Forward And Reverse Genetics Of B Cell Malignancies: From Insertional Mutagenesis To Crispr-Cas, Joanna C. Dawes, Anthony G. Uren
School of Biomedical Sciences
Cancer genome sequencing has identified dozens of mutations with a putative role in lymphomagenesis and leukemogenesis. Validation of driver mutations responsible for B cell neoplasms is complicated by the volume of mutations worthy of investigation and by the complex ways that multiple mutations arising from different stages of B cell development can cooperate. Forward and reverse genetic strategies in mice can provide complementary validation of human driver genes and in some cases comparative genomics of these models with human tumors has directed the identification of new drivers in human malignancies. We review a collection of forward genetic screens performed using …

Conservation Of The Ambystoma Jeffersonianum Complex, Morgan Motherwell
Theses
The Great Lakes region is home to the diverse genus Ambystoma, including the Blue Spotted Salamander (Ambystoma laterale) and Jefferson’s Salamander (Ambystoma jeffersonianum). These two species are relevant to conservation efforts because of unique hybrid populations. This hybridization creates offspring populations that are morphologically similar to the parental species. The hybrid populations are primarily all female lineages and are known as the Ambystoma jeffersonianum complex. Conservation interest in this hybrid complex stems from the fact that hybrid populations are not taxonomically recognized as separate species and therefore are not protected under the Endangered Species Act. To assess the contribution of …
Stigma And Social-Emotional Health In Youth With Learning Differences, Kelsey Maki
Stigma And Social-Emotional Health In Youth With Learning Differences, Kelsey Maki
Doctoral Dissertations
Youth diagnosed with specific learning disorder (SLD) and/or attention deficit hyperactivity disorder (ADHD), defined collectively for the purposes of this study as youth with learning differences, experience poorer social-emotional health outcomes in comparison to their typically developing peers. These youth additionally experience stigma from their peers, teachers, and broader community, which may impact social-emotional health. As a secondary data analysis of a larger study, the present study investigated the role of stigma consciousness alongside demographic variables, self-esteem, and peer relationships in social-emotional health outcomes. Due to the possible differences between youth diagnosed with SLD, ADHD, and comorbid SLD/ADHD, group differences …

Characterization Of Metabolic Gene Expression In The Raw 264.7 Monocyte Cell Line, Lauren Macaisa
Characterization Of Metabolic Gene Expression In The Raw 264.7 Monocyte Cell Line, Lauren Macaisa
PANDION: The Osprey Journal of Research and Ideas
Immune cells undergo changes in gene expression and metabolism when activated by a foreign molecule. Although the changes are short-lived in response to an acute infection, they persist in disorders with underlying chronic inflammation. To understand how changes in gene expression relate to changes in metabolism, the expression of metabolic transporter proteins, was analyzed in response to chronic inflammation in a mouse monocyte cell line. Specifically, Glucose Transporter-1 (GLUT1), Monocarboxylate Transporter-1 (MCT1), Monocarboxylate Transporter-4 (MCT4), and Glutamate Aspartate Transporter (GLAST) were analyzed, and it was hypothesized that all the transporters would increase in expression in response to a prolonged inflammatory …
Nuclear Displacement: Effects Of America's Nuclear Tests On Pacific Islanders, Jessica Faucher
Nuclear Displacement: Effects Of America's Nuclear Tests On Pacific Islanders, Jessica Faucher
PANDION: The Osprey Journal of Research and Ideas
The United States conducted a series of nuclear tests in the Pacific that permanently displaced Marshall Islanders, specifically the indigenous population of Bikini Atoll. In addition to rendering their native environment uninhabitable, the U.S. used the bodies of the Rongelapese that they exposed to radioactive fallout as a living laboratory to study the effects of radiation. Nuclear Displacement in the Pacific has not received very much attention in historical scholarship, but nevertheless represents a well documented case of imperialism in U.S. History.

Thermal Properties Of 18f-Fdg Uptake And Imaging In Positron Emission Tomography Scans Of Cancerous Cells, Carleigh R. Eagle
PANDION: The Osprey Journal of Research and Ideas
Positron Emission Tomography (PET) scans can utilize a radioactive tracer, in this case 2-deoxy2-[fluorine-18] fluoro-D-glucose (18F-FDG), to visualize malignant tumors in cancer patients. The uptake was compared to glucose to understand the difference in thermal properties, which contribute to the ability to image the cancerous cells. The uptake of 18F-FDG by cancer cells and the imaging process of positron emission tomography were reviewed from a thermodynamic perspective. Gastrointestinal and neurological imaging techniques were reviewed to understand the role of PET imaging in different areas of the human body.

Embodied Injustices: Covid-19, Race, And Epigenetics, Maria Encinosa
PANDION: The Osprey Journal of Research and Ideas
The co-occurrence of the COVID-19 pandemic with the long-lasting effects of systemic racism has been devastating, and results in vast inequities in infection and mortality rates within communities of color. In this article, I analyze the potential for epigenetic research to operationalize the social science theory of embodiment, which describes how the social and material worlds manifest in our physical bodies. Epigenetic modifications can be triggered by environmental stressors, to which minority populations are more likely to be exposed. In turn, these stressors are linked to disorders that increase COVID-19 susceptibility. Thus, epigenetic modifications provide an avenue by which racialized …

Background: TAR DNA-binding protein 43 (TDP-43) can be found within the cell nucleus in most tissues and is a fundamental component to protein production, as it works to slice and reconfigure mRNA molecules. Recently, TDP-43 inclusions have been identified as a prevalent proteinopathy in the brains of individuals diagnosed with Alzheimer's Disease (AD). However, despite the growing body of evidence demonstrating the important role of TDP-43 in AD pathogenesis, whether and how TDP-43 proteinopathy and other AD pathological hallmarks interact remain largely unknown. Furthermore, TDP-43 has a high propensity to undergo liquid-liquid phase separation (LLPS), a biological process necessary for …

Systematic Review Of Preclinical Surgical Meshes For Hernia Repair, Andrew Hedstrom, Nathan Bills, Cynthia M. Schmidt, Mark A. Carlson
Posters: 2021 Summer Undergraduate Research Program
Hernia repair is one of the most common procedures performed around the world. Surgical mesh materials are used to reinforce hernia repairs and improve surgical outcomes. However, current commercial mesh materials produce problems such as infection, rejection, shrinkage, adhesion, and mechanical failure. These problems contribute to a high rate of hernia recurrence and mesh removal. With the number of hernias expected to rise in the future due to increases in obesity and diabetes, the need for more effective mesh materials is critical. This review aims to identify, evaluate, and summarize experimental surgical mesh materials used in the preclinical stages of …

Temporal And Spatial Variations In Three-Dimensional Seismic Oceanography, Zheguang Zou, Parsa Bakhtiari Rad, Leonardo Macelloni, Likun Zhang
Faculty and Student Publications
Seismic oceanography is a new cross-discipline between geophysics and oceanography that uses seismic reflection data to image and study the oceanic water column. Previous work on seismic oceanography was largely limited to two-dimensional (2D) seismic data and methods. Here we explore and quantify temporal and spatial variations in three-dimensional (3D) seismic oceanography to address whether 3D seismic imaging is meaningful in all directions and how one can take advantage of the variations. From a 3D multichannel seismic survey acquired for oil and gas exploration in the Gulf of Mexico over a 6-month period, a 3D oceanic seismic volume was derived. …

Department of Nutrition and Health Sciences: Faculty Publications
We sought to characterize the lipid profile of skeletal muscle cell-derived Extracellular Vesicles (EVs) to determine if a hypertrophic stimulus would affect the lipid composition of C2C12 myotube-derived EVs. Analyses included C2C12 murine myoblasts differentiated into myotubes and treated with Insulin-Like Growth Factor 1 (IGF-1) for 24 h to induce hypertrophic growth. EVs were isolated from cell culture media, quantified using Nanoparticle Tracking Analysis (NTA) and analyzed using Transmission Electron Microscopy (TEM). EVs were homogenized and lipids extracted for quantification by Mass Spectrometry followed by downstream lipid class enrichment and lipid chain analysis. IGF-1 treatment elicited an increase in CD63 …

Pancreatic cancer is a highly aggressive human cancer and the third leading cause of death due to cancer. Cancer stems cells (CSC) are a small population of cancer cells that mediate tumorigenesis, metastasis, and resistance to standard treatments. By specifically identifying and targeting CSC maintenance genes, the efficiency of treatment modalities can be improved. PAF1 (RNA Polymerase II-Associated Factor 1), also known as PD2 (Pancreatic Differentiation 2) maintains pluripotency of stem cells and is a marker of pancreatic cancer stem cells. It is upregulated in poorly differentiated pancreatic cancer cells. Gemcitabine is a novel deoxycytidine analogue which has been developed …

Many chemotherapy drugs cause female infertility by permanently shutting down the ovaries. In pre-pubescent girls, this can also result in abnormal sexual development. It is known that many of these harmful chemotherapy drugs bind to cKIT receptors throughout the body. cKIT receptors exist in particularly high concentrations in the ovaries and when they are blocked by chemotherapy drugs, premature ovarian failure results. Our experiment uses a transgenic mouse model to evaluate the effects of cKIT receptors on oocyte formation and development. This experiment is part of a larger effort to understand the mechanisms by which chemotherapy drugs cause premature ovarian …

Background/Objective: Comorbidities can contribute to increased risk for mortality and disability in individuals with rheumatoid arthritis (RA)1,2. The Rheumatic Disease Comorbidity Index (RDCI) assesses 11 comorbidities and produces a weighted score (0-9) that accurately predicts several health outcomes3. The RDCI was developed with self-report data and later validated with ICD-9-CM codes collected from administrative data3,4. On October 1, 2015, the U.S. transitioned to ICD-10-CM, resulting in a nearly five-fold increase in the number of codes available to classify conditions5. Our objective was to update the RDCI by translating it into ICD-10-CM.

Lung cancer is the leading cause of cancer deaths accounting for about 22% of all cancer related cases in both males and females. Lung cancers are broadly grouped into two types mainly small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC) with SCLC accounting for about 15% of all lung cancer cases. SCLC is different from NSCLC because in most cases it originates centrally in the bronchi and is frequently seen in smokers. SCLC is aggressive and one of the most malignant forms of tumor characterized by uncontrolled rapid growth of certain cells in the lungs. SCLC displays …

Phosphatidylethanolamine N-methyltransferase (PEMT) is an enzyme that catalyzes the successive transfer of 3 methyl groups from S-adenosylmethionine (SAM) to phosphatidylethanolamine (PE) to generate phosphatidylcholine (PC). PC is vital for exporting fat out of the liver, ultimately preventing hepatic steatosis. Alcohol also induces steatosis partly through damaging this pathway, so the purpose of this study was to investigate the relationship between alcohol and PEMT in the liver. PEMT -/- (KO) and wild-type (WT) mice were subjected to a chronic + binge alcohol treatment, and both serum and liver samples were analyzed. Triglyceride quantification, SAM and S-adenosylhomocysteine (SAH) levels, and histological analyses …
