Proper Hedgehog (HH) signaling is essential for embryonic development, while aberrant HH signaling drives pediatric and adult cancers. HH signaling is frequently dysregulated in pancreatic cancer, yet its role remains controversial, with both tumor-promoting and tumor-restraining functions reported. Notably, the GLI family of HH transcription factors (GLI1, GLI2, GLI3), remain largely unexplored in pancreatic cancer. We therefore investigated the individual and combined contributions of GLI1-3 to pancreatic cancer progression. At pre-cancerous stages, fibroblast-specific Gli2/Gli3 deletion decreases immunosuppressive macrophage infiltration and promotes T cell infiltration. Strikingly, combined loss of Gli1/Gli2/Gli3 promotes macrophage infiltration, indicating that subtle changes in Gli expression differentially …

Using Community-Based Participatory Research To Tailor A Diabetes Education Program To An At-Risk Black Church Congregation In The Mid-Atlantic, Samantha A. Mielke

Doctor of Nursing Practice Projects

Background: Prediabetes and diabetes continue to have a high disease burden in the United States (Lin et al., 2018). Many individuals are unaware of their glucose state or the impact impaired glucose can have on their quality adjusted life years and cost of living. The Centers for Disease Control and Prevention (CDC) has underscored the importance of decreasing the incidence of diabetes and has resources available. Still, many communities, particularly minority populations, can benefit from tailoring programs to their specific populations, where typical approaches may not be as beneficial (Joo & Liu, 2020). Additionally, Black American (BA) congregations often have …

Renal Cell Carcinoma Of Pelvic Kidney With Atypical Nodal Metastasis: A Diagnostic Challenge, Roshan-E-Shahid Rana, Muhammad Hammad Ather Jul 2022

Renal Cell Carcinoma Of Pelvic Kidney With Atypical Nodal Metastasis: A Diagnostic Challenge, Roshan-E-Shahid Rana, Muhammad Hammad Ather

Section of Urology

Ectopic pelvic kidney is a known congenital anomaly; however, the presence of renal cell carcinoma (RCC) in an ectopic kidney is rare with the evidence available in the form of a few case reports only. In this case report, we present a case of metastatic RCC in the pelvic kidney which became a diagnostic challenge because of atypical contrast-enhanced computed tomographic (CT) characteristics and unusual pattern of lymph node involvement including cervical lymph node in the absence of visceral metastasis. Because of its unusual location and uncertain vascular anatomy, ectopic kidney poses a surgical challenge. Owing to the rarity of …

Mechanisms Involved In Mitoquinone-Mediated Protection Of H9c2 Cells Against Anti-Cancer Drug Doxorubicin-Induced Cardiotoxicity, Kelly Annie Mercado

PCOM Biomedical Studies Student Scholarship

INTRODUCTION: Due to cardiotoxicity, doxorubicin (DOX), a chemotherapeutic agent, has limited use. Our lab has shown that Mitoquinone (MitoQ) given as a pre-treatment exerted better cardioprotection against DOX-induced cell damage than co-treatment.

OBJECTIVE: This study investigated the underlying mechanisms associated with pre-treating cardiomyocytes with MitoQ and why it demonstrated more potent protection against Doxorubicin-induced cardiotoxicity when compared to co-treatment.

METHODS: Cultured H9c2 cells were dose-dependently treated with DOX alone, MitoQ alone, MitoQ given concurrently with DOX (i.e., co-treatment), or MitoQ given 24 hours prior to DOX (i.e., pre-treatment). Various factors were explored such as cell viability via CCK assay, DOX …

Obstetrics and Gynaecology, East Africa

Background: Maternal psychological stress has been linked to preterm birth. However, the dierential contribution of psychological stress versus stress hormones is not clear. Studies focus primarily on perceived stress and cortisol, with few assessing its inter-convertible hormone cortisone. Furthermore, little is known about the potential moderating roles of obstetric risk and fetal sex in the relationship between maternal stress and gestational length. This gap in knowledge is particularly evident for rural women who typically experience chronic multiple stressors during pregnancy. We explored the relationship of hormonal and psychological stress to gestational length and the eects of obstetric risks and …

Differences In Hiv, Sti And Other Risk Factors Among Younger And Older Male Sex Workers Who Have Sex With Men In Nairobi, Kenya, Nicholas Muraguri, Jerry Okoth Okal, Marleen Temmerman, Helgar Musyoki, Peter Gichangi

Obstetrics and Gynaecology, East Africa

Introduction: Previous surveys of male sex workers (MSW) in sub-Saharan Africa have not fully documented the HIV and sexually transmitted infections (STIs) rates and vulnerabilities by age category.

Methods: The bio-behavioral survey of MSW in Nairobi, Kenya, utilized respondent-driven sampling to recruit MSW. Structured interviews captured MSW's behavioral aspects, and biological tests for HIV and other STIs.

Results: Analysis of the two age categories, 18–24 years (younger MSW) and 25 years and above (older MSW), shows that of all participants, a significantly higher proportion of younger MSW (59.6% crude, 69.6% RDS-adjusted) were recruited compared to older MSW …

Aberrant transcriptional activity of androgen receptor (AR) is one of the dominant mechanisms for developing of castration-resistant prostate cancer (CRPC). Analyzing AR-transcriptional complex related to CRPC is therefore important towards understanding the mechanism of therapy-resistance. While studying its mechanism, we observed that a transmembrane protein called neuropilin-2 (NRP2) plays a contributory role in forming a novel AR-transcriptional complex containing nuclear pore proteins. Using immunogold electron microscopy, high-resolution confocal microscopy, chromatin immunoprecipitation, proteomics, and other biochemical techniques, we delineated the molecular mechanism of how a specific splice variant of NRP2 becomes sumoylated upon ligand stimulation and translocates to the inner nuclear …

Experimental Characterization Of Additively Manufactured Multi-Feather Wingtip Devices, Patricio Garzon Jul 2022

Experimental Characterization Of Additively Manufactured Multi-Feather Wingtip Devices, Patricio Garzon

Doctoral Dissertations and Master's Theses

Soaring birds have evolved to fly for long periods of time without flapping their wings. Inspired by the flight of these birds, the proposed thesis presents an experimental investigation focused on wingtip devices designed based on biomimicry. The overarching engineering objective was to reduce the induced drag as a means to improve the fuel efficiency via these experimental wingtips. An associated secondary objective was to establish a method for manufacturing complex structures suitable for wing tunnel testing. A manufacturing technique that involved using composite weaves to reinforce additively manufactured structures was developed. This technique has the potential to reduce manufacturing …

Gender Norms And Gendered Traits, Rowan Bell Jul 2022

Gender Norms And Gendered Traits, Rowan Bell

Dissertations - ALL

Gender oppression is sustained in part through enforcement of and compliance with gender norms. Understanding how they work is therefore salient to the goal of gender liberation.According to the category-based view, which is common in analytic feminist philosophy, gender norms are assigned to individuals based on their assigned gender category, such as woman or man. I argue that this is insufficient, because it ignores the experiences of those who are marginalized or excluded from those categories. On a category-based view, individual responsiveness to gender norms will track gender category assignment; only individuals assigned the category woman will be responsive to …

Preparation Of Loaded Liposomes By Extrusion And Methods Of Characterization, Maryna Kucheriava, Nicholas Bell, Matt Pitts, Daniel Robertson, Jacob Kammer, Daniel Fologea (Mentor)

Idaho Conference on Undergraduate Research

Adverse drug effects from chemotherapeutic agents are a well-known deterrent and a potential limiting factor for appropriate treatment regimens of tumors. To circumvent these limitations several approaches for precision chemotherapy have been proposed. Among those, liposomes are the most advanced carriers for drugs to tumor sites. Liposomes are spherical lipid structures that mimic the cell membrane, and their ability to protect and transport biologically active molecule is exploited for the purpose of targeted drug delivery. In this respect, our work describes production and characterization of liposomes via extrusion.

We produced liposomes from mixtures of lipids that included Polyethylene glycol (PEG) …

Management Faculty Publications and Presentations

This research aims to examine how the prior experiences of the chief executive officer (CEO) and board influence the focal firm’s Corporate Social Responsibility (CSR) activities. Further, the present study examines how CEO overconfidence influences the diffusion of CSR activities. The authors theorize that overconfident CEOs are influenced more by the corporate strategies they experienced on other boards and less by the corporate strategies experienced by other directors. Through longitudinal analyses of the CSR profiles a sample of S&P 500 companies for the period 2006-2013, the study shows that CEO and board prior CSR experience are positively related to the …

Simulating The Impact Of Crop–Livestock Interaction On Crop Performance Using Dssat, Teerath Rai, Sandeep Kumar, Thandiwe Nleya, Gerrit Hoogenboom

Agronomy, Horticulture and Plant Science Faculty Publications

An integrated crop–livestock system (ICL) can help with enhancing soil quality and crop productivity, ultimately increasing farm income. Field experiments that evalu- ated the impact of cover crops (CCs) and grazing on crop productivity have shown mixed results primarily because of a relatively short duration of the crop–livestock system. Dynamic crop models can help to simulate the long-term impact of soils and crop management on crop yield. The objectives of the current study were to develop a simple simulation methodology for crop–livestock interaction using the Decision Support System for Agrotechnology Transfer (DSSAT) and to evaluate the perfor- mance of CC …

Better Colloidal Lithography: Tilt-Rotate Evaporation Overcomes The Limits Of Plasma Etching, Macayla J. Caso, Michael G. Benton, Kevin M. Mcpeak Jul 2022

Better Colloidal Lithography: Tilt-Rotate Evaporation Overcomes The Limits Of Plasma Etching, Macayla J. Caso, Michael G. Benton, Kevin M. Mcpeak

Faculty Publications

Colloidal lithography (CL) is a promising method for large-area fabrication of nanohole and nanodot arrays with applications in optical biosensing, separations, and magnetic data storage. However, reducing the diameter of the polystyrene sphere mask by plasma etching unavoidably increases their coefficient of variation (CV) and deforms their shape, thereby limiting the pitch-to-hole-diameter ratio of the resulting nanohole array to less than 3:1 and the minimum hole size to 200 nm with a 10% or better CV. We show that tilt-rotate evaporation colloidal lithography (TRE-CL) breaks the trade-off between hole diameter and polydispersity by leveraging glancing angle evaporation, not plasma etching, …

Sanders-Brown Center on Aging Faculty Publications

Approximately 30% of elderly adults are cognitively unimpaired at time of death despite the presence of Alzheimer's disease neuropathology at autopsy. Studying individuals who are resilient to the cognitive consequences of Alzheimer's disease neuropathology may uncover novel therapeutic targets to treat Alzheimer's disease. It is well established that there are sex differences in response to Alzheimer's disease pathology, and growing evidence suggests that genetic factors may contribute to these differences. Taken together, we sought to elucidate sex-specific genetic drivers of resilience.

We extended our recent large scale genomic analysis of resilience in which we harmonized cognitive data across four cohorts …

Molecular Theoretical Model For Lipid Bilayers: Adsorption Of Lipidated Proteins On Lipid Bilayers As A Function Of Bilayer Composition And Curvature, Shauna Celeste Kennard Jul 2022

Molecular Theoretical Model For Lipid Bilayers: Adsorption Of Lipidated Proteins On Lipid Bilayers As A Function Of Bilayer Composition And Curvature, Shauna Celeste Kennard

Theses and Dissertations

Protein localization on biological membranes is motivated by either highly selective recognition of specific target membrane components or nonspecific attraction to general physical properties of the membrane, such as charge, lipid heterogeneity, and curvature. Here we discuss the interaction between lipidated proteins and lipid bilayer membranes from a comprehensive examination of how features of the membrane and its lipid constituents, including lipid composition, headgroup size, degree of tail saturation, tail length (bilayer thickness), and membrane geometry, affect the adsorption ability of the proteins. Of key importance is the strong interconnection among these compositional and morphological elements of the membrane in …

Culturally Tailored Diabetes Self-Management Educational (Dsme) Program And Its Impact On Motivation And Self-Efficacy Among Hispanic Adults With T2dm: A Pilot Study, Andrellie P. Van Wageningen

Doctoral Projects

Type 2 diabetes mellitus in the United States is an epidemic that disproportionately affects Hispanics in the state of California. Many factors have been attributed to this phenomenon because of their unique characteristics. As recent studies have pointed out, their social determinants of health, cultural influences, and genetic predisposition often lead to poor health outcomes and impaired quality of life. A targeted approach is critical in promoting increased diabetes knowledge, enabling skills to master self-care activities, and positive self-care behaviors to avert poor outcomes. The purpose of this doctoral project is to examine whether culturally tailored DSME influences an individual’s …

Project Ankle-Brachial Index: Implementation Of An Evidence-Based Peripheral Artery Disease Protocol, Melaney Banks Jul 2022

Project Ankle-Brachial Index: Implementation Of An Evidence-Based Peripheral Artery Disease Protocol, Melaney Banks

Doctor of Nursing Practice Projects

Background: Peripheral artery disease (PAD) is a narrowing or blocking of the arteries due to plaque buildup, which prevents oxygen, blood, and other nutrients from getting to the legs. When this buildup reaches a certain point, it becomes atherosclerosis, which is more prevalent in a smoker. Smoking intensifies the problem of plaque buildup if an individual already suffers from PAD. When an individual smokes, the nicotine causes the arteries to constrict and narrow, reducing blood flow further. The ankle-brachial index (ABI) is a simple, cost-effective diagnostic test to identify the presence of PAD which will improve patient outcomes.

Purpose: This …
