This document analyzes American composer Todd Goodman's 2012 work for solo tuba titled Tuba Concerto. It contains 10 chapters, including one for each of the three movements of the work: an introduction providing biographical information on the composer and context for the work within the late twentieth century and early twenty-first century; a performer’s analysis on the solo tuba part; and the biographical and performance information of composers also performed on the accompanying recital. Increasing mortality from opioid overdose and low treatment engagement are significant public health concerns. Along with increasing health care and criminal justice enforcement costs, there is an urgent need to study the factors associated with treatment retention in opioid use disorder. The study investigated the relative impact of the biomarker cholesterol on treatment retention in an opioid treatment program (OTP) clinic. Further, it examined the medical comorbidities, treatment, and socio-demographic variables that impact opioid use disorder treatment retention. Understanding the pollination requirements of threatened species is critical for the success of long-term conservation strategies. In a series of greenhouse experiments, we assessed the effects of different pollination treatments, including autogamy, facilitated autogamy, and cross-pollination on fruit and seed set for 2 generations of a rare island endemic plant. Santa Cruz Island rockcress, Sibara filifolia (Brassicaceae), is a rare, endangered annual herb that is endemic to the California Channel Islands. While efforts are underway to monitor and conserve this rare plant species, little is known about its pollination biology. We quantified the index of self-compatibility and multiplicative fitness. Hyperphosphorylated microtubule-associated protein tau has been implicated in dementia, epilepsy, and other neurological disorders. In contrast, site-specific phosphorylation of tau at threonine 205 (T205) by the kinase p38γ was shown to disengage tau from toxic pathways, serving a neuroprotective function in Alzheimer's disease. Using a viral-mediated gene delivery approach in different mouse models of epilepsy, we show that p38γ activity-enhancing treatment reduces seizure susceptibility, restores neuronal firing patterns, reduces behavioral deficits, and ameliorates epilepsy-induced deaths. Furthermore, we show that p38γ-mediated phosphorylation of tau at T205 is essential for this protection in epilepsy, as a lack of this critical interaction reinstates …

In the aftermath of World War II, the US was faced with the monumental task of finding and identifying over 405,000 service members who did not survive the conflict (McDermott, 2005, p. 1). Of these 405,000, 81,000 remain missing and 2,498 remain unidentified in cemeteries across Europe alone (American Battle Monuments Commission, 2022). Often, these individuals were interred and disinterred multiple times, crossing the continent in the journey from loss incident or battlefield to their final resting place. Protein design is a complex biomolecular and computational problem. Working on increasingly large protein folding problems requires an improvement in current analysis methods available. This work first discusses various methods of protein design, including de novo protein design, which is the primary focus of this thesis. Then, a new approach utilizing a B+ tree to effectively store and query a graph of keys and vertices is proposed in order to store the number of times two polypeptides are considered to be similar. UNLABELLED: Tumor suppressor mutations in head and neck squamous cell carcinoma (HNSCC) dominate the genomic landscape, hindering the development of effective targeted therapies. Truncating and missense mutations in NOTCH1 are frequent in HNSCC, and inhibition of PI3K can selectively target NOTCH1 mutant (NOTCH1MUT) HNSCC cells. In this study, we identify several proteins that are differentially regulated in HNSCC cells after PI3K inhibition based on NOTCH1MUT status. Neurons harbor high levels of single-strand DNA breaks (SSBs) that are targeted to neuronal enhancers, but the source of this endogenous damage remains unclear. Using two systems of postmitotic lineage specification-induced pluripotent stem cell-derived neurons and transdifferentiated macrophages-we show that thymidine DNA glycosylase (TDG)-driven excision of methylcytosines oxidized with ten-eleven translocation enzymes (TET) is a source of SSBs. Although macrophage differentiation favors short-patch base excision repair to fill in single-nucleotide gaps, neurons also frequently use the long-patch subpathway. Disrupting this gap-filling process using anti-neoplastic cytosine analogs triggers a DNA damage response and neuronal cell death, which is dependent on TDG. …

This cross-sectional investigation of 6,931 U.S. adults examined the relationship between sitting time and insulin resistance. A primary objective was to evaluate how this relationship was mediated by the following variables: age, sex, race, year of assessment, cigarette smoking, physical activity, body mass index (BMI), and waist circumference. Self-reported sitting time, measured in minutes per day, was the exposure variable. Insulin resistance, indexed by the homeostatic model assessment of insulin resistance (HOMA-IR), was the outcome variable. Data were used from the 2011-2018 National Health and Nutrition Examination Survey (NHANES). Results showed a strong, positive association between sitting time and insulin …

UNLABELLED: Clonal hematopoiesis resulting from the enhanced fitness of mutant hematopoietic stem cells (HSC) associates with both favorable and unfavorable health outcomes related to the types of mature mutant blood cells produced, but how this lineage output is regulated is unclear. Using a mouse model of a clonal hematopoiesis-associated mutation, DNMT3AR882/+ (Dnmt3aR878H/+), we found that aging-induced TNFα signaling promoted the selective advantage of mutant HSCs and stimulated the production of mutant B lymphoid cells. The genetic loss of the TNFα receptor TNFR1 ablated the selective advantage of mutant HSCs without altering their lineage output, whereas the loss of TNFR2 resulted …

This research examines how the characteristics of terrorist attacks predict the chance of an attack succeeding, where an attack is defined as successful if the intended attack type is carried out. Data was analyzed across three geographical missions within Operation Enduring Freedom: Trans-Sahara, Horn of Africa, and the Philippines. Using predicted probabilities of success obtained from logistic regression models, the medians were plotted to compare the characteristics of terrorist attacks across missions. While a great deal is known about instructor response to student writing—from commenting practices to student perceptions—less is known about how feedback impacts students’ writing and writerly development. While we set out to study students’ explicit engagement with written instructor feedback, our initial experimental design was disrupted by the COVID-19 pandemic. Accordingly, we describe the dialogic collaborative process that emerged as we considered both the data we were able to collect and, in turn, feedback anew. Description: A weighted vest is a vest that typically has 10% of a person’s body weight evenly distributed around the vest (Olson et al., 2004). A weighted vest can provide sensory input that is believed to alleviate dysregulation, inattentiveness, and stereotypic behaviors (Carter et al., 2009).

Goal: Our community group is specifically targeting children with sensory processing impairments who are attending speech therapy sessions at the RiteCare clinic. Sensory behaviors exhibited by the clients are most commonly shown through dysregulation, outbursts, inattentiveness, and hyperactivity. Despite the importance of high-latitude surface energy budgets (SEBs) for land-climate interactions in the rapidly changing Arctic, uncertainties in their prediction persist. Here, we harmonize SEB observations across a network of vegetated and glaciated sites at circumpolar scale (1994–2021). Our variance-partitioning analysis identifies vegetation type as an important predictor for SEB-components during Arctic summer (June-August), compared to other SEB-drivers including climate, latitude and permafrost characteristics. Differences among vegetation types can be of similar magnitude as between vegetation and glacier surfaces and are especially high for summer sensible and latent heat fluxes. Rising greenhouse gas emissions are causing climate change, and the world’s focus has shifted to the need to reduce our reliance on fossil fuels. There has been a rise in the published literature on the utilization of crops for bioenergy production in Louisiana. In 2020, nearly half of Texas’s 385,000 public high school graduates were unprepared for college-level reading or math. Limited research reveals K-12 faculty perceive limited roles and responsibilities in the college process, relying heavily on guidance counselors and college admissions counselors for preparing students for college and careers. The purpose of this study was to help fill this gap by answering the central research question: How do high school and college-level faculty and staff prepare high school students for college and careers?
