Activation Domains Can Decouple The Mean And Noise Of Gene Expression, Kaiser Loell, Yawei Wu, Max V Staller, Barak Cohen

2020-Current year OA Pubs

Regulatory mechanisms set a gene's average level of expression, but a gene's expression constantly fluctuates around that average. These stochastic fluctuations, or expression noise, play a role in cell-fate transitions, bet hedging in microbes, and the development of chemotherapeutic resistance in cancer. An outstanding question is what regulatory mechanisms contribute to noise. Here, we demonstrate that, for a fixed mean level of expression, strong activation domains (ADs) at low abundance produce high expression noise, while weak ADs at high abundance generate lower expression noise. We conclude that differences in noise can be explained by the interplay between a TF's nuclear …

Floc Size Distributions Of Cohesive Sediment In Homogeneous Isotropic Turbulence, M Yu, X Yu, S Balachandar, Aj Manning

School of Biological and Marine Sciences

Floc size distribution is one of the key parameters to characterize flocculating cohesive sediment. An Eulerian–Lagrangian framework has been implemented to study the flocculation dynamics of cohesive sediments in homogeneous isotropic turbulent flows. Fine cohesive sediment particles are modeled as the dispersed phase by the discrete element method, which tracks the motion of individual particles. An adhesive contact model with rolling friction is applied to simulate the particle–particle interactions. By varying the physicochemical properties (i.e., stickiness and stiffness) of the primary particles, the dependence of the mathematical form of the floc size distribution on sediment properties is investigated. At the …

This initiative examined systematically the extent to which a large set of archival research findings generalizes across contexts. We repeated the key analyses for 29 original strategic management effects in the same context (direct reproduction) as well as in 52 novel time periods and geographies; 45% of the reproductions returned results matching the original reports together with 55% of tests in different spans of years and 40% of tests in novel geographies. Some original findings were associated with multiple new tests. Reproducibility was the best predictor of generalizability—for the findings that proved directly reproducible, 84% emerged in other available time …

Exchange Broadening Underlies The Enhancement Of Ide-Dependent Degradation Of Insulin By Anionic Membranes, Qiuchen Zheng, Bethany Lee, Micheal T. Kebede, Valerie A. Ivancic, Merc M. Kemeh, Henrique Lemos Brito, Donald E. Spratt, Noel D. Lazo

Chemistry

Insulin-degrading enzyme (IDE) is an evolutionarily conserved ubiquitous zinc metalloprotease implicated in the efficient degradation of insulin monomer. However, IDE also degrades monomers of amyloidogenic peptides associated with disease, complicating the development of IDE inhibitors. In this work, we investigated the effects of the lipid composition of membranes on the IDE-dependent degradation of insulin. Kinetic analysis based on chromatography and insulin's helical circular dichroic signal showed that the presence of anionic lipids in membranes enhances IDE's activity toward insulin. Using NMR spectroscopy, we discovered that exchange broadening underlies the enhancement of IDE's activity. These findings, together with the adverse effects …

Magnetically Recoverable Silica-Decorated Ferromagnetic-Nanoceria Nanocatalysts And Their Use With O- And N-Butyloxycarbonylation Reaction Via Solvent-Free Condition, Shripad M Patil, Runjhun Tandon, Nitin Tandon

Faculty, Staff and Student Publications

Silica-decorated ferrite nanoparticles, a new kind, coated with ceric ammonium nitrate (CAN), have been prepared successfully by simple coprecipitation techniques. Powder X-ray diffraction spectroscopy (PXRD), Fourier transform-infrared spectroscopy (FT-IR), field emission-scanning electron microscope (FE-SEM), wavelength-dispersive X-ray spectroscopy (WDX), energy-dispersive spectroscopy (EDS), inductive coupled plasma-optical emission spectroscopy (ICP-OES), and thermogravimetric analysis (TGA) techniques were used to characterize these nanoparticles. The catalysts are further studied for catalytic activity in solvent-free conditions. Importantly, these nanoparticles have been collected from the reaction mixture using an external magnet and recycled up to minimum of 15 cycles with no substantial loss of catalytic characteristics.

The Process Of Establishing A Blooming Chemistry Tool For Use In Undergraduate Chemistry Education And Research, Emryse Geye

Dissertations and Theses

While it is still common in college chemistry to assess student learning and skill with summative assessments, the CER community does not currently have a simple tool to determine and communicate whether an assessment is actually aligned with the outcomes of interest. In particular, as so-called evidence-based teaching practices and active learning strategies gain a foothold in college chemistry classrooms, the ability to communicate whether those (often labor-, cost-, and time-intensive) interventions are not only aligned with course outcomes, but also provide measurable benefit to students becomes more imperative. While college chemistry has made some strides in the area of …

Parochial altruism refers to the propensity to direct prosocial behavior toward members of one's own ingroup to a greater extent than toward those outside one's group. Both theory and empirical research suggest that parochialism may be linked to political ideology, with conservatives more likely than liberals to exhibit ingroup bias in altruistic behavior. The present study, conducted in the United States and Italy, tested this relationship in the context of the COVID-19 pandemic, assessing willingness to contribute money to charities at different levels of inclusiveness—local versus national versus international. Results indicated that conservatives contributed less money overall and were more …

The Link Between Intracellular Calcium Signaling And The Nitric Oxide-Dependent Release Of Chloride In Retinal Amacrine Cells, Li Zhong Jul 2022

The Link Between Intracellular Calcium Signaling And The Nitric Oxide-Dependent Release Of Chloride In Retinal Amacrine Cells, Li Zhong

LSU Doctoral Dissertations

Regulation of cytosolic Cl- is an important determinant in synaptic function. Our lab has discovered that cytosolic Cl- is regulated not only by transport across the plasma membrane but that Cl- can also be released from internal Cl- stores (Hoffpauir et al., 2006) in amacrine cells (ACs). Nitric oxide (NO) is a small molecule neurotransmitter that has been demonstrated to be involved in cytosolic Ca2+ elevations and Cl- regulation via activation of the cystic fibrosis transmembrane regulator (CFTR) (Krishnan et al., 2017). Here, I test the hypothesis that intracellular Ca2+ signaling mediates the …

Epigenetic dysregulation is a universal feature of cancer that results in altered patterns of gene expression that drive malignancy. Brain tumors exhibit subtype-specific epigenetic alterations; however, the molecular mechanisms responsible for these diverse epigenetic states remain unclear. Here, we show that the developmental transcription factor Sox9 differentially regulates epigenomic states in high-grade glioma (HGG) and ependymoma (EPN). Using our autochthonous mouse models, we found that Sox9 suppresses HGG growth and expands associated H3K27ac states, while promoting ZFTA-RELA (ZR

A universal oncogenic driver of basal-like breast cancer (BLBC) has resisted identification. We show that continuous transcriptional coactivator with PDZ-binding motif (TAZ) activation in precancerous murine luminal cells generates luminal cancers that later become BLBCs. Subsequent TP53 alteration, a feature of invasive human BLBCs, accelerates tumor progression. Because BLBC development is inhibited by TAZ inactivation in vivo, our work provides a sound rationale for targeting Hippo-TAZ signaling as therapy for human BLBC. Our mouse model of BLBC represents a powerful tool for evaluating such drugs.

Changes in the physiological health of species are an essential indicator of changing conditions and environmental challenges. Reponses to environmental challenges can often induce stress, influence physiology, and change metabolism in organisms. Here we tested blood chemistry parameters indicative of stress and metabolic activity using an i-STAT point-of-care blood analyzer in seven populations of free-ranging rock iguanas exposed to varying levels of tourism and supplemental feeding. We found significant differences in blood chemistry (glucose, oxygen, carbon dioxide, hematocrit, hemoglobin, calcium, potassium, and biliverdin levels) among populations exposed to varying levels of tourism, and also some variation between sexes and reproductive …

War Is The Storm—Clausewitz, Chaos, And Complex War Studies, B. A. Friedman

Naval War College Review

Complexity theories indeed do apply to war, and Clausewitz’s theories were the first to grapple with them. War is nonlinear, but the parallels between war and complexity science, and between complexity science and Clausewitz, do not stop with nonlinearity. Clausewitz’s theory of war does not just allude to complexity; rather, complexity is at its very core.

Unravelling Inventorship, Toshiko Takenaka Jul 2022

Unravelling Inventorship, Toshiko Takenaka

Chicago-Kent Journal of Intellectual Property

Inventorship, who made an invention, is one of the most important concepts under the U.S. patent system. Incorrect inventorship determinations result in patent invalidity not only because U.S. Constitution requires granting patents to true inventors, but also first-inventor- to-file novelty inherited many aspects of first-to-invent novelty which depended on inventorship whether to include prior inventions as prior art. Correcting inventorship may result in sharing patent exclusivity with competitors, which forfeits profits necessary to recover expensive development costs. However, the standard to determine inventorship has been called muddy by judges and commentators because neither the Patent Act nor case law provide …

Ethanol-Induced Impacts On Nervous System Development And Dopamine Transporter Characterization In Schmidtea Mediterranea Planaria, Brennen Covely

Theses and Dissertations

Fetal alcohol spectrum disorders (FASD) are diagnosed in 2-5% of newborns, but the biology underlying FASD is poorly understood and challenging to study with existing model organisms. A new model for FASD research is Schmidtea mediterranea (Smed) planaria. Smed have a remarkable ability to regenerate their central nervous system (CNS), and possess a well-studied, simple genome. Previous studies have shown that ethanol exposure delays this regeneration, yet this relationship is not fully understood. Here, we show that alcohol exposure affects Smed in a dose-dependent manner, eliciting characteristic withdrawal-like behaviors and impacting cognitive function. Interestingly, prior exposure does not alter subsequent …
