Unbiased Automated Quantitation Of Ros Signals In Live Retinal Neurons Of Drosophila Using Fiji/Imagej, Prajakta Deshpande, Neha Gogia, Anuradha Venkatakrishnan Chimata, Amit Singh

Biology Faculty Publications

Numerous imaging modules are utilized to study changes that occur during cellular processes. Besides qualitative (immunohistochemical) or semiquantitative (Western blot) approaches, direct quantitation method(s) for detecting and analyzing signal intensities for disease(s) biomarkers are lacking. Thus, there is a need to develop method(s) to quantitate specific signals and eliminate noise during live tissue imaging. An increase in reactive oxygen species (ROS) such as superoxide (O2•-) radicals results in oxidative damage of biomolecules, which leads to oxidative stress. This can be detected by dihydroethidium staining in live tissue(s), which does not rely on fixation and helps prevent stress on tissues. However, …

Enhancing Our Understanding Of Plant Cell-To-Cell Interactions Using Single-Cell Omics, Sandra Thibivilliers, Marc Libault

Department of Agronomy and Horticulture: Faculty Publications

Plants are composed of cells that physically interact and constantly adapt to their environment. To reveal the contribution of each plant cells to the biology of the entire organism, their molecular, morphological, and physiological attributes must be quantified and analyzed in the context of the morphology of the plant organs. The emergence of single-cell/nucleus omics technologies now allows plant biologists to access different modalities of individual cells including their epigenome and transcriptome to reveal the unique molecular properties of each cell composing the plant and their dynamic regulation during cell differentiation and in response to their environment. In this manuscript, …

Tandem Duplicate Expression Patterns Are Conserved Between Maize Haplotypes Of The Α-Zein Gene Family, Preston Hurst, James C. Schnable, David R. Holding Aug 2021

Tandem Duplicate Expression Patterns Are Conserved Between Maize Haplotypes Of The Α-Zein Gene Family, Preston Hurst, James C. Schnable, David R. Holding

Department of Agronomy and Horticulture: Faculty Publications

Tandem duplication gives rise to copy number variation and subsequent functional novelty among genes as well as diversity between individuals in a species. Functional novelty can result from either divergence in coding sequence or divergence in patterns of gene transcriptional regulation. Here, we investigate conservation and divergence of both gene sequence and gene regulation between the copies of the α-zein gene family in maize inbreds B73 and W22. We used RNA-seq data generated from developing, self-pollinated kernels at three developmental stages timed to coincide with early and peak zein expression. The reference genome annotations for B73 and W22 were modified …

Identifying Transfer Of Care Gaps: Electronic Health Record Capture Of Perioperative Handoff Communications, Sharon Giarrizzo-Wilson Aug 2021

Identifying Transfer Of Care Gaps: Electronic Health Record Capture Of Perioperative Handoff Communications, Sharon Giarrizzo-Wilson

Dissertations

Transitions in patient care are held together by interdisciplinary handoff communications intended to coordinate the patient's ongoing care requirements. Patients with complexity in care encumber the transfer of care process requiring a higher level of care coordination between the interdisciplinary team (Coleman, 2003; Naylor et al., 2004). While the literature is abundant on the characteristics and quality of handoff communications, it is limited on the requirements of what data is necessary for ongoing care following transfer communications (Galatzan & Carrington, 2018).

Beauveria bassiana is endophytic in many plant species and has been shown to protect host plants against insect pests and plant pathogens. However, less is known about its activity against plant-parasitic nematodes. In vitro and plant assays were conducted to determine the effect of B. bassiana 11-98 (Bb) on Meloidogyne incognita (root-knot nematode; RKN). Beauveria bassiana was confirmed as an endophyte in ‘Rutgers’ tomato and colonization patterns of Bb in ‘Rutgers’ (highly susceptible to RKN) were compared with those in ‘Mountain Spring’ (less susceptible to RKN). In greenhouse tests with ‘Rutgers’ at 30 and 60 days after treatment (DAT) with …

Immune Cell Residency In The Nasal Mucosa May Partially Explain Respiratory Disease Severity Across The Age Range., Konner Winkley, Dithi Banerjee, Todd Bradley, Boryana Koseva, Warren A. Cheung, Rangaraj Selvarangan, T Pastinen, Elin Grundberg

Manuscripts, Articles, Book Chapters and Other Papers

Previous studies focusing on the age disparity in COVID-19 severity have suggested that younger individuals mount a more robust innate immune response in the nasal mucosa after infection with SARS-CoV-2. However, it is unclear if this reflects increased immune activation or increased immune residence in the nasal mucosa. We hypothesized that immune residency in the nasal mucosa of healthy individuals may differ across the age range. We applied single-cell RNA-sequencing and measured the cellular composition and transcriptional profile of the nasal mucosa in 35 SARS-CoV-2 negative children and adults, ranging in age from 4 months to 65 years. We analyzed …

Molten-Salt-Assisted Annealing For Making Colloidal Znga2o4:Cr Nanocrystals With High Persistent Luminescence, Bhupendra B. Srivastava, Santosh K. Gupta, Swati Mohan, Yuanbing Mao

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Chemistry)

Persistent luminescent nanocrystals (PLNCs) in the sub-10 nm domain are considered to be the most fascinating inventions in lighting technology owing to their excellent performance in anti-counterfeiting, luminous paints, bioimaging, security applications, etc. Further improvement of persistent luminescence (PersL) intensity and lifetime is needed to achieve the desired success of PLNCs while keeping the uniform sub-10 nm size. In this work, the concept of molten salt confinement to thermally anneal as-synthesized ZnGa2O4:Cr3+ (ZGOC) colloidal NCs (CNCs) in a molten salt medium at 650 °C is introduced. This method led to significantly monodispersed and few agglomerated NCs with a much improved …

An End-To-End Cnn With Attentional Mechanism Applied To Raw Eeg In A Bci Classification Task, Elnaz Lashgari, Jordan Ott, Akima Connelly, Pierre Baldi, Uri Maoz

Psychology Faculty Articles and Research

Objective. Motor-imagery (MI) classification base on electroencephalography (EEG) has been long studied in neuroscience and more recently widely used in healthcare applications such as mobile assistive robots and neurorehabilitation. In particular, EEG-based motor-imagery classification methods that rely on convolutional neural networks (CNNs) have achieved relatively high classification accuracy. However, naively training CNNs to classify raw EEG data from all channels, especially for high-density EEG, is computationally demanding and requires huge training sets. It often also introduces many irrelevant input features, making it difficult for the CNN to extract the informative ones. This problem is compounded by a dearth of training …

A Patient’S Narrative Of Engaging Hiv Care: Lessons Learned To Harness Resources And Improve Access To Care, David Lessard, Serge Vicente, Patrick Keeler, Bertrand Lebouché

Patient Experience Journal

In Canada and the USA, about 30% of people with HIV are uninsured or not covered by government-subsidized health insurance. This paper presents a patient’s narrative of his experience being diagnosed with HIV and accessing care in the midst of his process of immigrating to and studying in Canada. The narrative explores how Vincent Croft (pseudonym) has coped with the chronicity of the infection and its associated social stigma, and the temporary solutions he found to access treatment. Engaging with healthcare providers, researchers, and other people living with HIV has allowed Croft to share his experience, including the barriers he …

Total Flavonoid Content, Total Phenolic Content, Antimicrobial And Antioxidant Activities Of Acacia Rigidula Leaf Extracts, Pablo Arenaz

Theses and Dissertations

The growing incidence of multidrug resistant bacteria has prompted a need for alternatives to traditional antibiotics. The Fabaceae family including the Acacia species have been reported to contain secondary metabolites known to exhibit antibacterial and antioxidant activities. Specifically, A. ridigula was previously shown to be effective against a range of bacterial species. When extracting secondary metabolites from plants, the method and choice of solvent is important in order to efficiently extract a high yield. This study aims to: 1) determine whether Soxhlet or maceration is the more efficient extraction method; 2) determine whether acetone and ethanol will yield more bioactive …

Biochemical Evaluation Of Drug Inhibition Of Abc Transporters, Maisa Correa De Oliveira Aug 2021

Biochemical Evaluation Of Drug Inhibition Of Abc Transporters, Maisa Correa De Oliveira

Biological Sciences Theses and Dissertations

Multidrug resistant (MDR) cancers often become resistant to pharmacologically and structurally diverse drugs by overexpressing members of the family of ATP Binding Cassette (ABC) transporters. These membrane pumps protect normal cells by actively exporting a large range of toxins out of the cells. Classic MDR is often correlated with the overexpression of three well-studied mammalian ABC transporters: P-glycoprotein (P-gp), Breast Cancer Resistant Protein (BCRP), and Multidrug Resistant Protein 1 (MRP1).

P-gp is the most studied ABC transporter, and the search for P-gp inhibitors as co-therapeutics to combat MDR has had little success after decades of studies. We previously used high-throughput …

The Zooarchaeological Dimension Of Preceramic Human-Environment Dynamics In The Highlands Of Southwestern Honduras, Alejandro J. Figueroa Aug 2021

The Zooarchaeological Dimension Of Preceramic Human-Environment Dynamics In The Highlands Of Southwestern Honduras, Alejandro J. Figueroa

Anthropology Theses and Dissertations

In this dissertation I evaluate different hypotheses regarding human-environment dynamics in the Mesoamerican neotropics during the Preceramic period (ca. 11000-7400 cal B.P.) by examining the largest extant faunal assemblage dated to this time. The Preceramic was characterized by major climatic and ecological changes following the end of the Pleistocene, including the extinction of megafauna and the expansion of tropical forests. This period ended with a series of behavioral adaptations suited to this transformed landscape such as increased territoriality, sedentism, agriculture, and domestication. Three hypotheses have been proposed to explain these dynamics: the Broad-Spectrum Revolution hypothesis suggests post-Pleistocene resource uncertainty and …

Quantitative Soundscape Analysis To Understand Multidimensional Features, Dylan Charles Wilford, Jennifer L. Miksis-Olds, S. Bruce Martin, Daniel R. Howard, Kim Lowell, Anthony P. Lyons, Michael James Smith

Center for Coastal and Ocean Mapping

A methodology for the analysis of soundscapes was developed in an attempt to facilitate efficient and accurate soundscape comparisons across time and space. The methodology consists of a collection of traditional soundscape metrics, statistical measures, and acoustic indices that were selected to quantify several salient properties of marine soundscapes: amplitude, impulsiveness, periodicity, and uniformity. The metrics were calculated over approximately 30 h of semi-continuous passive acoustic data gathered in seven unique acoustic environments. The resultant metric values were compared to a priori descriptions and cross-examined statistically to determine which combination most effectively captured the characteristics of the representative soundscapes. The …

Chemistry Theses and Dissertations

SYNTHESIS OF P-GLYCOPROTEIN INHIBITOR COMPOUNDS Advisor: Professor Alexander R. Lippert Doctor of Philosophy conferred: August 4th, 2021 Dissertation completed: August 4th, 2021 A problem with cancer treatment is that many cancers develop resistance to chemotherapeutic agents, causing them to fail to accumulate in resistant cancer cells long enough to have any effect. This is due to the overexpression of a plasma membrane protein called P-glycoprotein (P-gp). Generally, the role of P-gp is to protect the cells from any toxins or foreign substances by pumping these toxins (including chemotherapeutic drugs) out of the cell. I am collaborating with the Wise-Vogel laboratory …
