The “magic spot” alarmones (pp)pGpp, previously implicated in Clostridioides difficile antibiotic survival, are synthesized by the RelA-SpoT homolog (RSH) of C. difficile (RSHCd) and RelQCd. These enzymes are transcriptionally activated by diverse environmental stresses. RSHCd has previously been reported to synthesize ppGpp, but in this study, we found that both clostridial enzymes exclusively synthesize pGpp. While direct synthesis of pGpp from a GMP substrate, and (p)ppGpp hydrolysis into pGpp by NUDIX hydrolases, have previously been reported, there is no precedent for a bacterium synthesizing pGpp exclusively. Nonprofits are a growing part of America’s economy and play an import role in fulfilling the needs of society in addition to private firms and government agencies. Economists haven’t researched nonprofits as much as other firms in the past, but this is starting to change. This project looks at the efficiency of 20 food banks in the United States using Data Envelopment Analysis. The results show that American Food Banks appear to be using their funds efficiently, but others could learn to use their inputs and outputs in a more efficient manner. Environmental factors such as temperature are substantial determinants of the spongy moth, L. dispar, distribution, reproduction, and growth. Accumulating energy reserves at the larval stage is particularly important to L. dispar, since the larvae metamorphose into a fully-grown, non-feeding adult. As non-feeding adults, the energy balance of the pupae must be adequate, to ensure enough energy reserves for adult dispersal, egg maturation and overall persistence of the species in the region. The aim of the present study is to examine the mediating effect of social support on the relationship between internal locus of control (LOC) and engagement in activity, treating general physical activity and deliberate exercise as specific and separate domains. Two models will be investigated: a domain-general model and an exercise-specific model, the former examining the mediating effect of general social support on the relationship between health locus of control and general physical activity and the latter examining the mediating effect of exercise-specific social support on the relationship between exercise LOC and deliberate exercise engagement. The current study examined pre-service teachers’ knowledge of behavior management strategies, early intervention, and developmental delay. Data were collected from 107 participants. Thirty-two were pre-service early education teachers currently enrolled in educator prep courses and 75 were in-service early education teachers. Participants completed an online questionnaire that included the following topics: problem behaviors, evidence-based practices, preschool expulsion, and educator prep courses. In-service teachers were better than pre-service teachers at identifying the top three commonly observed preschool student problem behaviors. Purpose: Lasers have been used for treatment of dentinal hypersensitivity and for various bacterial reduction indications in periodontology. Their effectiveness in killing oral bacteria is not well known. The compounding effect of the combination of a laser treatment and adjunct antimicrobial agent use on bacterial viability is still evolving. The purpose of this in vitro study was to evaluate the effect of three lasers commonly used in dentistry in conjunction with chlorhexidine (CHX), hydrogen peroxide (H2O2), sodium hypochlorite (NaOCl), or sodium fluoride (NaF) on viability of oral bacteria associated with root caries.

This dissertation study examined the impacts of the COVID-19 Pandemic on the K-12 School Principalship and attempted to forecast the future of the principalship post-Pandemic. Using a forecasting method known as the Delphi Technique, I solicited public K-12 principals working in the Commonwealth of Virginia with at least 3 years of administrative experience to serve as participants. In total, 26 practicing principals completed three questionnaires offering their expert opinions on pre-Pandemic roles and responsibilities of the school principal and how those roles were impacted throughout the Pandemic. Previous literature documents caregivers’ difficulties navigating the Neonatal Intensive Care Unit, showing that this problem has persisted for close to 20 years. Tummy time or awake, supervised, prone play is an important developmental play position in infancy. Engaging in more than 15 minutes of tummy time per day is associated with a lower risk of plagiocephaly, lower body mass index, and achievement of higher motor scores in full-term and at-risk preterm infants. However, there are significant gaps in the measurement of tummy time in current literature. Understanding the characteristics of research participants who provide DNA is crucial to ensuring sample representativeness and generalizability of findings of genetics studies of substance use disorders. Using anonymous survey data, the present study had a unique opportunity to compare participants who provided DNA (n=2,414) with those that did not provide DNA (n=1,515). The present study found numerous differences between groups across demographic, substance use, and personality domains. Using multivariate regression, the most parsimonious model found that being male, a non-smoker, and endorsing perseverance was associated with providing DNA. Binge drinking is the most common form of excessive alcohol use. Repeated episodes of binge drinking cause multiple organ injuries, including liver damage. We previously demonstrated that chronic ethanol administration causes a decline in the intrahepatic ratio of S-adenosylmethionine (SAM) to S-adenosylhomocysteine (SAH). This decline causes impairments in essential methylation reactions that result in alcohol-induced fatty liver (steatosis) and other features of alcohol-associated liver disease (ALD). Co-treatment with betaine during chronic ethanol feeding, normalizes hepatocellular SAM:SAH ratio and alleviates many features of liver damage including steatosis. This Essay explores how menopausal bodies are managed and monitored through both menopausal hormone therapy (MHT) and the burgeoning market for technology-driven menopause products and services. While each of these allegedly improves the menopause experience, a closer investigation reveals a more complex interaction of profit motives and traditional notions of gender identity. The Essay identifies problems with—and suggests some solutions for reforming—current practices of monitoring and managing the menopausal body.
Careful consideration of menopause brings this Essay into ongoing conversations about theorizing beyond the gender binary and stereotypical notions of femininity. Purveyors of both MHT and menopause-related digital products and …

Background: Mothers’ perceptions of their children’s weight remain mostly unknown in oil-rich countries of the Middle East.
Aims: To compare maternal perceptions of their children’s weight with actual body weight.
Methods: We used the Kuwait nutritional surveillance data for 5 consecutive years (2015-2019) (N = 5119), which were collected through face-to-face interviews with mothers of children aged 2-5 years attending vaccination centres.
Results: Of the 5119 mothers in the study, 163 (3.18%) and 332 (6.48%) had obese or overweight children, respectively. Among 4624 mothers who had normal weight children, 1350 (29.20%) perceived their children’s weight as …

Objective To examine how living arrangement as a social contextual factor can affect Chinese elders’ cognitive function.
Setting and participants Our sample consists of 2486 Chinese elders from two waves (2014 and 2018) of the Chinese Longitudinal Healthy Longevity Survey (CLHLS) that was administered in 22 of China’s 31 provinces using a multi-stage, disproportionate, purposive random sampling method. The CLHLS aims to better understand the determinants of healthy longevity in China and collects extensive data on a large population of fragile elders aged 80–112 in China.

The concept of the “family farm” holds powerful sway within the American narrative, embodying both nostalgia for an imagined past and anxiety for a future perceived to be under threat. Since the founding of the United States, this cultural ideal has been invoked in support of a rosy vision of agrarian democracy while obscuring the ways in which the U.S. Department of Agriculture’s codified definition of “family farm” has unfairly aggregated advantages for the benefit of a particular kind of family (nuclear) and farmer (white, male, straight). Background: There is a paucity of data on the secular trends of stunting and overweight among children aged < 5 years in oil-rich countries in the Middle East.
Aims: To examine the secular trends of stunting, underweight and overweight in children aged < 5 years in Kuwait between 2007 and 2019.
Methods: We used large individual data records (n=48 108) from the Kuwait Nutritional Surveillance System (KNSS) to calculate height/length-for-age z score (HAZ), weight-for-age z score and body mass index (BMI)-for-age z score using World Health Organization growth references. The assessment of patients presenting with chest pain or symptoms indicative of cardiac ischemia remains a diagnostic challenge. Many types of research have focused on the search for ideal biological markers for the rapid detection of cardiac cell injuries. Markers of inflammation and oxidative stress are the way forward. At present, the biomarker most widely used for diagnosing acute coronary syndrome is cardiac troponin though it has some limitations. Within-subject training models, whereby researchers apply an exercise condition to one limb, and a separate exercise condition to the opposing limb, have become routine amongst the exercise literature. However, no study has directly tested whether exercising one limb with a high-load condition will influence strength adaptations within the opposing limb, even when the opposite limb is training. Furthermore, muscle post-activation performance enhancement (PAPE) is representing local level changes, however, more research is warranted to understand if this can be differentially impacted with different types (e.g. low load vs. high load) of resistance training. One of the most pressing concerns in mental healthcare is the limited use of mental health services across the country. Research has indicated that individuals from racial/ethnic minority groups are significantly less likely to seek mental health services, which has led many to examine specific barriers to treatment-seeking. Given research that has suggested that barriers to treatment-seeking may be structural, attitudinal, or cultural in nature, this study aimed to gain a better understanding of the relationship between these barriers and willingness to seek psychological help. Bigheaded carp have spread rampantly throughout the Mississippi watershed and continue to spread by navigating through rivers, locks, and dams. The long-term effects these species will have on our ecosystems and natural resources is unknown. Their movements, behavior, and general seasonal patterns are well studied within rivers. However, their movements and behavior within reservoirs is poorly understood due to their initial numbers within reservoirs being lower than in rivers. Objective: Vanishing white matter (VWM) is a genetic brain white matter disorder caused by mutations in eIF2B. eIF2B is central in the integrated stress response (ISR), during which its activity is inhibited by various cellular stresses. VWM is a chronic progressive disease with episodes of rapid neurological deterioration provoked by stresses. VWM patients and VWM mouse models show ISR deregulation in brain, correlating with chronic disease development. ISR inhibition ameliorates the chronic disease in VWM mice. The subacute deteriorations have not been modeled yet. We hypothesized that ISR activation could worsen disease progression in mice and model the episodic neurological …
