Hermite Gaussian beams are the solutions of the scalar paraxial wave equation in Cartesian coordinates. A method was developed to modulate the intensity profile of non-diffracting Hermite Gaussian (HG) beams. The original HG beamâ??s intensity profile consists of high intense corner lobes and low intense central lobes which is not ideal for structured illumination in light-field microscopy. The modulated HG beams were generated by multiplying the original HGâ??s beam envelope by a super-Gaussian envelope to modify the intensity profile to attain equal intensity lobes. Pretrial risk assessments are used to divert defendants from pretrial detention by estimating risk of pretrial specific outcomes (i.e., failure to appear, rearrest). Ongoing validation of this tool is recommended to assess accuracy and ensure that there is no bias against specific subgroups (e.g., gender, race/ethnicity, or age) of defendants. The present study evaluates the utility of a locally developed instrument in El Paso County - a predominantly Latinx county. Area Under the Curve (AUC) Receiving Operator Condition (ROC) analyses indicate statistically â??fairâ?? predictive utility for the tool. Serotonin reuptake inhibitors and receptor agonists are used to treat obesity, anxiety and depression. Here we studied the role of the serotonin 2C receptor (5-HT2CR) in weight regulation and behavior. Using exome sequencing of 2,548 people with severe obesity and 1,117 control individuals without obesity, we identified 13 rare variants in the gene encoding 5-HT2CR (HTR2C) in 19 unrelated people (3 males and 16 females). Eleven variants caused a loss of function in HEK293 cells. All people who carried variants had hyperphagia and some degree of maladaptive behavior. Genetic screens are widely exploited to develop novel therapeutic approaches for cancer treatment. With recent advances in single-cell technology, single-cell CRISPR screen (scCRISPR) platforms provide opportunities for target validation and mechanistic studies in a high-throughput manner. Here, we aim to establish scCRISPR platforms which are suitable for immune-related screens involving multiple cell types. We integrated two scCRISPR platforms, namely Perturb-seq and CROP-seq, with both in vitro and in vivo immune screens. By leveraging previously generated resources, we optimized experimental conditions and data analysis pipelines to achieve better consistency between results from high-throughput and individual validations. Myelin is essential to nervous system function, playing roles in saltatory conduction and trophic support. Oligodendrocytes (OLs) and Schwann cells (SCs) form myelin in the central and peripheral nervous systems respectively and follow different developmental paths. OLs are neural stem-cell derived and follow an intrinsic developmental program resulting in a largely irreversible differentiation state. During embryonic development, OL precursor cells (OPCs) are produced in distinct waves originating from different locations in the central nervous system, with a subset developing into myelinating OLs. OPCs remain evenly distributed throughout life, providing a population of responsive, multifunctional cells with the capacity to remyelinate …

Background/aim: Chimeric antigen receptor (CAR) T cells with tumor specificity are being increasingly investigated. Phase I trials are the first step of testing for safety of novel CAR-T therapy to determine the maximum tolerated dose (MTD). Several dose escalation methods have been developed over time including rule-based, model-based, and model-assisted designs. Reconstruction of midfacial traumatic defects secondary to a gunshot wound (GSW) is one of the most challenging head surgeries. The high impact on the functional and aesthetic quality of life, and the small margin of error to achieve a successful outcome are significant hurdles in the surgical treatment of these cases. Here we report a 33-years old patient who suffered a GSW with an entrance wound penetrating between the soft and hard palate and an exit wound on the right malar region. BACKGROUND: Aneurysmal subarachnoid hemorrhage (aSAH) leads to a robust systemic inflammatory response. We hypothesized that an early systemic glycolytic shift occurs after aSAH, resulting in a unique metabolic signature and affecting systemic inflammation.
METHODS: Control patients and patients with aSAH were analyzed. Samples from patients with aSAH were collected within 24 h of aneurysmal rupture. Mass spectrometry-based metabolomics was performed to assess relative abundance of 16 metabolites involved in the tricarboxylic acid cycle, glycolysis, and pentose phosphate pathway. BACKGROUND: Non-metastatic muscle invasive urothelial bladder cancer (MIBC) has a poor prognosis and standard of care (SOC) includes neoadjuvant cisplatin-based chemotherapy (NAC) combined with cystectomy. Patients receiving NAC have at best
METHODS: We optimized engraftment conditions for primary MIBC tumors using the CAM-PDX model and tested concordance between cisplatin-based chemotherapy response of patients to matching PDX tumors using tumor growth coupled with immunohistochemistry markers of proliferation and apoptosis. We also tested select kinase inhibitor response on chemotherapy-resistant bladder cancers on the CAM-PDX using tumor growth measurements and immuno-detection of proliferation marker, Ki-67.

This dissertation conducts a county-level analysis of human appropriation of net primary production (HANPP) harvested from crops, timber and grazing in the conterminous United States in the years 1997, 2002, 2007 and 2012. This study consists of three manuscripts (Chapters 2, 3, and 4). The first study quantifies net primary production in US counties in 1997, 2002, 2007 and 2012. The detailed HANPP study conducted concludes that HANPP is a valuable footprint tool for analyzing land use intensity and agricultural ecosystems. Climate patterns affect where plants can grow and survive. In tropical areas, the main form of precipitation is rain, and rainfall has been demonstrated to influence tree species distributions. For this thesis, I examined the relationship between rainfall and the diversity of animal-dispersed tropical shrubs. To do this, I used data on shrub diversity collected in the summer of 2017. Plots were established across the Isthmus of Panama, from the drier Pacific side to the wetter Atlantic side, to survey animal-dispersed shrubs. Ferruginous Hawks are a bird of prey species that nest in sagebrush steppe and grassland habitat. These birds are threatened by habitat loss and fragmentation from urbanization, rural development, oil and natural gas extraction, habitat treatment projects, and wildfires, and experiencing widespread population declines across their breeding range. Because of this, Ferruginous Hawks (FEHAs) have been deemed a Species of Greatest Conservation Need by state management agencies in Utah, Idaho, Wyoming and Nevada. Mental health treatments can be delivered in many ways. One approach is to use storytelling to communicate healthy practices. While societies across the world have engaged in storytelling for thousands of years, these practices have been used less in the mental health field. Native American students in higher education are often asked to find a sense of belonging in places and spaces that do not reflect their cultures or worldviews. This can lead to isolation and a feeling of having to choose between themselves and their identities as Indigenous peoples. This contributes to poorer mental health, loss of well-being, and decreased academic success. Bees are important for agricultural and non-agricultural ecosystems because they pollinate both wild plants and commercial crops. Flowers provide pollen and nectar resources that bees use to survive and reproduce. Measuring the relationship between the floral community and bee community may help apiarists and land managers to make informed decisions in managing wild and domesticated bee species. Computerized obstacle detection for moving vehicles is becoming more important as vehicle manufacturers make their systems more autonomous and safe. However, obstacle detection must operate quickly in dynamic environments such as driving at highway speeds. A unique obstacle detection system using 3D changes in the environment is proposed. Furthermore, these 3D changes are shown to contain sufficient information for avoiding obstacles. To make the system easy to integrate onto a vehicle, additional processing is implemented to remove unnecessary dependencies. This system provides a method for obstacle detection that breaks away from typical systems to be more efficient.

As humans harvest increasing amounts of biomass, it is crucial to gain an understanding of how much energy is being appropriated and the impact that this could have on ecosystems and biodiversity. The primary way in which humans impact biodiversity loss is through land use change. One way of quantifying the impact of land use change is through human appropriation of net primary production (HANPP). A heuristic is the simplified approximations that helps guide a planner in deducing the best way to move forward. Heuristics are valued in many modern AI algorithms and decision-making architectures due to their ability to drastically reduce computation time. Geologists use various “rock-clocks” in order to interpret the timing and tempo of events in Earth’s history. These include global geochemical events in the oceans (chemostratigraphy), the evolution of certain fossil species (trilobite biostratigraphy), and the radiogenic decay of uranium to lead in certain mineral such as zircon (U-Pb geochronology). Integrating these methods in the stratigraphic record in one location can provide a well-constrained timeline for a select part of the Geologic, or Deep Time, record. The wetlands of Great Salt Lake once supported hundreds of thousands of nesting ducks each year. In recent years, the number of nesting ducks in the same area was a fraction of those historic numbers. While many species of ducks do not rely on these wetlands for primary nesting habitat, cinnamon teal (Spatula cyanoptera) do. Great Salt Lake and its associated wetlands are in the heart of the cinnamon teal breeding range, and once supported half of the continental population.

Intertemporal choices are decisions between outcomes occurring at different times. For example, people may choose to quit smoking cigarettes for the delayed health-related benefits associated with abstention, or they may continue to smoke for the immediate gratification associated with smoking now. Importantly, patterns of intertemporal choices among people are associated with a number of maladaptive behaviors (e.g., cigarette smoking). In the present set of studies, I examine a facet of intertemporal choice: preference reversals. Leishmaniasis is an understudied disease found predominantly in high heat and humidity areas. The disease is transmitted by sandflies which are blood-feeding, biting insects. There is currently no known vaccine for any form of leishmaniasis and treatment imposes a significant economic impact on already poor and marginalized populations, as well as severe, lingering side effects for afflicted individuals. Annually, there are millions of people around the world afflicted with a form of Leishmaniasis that is transmitted by species of sandflies. Investing resources into reproduction can limit energy available to other competing demands, such as fighting off an infection; yet, both processes are necessary for organisms to survive and pass on their genes to the next generation. These strategies often follow patterns associated with lifespan, such that shorter-lived animals are more likely to invest more resources into reproduction over survival, and vice versa in long-lived animals. However, environmental change caused by urbanization can disrupt these relationships, and the within- and transgenerational costs of urbanization on females and offspring are unknown. This thesis examines religion as practiced in the Latter-day Saint home during a period of religious transition between 1890 and 1930. Using the family of June A. Bushman and Hyrum Smith as subjects, we examine how families managed the religious reforms of the Church of Jesus Christ of Latter-day Saints during this period. As individuals who came of age at the turn of the twentieth century, June and Hyrum’s lives intersected with their church’s transition from an isolated religion to a modern, American church.
Administrative modernization, priesthood reforms, reimagined family relationships, and other ecclesiastical changes came into tension with the …
