A Novel Computational Network Methodology For Discovery Of Biomarkers And Therapeutic Targets, Qing Ye
Graduate Theses, Dissertations, and Problem Reports (ETD)
Lung cancer has the second highest cancer incidence rate and the top cancer-related mortality worldwide. An estimate from the American Cancer Society shows that, in 2022, there will be about 236,740 lung cancer cases (117,910 men and 118,830 women) in the US. To date, there are no prognostic/predictive biomarkers to select chemotherapy, immunotherapy, and radiotherapy in individual non-small cell lung cancer (NSCLC) patients. There is an unmet clinical need to identify patients with early-stage NSCLC who are likely to develop recurrence and to predict their therapeutic responses. This dissertation developed a novel computational methodology for modeling molecular gene association networks …

Emergency medical technicians (EMT) and firefighters respond to critical incidents and provide emergent care. As the opioid crisis continues and drug overdose rates increase, first responders administer first aid to substance users, including Naloxone, more frequently. This study examined the influence of repeated exposure to drug-related critical incidents and provision of care to substance users on EMTs' and firefighters' secondary traumatic stress (STS) level, world assumptions, and substance use stigma. First responders' world assumptions, STS, burnout, and exposure to drug-related overdoses were analyzed for their power in predicting substance use stigma. Results suggest that occupational burnout, rather than STS, is …
Influences Of Disrupted Circadian Rhythms On Stroke Outcome, Jennifer A. Liu
Influences Of Disrupted Circadian Rhythms On Stroke Outcome, Jennifer A. Liu
Graduate Theses, Dissertations, and Problem Reports (ETD)
The circadian system is composed of a subset of temporal oscillators that function through a transcriptional and post-translational molecular and functional negative feedback loop cycling approximately every 24 hours. The central clock located in the suprachiasmatic nucleus is responsible for entrainment using light as the key timekeeper (zeitgeber); it is responsible for synchronizing and optimizing physiological behavior and function to the environment. Exogenous information, such as day length and light-dark cycles, provide critical temporal cues for adjusting to environmental conditions. Proper alignment to natural light dark cycles and circadian rhythms is optimal for vital health, fitness, and survival in organisms. …
Aging, A Pathological Factor In Neurological Injury, Aruvi Vijikumar
Aging, A Pathological Factor In Neurological Injury, Aruvi Vijikumar
Graduate Theses, Dissertations, and Problem Reports (ETD)
One of the main reasons for CNS drugs to fail in clinical development is not considering age as a risk factor while studying chronic age-related neurological/neurodegenerative diseases in preclinical studies. We first set out to gain a comprehensive understanding of the impact of age on various aspects (anatomical, immunological, and biochemical) in rodents that play a key role in determining the onset, progression, and evolution of disease severity. With advancing age, the vascular structure and function are compromised which is hypothesized to accelerate cognitive decline. The initial step toward developing novel therapeutics is to characterize the age-related vascular modifications. Utilizing …

Graduate Theses, Dissertations, and Problem Reports (ETD)
In recent years the mass spectrometry (MS) area of field and/or direct analysis has grown dramatically. As a result, field-portable and miniaturized mass spectrometers, introduced only a few years ago, are proliferating. A highly desired feature for field-portable MS, or in-field analysis, is the ability to use ionization techniques requiring very little sample preparation as well as an ability to generate the ions under ambient conditions. Recently, a new ambient ionization technique termed vibrating sharp-edge spray ionization (VSSI) has been introduced which overcomes the field-portable limitations of other spray-based methods including the requirements for auxiliary components (e.g., nebulizing gas and …
Implementation Of Stress Management And Resiliency Training To Address Nursing Burnout In A Neuroscience Critical Care Unit, Jessica May Marie Hughes
Implementation Of Stress Management And Resiliency Training To Address Nursing Burnout In A Neuroscience Critical Care Unit, Jessica May Marie Hughes
Graduate Theses, Dissertations, and Problem Reports (ETD)
In the United States, over one-third of nurses experience symptoms of burnout (Reith, 2018). If left unchecked, poor stress management and resiliency skills can contribute to nursing burnout, leading to costly employee turnover. Stress management and resiliency training (SMART) provides the tools necessary to develop resiliency, improve stress management and decrease burnout (Resilient Option, 2020). Nursing staff under the age of 36, who work in a Neurosciences Critical Care Unit (NCCU) demonstrate lower resiliency scores and a higher risk of burnout. This project aimed to evaluate: (a) the effects of a SMART program on the wellbeing of nursing staff in …
Standardized Postoperative Handoff Tool To Improve Handoff Quality, Haley Durkacz
Standardized Postoperative Handoff Tool To Improve Handoff Quality, Haley Durkacz
Graduate Theses, Dissertations, and Problem Reports (ETD)
Abstract
Effective communication between healthcare providers is an essential element in the delivery of safe patient care. Loss of relevant information can occur during all phases of patient care but is most inclined to occur during the handoff from the operating room to the postoperative anesthesia care unit (PACU). Evidence has indicated that the transfer of patients by anesthesia providers remains informal and brief despite practice guidelines. When pertinent details are omitted during the anesthesia handoff there is a potential increase for medical errors, delayed treatment, and patient harm. The purpose of this project was to analyze the use of …
A Quality Improvement Initiative Regarding Ondansetron In The Prevention Of Spinal Anesthesia-Induced Hypotension, Kassidy Dell Nutter
A Quality Improvement Initiative Regarding Ondansetron In The Prevention Of Spinal Anesthesia-Induced Hypotension, Kassidy Dell Nutter
Graduate Theses, Dissertations, and Problem Reports (ETD)
Spinal anesthesia is an excellent choice as the primary anesthetic for lower abdominal, perineal, and lower extremity procedures. Spinal anesthesia boasts several distinct advantages over general anesthesia. However, it is important to note that spinal anesthesia does not come without risk. The most common adverse reaction of spinal anesthesia is hypotension. Anesthesia providers use several methods to combat the hypotension that is so commonly associated with spinal anesthesia. One emerging trend to prevent spinal anesthesia-induced hypotension (SAIH) is the administration of ondansetron, a serotonin 5-hydroxytryptamine3 antagonist. Evidence has shown that the administration of ondansetron just prior to spinal anesthesia administration …

Mechanical Engineering and Applied Mechanics
This work looks to provide a review of modern stents by evaluating the need for them as well as providing a detailed engineering analysis of the conditions they are subjected to. It is estimated that over two million of these devices are implanted in patients annually. Of those implanted, a majority are used in the treatment of arteriosclerosis. This disease causes arterial walls to thicken and toughen leading to restricted blood flow and in extreme cases, complete loss of circulation. Arteriosclerosis and complications arising from it form a leading cause of death in the developed world. Although there are a …
An Experimental Study Of Fluid-Structure Interaction In Collapsible Vessels, Jennifer Schmeling
An Experimental Study Of Fluid-Structure Interaction In Collapsible Vessels, Jennifer Schmeling
Mechanical Engineering and Applied Mechanics
The fluid-structure interaction in collapsible thin-walled vessels is an important topic of research to better understand the physical mechanisms behind many physiological processes and diseases. In this work, we established an experimental setup to study the collapsible tube deformation and fluid-structure interaction within a thin-walled collapsible vessel. The effects of transmural pressure and flow rate were characterized experimentally using high-frequency pressure transducers and optical measurements. Various transmural pressures were also simulated through finite element analysis. Results suggest that the deformation of thin-wall vessel follows the pattern described by Shapiro’s tube law. The collapsed pattern and cross-sectional area changes with different …

Low- And High-Wattage Vaping During Pregnancy Influence Aortic Structure And Function In Rat Offspring, James Ian Frazier
Graduate Theses, Dissertations, and Problem Reports (ETD)
Introduction: Electronic cigarettes (e-cigs) are being marketed as a healthier alternative to traditional cigarettes, yet the health consequences of e-cig usage (also called vaping) are still being investigated. E-cigs are easily customized for user preferences. For example, the choice of flavors, nicotine concentration, and/or the temperature (i.e., wattage) used to heat the e-liquid can be selected in many devices. Emerging evidence suggests that vaping during pregnancy impairs cerebral vascular function in rodent offspring. In this study, we sought to investigate the potential effects of changes to e-cig device wattage (i.e., 5W vs. 30W) with maternal vaping during pregnancy on vascular …

Graduate Theses, Dissertations, and Problem Reports (ETD)
The specificity and efficiency with which enzymes catalyze selective chemical reactions far exceeds the performance of traditional heterogeneous catalysts that are predominant in industrial applications such as conversion of commodity chemicals to value-added products, fuel cells, and petroleum refinement. Moreover, biocatalysts exhibit exceptionally high product turnover at ambient conditions with little health and environmental burden. These advantageous qualities have led to the prolific use of enzyme catalysis in pharmaceutical, detergents, and food preservation industries wherein their use has greatly reduced waste generation, Unfortunately, the full slate of benefits that enzymes can impart to a broader range of chemical processes is …
A Domain Adaptation Approach For Segmenting Cell Instances In Microscopy Data, Matthew R. Keaton
A Domain Adaptation Approach For Segmenting Cell Instances In Microscopy Data, Matthew R. Keaton
Graduate Theses, Dissertations, and Problem Reports (ETD)
Automated cellular instance segmentation is a process that has been utilized for accelerating biological research since before the deep learning era, and recent advancements have produced higher quality results with less effort from the biologist. Most current endeavors focus on completely cutting the researcher out of the picture by generating highly generalized models. However, these models invariably fail when faced with novel data and effectively opt to miss out on the full capabilities of deep learning in pursuit of this goal. In our work, we demonstrate how, with even a minimal amount of annotated data, dominant approaches in this space …
Development Of An Entry-Scale Modeling Methodology To Provide Ground Reaction Curves For Longwall Gateroad Support Evaluation, Haochen Zhao
Development Of An Entry-Scale Modeling Methodology To Provide Ground Reaction Curves For Longwall Gateroad Support Evaluation, Haochen Zhao
Graduate Theses, Dissertations, and Problem Reports (ETD)
Longwall gateroad entries are generally subjected to severe mining-induced stress changes and deformations; they are prone to large roof falls, floor heaves, and rib sloughing. The standing support system in underground mines needs to be carefully designed to compensate for the mining-induced deformation of the surrounding rock mass and consecutively protect the integrity of gateroad entries. Local geology, together with mining-induced stress redistribution, influences the ground response and plays a major role in the performance of a standing support design. In the US, Support Technology Optimization Program (STOP) and its Ground Reaction Curve (GRC) design criteria have been applied to …

Graduate Theses, Dissertations, and Problem Reports (ETD)
Freshwater use for power generation represents the second-largest water use globally. In the United States, freshwater withdrawals for thermoelectric power accounted for 38% of the total freshwater withdrawals for all uses in 2010. Cooling systems are the most water-intensive part of the thermoelectric generation process. For instance, one 300 MW power generator required about 20,000 m3/h circulating cooling water. The cooling operation discharges a large volume of wastewater in the form of blowdown water (10-20% of the consumed water) that requires treatment for reuse or surface discharge. Produced water (PW), the fluid which returns to the surface from …
