There are significant physiological and anatomical differences in the nasal cavity, olfactory area, and airflow reaching the olfactory epithelium between humans and experimentally studied species that should be considered when extrapolating experimental results to humans. Mucus, transporters, and tight junction proteins present barriers to material translocation across the olfactory epithelium. Rhodopseudomonas palustris CGA009 is a metabolically robust microbe that can utilize lignin breakdown products to produce polyhydroxyalkanoates (PHAs), biopolymers with the potential to replace conventional plastics. Our recent efforts suggest PHA granule formation is a limiting factor for maximum production of the bioplastic poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) by R. palustris. The Phap1 phasin (phaP1) from the PHBproducing model bacterium Cupriavidus necator H16 was expressed in R. palustris with the aim of overproducing PHBV from the lignin breakdown product p-coumarate by fostering smaller and more abundant granules. Undergraduate and graduate students in courses designed to introduce research techniques in exercise physiology were provided the novel opportunity through Project-based Learning (PBL) to utilize a prototype device to measure energy expenditure (EE). This report summarizes how EE measurements were incorporated via PBL into course-required experiments and determined perceived understanding of exercise and metabolism. Undergraduate experiments included measurement of EE following high intensity cycling preceded by a motivating yell, and EE after upper-body and lower-body exercise performed at simulated altitude.
