Maintenance of proper blood glucose homeostasis is governed by the endocrine cells of the pancreas. Dysregulation of blood glucose is characteristic of the disease, diabetes mellitus. Relatively little is known about the molecular mechanisms that underlie diabetes. The genes associated with more common forms of diabetes remain more enigmatic and likely have less direct roles in blood glucose homeostasis such as glucose sensing, insulin secretion, and β cell maintenance. GLIS3, a transcription factor involved in pancreatic development as well as the transcriptional regulation of insulin has been identified as a risk locus for type 1 and type 2 diabetes …

In the spring of 2020, the COVID-19 pandemic brought about a multitude of challenges around the world. Mass closures of public places seemed to happen overnight and instantaneously. Essential workers kept the world afloat to the best of their abilities, but the harsh reality quickly settled in that education was not considered essential in the view of a worldwide pandemic. Identifying students with literacy disabilities, such as dyslexia, efficiently in public schools is crucial for early intervention. Literacy and musical skills share common neurological pathways, so music educators possess a skill set that could be advantageous in screening students. This study utilized a descriptive research design to identify music educators’ roles in the identification of students with dyslexia/reading disabilities in Kentucky’s public schools, and their attitudes about those roles. Data was collected through a survey completed by Kentucky public school music teachers. Results indicated that 92% of respondents had not participated in the screening process for children with literacy disorders/dyslexia …

College students in the United States face unique health risks such as problematic alcohol use, poor nutrition, low sleep quality, and low rates of exercise. Research increasingly shows interventions utilizing individualized approaches lead to longer term pro-health behavior change. Values-centered Acceptance and Commitment Therapy (ACT) is a useful framework to for development of personalized, effective interventions. One reason for this may be that impersonalized interventions and researcher-generated target behaviors common in other literatures may not hold the same evocative effect and connection to health behaviors for each or every participant. This phenomenological research study explored industry certifications in career and technical education. A grand tour question and three research questions guided this study. Data collected from interviews with graduate of career pathway programs, career and technical education instructors, and industry employers provided a thorough understanding of the issues experienced in career and technical education. This study revealed a need for appropriate industry certifications in career and technical education programs and the workplace. In addition, the participants expressed a desire for students to start preparing earlier for their careers and be provided with alternatives to college for post-secondary education.

Service animals and emotional support animals are gaining a greater presence in many public spaces. The purpose of the following study is to explore attitudes towards service animals and emotional support animals in a more rigorous fashion than previous research and to explore possible predictors of people’s attitudes towards service animals and emotional support animals. Specifically, this study examined the following predictors: public and self-stigma against mental health, ableism, and attitudes towards pets or pet ownership. The Personality Assessment Inventory (PAI) is a well-known self-report measure that aids in treatment planning and evaluation outcomes. The PAI short-form (PAI-SF) consists of the first 160 items from the full-form’s 344 items. The current study examined extra-test relationships/ correlates of the PAI-SF interpersonal scales dominance (DOM) and warmth (WRM), as well as internal correlations between the treatment rejection scale (RXR) and the warmth scale in a forensic sample. Data were collected from archival evaluations, from a private practice, of adults ages 18-69 years old. Bigheaded carp have spread rampantly throughout the Mississippi watershed and continue to spread by navigating through rivers, locks, and dams. The long-term effects these species will have on our ecosystems and natural resources is unknown. Their movements, behavior, and general seasonal patterns are well studied within rivers. The ENIGMA group on Generalized Anxiety Disorder (ENIGMA-Anxiety/GAD) is part of a broader effort to investigate anxiety disorders using imaging and genetic data across multiple sites worldwide. The group is actively conducting a mega-analysis of a large number of brain structural scans. In this process, the group was confronted with many methodological challenges related to study planning and implementation, between-country transfer of subject-level data, quality control of a considerable amount of imaging data, and choices related to statistical methods and efficient use of resources. The goal of the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Stroke Recovery working group is to understand brain and behavior relationships using well-powered meta- and mega-analytic approaches. ENIGMA Stroke Recovery has data from over 2,100 stroke patients collected across 39 research studies and 10 countries around the world, comprising the largest multisite retrospective stroke data collaboration to date. Growth faltering under 5 years of age is unacceptably high worldwide, and even more children, while not stunted, fail to reach their growth potential. The time between conception and 2 years of age is critical for development. Development of evidence-based practice requires practice-based evidence, which can be acquired through analysis of real-world data from electronic health records (EHRs). The EHR contains volumes of information about patients-physical measurements, diagnoses, exposures, and markers of health behavior-that can be used to create algorithms for risk stratification or to gain insight into associations between exposures, interventions, and outcomes. The Philadelphia (Ph) chromosome was the first translocation identified in leukemia. It is supposed to be generated by aberrant ligation between two DNA double-strand breaks (DSBs) at the BCR gene located on chromosome 9q34 and the ABL1 gene located on chromosome 22q11. Thus, mimicking the initiation process of translocation, we induced CRISPR/Cas9-mediated DSBs simultaneously at the breakpoints of the BCR and ABL1 genes in a granulocyte-macrophage colony-stimulating factor (GM-CSF) dependent human leukemia cell line. With the increasing number of sequencing projects involving families, quality control tools optimized for family genome sequencing are needed. However, accurately quantifying contamination in a DNA mixture is particularly difficult when genetically related family members are the sources. We developed TrioMix, a maximum likelihood estimation (MLE) framework based on Mendel's law of inheritance, to quantify DNA mixture between family members in genome sequencing data of parent-offspring trios. TrioMix can accurately deconvolute any intrafamilial DNA contamination, including parent-offspring, sibling-sibling, parent-parent, and even multiple familial sources. CD36 mediates the uptake of long-chain fatty acids (FAs), a major energy substrate for the myocardium. Under excessive FA supply, CD36 can cause cardiac lipid accumulation and inflammation while its deletion reduces heart FA uptake and lipid content and increases glucose utilization. As a result, CD36 was proposed as a therapeutic target for obesity-associated heart disease. However, more recent reports have shown that CD36 deficiency suppresses myocardial flexibility in fuel preference between glucose and FAs, impairing tissue energy balance, while CD36 absence in tissue macrophages reduces efferocytosis and myocardial repair after injury. Injured sensory neurons activate a transcriptional program necessary for robust axon regeneration and eventual target reinnervation. Understanding the transcriptional regulators that govern this axon regenerative response may guide therapeutic strategies to promote axon regeneration in the injured nervous system. Here, we used cultured dorsal root ganglia neurons to identify pro-regenerative transcription factors. Using RNA sequencing, we first characterized this neuronal culture and determined that embryonic day 13.5 DRG (eDRG) neurons cultured for 7 days are similar to e15.5 DRG neurons

BACKGROUND PURPOSE: A substantial number of patients with acute ischemic stroke (AIS) experience multiple acute lesions (MAL). We here aimed to scrutinize MAL in a large radiologically deep-phenotyped cohort.
MATERIALS AND METHODS: Analyses relied upon imaging and clinical data from the international MRI-GENIE study. Imaging data comprised both Fluid-attenuated inversion recovery (FLAIR) for white matter hyperintensity (WMH) burden estimation and diffusion-weighted imaging (DWI) sequences for the assessment of acute stroke lesions. OBJECTIVE: To evaluate the association between cumulative fentanyl dose during neonatal intensive care and 5-year neurodevelopmental and socioemotional outcomes in very preterm infants.
MATERIALS AND METHODS: Patient demographics and clinical factors during the perinatal and neonatal course were collected in 84 patients born between 23- and 30-weeks gestational age (GA). Cumulative fentanyl dose during neonatal intensive care was calculated. Tailored interventions have been shown to be effective and tailoring is a popular process with intuitive appeal for researchers and practitioners. However, the concept and process are ill-defined in implementation science. Descriptions of how tailoring has been applied in practice are often absent or insufficient in detail. INTRODUCTION: Cardiac immunology studies in murine models have identified a sizeable population of myocardial B-cells and have shown that its modulation represents a promising strategy to develop novel therapies for heart failure. However, scarce data on B-cells in the human heart leaves unclear whether findings in rodents are relevant to human biology.
METHODS: We performed immunohistochemical stains to characterize the amount and distribution of B-cells in human hearts, analyzing both fresh and post-mortem tissue.
