Effects Of Glucose On Early Development In Zebrafish Embryos, Angelica Manuel Fernando
Effects Of Glucose On Early Development In Zebrafish Embryos, Angelica Manuel Fernando
Undergraduate Honors Theses
In women with gestational diabetes, various hormones are secreted during pregnancy, such as estrogen and cortisol, which blocks insulin from activating the receptors to let glucose travel through the cells. To help adjust to the high glucose levels in the blood, pancreatic beta cells are used to uptake glucose to release insulin, to lower the concentration of the amount of glucose in the blood. When a fetus is indirectly exposed to high glucose throughout the bloodstream, it can lead to early-onset Type II diabetes in children. Zebrafish are an excellent animal model to use as they are aquatic and make …

Curated databases of species interactions are instrumental to exploring and understanding the spatial distribution of species and their biotic interactions. In the process of conducting such projects, data development and curation efforts may give rise to a data product with utility beyond the scope of the original work, but which becomes inaccessible over time. Data describing insect host–pathogen interactions are fairly rare, and should thus be preserved and curated with appropriate metadata. Here, we introduce the insectDisease R package, a mechanism for curating, updating and distributing data from the Ecological Database of the World's Insect Pathogens, a database of insect …

HIV vaccine mediated efficacy, using an expanded live attenuated recombinant varicella virus-vectored SIV rSVV-SIVgag/env vaccine prime with adjuvanted SIV-Env and SIV-Gag protein boosts, was evaluated in a female rhesus macaques (RM) model against repeated intravaginal SIV challenges. Vaccination induced anti-SIV IgG responses and neutralizing antibodies were found in all vaccinated RMs. Three of the eight vaccinated RM remained uninfected (vaccinated and protected, VP) after 13 repeated challenges with the pathogenic SIVmac251-CX-1. The remaining five vaccinated and infected (VI) macaques had significantly reduced plasma viral loads compared with the infected controls (IC). A significant increase in systemic central memory CD4+ T …

Fitness Of Interspecific Hybrids In The Genus Cyprinella: An Evaluation Of Swimming Performance In North American Stream Fishes, Jessica Wilks
Biology Theses
Due to anthropogenic pressures, freshwater ecosystems are being rapidly destroyed worldwide. Accordingly, human impacts have also resulted in the loss of native aquatic biodiversity. In particular, the introduction of non-native species to aquatic habitats is of great concern. Historically, the introductions of many non-native freshwater fishes are the result of commercial baitfish aquaculture, food industries, and private aquarium release. The red shiner (Cyprinella lutrensis) is a small stream fish that is endemic to the Central U.S., and its natural range is restricted to East of the Mississippi River. Since the 1950’s, red shiner have been commercially raised and transported …

Prenatal Opioid Use And Neonatal Abstinence Syndrome: A Review Of The Neurophysiological, Neuropsychological, And Behavioral/Emotional/Social Impacts In The Pediatric Population, Candice Gore
Dissertations
The opioid epidemic over the past two decades has raised concerns regarding the developmental fetal impact of prenatal opioid use. Research in this area continues to grow, but largely has focused on treatment for neonates experiencing withdrawal symptoms postnatally. Long term clinical implications for this at-risk population have not been studied extensively leaving many gaps in research and highlighting the need for future empirical studies. This literature review will examine the neurophysiological, neuropsychological, and the behavioral/social/emotional impacts on infants, toddlers, and school aged children who were prenatally exposed to opioids with or without the diagnosis of neonatal abstinence syndrome. Providing …

Investigators traditionally use randomized designs and corresponding analysis procedures to make causal inferences about the effects of interventions, assuming independence between an individual's outcome and treatment assignment and the outcomes of other individuals in the study. Often, such independence may not hold. We provide examples of interdependency in model organism studies and human trials and group effects in aging research and then discuss methodologic issues and solutions. We group methodologic issues as they pertain to (1) single-stage individually randomized trials; (2) cluster-randomized controlled trials; (3) pseudo-cluster-randomized trials; (4) individually randomized group treatment; and (5) two-stage randomized designs. Although we present …

Serotonin reuptake inhibitors and receptor agonists are used to treat obesity, anxiety and depression. Here we studied the role of the serotonin 2C receptor (5-HT2CR) in weight regulation and behavior. Using exome sequencing of 2,548 people with severe obesity and 1,117 control individuals without obesity, we identified 13 rare variants in the gene encoding 5-HT2CR (HTR2C) in 19 unrelated people (3 males and 16 females). Eleven variants caused a loss of function in HEK293 cells. All people who carried variants had hyperphagia and some degree of maladaptive behavior. Knock-in male mice harboring a human loss-of-function HTR2C variant developed …
Neurocognitive And Momentary Assessment Of Fear-Based Mechanisms Underlying The Transition From Partial To Full Remission In Anorexia Nervosa., Caroline Christian
Neurocognitive And Momentary Assessment Of Fear-Based Mechanisms Underlying The Transition From Partial To Full Remission In Anorexia Nervosa., Caroline Christian
Electronic Theses and Dissertations
Background. Anorexia nervosa (AN) is a serious psychiatric illness, with high mortality, chronicity, and societal costs. Many individuals with AN that are weight restored do not to fully remit from cognitive-affective symptoms (e.g., fear of weight gain) maintaining the disorder, contributing to the poor outcomes. This critical stage of AN is called partial remission. AN is characterized by high levels of fear and avoidance, which may prevent full remission; however, the role of fear-based mechanisms (i.e., fear approach; avoidance) in partially-remitted AN remains untested. Methods. In the current study (N = 55 partially-remitted AN), participants completed two behavioral paradigms …

Death Of A Bacterium: Exploring The Inhibition Of Staphylococcus Aureus By Burkholderia Cenocepacia., Tiffany Brandt
Death Of A Bacterium: Exploring The Inhibition Of Staphylococcus Aureus By Burkholderia Cenocepacia., Tiffany Brandt
Electronic Theses and Dissertations
Antimicrobial resistance is a phenomenon of increasing concern as antimicrobial overuse and misuse eliminate current therapeutic options, ushering society into a post-antimicrobial era. Antibiotic discovery and synthesis efforts are urgently needed to counter the increasing burden of antimicrobial resistance. Staphylococcus aureus is a causative agent of a variety of clinical manifestations including bacteremia, endocarditis, soft tissue infection, osteomyelitis, and device-related infections. S. aureus infection presents additional treatment challenges due to its capacity for biofilm formation, which is a mode of growth that confers protection from antibiotics and physical elimination, and the emergence of antibiotic resistant strains, including methicillin-resistant S. aureus …
Bayesian Methods For Graphical Models With Neighborhood Selection., Sagnik Bhadury
Bayesian Methods For Graphical Models With Neighborhood Selection., Sagnik Bhadury
Electronic Theses and Dissertations
Graphical models determine associations between variables through the notion of conditional independence. Gaussian graphical models are a widely used class of such models, where the relationships are formalized by non-null entries of the precision matrix. However, in high-dimensional cases, covariance estimates are typically unstable. Moreover, it is natural to expect only a few significant associations to be present in many realistic applications. This necessitates the injection of sparsity techniques into the estimation method. Classical frequentist methods, like GLASSO, use penalization techniques for this purpose. Fully Bayesian methods, on the contrary, are slow because they require iteratively sampling over a quadratic …
A New Direction For Public Understanding Of Science: Toward A Participant-Centered Model Of Science Engagement., Christopher Rickels
A New Direction For Public Understanding Of Science: Toward A Participant-Centered Model Of Science Engagement., Christopher Rickels
Electronic Theses and Dissertations
Engaging the public with science is not an easy task. When presented, scientific findings, public health recommendations, and other scientific information filter through the personal values, beliefs, and biases of members of the public. Science communicators must contend with these differences in order to be effective in cultivating a public understanding of science. Given the importance of scientific understanding for living well in a complex world, increasing science understanding through science engagement is imperative. The field of public engagement with science is dichotomized by a public information deficit approach and a contextualist approach. The deficit approach prizes the factual content …
Metabolic Foundations Of Exercise-Induced Cardiac Growth., Kyle Fulghum
Metabolic Foundations Of Exercise-Induced Cardiac Growth., Kyle Fulghum
Electronic Theses and Dissertations
Regular aerobic exercise promotes physiological cardiac growth, which is an adaptive response thought to enable the heart to meet higher physical demands. Cardiac growth involves coordination of catabolic and anabolic activities to support ATP generation, macromolecule biosynthesis, and myocyte hypertrophy. Although previous studies suggest that exercise-induced reductions in cardiac glycolysis are critical for physiological myocyte hypertrophy, it remains unclear how exercise influences the many interlinked pathways of metabolism that support adaptive remodeling of the heart. In this thesis project, we tested the general hypothesis that aerobic exercise promotes physiological cardiac growth by coordinating myocardial metabolism to promote glucose-supported anabolic pathway …

The Impact Of Pegylation On Cellular Uptake And In Vivo Biodistribution Of Gold Nanoparticle Mri Contrast Agents., Nagwa El-Baz
Electronic Theses and Dissertations
Gold nanoparticles (GNPs) have become a pivotal platform for the delivery of pharmaceutical and diagnostic agents as well as for general therapeutic purposes. Despite their potential for use in biomedicine, their interaction with serum proteins is crucial as it could change their biological profile due to the formation of a protein corona, which can affect their delivery to target tissues in the body. Grafting the GNPs with polyethylene glycol (PEG) is widely used in research to decrease opsonization of the particles by serum proteins and decrease the uptake by the reticuloendothelial system. Here in this dissertation, we have developed a …

Re-Engineering Cardio-Oncology Testing Using Biomimetic Heart Slice Cultures., Jessica Miller
Electronic Theses and Dissertations
28% of drug withdrawal from the market are due to unforeseen disruptions in cardiomyocyte contractility and electrophysiology. The most commonly used platforms for drug testing are in vivo animal models and in vitro cell culture models. While both have been of paramount importance for the discovery and detection of many cardiotoxicities and mechanisms of action, they lack the ability to model an intact human myocardium. This work aims to establish cardiac tissue slices, which are 300-micron thin tissue sections taken from the left ventricular myocardium, as an alternative platform for cardio-oncology studies, specifically cardiotoxicity testing. Additionally, this work aims to …

Theses and Dissertations
Medical nonadherence is a common and formidable problem that many pediatric healthcare providers face when treating children with acute and chronic medical conditions because adhering to a physicians’ treatment recommendations is beneficial for successful treatment, disease prevention, and healthy promotion (Dawood et al., 2010). It was estimated that at least one third of patients are unable to complete or adhere to relatively short treatment regimens (Dawood et al., 2010). Reasons for nonadherence to a physicians’ recommendations range from extended duration of treatment, multiple prescribed medications, and periods of symptom remission (Dawood et al., 2010). Despite a parent’s or family’s reason …
