Plants are the primary source of food for world’s population. Diseases in plants can cause yield loss, which can be mitigated by continual monitoring. Monitoring plant diseases manually is difficult and prone to errors. Using computer vision and artificial intelligence (AI) for the early identification of plant illnesses can prevent the negative consequences of diseases at the very beginning and overcome the limitations of continuous manual monitoring. The research focuses on the development of an automatic system capable of performing the segmentation of leaf lesions and the detection of disease without requiring human intervention. Chronic pain often leads to depression, increasing patient suffering and worsening prognosis. While hyperactivity of the anterior cingulate cortex (ACC) appears to be critically involved, the molecular mechanisms underlying comorbid depressive symptoms in chronic pain remain elusive. T cell lymphoma invasion and metastasis 1 (Tiam1) is a Rac1 guanine nucleotide exchange factor (GEF) that promotes dendrite, spine, and synapse development during brain development. Here, we show that Tiam1 orchestrates synaptic structural and functional plasticity in ACC neurons via actin cytoskeleton reorganization and synaptic N-methyl-d-aspartate receptor (NMDAR) stabilization. This Tiam1-coordinated synaptic plasticity underpins ACC hyperactivity and drives chronic pain-induced depressive-like behaviors. …

Glioblastoma (GBM) is the most common primary adult brain tumor diagnosed, with a 15-month prognosis using current standard of care treatment which includes chemotherapy with temozolomide (Temodar, TMZ). Chronotherapy, the practice of considering time of day in treating a disease, has been shown to improve outcome in several cancers, but had not been applied to GBM. Previous work in the lab on a cultured murine mesenchymal GBM cell line showed a time-of-day dependent maximum in sensitivity to TMZ corresponding with the daily peak of Bmal1 and trough of Per2 expression, two core clock genes. A big question in biology is how organisms compete in an environment of competitors for scarce resources. Part of the answer lies in distinguishing friend from foe and in forging cooperative bonds in the face of cheaters. The social amoeba – bacteria system I have studied here is an excellent place to explore these tensions. The first part of my thesis research involves a review of cooperation and conflict in the social amoeba Dictyostelium discoideum and a study of the limits on obligate social cheating in this species. This thesis describes research under the rubric of the Center for Sustainable Polymers that is aimed at two separate goals. The goal of the first project (Chapters 1 and 2) was to develop a deeper mechanistic understanding of a method for the synthesis of linear α-olefins, while the second aimed at synthesizing statistical copolymers that incorporate olefin-containing monomers through ring-opening transesterification polymerization and showing that these copolymers could be functionalized. The N-methyl-D-aspartate receptor (NMDAR) plays a variety of important roles in the development and function of primate central nervous systems. This thesis describes three nonhuman primate studies that, together, demonstrate that pharmacologically reducing NMDAR activity not only causes dramatic changes in neural activity in cortex, but also changes the relationship between that activity and a key signal often used as its proxy in human studies, the blood-oxygen-level-dependent (BOLD) signal. The first study reveals that NMDA antagonism is sufficient to induce powerful low-frequency modulation of spiking activity and extracellular local field potential (LFP) and discusses the implications of this finding for …

Ultrasensitive detection and quantification of soluble, secreted and cell surface-bound proteins is critical for advancing our understanding of cellular systems, enabling effective drug development, novel therapies, and bio-diagnostics. However, exiting technologies are largely limited by their sensitivity, making the detection and quantification of low-abundant proteins extremely challenging. This forms a major barrier in various fields of biology and biomedical sciences. The circadian clock and inflammation have a well-described bidirectional relationship in the peripheral immune system: the circadian clock regulates inflammatory responses, but inflammation also impacts oscillatory gene transcription controlled by the clock. The purpose of this thesis work is to understand if manipulation of the circadian clock can impact Alzheimer’s disease pathology, and if pathology itself can alter the circadian clock and its outputs. Here, I show that astrocyte-specific disruption of the circadian clock via deletion of the clock gene Bmal1 surprisingly decreased intra-neuronal protein pathologies. The Williams Syndrome Critical Region (WSCR) at chromosome 7q11.23 provides a unique opportunity to untangle the relationship between genotype and phenotype in complex behaviors, from fear and anxiety to sociability and sensorimotor processing. Copy number variations (CNVs) in this region result in two syndromes, Williams Syndrome (WS) and Duplication 7q11.23 Syndrome (Dup7), which display phenotypes that may align, indicating a common disruption of a system, or diverge, reflecting an underlying gene dosage-dependent effect. Humans require a shared understanding of others’ emotions for adaptive social functioning. For instance, if someone tells a joke and another person laughs, we assume the laughing person was happy because we have a shared concept for such positive interactions. Children develop and refine these concepts as they grow up, but the underlying neural processing that supports this development is unknown. Identifying the neurodevelopment underlying emotion processing could provide important insight to emotional disorders. Recent human genetic studies have associated mutations in a gene called Myelin Transcription Factor 1 Like (MYT1L) with neurodevelopmental disorders (NDDs). Patients with MYT1L loss of function (LoF) mutations (MYT1L Syndrome patients) demonstrate shared symptoms such as microcephaly, attention deficit and hyperactivity disorder (ADHD), and obesity. Despite prior studies showing MYT1L overexpression facilitates neuronal differentiation in vitro, its functions in vivo, especially in the mammalian brain, and how its mutation leads to human disease pathology remains poorly understood. This dissertation focuses on the impact of anglophilia on identity formation in the British empire. Since Britishness was an identity unavailable to non-white citizens of the empire, this attachment was necessarily unrequited. Analyzing works by Cornelia Sorabji, E.R. Braithwaite, V.S. Naipaul, and Tsitsi Dangarembga, I argue that the critical significance of anglophilia does not lie in the postcolonial subject’s success or failure in achieving a British-coded identity, but in the ways in which an affiliation to Britishness can be mobilized to accomplish a variety of personal, political, and social ends. Mechanosensitive (MS) ion channels are an evolutionarily conserved way for cells to sense mechanical forces and transduce them into ionic signals. A plasma membrane-localized MS channel from Arabidopsis thaliana, MscS-Like (MSL)10, senses cell swelling and initiates a signaling cascade that triggers programmed cell death. Whereas the channel properties of MSL10 have been well studied, how MSL10 signals remains largely unknown. I worked collaboratively to show that important lesions for cell death signaling in the cytosolic N- and C-terminal domains of MSL10 interact genetically. I also helped show that ionic flux through MSL10 is dispensable for signaling, which suggested that MSL10 …

Towards the end of the Late Archaic period (ca. 4800-3000 cal BP), between 3,600 and 3,300 years ago, Native Americans engineered a colossal earthwork complex that covers approximately 200 hectares in northeast Louisiana. Today, it is a UNESCO World Heritage site known as Poverty Point and the namesake for a material culture pattern documented to varying degrees at sites throughout the Lower Mississippi Valley (LMV). The mammalian gut is a complex organ which changes over the course of development and is influenced by the presence of a dense community of gut-resident microbes. Enteric viruses infect the gastrointestinal tract in the context of the commensal microbiota and host immunity. Here, we dissect the influence of host developmental stage and the gut microbiota in norovirus infection. From this work, we determine that murine norovirus (MNoV) infection in early life is restricted by an absence of the target MNoV cell tuft cells and intact interferon (IFN) signaling. This paper presents an integrated review of 144 articles published on green advertising (GA) between 1991-2020. The extant literature is reviewed to aid taxonomical analysis of definitions, methodologies, and theoretical frameworks. The review suggests that the GA literature tracks the stimuli-moderators-outcome template of the general advertising literature. Consequently, advertising claims and appeals, consumer related factors, and product related factors are the organizing themes of the review. Emerging trends and possible research gaps for each of these three categories are identified. At many academic libraries, library workers run the teaching, general reference consultations, technical troubleshooting, and software and licensing maintenance in Geographic Information Systems (GIS) for their institutions. This is very much the case in the Data Services unit of Johns Hopkins University’s Sheridan Libraries, where staff receive requests for help with a wide variety of mapping projects every semester. Sometimes they are straightforward requests for technical assistance, but sometimes they underpin much deeper investigations into how to situate people and significant events through time and geographic settings. Rapid globalization has slowly eroded the survival of the Baliatn ritual of the Dayak Kanayatn. Nowadays, the locals only rarely perform this ritual which is a manifestation of their culture. This study describes the Baliatn ritual pointing out it is pregnant with the meaningful philosophy of the ancestors. This tradition sets out the relationship between the people and nature; an example of gratitude and spirituality in this modern era. It also discusses the importance of cultural conceptualization through philosophy, art, and spirituality. Mentawaians sing ritual songs enshrined in archaic texts referring to particular primates and birds, while ritual and traditional dances imitate how gibbons, sea eagles, and other animals live in the natural world. Mentawaians craft sculptures of endemic primates and unique birds. The bilou gibbon ape and various other animals also symbolize specific sacred knowledge within the sabulungan spiritual belief system and traditional cosmology of Mentawai society. Although some do succeed in surviving, many older traditions have faded away. As an island community, the culture of the Bandanese is closely linked to their natural maritime environment. Not only is fishery the main source of income for many Bandanese, their cultural practices also include the creation of traditional boats and songs which tie the people, their environment and history together. These boats, locally referred to as belang or kora-kora, feature symbolic decorations and take part in an annual competition in which competing villages chant about their oral stories, known as kabata. This research is an attempt to delve into understanding the process of creative imagination of the sacred which is revealed in the intertwining of culture and nature in Geriana Kauh, Karangasem, Bali. This study aims to investigate the relationship between the individual, the social and ecology, as well as the transformation of individual consciousness into a collective awareness sharing a communal reality. In earlier publications I have argued that ancient Javanese poets imagined the world to be one marked by distinctions between a social world consisting of palace (kaḍatwan) and countryside (thāni-ḍusun) and a wilderness of seashores and forested mountains (pasir-wukir). The social world was characterized by the presence of an effective royal authority; the wilderness by its absence. A distinction was also drawn between this world inhabited by human beings and a world in which gods, ancestral spirits, and other divine beings dwelt (kedewatan). A bstract There are about 60 panels with narrative reliefs from the area of Trowulan in East Java depicting mountains, rivers, bridges, trees, fields, dwellings, pendopo, and palaces. A male figure wearing a cap-like headdress accompanied by a little panakawan-like figure, walks along a cobbled path through a beautiful landscape. The figures are carved in the simple style of East Javanese reliefs on Majapahit temples. The predominance of nature, in amazing detail, is unusual compared to other narrative temple reliefs. The exact provenance is difficult to determine. The artefacts are scattered in museums all over the world: the majority in …
