR)-T cell therapy are potentially cuarative options of diffuse large B-cell lymphoma (DLBCL) relapsing after an autologous (auto)HCT. Although the Center for International Blood and Marrow Transplant Research (CIBMTR) prognostic model can predict outcomes of alloHCT in DLBCL after autoHCT failure, corresponding models of CAR-T treatment in similar patient populations are not available. We performed next-generation sequencing in patients with familial steroid-sensitive nephrotic syndrome (SSNS) and identified a homozygous segregating variant (p.H310Y) in the gene encoding clavesin-1 (CLVS1) in a consanguineous family with 3 affected individuals. Knockdown of the clavesin gene in zebrafish (clvs2) produced edema phenotypes due to disruption of podocyte structure and loss of glomerular filtration barrier integrity that could be rescued by WT CLVS1 but not the p.H310Y variant. Analysis of cultured human podocytes with CRISPR/Cas9-mediated CLVS1 knockout or homozygous H310Y knockin revealed deficits in clathrin-mediated endocytosis and increased susceptibility to apoptosis that could be rescued with corticosteroid treatment, mimicking …

Cereal plant age at harvesting and the proportion of vetch in the harvested forage from cereal-common vetch mixtures might influence total forage quality. The objectives were to determine forage of a forage mixture cereal-vetch, from cereal harvested at two development stages and vetch at different proportion. Cereals were oats and triticale, cultivars: Chihuahua, Bicentenario and Siglo XXI, the last two were triticale; cereal development stages at harvest were: 50% flowering and hard grain. Forage oat (Avena sativa L.) for livestock in Qinghai-Tibet Plateau may be used biomass under widely different altitude conditions. To assess the effect of altitude on the biomass of oats, we conducted an experiment to explore the adaptability of forage oat Jiayan No. 2 in different altitudes for better agronomic traits and higher forage yield. We aimed at evaluating the microclimate, canopy structure, and grazing behaviour of cattle on Brachiaria sp. pastures within continuous pasture (CON), integrated crop-livestock (ICL), and crop-livestock-forestry (ICLF) systems in Brazilian dry season. Forage accumulation was monthly assessed in four paddocks per system from May to August 2019. To characterize the microclimate, air temperature and relative humidity were measured every hour and temperature-humidity index calculated. Grazing behaviour of 12 Nellore heifers per system (mean bodyweight 355 standard deviation±32.6 kg) was recorded by chewing sensors on nine continuous days (1 d adaptation + 8 d measurement) of three 15-d periods. In the past, tall fescue ecotypes were compared under intermediate management defoliation frequency, but now has station, Argentina (-33 ° 56 'S, -60 ° 33' W) in autumn 2016, on a typical Argiudoll soil. In recent years walnut orchards implemented cover crops in between rows to improve soil’s quality, lessen soil’s erosion, increase organic matter, manage nutrient movement and availability, enhance water retention, and expand microbe, insect, and flora diversity. Commonly selected cover crops in California are from families Poaceae, Brassicaceae, and Fabaceae. Considerations should be made when choosing a particular cover crop mixture to enhance multiple benefits and improve sustainable practices in orchard settings.
