Cholinergic dysfunction has been implicated in the pathophysiology of psychosis and psychiatric disorders such as schizophrenia, depression, and bipolar disorder. The basal forebrain (BF) cholinergic nuclei, defined as cholinergic cell groups Ch1-3 and Ch4 (Nucleus Basalis of Meynert; NBM), provide extensive cholinergic projections to the rest of the brain. Here, we examined microstructural neuroimaging measures of the cholinergic nuclei in patients with untreated psychosis (~31 weeks of psychosis, <2 defined daily dose of antipsychotics) and used magnetic resonance spectroscopy (MRS) and transcriptomic data to support our findings. We used a cytoarchitectonic atlas of the BF to map the nuclei and obtained measures of myelin (quantitative T1, or qT1 as myelin surrogate) and microstructure (axial diffusion; AxD). In a clinical sample (n = 85; 29 healthy controls, 56 first-episode psychosis), we found significant correlations between qT1 of Ch1-3, left NBM and MRS-based dorsal anterior cingulate choline in healthy controls while this relationship was disrupted in FEP (p > 0.05). Case-control differences in qT1 and AxD were observed in the Ch1-3, with increased qT1 (reflecting reduced myelin content) and AxD (reflecting reduced axonal integrity). We found clinical correlates between left NBM …

The human brain entertains rich spatiotemporal dynamics, which are drastically reconfigured when consciousness is lost due to anaesthesia or disorders of consciousness (DOC). Here, we sought to identify the neurobiological mechanisms that explain how transient pharmacological intervention and chronic neuroanatomical injury can lead to common reconfigurations of neural activity. We developed and systematically perturbed a neurobiologically realistic model of whole-brain haemodynamic signals. By incorporating PET data about the cortical distribution of GABA receptors, our computational model reveals a key role of spatially-specific local inhibition for reproducing the functional MRI activity observed during anaesthesia with the GABA-ergic agent propofol. Additionally, incorporating …

Robust frontoparietal connectivity is a defining feature of primate cortical organization. Whether mammals outside the primate order, such as rodents, possess similar frontoparietal functional connectivity organization is a controversial topic. Previous work has primarily focused on comparing mice and rats to primates. However, as these rodents are nocturnal and terrestrial, they rely much less on visual input than primates. Here, we investigated the functional cortical organization of grey squirrels which are diurnal and arboreal, thereby better resembling primate ecology. We used ultra-high field resting-state fMRI data to compute and compare the functional connectivity patterns of frontal regions in grey squirrels …

Background: Clinical endpoints for upcoming therapeutic trials in frontotemporal dementia (FTD) are increasingly urgent. Cognitive composite scores are often used as endpoints but are lacking in genetic FTD. We aimed to create cognitive composite scores for genetic frontotemporal dementia (FTD) as well as recommendations for recruitment and duration in clinical trial design. Methods: A standardized neuropsychological test battery covering six cognitive domains was completed by 69 C9orf72, 41 GRN, and 28 MAPT mutation carriers with CDR® plus NACC-FTLD ≥ 0.5 and 275 controls. Logistic regression was used to identify the combination of tests that distinguished best between each mutation carrier …

We report results from serologic surveillance for exposure to SARS-CoV-2 among 1,237 wild rodents and small mammals across Europe. All samples were negative, with the possible exception of 1. Despite suspected potential for human-to-rodent spillover, no evidence of widespread SARS-CoV-2 circulation in rodent populations has been reported to date.

Esitämme tulokset serologisesta tutkimuksesta, jossa seulottiin SARS-CoV-2 tartuntojen varalta 1,237 luonnonvaraista jyrsijää ja piennisäkästä eri puolilta Eurooppaa. Kaikki näytteet olivat negatiivisia, yhtä näytettä lukuun ottamatta. SARS-CoV-2:n läikkymisen ihmisistä jyrsijöihin on arveltu olevan mahdollista, mutta todisteet viruksen laajamittaisesta leviämisestä jyrsijäpopulaatioissa puuttuvat.

Blind Spot: Implicit Bias In Health Care And Its Awareness Among Nursing Students At The University Of Southern Mississippi, Debora Skaliks

Doctoral Projects

While prejudice and bias are eminent topics in our society, implicit bias or unconscious bias is not as easily detected. Even though implicit bias may be a familiar concept to many, numerous healthcare workers frequently do not recognize the extent of harm such biases can impose on patients' health and outcomes. The Blind Spot DNP project exposed in this research paper explores the relentless question of the correlation between implicit bias awareness, implicit bias education, and its impact on health care. This Doctor of Nursing Project (DNP) project aimed to analyze whether implicit bias education and testing could increase unconscious …

An Ultrasensitive Bacterial Detection Platform For Culture-Free Diagnosis Of Infections, Xuyang Shi Dec 2022

An Ultrasensitive Bacterial Detection Platform For Culture-Free Diagnosis Of Infections, Xuyang Shi

ETD Archive

The current methods of the diagnosis of bloodstream infections are based on bacterial culture growth, a process that requires considerable time, e.g., 12-16 hours, to obtain a result. This long wait time for the result creates many problems, including the generation of multi-drug resistant organisms (MDROs). At the same time, infected bloodstream usually contains a very low concentration of bacteria, i.e., lower than 5 CFU/mL. The long diagnosis time and the extremely low concentration of bacteria in the infected bloodstream make such infections difficult to diagnose. Here, we demonstrate a culture-free approach for the diagnosis of bloodstream infections using a …

Lipids Affect The Diffusion Of Estradiol In Osteonal Bone, Nicholas Anthony Cimino Dec 2022

Lipids Affect The Diffusion Of Estradiol In Osteonal Bone, Nicholas Anthony Cimino

ETD Archive

In healthy osteonal bone, embedded osteocytes form an intercellular communication network through overlapping cell membrane extensions, possibly defining an overlooked and clinically relevant lipid-mediated transport pathway for nonpolar molecules. Previous techniques evaluating solute transport in cortical bone limit tissue analysis to microscale areas (less than 1 mm2) using tracers and assumptions that diminish clinical relevance, presenting the need for an improved method to evaluate solute diffusion in macroscale areas (greater than 1 mm2) of osteonal bone. A new diffusion system - constructed of glass and polytetrafluoroethylene - was designed and validated for this purpose, exhibiting minimal adsorption of solutes and …

Complex morphological traits are the product of many genes with transient or lasting developmental effects that interact in anatomical context. Mouse models are a key resource for disentangling such effects, because they offer myriad tools for manipulating the genome in a controlled environment. Unfortunately, phenotypic data are often obtained using laboratory-specific protocols, resulting in self-contained datasets that are difficult to relate to one another for larger scale analyses. To enable meta-analyses of morphological variation, particularly in the craniofacial complex and brain, we created MusMorph, a database of standardized mouse morphology data spanning numerous genotypes and developmental stages, including E10.5, E11.5, …

Sleep Medicine Articles

BACKGROUND: Prevention of major depressive disorder (MDD) is a public health priority. Strategies targeting individuals at elevated risk for MDD may guide effective preventive care. Insomnia is a reliable precursor to depression, preceding half of all incident and relapse cases. Thus, insomnia may serve as a useful entry point for preventing MDD. Cognitive-behavioral therapy for insomnia (CBT-I) is recommended as the first-line treatment for insomnia, but widespread implementation is limited by a shortage of trained specialists. Innovative stepped-care approaches rooted in primary care can increase access to CBT-I and reduce rates of MDD.

METHODS/DESIGN: We propose a large-scale stepped-care clinical …

Biology, Monitoring, Sampling, And Management Of Rice Billbug, Sphenophorus Pertinax, In Furrow-Irrigated Rice, Chase Alan Floyd

Graduate Theses and Dissertations

Furrow-irrigated rice hectares have increased annually for the last five years in the predominant long-grain rice producing states. Initial implementation of the system began in the 1980’s, but lack of rice blast (Magnaporthe grisea, Hebert) resistant cultivars made the system a nonviable option for rice producers. Hybridization of rice cultivars with less susceptibility has begun to increase the interest of producers to implement a furrow-irrigated rice production system due to the cost saving opportunities that it may provide. Elimination of the flood from large sections of the rice fields in a furrow-irrigated system alters management strategies and pest complexes compared …

Evaluation Of Biological And Chemical Seed Treatments For Management Of Rice Diseases, Sherif Adam Sharfadine

Graduate Theses and Dissertations

Rice is the primary staple for more than half of the global population and is the second most important cereal worldwide. In the US, rice is primarily grown in the southern states, with Arkansas leading production and responsible for 47% of the total rice production in the country. Diseases cause significant yield losses in rice. Sheath blight, rice blast and bacterial panicle blight are the main diseases in rice and cause significant yield losses. Sheath blight alone could cause up 50% yield loss in heavily damaged fields with highly susceptible cultivars. Additionally, rice seed and seedling diseases primarily caused by …

Flood Hazard And Risk Analyses In The Republic Of Panama: A Case Study From The Juan Diaz River Watershed In Panama City, Virgilio De Jesus Quintero

Graduate Theses and Dissertations

Flooding is the natural hazard with most occurrences in Panama. Its frequency and magnitude have increased over the years. This dissertation analyzes Panama’s flood activity in order to better understand flood hazards, the current evolution of Panamanian perceptions of flood risk, and the incorporation of indigenous knowledge used to mitigate flood hazards. The first chapter developed a baseline of past and current flood inventory in Panama, which required the use of flood historical data, thematic cartography, and Geographic Information Science (GIS). This chapter shows Panama has experienced floods in varying degrees. Through the spatial and temporal distribution of floods from …

Cardiac contractile strength is recognised as being highly pH-sensitive, but less is known about the influence of pH on cardiac gene expression, which may become relevant in response to changes in myocardial metabolism or vascularization during development or disease. We sought evidence for pH-responsive cardiac genes, and a physiological context for this form of transcriptional regulation. pHLIP, a peptide-based reporter of acidity, revealed a non-uniform pH landscape in early-postnatal myocardium, dissipating in later life. pH-responsive differentially expressed genes (pH-DEGs) were identified by transcriptomics of neonatal cardiomyocytes cultured over a range of pH. Enrichment analysis indicated “striated muscle contraction” as a …

How Disgust In Germ Averse Individuals Biases Avoidance Decision-Making, Wesley Stuart, Timothy Schoenfeld Phd Dec 2022

How Disgust In Germ Averse Individuals Biases Avoidance Decision-Making, Wesley Stuart, Timothy Schoenfeld Phd

Science University Research Symposium (SURS)

The present study investigates the relationship between germ aversion and avoidant decision making. Individuals in the general population who rate highly in germ-aversion and disgust sensitivity may act maladaptively in order to avoid potentially anxiety inducing scenarios. If encountered, these scenarios will cause an increase in physiological arousal and a suppression of salivatory cortisol in the individual. We hypothesize that this stress response biases decision making in an avoidant manner. Participants for this study, consisting of 60 undergraduate students at Belmont University, started by taking a perceived vulnerability to disease scale. Then, those in the experimental group performed a task …

(R2022) Mathematical Modelling Of Tuberculosis And Covid-19 Co-Infection In India: A Real Data Analysis On Concomitant Diseases, Vijai Shanker Verma, Harshita Kaushik, Archana Singh Bhadauria

Applications and Applied Mathematics: An International Journal (AAM)

In this paper, we have proposed an epidemiological model to study the dynamics of two concomitant diseases Tuberculosis (TB) and COVID-19. Here, we have formulated a deterministic compartmental model as an extended form of the classical SIS model. First, the basic reproduction number R0 is derived and then stability analysis of the model is done. It is observed that the disease-free equilibrium is stable when R0 is less than one and the endemic equilibrium is stable only when R0 is greater than one. Numerical simulation is carried out to illustrate the theoretical findings and to study the …

The Development Of A Novel Peptide-Drug Conjugate For Treating Triple-Negative Breast Cancer, Phi-Phung Than Dec 2022

The Development Of A Novel Peptide-Drug Conjugate For Treating Triple-Negative Breast Cancer, Phi-Phung Than

Pharmaceutical Sciences (MS) Theses

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with poor prognosis and is difficult to treat. A diagnosis of TNBC means that the three most common types of receptors known to fuel most breast cancer growth–estrogen, progesterone, and HER2– are not present in the cancer tumors. Since the tumor cells lack necessary receptors, common treatments like hormone therapies and targeted HER2 drugs are ineffective. The use of chemotherapies in treating TNBC is an effective option instead. However, chemotherapies are not selective, thus they cause severe side effects such as cardiotoxicity, myelosuppression, alopecia and gastrointestinal problems. A peptide-drug conjugate …

(R1522) Modelling The Influence Of Desertic Aerosols On The Transmission Dynamics Of Neisseria Meningitidis Serogroup A, Francis Signing, Berge Tsanou, Samuel Bowong

Applications and Applied Mathematics: An International Journal (AAM)

This paper assesses the role of desert aerosols and vaccine on the transmission dynamics of Neisseria Meningitis serogroup A (NmA). It is biologically well-documented that the inhalation of aerosol dust and its presence in the nasal cavity weakens the nasopharyngeal mucosa by damaging the mucosal barrier and inhibiting the mucosal immune defenses of susceptible and vaccinated individuals. We address the latter by proposing and analyzing a mathematical model for the dynamics of NmA that specifically accounts for the fast progression of susceptible and vaccinated individuals to the invasive stage of the disease. We compute the basic reproduction number and use …

Environmental Influences On The Growth Of Inland Cisco Populations In Three Minnesota Sentinel Lakes, Edward Carlson Dec 2022

Environmental Influences On The Growth Of Inland Cisco Populations In Three Minnesota Sentinel Lakes, Edward Carlson

Biology Graduate Theses

Cisco Coregonus artedi are a pelagic cold-water fish that are widely distributed throughout many inland lakes across the northern Midwest and play an important role as forage for large piscivores. Inland Cisco populations have been observed to vary in their overall size and recruitment potential based on the system in which they are found. Cisco are sensitive to oxythermal stress and with climate change, land use, and invasive species effects becoming more prevalent in Minnesota, aspects of lake ecosystems are changing, ultimately making Cisco more susceptible to these stressors. To investigate how different factors were affecting the population dynamics of …
