Dietary protein restriction is increasingly recognized as a unique approach to improve metabolic health, and there is increasing interest in the mechanisms underlying this beneficial effect. Recent work indicates that the hormone FGF21 mediates the metabolic effects of protein restriction in young mice. Here we demonstrate that protein restriction increases lifespan, reduces frailty, lowers body weight and adiposity, improves physical performance, improves glucose tolerance, and alters various metabolic markers within the serum, liver, and adipose tissue of wildtype male mice. Autophagy and telomere maintenance are two cellular survival processes that show a strong correlation during human ageing and cancer growth, however, their causal relationship remains unclear. In this study, using an unbiased transcriptomics approach, we uncover a novel role of autophagy genes in regulating telomere extension and maintenance pathways. Concomitantly, the pharmacological inhibition of ULK1 (Unc-51 like autophagy activating kinase 1) attenuated human telomerase reverse transcriptase (hTERT) gene expression and telomerase activity in HepG2 cells. Furthermore, the suppression of telomerase activity upon ULK1 inhibition was associated with telomere shortening and onset of cellular senescence in HepG2 cells. These results, thus, …

Geolocators are small devices that record and store time-stamped light levels that researchers typically use to approximate the latitude and longitude of small birds across the annual cycle. However, when geolocators are affixed to leg bands of larger-bodied birds, nest incubation by females interrupts the daily pattern of light and darkness. Thus, geolocators can provide information on nesting propensity, nest success, and renesting intensity; these demographic parameters are both difficult to measure unobtrusively and are critically important in determining population dynamics of birds, especially ducks. Climate change is already increasing the severity of extreme weather events such as with rainfall during hurricanes. But little research to date investigates if, and to what extent, there are social inequalities in climate change-attributed extreme weather event impacts. Here, we use climate change attribution science paired with hydrological flood models to estimate climate change-attributed flood depths and damages during Hurricane Harvey in Harris County, Texas. Using detailed land-parcel and census tract socio-economic data, we then describe the socio-spatial characteristics associated with these climate change-induced impacts. Background: To date, no medication has slowed the progression of Parkinson’s disease (PD). Preclinical, epidemiological, and experimental data on humans all support many benefits of endurance exercise among persons with PD. The key question is whether there is a definitive additional benefit of exercising at high intensity, in terms of slowing disease progression, beyond the well-documented benefit of endurance training on a treadmill for fitness, gait, and functional mobility. This study will determine the efficacy of high-intensity endurance exercise as first-line therapy for persons diagnosed with PD within 3 years, and untreated with symptomatic therapy at baseline. Cancer cachexia is a common, debilitating condition with limited therapeutic options. Using an established mouse model of lung cancer, we find that cachexia is characterized by reduced food intake, spontaneous activity, and energy expenditure accompanied by muscle metabolic dysfunction and atrophy. We identify Activin A as a purported driver of cachexia and treat with ActRIIB-Fc, a decoy ligand for TGF-β/activin family members, together with anamorelin (Ana), a ghrelin receptor agonist, to reverse muscle dysfunction and anorexia, respectively. Ana effectively increases food intake but only the combination of drugs increases lean mass, restores spontaneous activity, and improves overall survival. These beneficial …

There have been multiple instances of novel pathogen emergence that have affected the health and security of the global community. To highlight that these novel pathogens presented a clear danger to public health, the WHO included “Disease X” on their list of priority pathogens in 2018. Indeed, since the emergence of SARS-CoV-2, Disease X has been pointed to as the looming threat of “the next big thing.” However, developing surveillance and preparedness plans with Disease X as the linchpin is too narrow and ignores a large swath of potential threats from already identified, often neglected diseases. Uterine lumen fluid (ULF) is central to successful pregnancy establishment and maintenance, and impacts offspring wellbeing into adulthood. The current dogma is that ULF composition is primarily governed by endometrial glandular epithelial cell secretions and influenced by progesterone. To investigate the hypothesis that ULF is metabolically semi-autonomous, ULF was obtained from cyclic heifers, and aliquots incubated for various durations prior to analysis by untargeted semi-quantitative metabolomic profiling. Metabolite flux was observed in these ULF isolates, supporting the idea that the biochemical makeup of ULF is semi-autonomously dynamic due to enzyme activities. There is considerable uncertainty regarding which ecosystems are most vulnerable to warming. Current understanding of organismal sensitivity is largely centred on species-level assessments that do not consider variation across populations. Here we used meta-analysis to quantify upper thermal tolerance variation across 305 populations from 61 terrestrial, freshwater, marine and intertidal taxa. We found strong differentiation in heat tolerance across populations in marine and intertidal taxa but not terrestrial or freshwater taxa. This is counter to the expectation that increased connectivity in the ocean should reduce intraspecific variation. Probiotics are commonly added to yogurt to provide many health benefits for the consumer. A description is provided for some commonly used probiotics in yogurt. A GRAS (generally recognized as safe) list of probiotic bacteria that can be added to yogurt or similar types of products is provided. Additionally, prebiotics, synbiotics (combination of prebiotics and probiotics), postbiotics, paraprobiotics, and psychobiotics can be added to yogurt. Probiotic yogurt can come in various forms in addition to spoonable yogurt, and yogurt can be used as an ingredient in other food products. This paper reports on a three-year initiative to enhance the undergraduate Extension Education minor at a state land-grant university. Specific initiatives to improve the undergraduate Extension Education minor curriculum involved (a) revising the Extension Education minor due to varied curriculum and faculty changes and (b) aligning eight Extension Education courses’ units of study with Extension professional competency domains. The described processes helped understand the Extension Education curriculum by mapping competency domains and showing which domains were and were not taught in the eight courses. The rise in popularity of farmers markets in the United States reflects consumers’ negative response to more traditional food distribution systems. Farmers markets provide consumers with a more local and often more personal food purchasing experience. The purpose of this study was to examine consumer motivations to patronize farmers markets through the lens of social, spatial, and natural embeddedness. A qualitative approach was employed utilizing semi-structured, in-depth interviews. These interviews were conducted in person using a set of predetermined questions and revealed nine themes. Meloidogyne enterolobii is an aggressive root-knot nematode (RKN) species that has emerged as a significant pathogen of sweetpotato in the Southeastern US. Meloidogyne enterolobii is spread through the movement of infected ‘seed’ sweetpotatoes used for propagation, and the RKN resistance in commercial sweetpotato cultivars has proven ineffective against this nematode. This has prompted regulatory agencies to impose quarantines on regions infected with M. enterolobii. Sweetpotato storage roots can be rendered unmarketable due to M. enterolobii infection, resulting in potential overall yield loss. Background and Objectives
While somatic mutations have been well-studied in cancer, their roles in other complex traits are much less understood. Our goal is to identify somatic variants that may contribute to the formation of saccular cerebral aneurysms.
Methods
We performed whole-exome sequencing on aneurysm tissues and paired peripheral blood. RNA sequencing and the CRISPR/Cas9 system were then used to perform functional validation of our results.
Results

Heavy reliance on synthetic pesticides has led to insecticide resistance of pests such as the onion armyworm, leading to the use of alternative strategies such as biopesticides. While studies on the efficacy of these biopesticides on pest control and yield exist, their subsequent effects on the quality and postharvest behavior of bulb onions during cold storage need to be determined. Camelina is an alternative low-input oil plant grown in marginal areas in Turkey. This study aimed to determine some quality characteristics of 39 camellia genotypes in ecological conditions in Bolu, Turkey. The field experiments were carried out in an augmented design with randomized blocks. In this work, some important properties such as thousand seed weight, oil content, fatty acid composition, and protein ratio were investigated. Results showed that mean data for thousand seed weight, oil ratio, and protein ratio were 0.63 – 1.5 g, 26.69 – 39.17%, and 21.81 – 29.81%, respectively. This study evaluated the safety of Bignay (Antidesma bunius (L.) Spreng cv. ‘Kalabaw’) fruits through a sub-acute oral toxicity test in male and female ICR mice. The evaluation was done through oral gavage of the aqueous fruit sample at 2,000 mg kg-1 body weight dose for 28 d. Results showed that there were no significant changes in feed and water intake of male and female ICR mice as compared to the control groups that received distilled water (p < 0.05). Blood chemistry and hematology values for all mice groups were comparable to the controls and published values. All mice groups also had appreciable body weight gain (10.89% – 21.69%) with zero morbidity and mortality. Lastly, the gross and microscopic examination of the brain, heart, lungs, GI tract, liver, spleen, and kidneys showed normal architecture and histology, suggesting that the fruits did not induce any morphological abnormalities. Results of this study showed that Bignay (cv. ‘Kalabaw’) fruits are safe for consumption with an LD50 of greater than 2,000 mg kg-1 body weight.

Since its introduction, the amine-thiol cosolvent system has been successfully utilized for the deposition of various thin-film devices, but its mechanism of action is still uncertain. Herein, we have attempted to dissect some of the chemical characteristics of amine-thiol cosolvents, with a special interest taken towards a mixture of ethylenediamine (en) and mercaptoethanol (ME). Conductivity was measured for multiple amine-thiol combinations at different ratios to determine extent of ionization in solution, with en-ME having one of the highest solution conductivities. Type-1 diabetes mellitus (T1DM) has been repeatedly linked to executive functioning (EF) deficits in children and adolescents but the mechanisms underlying this association are not well understood. Chronic hyperglycemia — frequently measured as glycosylated hemoglobin A1c (HbA1c) levels — and increased glycemic variability have both been proposed as risk factors for EF difficulties in this population, but it is unclear whether they each uniquely reflect T1DM-related risk for EF deficits. Furthermore, few studies have considered interactions between glycemic factors and family-level risk factors in shaping children’s EF development.
