Voltage-gated Kv1.3 potassium channels are key regulators of T lymphocyte activation, proliferation and cytokine production, by providing the necessary membrane hyper-polarization for calcium influx following immune stimulation. It is noteworthy that an accumulating body of in vivo and in vitro evidence links these channels to multiple sclerosis pathophysiology. Here we studied the electrophysiological properties and the transcriptional and translational expression of T lymphocyte Kv1.3 channels in multiple sclerosis, by combining patch clamp recordings, reverse transcription polymerase chain reaction and flow cytometry on freshly isolated peripheral blood T lymphocytes from two patient cohorts with multiple sclerosis, as well as from healthy …

Chromosome translocations involving mixed lineage leukemia (MLL) gene cause acute leukemia with a poor prognosis. MLL is frequently fused with transcription cofactors AF4 (~35%), AF9 (25%) or its paralog ENL (10%). The AHD domain of AF9/ENL binds to AF4, its paralog AFF4, or histone-H3 lysine-79 (H3K79) methyltransferase DOT1L. Formation of AF9/ENL/AF4/AFF4-containing super elongation complexes (SEC) and the catalytic activity of DOT1L are essential for MLL-rearranged leukemia. Protein-protein interactions (PPI) between AF9/ENL and DOT1L/AF4/AFF4 are therefore a potential drug target.

Methods: Compound screening followed by medicinal chemistry was used to find inhibitors of such PPIs, which were examined for their biological …

Lrrtm4 Is A Member Of The Transsynaptic Complex Between Rod Photoreceptors And Bipolar Cells, Melina A Agosto, Theodore G Wensel Jan 2021

Lrrtm4 Is A Member Of The Transsynaptic Complex Between Rod Photoreceptors And Bipolar Cells, Melina A Agosto, Theodore G Wensel

Faculty, Staff and Students Publications

Leucine rich repeat transmembrane (LRRTM) proteins are synaptic adhesion molecules with roles in synapse formation and signaling. LRRTM4 transcripts were previously shown to be enriched in rod bipolar cells (BCs), secondary neurons of the retina that form synapses with rod photoreceptors. Using two different antibodies, LRRTM4 was found to reside primarily at rod BC dendritic tips, where it colocalized with the transduction channel protein, TRPM1. LRRTM4 was not detected at dendritic tips of ON-cone BCs. Following somatic knockout of LRRTM4 in BCs by subretinal injection and electroporation of CRISPR/Cas9, LRRTM4 was abolished or reduced in the dendritic tips of transfected …

Ketamine: Neuroprotective Or Neurotoxic?, Divya Choudhury, Anita E Autry, Kimberley F Tolias, Vaishnav Krishnan

Faculty, Staff and Students Publications

Ketamine, a non-competitive N-methyl-D-aspartate receptor (NMDAR) antagonist, has been employed clinically as an intravenous anesthetic since the 1970s. More recently, ketamine has received attention for its rapid antidepressant effects and is actively being explored as a treatment for a wide range of neuropsychiatric syndromes. In model systems, ketamine appears to display a combination of neurotoxic and neuroprotective properties that are context dependent. At anesthetic doses applied during neurodevelopmental windows, ketamine contributes to inflammation, autophagy, apoptosis, and enhances levels of reactive oxygen species. At the same time, subanesthetic dose ketamine is a powerful activator of multiple parallel neurotrophic signaling cascades …

Asthma remains one of the most important challenges to pediatric public health in the US. A large majority of children with persistent and chronic asthma demonstrate aeroallergen sensitization, which remains a pivotal risk factor associated with the development of persistent, progressive asthma throughout life. In individuals with a tendency toward Type 2 inflammation, sensitization and exposure to high concentrations of offending allergens is associated with increased risk for development of, and impairment from, asthma. The cascade of biological responses to allergens is primarily mediated through IgE antibodies and their production is further stimulated by IgE responses to antigen exposure. In …

Case Report: Dupilumab Successfully Controls Severe Eczema In A Child With Elevated Ige Levels And Recurrent Skin Infections, Tejas P Joshi, Sara Anvari, Meera R Gupta, Carla M Davis, Joud Hajjar

Faculty, Staff and Students Publications

The efficacy of dupilumab in pediatric patients with severe eczema presenting in the setting of elevated immunoglobulin E (IgE) levels and recurrent bacterial skin infections is not well-understood. Here we present the case of a child with elevated IgE levels in whom dupilumab treatment led to remarkable control of his eczema and recurrent skin infections. We also review the use of dupilumab in other patients with molecularly proven cases of hyper IgE (HIGE) syndrome. Our case supports the notion that dupilumab may have a seminal application in treating severe eczema that occurs in the setting of elevated IgE levels and …

Data Are In The Eye Of The Beholder: Co-Creating The Value Of Personal Data, Marta Stelmaszak Rosa, Glenn Parry Jan 2021

Data Are In The Eye Of The Beholder: Co-Creating The Value Of Personal Data, Marta Stelmaszak Rosa, Glenn Parry

Business Faculty Publications and Presentations

The value of personal data has traditionally been understood in economic terms, but recent scholarship casts the value of data as multi-faceted, dynamic, emergent and co-created by stakeholders. The dynamics of the co-creation of value with personal data lacks empirical study. We conduct a case study of the development of a personalised e-book and find different perceptions of the value of personal data exist from the firm, intermediary and customer perspective: means to an end, medium of exchange and net benefit. The different data perspectives highlight ontological differences in the perception of what data are. This creates epistemological tension and …

Salt Marsh Hydrogeology: A Review, Julia Guimond, Joseph Tamborski Jan 2021

Salt Marsh Hydrogeology: A Review, Julia Guimond, Joseph Tamborski

OES Faculty Publications

Groundwater–surface water exchange in salt marsh ecosystems mediates nearshore salt, nutrient, and carbon budgets with implications for biological productivity and global climate. Despite their importance, a synthesis of salt marsh groundwater studies is lacking. In this review, we summarize drivers mediating salt marsh hydrogeology, review field and modeling techniques, and discuss patterns of exchange. New data from a Delaware seepage meter study are reported which highlight small-scale spatial variability in exchange rates. A synthesis of the salt marsh hydrogeology literature reveals a positive relationship between tidal range and submarine groundwater discharge but not porewater exchange, highlighting the multidimensional drivers of …

Microbially Induced Sedimentary Structures In Clastic Deposits: Implication For The Prospection For Fossil Life On Mars, Nora Noffke Jan 2021

Microbially Induced Sedimentary Structures In Clastic Deposits: Implication For The Prospection For Fossil Life On Mars, Nora Noffke

OES Faculty Publications

Abundant and well-preserved fossil microbenthos occurs in siliciclastic deposits of all Earth ages, from the early Archean to today. Studies in modern settings show how microbenthos responds to sediment dynamics by baffling and trapping, binding, biostabilization, and growth. Results of this microbial-sediment interaction are microbially induced sedimentary structures (MISS). Successful prospection for rich MISS occurrences in the terrestrial lithological record requires unraveling genesis and taphonomy of MISS, both of which are defined only by a narrow range of specific conditions. These conditions have to coincide with high detectability which is a function of outcrop quality, bedding character, and rock type. …

OES Faculty Publications

Glacial meltwater from the western Antarctic Ice Sheet is hypothesized to be an important source of cryospheric iron, fertilizing the Southern Ocean, yet its trace-metal composition and factors that control its dispersal remain poorly constrained. Here we characterize meltwater iron sources in a heavily glaciated western Antarctic Peninsula (WAP) fjord. Using dissolved and particulate ratios of manganese to iron in meltwaters, porewaters, and seawater, we show that surface glacial melt and subglacial plumes contribute to the seasonal cycle of iron and manganese within a fjord still relatively unaffected by climate-change-induced glacial retreat. Organic ligands derived from the phytoplankton bloom and …

Designing Disruption In Pharmaceuticals, Robin C. Feldman Jan 2021

Designing Disruption In Pharmaceuticals, Robin C. Feldman

Antitrust is atomistic: deliberately focused on trees, not forests. It pays attention to the consequences of individual acts alleged to be anticompetitive. That focus is misplaced. Companies and markets don't focus on one particular act to the exclusion of all else. Business strategy em phasizes holistic, integrated planning. And market outcomes aren't determined by a single act, but by the result of multiple acts by multiple parties in the overall context of the structure and character istics of the market. The atomistic nature of modern antitrust law causes it to miss two important classes of potential competitive harms. First, the …

Covid-19 Vaccine Uptake Through The Lived Experiences Of Health Care Personnel: Policy And Legal Considerations, Rachel Gur-Arie, Zackary Berger, Dorit Rubinstein Reiss Jan 2021

Covid-19 Vaccine Uptake Through The Lived Experiences Of Health Care Personnel: Policy And Legal Considerations, Rachel Gur-Arie, Zackary Berger, Dorit Rubinstein Reiss

Faculty Scholarship

Purpose: To investigate whether coronavirus disease 2019 (COVID-19) vaccination campaigns targeted at health care personnel (HCP) in the United States have addressed the lived experiences of HCP on the frontlines of the COVID-19 pandemic and to analyze policy and legal considerations for improving COVID-19 vaccine uptake among HCP. Methods: We conducted a literature and policy review to explore the lived experiences of different occupational groups of HCP on the frontlines of the COVID-19 pandemic—physicians, nurses, trainees, and nonclinical essential workers—in relation to ongoing COVID-19 vaccination campaigns. Finally, we discuss policy and legal considerations to improve the state of HCP COVID-19 …

Impact Of Neutron Energy On Asteroid Deflection Performance, Lansing S. Horan Iv, Darren E. Holland, Megan Bruck Syal, James E. Bevins, Joseph W. Wasem

Faculty Publications

In the future, a hazardous asteroid will find itself on a collision course with Earth. For asteroids of moderate size or larger, a nuclear device is one of humanity's only technologies capable of mitigating this threat via deflection on a timescale of less than a decade. This work examined how the output neutron energy from a nuclear device standoff detonation affects the deflection of a notional asteroid that is 300 meters in diameter and composed of silicon dioxide at a bulk density of 1.855 g/cm3. 14.1 MeV and 1 MeV neutron energy sources were modeled in MCNP to …

Government Activism In Bankruptcy, Jared A. Ellias, George Triantis Jan 2021

Government Activism In Bankruptcy, Jared A. Ellias, George Triantis

Faculty Scholarship

It is widely recognized that bankruptcy law can stymie regulatory enforcement and present challenges for governments when regulated businesses file for Chapter 11. It is less-widely understood that bankruptcy law can present governments with opportunities to advance policy goals if they are willing to adopt tactics traditionally associated with activist investors, a strategy we call “government bankruptcy activism.” The bankruptcy filings by Chrysler and General Motors in 2009 are a famous example: the government of the United States used the bankruptcy process to help both auto manufacturers resolve their financial distress while promoting the policy objectives of protecting union workers …

Congress's Power Over Militay Offices, Zachary S. Price Jan 2021

Congress's Power Over Militay Offices, Zachary S. Price

Faculty Scholarship

Although scholars have explored at length the constitutional law of office-holding with respect to civil and administrative offices, parallel questions regarding military office-holding have received insufficient attention. Even scholars who defend broad congressional authority to structure civil administration typically presume that the President, as Commander in Chief, holds greater authority over the military. For its part, the executive branch has claimed plenary authority over assignment of military duties and control of military officers. This pro-presidential consensus is mistaken. Although the President, as Commander in Chief, must have some form of directive authority over U.S. military forces in the field, the …

Incentives in patent law have driven innovation into spaces that are affirmatively harmful to patients, and patentees are discouraged from taking steps to improve the product so as to prevent adverse health outcomes.

States’ Merger Review Authority Is Associated With States Challenging Hospital Mergers, But Prices Continue To Increase, Brent D. Fulton, Jaime S. King, Daniel R. Arnold, Alexandra D. Montague, Samuel M. Chang, Thomas L. Greaney, Richard M. Scheffler Jan 2021

States’ Merger Review Authority Is Associated With States Challenging Hospital Mergers, But Prices Continue To Increase, Brent D. Fulton, Jaime S. King, Daniel R. Arnold, Alexandra D. Montague, Samuel M. Chang, Thomas L. Greaney, Richard M. Scheffler

Faculty Scholarship

States can challenge proposed hospital mergers by using antitrust laws to prevent anticompetitive harms. This observational study examined additional state laws—principally charitable trust, nonprofit corporation, health and safety, and certificate-of-need laws—that can serve as complements and substitutes for antitrust laws by empowering states to be notified of, review, and challenge proposed hospital mergers through administrative processes. During the period 2010–19, 862 hospital mergers were proposed, but only forty-two (4.9 percent) were challenged by states, including thirty-five by states without federal involvement, of which twenty-five (71.4 percent) originated in the eight states with the most robust merger review authority. The twenty-five …

A Deweyan Approach To The Dilemma Of Everyday Aesthetics, Thomas Leddy Jan 2021

A Deweyan Approach To The Dilemma Of Everyday Aesthetics, Thomas Leddy

Faculty Research, Scholarly, and Creative Activity

Everyday aesthetics is a new sub-discipline of aesthetic theory that has only been actively discussed since the 1980s. This paper addresses what many consider the central issue of the field, called "the dilemma of everyday aesthetics." I discuss three authors who address this issue: Yuriko Saito, Allen Carlson, and Paisley Livingston. Drawing on Dewey's anti-dualist stance, I argued for a continuity between the aesthetics of everyday life and the aesthetics of art. In course of my discussion, I question such dichotomies as that between the practical and the aesthetic, the ordinary and the extraordinary, and disinterestedness vs. engagement. In my …

Cross Border Regional Planning: Insights From Cascadia, Francesco Cappellano, Kathrine Richardson, Laurie Trautman

Faculty Research, Scholarly, and Creative Activity

This analysis focuses on different levels of Cross-Border Regional Planning (CBRP) processes in the Cascadia borderland. The region is home to the business-led initiative ‘Cascadia Innovation Corridor’ (CIC), designed to foster cross-border economic integration. The CIC strives to build a global innovation ecosystem in Cascadia, including a new high-speed train to connect Seattle and Vancouver. This paper focuses on the scope of the CIC as a CBRP case. The authors evaluate engagement of city governments and coherency between different planning scales to determine whether the CIC has been addressing the major challenges that may prevent tighter economicintegration in Cascadia. The …
