Distributed-volume atmospheric turbulence near the ground significantly limits the performance of incoherent imaging and coherent beam projection systems operating over long horizontal paths. Defense, military and civilian surveillance, border security, and target identification systems are interested in terrestrial imaging and beam projection over very long horizontal paths, but atmospheric turbulence can blur the imagery and aberrate the laser beam such that they are beyond usefulness. While many post-processing and adaptive optics techniques have been developed to mitigate the effects of turbulence, many of these techniques do not work as expected in stronger volumetric turbulence, or in many cases don't work …

Aberrant or constitutive activation of nuclear factor kappa B (NF-κB) contributes to various human inflammatory diseases and malignancies via the upregulation of genes involved in cell proliferation, survival, angiogenesis, inflammation, and metastasis. Thus, inhibition of NF-κB signaling has potential for therapeutic applications in cancer and inflammatory diseases. We reported previously that Nei-like DNA glycosylase 2 (NEIL2), a mammalian DNA glycosylase, is involved in the preferential repair of oxidized DNA bases from the transcriptionally active sequences via the transcription-coupled base excision repair pathway. Chromosome translocations involving mixed lineage leukemia (MLL) gene cause acute leukemia with a poor prognosis. MLL is frequently fused with transcription cofactors AF4 (~35%), AF9 (25%) or its paralog ENL (10%). The AHD domain of AF9/ENL binds to AF4, its paralog AFF4, or histone-H3 lysine-79 (H3K79) methyltransferase DOT1L. Formation of AF9/ENL/AF4/AFF4-containing super elongation complexes (SEC) and the catalytic activity of DOT1L are essential for MLL-rearranged leukemia. Protein-protein interactions (PPI) between AF9/ENL and DOT1L/AF4/AFF4 are therefore a potential drug target.

PURPOSE: To achieve the ultimate goal of personalized treatment of patients, accurate molecular diagnosis and precise interpretation of the impact of genetic variants on gene function is essential. With sequencing cost becoming increasingly affordable, the accurate distinguishing of benign from pathogenic variants becomes the major bottleneck. Although large normal population sequence databases have become a key resource in filtering benign variants, they are not effective at filtering extremely rare variants.

Insulin-like growth factor (IGF) is a potent mitogen that activates the IGF receptor (IGFR)/insulin receptor substrate (IRS) axis, thus stimulating growth in normal cells and uncontrolled cell proliferation in cancer. Posttranslational modifications of IRS such as ubiquitination tightly control IGF signaling, and we previously identified IRS-1 as a potential substrate for the E3 ubiquitin ligase TRAF4 using an unbiased screen. Here we provide evidence that TRAF4-mediated ubiquitination of IRS-1 is physiologically relevant and crucial for IGF signal transduction. Through site-directed mutagenesis we found that TRAF4 promotes an atypical K29-linked ubiquitination at the C-terminal end of IRS-1. Its depletion abolishes AKT …

Leucine rich repeat transmembrane (LRRTM) proteins are synaptic adhesion molecules with roles in synapse formation and signaling. LRRTM4 transcripts were previously shown to be enriched in rod bipolar cells (BCs), secondary neurons of the retina that form synapses with rod photoreceptors. Using two different antibodies, LRRTM4 was found to reside primarily at rod BC dendritic tips, where it colocalized with the transduction channel protein, TRPM1. LRRTM4 was not detected at dendritic tips of ON-cone BCs. Following somatic knockout of LRRTM4 in BCs by subretinal injection and electroporation of CRISPR/Cas9, LRRTM4 was abolished or reduced in the dendritic tips of transfected …

Combined Oxidative Phosphorylation Deficiency 23 (COXPD23) caused by mutations in GTPBP3 gene is a rare mitochondrial disease, and this disorder identified from the Chinese population has not been described thus far. Here, we report a case series of three patients with COXPD23 caused by GTPBP3 mutations, from a severe to a mild phenotype. The main clinical features of these patients include lactic acidosis, myocardial damage, and neurologic symptoms. Sea turtles are exposed to trace elements through water, sediment, and food. Exposure to these elements has been shown to decrease immune function, impair growth, and decrease reproductive output in wildlife. This study compares trace element concentrations in green turtles in captivity at Sea Life Park Hawaii (n = 6) to wild green turtles in Kapoho Bay, HI (n = 5 to 7). Blood and scute samples were collected and analyzed for eleven elements via inductively coupled plasma-mass spectrometry (ICP-MS). Selenium was significantly greater (p < 0.05) in the blood of captive turtles compared to wild turtles, while V, Ni, and Pb were significantly greater in the blood of wild turtles. In scute, V, Cu, Se, and Cr were significantly greater in captive turtles, while As was significantly greater in wild turtles. Pelleted food fed to the captive turtles and representative samples of the wild turtle diet were analyzed via ICP-MS to calculate trophic transfer factors and daily intake values. Wild turtles had greater estimated daily intake than captive turtles for all elements except Cu and Se. Because captive turtles are fed a diet very different from their wild counterparts, captive turtles do not represent control or reference samples for chemical exposure studies in wild turtles. No toxic thresholds are known for sea turtles, but rehabilitation and managed care facilities should monitor sea turtle elemental concentrations to ensure the animals’ health.

BACKGROUND AND AIMS: South Asians (SA) experience disproportionately higher rates of atherosclerotic cardiovascular disease (ASCVD) events than non-Hispanic whites (NHW) and several other Asian groups. The coronary artery calcium (CAC) Agatston score may not capture the unique characteristics of coronary plaque in SA. We therefore evaluated the prevalence and patterns of advanced CAC measures (specific coronary vessel involvement, CAC volume and density) in SA versus other race/ethnicities.

BACKGROUND: Cardiovascular prevention guidelines use estimated 10-year atherosclerotic cardiovascular disease (CVD) risk based on the pooled cohort equations to guide treatment decisions and engage patients in shared decision-making. We sought to determine patient perceived versus actual risk of atherosclerotic CVD and associations with willingness for preventive therapy.
METHODS: We evaluated calculated and perceived CVD risk among 4187 patients across 124 sites in the Patient and Provider Assessment of Lipid Management Registry. Hyperferritinemia is associated with poor outcomes in critically ill patients with sepsis, hemophagocytic lymphohistiocytosis (HLH), macrophage activation syndromes (MAS) and coronavirus disease 19 (COVID-19). Autopsies of hyperferritinemic patients that succumbed to either sepsis, HLH, MAS or COVID-19 have revealed disseminated microvascular thromboses with von Willebrand factor (VWF)-, platelets-, and/or fibrin-rich microthrombi. It is unknown whether high plasma ferritin concentration actively promotes microvascular thrombosis, or merely serves as a prognostic biomarker in these patients. BACKGROUND: Emerging data suggest that type 2 diabetes mellitus (T2D) is associated with an increased risk for fractures despite relatively normal or increased bone mineral density (BMD). Although the mechanism for bone fragility in T2D patients is multifactorial, whether glycemic control is important in generating this impairment in bone metabolism remains unclear. Waste generation is increasing, and a significant portion is being landfilled. In parallel, we are constantly pursuing cleaner fuels due to environmental and regulatory factors. To address both these challenges, torrefaction of wastes to produce clean fuels and feedstock for other thermochemical processes is one of the potential solutions. The atomic-scale Monte Carlo simulation study focuses on the intrinsic behaviors of a defect-free crystal that undergoes a cubic-to-tetragonal martensitic transformation. The quasi-spin variable associated with the lattice sites characterizes the local unit cells of the orientation variants of the ground-state martensite phase, which interact with each other through long-range elastic interactions. It is shown that the diffuse scattering in the premartensitic austenite state results from the spatial correlation of the atomic-scale heterogeneous lattice displacements and manifests the displacement short-range ordering. Atmospheric aerosols play a vital role in the Earth's energy budget-directly by scattering and absorbing solar radiation and indirectly by acting as cloud condensation nuclei and ice-nucleating particles [1, 2]. The cloud formation potential of aerosol is driven by multiple factors, including surface properties, size distribution, composition, mixing state, phase state, and morphology [3]. The interaction of aerosols with clouds alters the aerosol's physicochemical properties. This thesis concerns the generating functions $f_{\lambda, k}(q)$ for standard Young tableaux of shape $\lambda$ with precisely $k$ descents, aiming to find closed formulas for a general form given by Kirillov and Reshetikhin in 1988. Throughout, we approach various methods by which further closed forms could be found. In Chapter 2 we give closed formulas for tableaux of any shape and minimal number of descents, which arise as principal specializations of Schur functions. We provide formulas for tableaux with three parts and one more than minimal number of descents, and demonstrate that the technique is extendable to any number of …

Chapter 2 is based on the synthesis and study of the compounds of the bidentate ligand ((5-phenyl-1H-pyrazol-3-yl)methyl)phosphine oxide with molybdenum and cobalt as the transition metal. The complexes were analyzed via FTIR, NMR, UV-Vis, Fluorescence Spectroscopy, TGA, DFT, and XRD. Chapter 3 resulted in the synthesis of the complexes [Ni(II)SSRRL](PF6)2 and [Ni(II)SRSRL](Cl)(PF6) of which [Ni(II)SRSRL](Cl)(PF6) had not been previously analyzed. Within the last 40 years, academic research on disasters has focused on resilience as applied to individual adaptive capacities, rebuilding resources, and policy-driven solutions. While there has been an increased awareness of the many gendered dimensions of post-disaster recovery, women’s and mother’s agency in such situations is still largely ignored. Thus, this dissertation adopts a maternal focus, arguing that mothers are not merely vulnerable subjects but critical agents of post-disaster recovery for families, communities, and social systems more generally.

In the global transition to green energy in the transportation and power production sectors, lithium has emerged as a viable solution, and crucial component, for the effective storage of renewable resources. To avoid further degradation of the land due to mining for virgin material, methods focused on recovering lithium must be developed. This research project seeks to explore a new way to recover lithium by using the bacterium Paenibacillus polymyxa. The bacteria were grown and then applied to the lithium bearing mineral spodumene. Dam removal is a critical part of river and stream remediation projects around the world. Frequently dams are associated with impoundment ponds containing sediments that can act as a sink of various persistent organic pollutants (POPs) that can pose risks for human and aquatic wildlife. In this study, we deployed semi-permeable membrane devices (SPMDs) at multiple locations along a 25-mile stretch of the Boardman River in Traverse City Michigan to passively sample POP concentrations in this area that has recently undergone dam and impoundment pond sediment removal. Data on the variation of the direction and strength of Earth’s ancient magnetic field (absolute paleointensity) provide crucial information into the mechanisms of the geodynamo and the Earth’s thermal history. However, the use of conventional methods and instrumentation for absolute paleointensity determination has been hampered by physicochemical alteration of the samples caused by multiple high-temperature cycles and long experiment durations. Advanced fossil energy power generation plants require materials that withstand high temperatures and corrosive environments. One such material that is used in steam turbines is Nimonic 263. It is a nickel-base superalloy that is principally strengthened by gamma primephase (Ni3(Ti, Al)) and has an L12structure. At extended times and at turbine operating temperatures however, eta (Ni3Ti) phase is known to form at the expense of gamma prime. Atmospheric scientists and climate modelers are faced with uncertainty around the process of ice production in clouds. While significant progress has been made in predicting homogeneous and heterogeneous ice nucleation rates as a function of temperature, recent experiments have shown that ice nucleation rates can be enhanced without decreasing temperature, through various mechanical agitations. One hypothesis for these findings is that mechanisms of stretching water and thereby inducing negative pressure in the liquid could lead to an increase in freezing rate. The interaction of aerosol particles with solar radiation significantly contributes to the global radiation balance. The magnitude of this aerosol-radiation interaction, among other parameters, depends on different aerosol properties, including how readily these particles would act as cloud condensation nuclei (CCN). These properties are governed by the formation and scavenging processes of aerosol. This dissertation explores some of these scavenging processes.
Favorable humidity and preexisting aerosol particles acting as CCN are the sine qua non conditions to form cloud droplets in Earth’s atmosphere. Forming cloud droplets (known as activation), meanwhile, acts as a wet scavenging mechanism for those CCN. Given …

Denitrification and nitrogen fixation are nitrogen cycling processes that can occur simultaneously in streams but are rarely studied together. The overarching goal of this dissertation was to quantify temporal and spatial variation of these processes in streams across the USA, characterize the environmental drivers of that variation, and determine the role that denitrification plays in the carbon cycle. To characterize temporal variation in these processes, a 2-year study in the Pilgrim River in Michigan’s Upper Peninsula found no difference in rates among seasons but high day-to-day variation in rates of both processes (maximum daily change 4,390 μg N/m2/hr …
