Coupling 210Pb And 14C To Constrain Carbon Burial Efficiency Of Blue Carbon Ecosystems, Tynisha R. Martin Jul 2022

Coupling 210Pb And 14C To Constrain Carbon Burial Efficiency Of Blue Carbon Ecosystems, Tynisha R. Martin

USF Tampa Graduate Theses and Dissertations

Blue carbon ecosystems cover a small global area but have the potential to sequester large amounts of organic carbon (OC) from the coastal ocean in the sediment. Organic carbon is continually remineralized and exported in the dissolved form, which is currently only poorly accounted for in blue carbon budgets. Constraining carbon cycling in blue carbon systems is complicated by the range of carbon sources and sinks in the system and high export rates from the system. By coupling 14C and 210Pb chronometers to ascertain the amount of primary production stored within peat, it is possible to study carbon transport through …

Physiologically-Based Pharmacokinetics Modeling Of Colloidal Nanoparticles, Anh-Dung Le Jul 2022

Physiologically-Based Pharmacokinetics Modeling Of Colloidal Nanoparticles, Anh-Dung Le

Nanoscience and Microsystems ETDs

Physiologically-based Pharmacokinetics (PBPK) modeling—a mechanistic mathematical tool to predict biodistribution to inform drug development, requires knowledge of the physico-chemical properties of the drug in question as well as the anatomy and physiology of the organism. Unlike with molecular drugs, nanoparticle parameters often need to be estimated by fitting models to time-course data, which can result in large uncertainties in parameter estimates due to a high-dimensional parameter space. Our research addresses this challenge by offering a path to parameter insights using global sensitivity analysis. The first chapter is an overview of the PBPK model structure and mathematical framework used in our …

Hybrid-Control Arm Construction Using Historical Trial Data For An Early-Phase, Randomized Controlled Trial In Metastatic Colorectal Cancer, Chen Li, Katrina S Pedersen, Et Al

2020-Current year OA Pubs

Background: Treatment for metastatic colorectal cancer patients beyond the second line remains challenging, highlighting the need for early phase trials of combination therapies for patients who had disease progression during or following two prior lines of therapy. Leveraging hybrid control design in these trials may preserve the benefits of randomization while strengthening evidence by integrating historical trial data. Few examples have been established to assess the applicability of such design in supporting early phase metastatic colorectal cancer trials.

Methods: MORPHEUS-CRC is an umbrella, multicenter, open-label, phase Ib/II, randomized, controlled trial (NCT03555149), with active experimental arms ongoing. Patients enrolled were assigned …

Center for Medical Ethics and Health Policy Staff Publications

Background: The Childhood Cancer Research Network (CCRN) was established by the Children's Oncology Group (COG) as a resource for epidemiologic studies of childhood cancer. The objective of this study was to evaluate the representativeness of CCRN and identify factors associated with enrollment.

Method: The number of US childhood patients with cancer diagnosed < 20 years of age enrolled in CCRN (2008-2015) was compared to expected counts, calculated from Surveillance, Epidemiology, and End Results incidence rates and US Census population estimates. Observed-to-expected ratios and corresponding 95% confidence intervals (CI) were estimated across sex, race, diagnosis age, calendar year, and cancer diagnosis groups. Multivariable linear regression models were generated to evaluate the association between open COG phase 3 therapeutic trials and CCRN enrollment rates.

Result: The 43,110 cases enrolled in CCRN represented 36% of the expected childhood cancers diagnosed from 2008 to 2015 (N = 120,118). CCRN enrollment ratios [95% CI] were highest among males (0.38 [95% CI, 0.37-0.38]), non-Hispanics (0.35 [95% CI, 0.35-0.36]), and those diagnosed from 1 to 4 years …

Research outputs 2022 to 2026

Circulating tumour cells (CTC) from solid tumours are a prerequisite for metastasis. Isolating CTCs and understanding their biology is essential for developing new clinical tests and precision oncology. Currently, CellSearch is the only FDA (U.S. Food and Drug Administration)-approved method for CTC enrichment but possesses several drawbacks owing to a reliance on the epithelial cell adhesion molecule (EpCAM) and a resource-intensive nature. Addressing these shortcomings, we optimised an existing size-based method, MetaCell, to enrich CTCs from blood of colorectal cancer (CRC) patients. We evaluated the ability of MetaCell to enrich CTCs by spiking blood with CRC cell lines and assessing …

Cellular Mechanisms Underlying State-Dependent Neural Inhibition With Magnetic Stimulation, Hui Ye, Vincent Chiun-Fan Chen, Jenna Hendee Jul 2022

Cellular Mechanisms Underlying State-Dependent Neural Inhibition With Magnetic Stimulation, Hui Ye, Vincent Chiun-Fan Chen, Jenna Hendee

Engineering Science Faculty Publications

Novel stimulation protocols for neuromodulation with magnetic fields are explored in clinical and laboratory settings. Recent evidence suggests that the activation state of the nervous system plays a significant role in the outcome of magnetic stimulation, but the underlying cellular and molecular mechanisms of state-dependency have not been completely investigated. We recently reported that high frequency magnetic stimulation could inhibit neural activity when the neuron was in a low active state. In this paper, we investigate state-dependent neural modulation by applying a magnetic field to single neurons, using the novel micro-coil technology. High frequency magnetic stimulation suppressed single neuron activity …

Functional Morphology And Taphonomy Of Ordovician–Silurian Diploporan Echinoderms, Stephen Ashley Hill Jul 2022

Functional Morphology And Taphonomy Of Ordovician–Silurian Diploporan Echinoderms, Stephen Ashley Hill

USF Tampa Graduate Theses and Dissertations

Diploporan echinoderms were one of the most speciose groups of stemmed echinoderms during the Ordovician. Despite this widespread abundance, the fossil record of diploporan fauna consists primarily of incomplete specimens, with complete specimens being rare. While the theca, or body, is more commonly preserved, the feeding appendages and the attachment structures (i.e. stems or holdfasts) are seldomly preserved. Insight into these features is highly valuable to the understanding the paleoecology, like the feeding and respiratory strategies, of these enigmatic creatures.

One group of diploporan echinoderms which does have an extensive fossil record, is the Holocystites Fauna, primarily known from the …

Alphafold2 And Cryoem: Revisiting Cryoem Modeling In Near-Atomic Resolution Density Maps, Corey F Hryc, Matthew L Baker

Faculty, Staff and Student Publications

With the advent of new artificial intelligence and machine learning algorithms, predictive modeling can, in some cases, produce structures on par with experimental methods. The combination of predictive modeling and experimental structure determination by electron cryomicroscopy (cryoEM) offers a tantalizing approach for producing robust atomic models of macromolecular assemblies. Here, we apply AlphaFold2 to a set of community standard data sets and compare the results with the corresponding reference maps and models. Moreover, we present three unique case studies from previously determined cryoEM density maps of viruses. Our results show that AlphaFold2 can not only produce reasonably accurate models for …

Inflammation is central to several diseases. TLR4 mediates inflammation by recognising and binding to bacterial lipopolysaccharides and interacting with other proteins in the TLR4 signalling pathway. Although there is extensive research on TLR4-mediated inflammation, there are gaps in understanding its mechanisms. Recently, TLR4 co-localised with LPCAT2, a lysophospholipid acetyltransferase. LPCAT2 is already known to influence lipopolysaccharide-induced inflammation; however, the mechanism of LPCAT2 influencing lipopolysaccharide-mediated inflammation is not understood. This study combined computational analysis with biochemical analysis to investigate the influence of LPCAT2 on lysine acetylation in LPS-treated RAW264.7 cells. The results suggest for the first time that LPCAT2 influences lysine …

Use Of Power-To-Gas Technology With An Excess Of Electricity In The Network, N A. Abdivakhidova

Technical science and innovation

Electricity-to-gas conversion is a key area of interest for decarbonizing and increasing the flexibility of energy systems, as it can both absorb renewable electricity during periods of excess supply and provide backup energy during periods of excess demand. By combining the power-to-gas conversion with the natural gas network, the inherent flexibility of the line package can be exploited and some of the power variability can be transferred to the gas network. Also, as long as the gas injected into the gas grid is low-carbon, such as hydrogen from renewables to gas, then the overall greenhouse gas emissions from the gas …

Disaster Resilience Versus Ecological Resilience And The Proposed Second Causeway To South Padre Island, Juliet Vallejo, Katia Sanchez, Evelyn Roozee, Owen Temby

School of Earth, Environmental, & Marine Sciences Faculty Publications

The barrier island of South Padre is located off the coast of Texas’s southern tip in Cameron County and is a popular tourist destination with over 4 million annual visits. The only road access to and from the island is a four-lane causeway, 2.3 miles in length, that routinely experiences heavy traffic. Twenty years ago, a barge crashed into the Queen Isabella Causeway, destroying a portion of the bridge. It quickly became apparent how reliant South Padre Island (SPI) is on the causeway and raised questions regarding its lack of disaster resilience. Local boosters and government responded by proposing and …

Whole-organ mapping was used to study molecular changes in the evolution of bladder cancer from field effects. We identified more than 100 dysregulated pathways, involving immunity, differentiation, and transformation, as initiators of carcinogenesis. Dysregulation of interleukins signified the involvement of inflammation in the incipient phases of the process. An aberrant methylation/expression of multiple HOX genes signified dysregulation of the differentiation program. We identified three types of mutations based on their geographic distribution. The most common were mutations restricted to individual mucosal samples that targeted uroprogenitor cells. Two types of mutations were associated with clonal expansion and involved large areas of …

Kras Mutations As Essential Promoters Of Lymphangiogenesis Via Extracellular Vesicles In Pancreatic Cancer, Radu Pirlog, George A Calin

Faculty, Staff and Student Publications

Kirsten rat sarcoma virus (KRAS) gene mutations are present in more than 90% of pancreatic ductal adenocarcinomas (PDACs). KRASG12D is the most frequent alteration, promoting preneoplastic lesions and associating with a more aggressive phenotype. These tumors possess increased intratumoral lymphatic networks and frequent lymph node (LN) metastases. In this issue of the JCI, Luo, Li, et al. explored the relationship between the presence of the KRASG12D mutation and lymphangiogenesis in PDAC. The authors used in vitro and in vivo models and an elegant mechanistic approach to describe an alternative pathway for lymphangiogenesis promotion. KRASG12D induced SUMOylation of heterogenous nuclear ribonucleoprotein …

Primary graft dysfunction (PGD) is the leading cause of postoperative mortality in lung transplant recipients and the most important risk factor for development of chronic lung allograft dysfunction. The mechanistic basis for the variability in the incidence and severity of PGD between lung transplant recipients is not known. Using a murine orthotopic vascularized lung transplant model, we found that redundant activation of Toll-like receptors 2 and 4 (TLR2 and -4) on nonclassical monocytes activates MyD88, inducing the release of the neutrophil attractant chemokine CXCL2. Deletion of Itgam (encodes CD11b) in nonclassical monocytes enhanced their production of CXCL2 and worsened PGD, …

Un-Earthing Popular Climate Narratives: Maintenance And Prophecy In Settler World, Hannah Oxford

Communication ETDs

Climate change is fundamentally rooted in facts such as rising global temperatures, carbon emissions, losses in biodiversity, etc. However new information about our changing world is translated and incorporated within the narratives we live by and give form to our personal and collective worlds. While climate change is scientific, our mitigation efforts are entirely storied. This thesis looks at popular climate narratives that frames climate change as an issue of Earthly mechanics fixable through innovative technology. The goal of this thesis is to understand the ways in which settler colonialism, as a communicable mechanism of cultural production, organizes this particular …

Despite the significance of groundwater to the hydrological cycle and as a source of potable water, very little information exists on microplastics (MPs) in this environment. In the present study, MPs have been determined in ten well samples obtained from an alluvial aquifer in a semi-arid region (Shiraz, Iran) following filtration, digestion and inspection under a binocular microscope. A total of 96 MPs were identified, and concentrations ranged from 0.1 to 1.3 MP L−1 (mean and median = 0.48 and 0.43 MP L−1, respectively) and exhibited a complex distribution across the area that reflected differences in land use and local …

Recessive variants in GBA1 cause Gaucher disease, a prevalent form of lysosome storage disease. GBA1 encodes a lysosomal enzyme that hydrolyzes glucosylceramide (GlcCer) into glucose and ceramide. Its loss causes lysosomal dysfunction and increased levels of GlcCer. We generated a null allele of the Drosophila ortholog Gba1b by inserting the Gal4 using CRISPR-Cas9. Here, we show that Gba1b is expressed in glia but not in neurons. Glial-specific knockdown recapitulates the defects found in Gba1b mutants, and these can be rescued by glial expression of human GBA1. We show that GlcCer is synthesized upon neuronal activity, and it is transported …

The flocculation behavior of clay minerals in aquatic environments is an important process in estuarine and riverine dynamics, where strong gradients in salinity can locally occur. Various contradicting observations have been reported in the literature on the impact of salt concentration on the settling process of cohesive sediments. To address this issue in a systematic manner, we investigate the settling behavior of clay minerals as a function of the salt concentration of the ambient water. Specifically, we focus on montmorillonite as a prototype clay mineral with a high cation exchange capacity (CEC). To this end, we study suspensions of Wyoming …

Determining The 3d Structure And Dynamics Of Mouse Notch1 Epidermal Growth Factor-Like Repeat 26 Using Nuclear Magnetic Resonance Spectroscopy, Kendra D. Jenkins

LSU Doctoral Dissertations

NOTCH is a transmembrane protein that transmits signals between cells in direct contact and alters gene expression. The signaling pathway is important because it is involved in cell fate determination. The NOTCH1 extracellular domain has 36 epidermal growth factor-like (EGF) repeats that are responsible for ligand binding. Mutations in the EGF-like repeats cause various neurological and developmental disorders. NOTCH1 signaling initiates when DSL ligands bind to EGF11-12, the minimum Ligand Binding Domain (LBD). The Abruptex region of the NOTCH1 protein (EGF24-29) is a highly glycosylated, flexible region with calcium binding sites that regulates NOTCH1 signaling and binding by interacting with …

Natural products play a critical role in the discovery and development of numerous drugs for the treatment of various types of cancer. These phytochemicals have demonstrated anti-carcinogenic properties by interfering with the initiation, development, and progression of cancer through altering various mechanisms such as cellular proliferation, differentiation, apoptosis, angiogenesis, and metastasis. Treating multifactorial diseases, such as cancer with agents targeting a single target, might lead to limited success and, in many cases, unsatisfactory outcomes. Various epidemiological studies have shown that the steady consumption of fruits and vegetables is intensely associated with a reduced risk of cancer. Since ancient period, plants, …

Cervical cancer (CC) is a preventable disease using proven interventions, specifically prophylactic vaccination, pervasive disease screening, and treatment, but it is still the most frequently diagnosed cancer in women worldwide. Patients with advanced or metastatic CC have a very dismal prognosis and current therapeutic options are very limited. Therefore, understanding the mechanism of metastasis and discovering new therapeutic targets are crucial. New sequencing tools have given a full visualization of the human transcriptome’s composition. Non-coding RNAs (NcRNAs) perform various functions in transcriptional, translational, and post-translational processes through their interactions with proteins, RNA, and even DNA. It has been suggested that …

Evidence-informed decision-making is in increasing demand given growing pressures on marine environments. A way to facilitate this is by knowledge exchange among marine scientists and decision-makers. While many barriers are reported in the literature, there are also examples whereby research has successfully informed marine decision-making (i.e., ‘bright-spots’). Here, we identify and analyze 25 bright-spots from a wide range of marine fields, contexts, and locations to provide insights into how to improve knowledge exchange at the interface of marine science and policy. Through qualitative surveys we investigate what initiated the bright-spots, their goals, and approaches to knowledge exchange. We also seek …
