Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has killed more than 4 million humans globally, but there is no bona fide Food and Drug Administration–approved drug-like molecule to impede the COVID-19 pandemic. The sluggish pace of traditional therapeutic discovery is poorly suited to producing targeted treatments against rapidly evolving viruses. Here, we used an affinity-based screen of 4 billion DNA-encoded molecules en masse to identify a potent class of virus-specific inhibitors of the SARS-CoV-2 main protease (Mpro) without extensive and time-consuming medicinal chemistry. Four new lithophilic species of Scorpiops Peters, 1861 (Scorpiopidae) have been described from peninsular India using an integrated taxonomic approach. Three species Scorpiops vrushchik sp. n., S. nagphani sp. n. and S. neera sp. n. have been described from northern Western Ghats and S. lioneli sp. n. is described from Deccan Plateau. S. vrushchik sp. n. is closely related to S. tenuicauda and S. telbaila and differs by a raw genetic divergence of 9.6-9.8% from S. tenuicauda and 11% from S. telbaila. S. nagphani sp. n. is closely related to S. lioneli sp. n. and S. maharashtraensis and differs by …

Exercise-associated hyponatremia (EAH) has been reported to develop during endurance events such as triathlons and marathons. As these events become more popular, the incidence of developing EAH also increases. The development of EAH is commonly associated with the overconsumption of hypotonic fluids such as water and tends to be more prevalent in females. Resistance to anthracnose, dry matter yield (DMY) and seed yield (SY) was assessed for germplasm and breeding lines of Stylosanthes in Brazil, India and China. Overall, Stylosanthes guianensis produced higher DMY than S. scabra, S. capitata and S. macrocephala at most sites in Brazil. Data from China suggest that there are high yielding anthracnose resistant S. guianensis lines that can reduce the reliance on CIAT 184. S. seabrana might also prove successful. S. seabrana in India produced the highest DMY and SY and it can form nodules with native Bradyrhizobium strains. Regional differences in resistance within accessions stress the …

Background
Environmental fluctuation during embryonic and fetal development can permanently alter an organism’s morphology, physiology, and behaviour. This phenomenon, known as developmental plasticity, is particularly relevant to reptiles that develop in subterranean nests with variable oxygen tensions. Previous work has shown hypoxia permanently alters the cardiovascular system of snapping turtles and may improve cardiac anoxia tolerance later in life. The mechanisms driving this process are unknown but may involve epigenetic regulation of gene expression via DNA methylation. This paper examines university administration and theology based on the author’s growing appreciation for the writings of Christian theologian, Reinhold Niebuhr (1892-1971), while serving in the senior administrative roles of provost and chief of staff at Fayetteville State University in North Carolina. One of the leading Protestant theologians of the twentieth century, Niebuhr is perhaps best known as the writer of the “Serenity Prayer,” but his analyses of political and social events of the early and midtwentieth century influenced Martin Luther King, Jr., Jimmy Carter, and Barack Obama, among many others. Background: Few studies have examined a reciprocal parallel process longitudinal trajectory of adolescent substance use and mental health problems simultaneously, and none have described this parallel process for adolescents receiving outpatient substance use treatment. No studies have examined how social support may account for deviations outside the overall trajectories for substance use or mental health problems. Method: Secondary data analysis examined a longitudinal parallel process latent growth model between mental health and substance use problems among adolescents who received outpatient substance use treatment. Objectives People living with HIV (PLHIV) in the USA, particularly women, have a higher prevalence of food insecurity than the general population. Cigarette smoking among PLHIV is common (42%), and PLHIV are 6–13 times more likely to die from lung cancer than AIDS related causes. This study sought to investigate the associations between food security status and smoking status and severity among a cohort of predominantly low-income women of colour living with and without HIV in the USA.
Design Women enrolled in an ongoing longitudinal cohort study from 2013 to 2015.

BACKGROUND: Cerebral glucose hypometabolism is consistently observed in individuals with Alzheimer's disease (AD), as well as in young cognitively normal carriers of the Ε4 allele of Apolipoprotein E (APOE), the strongest genetic predictor of late-onset AD. While this clinical feature has been described for over two decades, the mechanism underlying these changes in cerebral glucose metabolism remains a critical knowledge gap in the field.
METHODS: Here, we undertook a multi-omic approach by combining single-cell RNA sequencing (scRNAseq) and stable isotope resolved metabolomics (SIRM) to define a metabolic rewiring across astrocytes, brain tissue, mice, and human subjects expressing APOE4.

Disrupted systemic copper (Cu) homeostasis underlies neurodegenerative diseases with early symptoms including olfactory dysfunction. This study investigated the impact of Cu dyshomeostasis on olfactory function, adult neurogenesis, and neurochemical balance. Models of Cu deficiency (CuD) and Cu overload (CuO) were established by feeding adult rats with Cu-restricted diets plus ip. injection of a Cu chelator (ammonium tetrathiomolybdate) and excess Cu, respectively. CuD reduced Cu levels in the olfactory bulb (OB), subventricular zone (SVZ), rostral migratory stream (RMS), and striatum, while CuO increased Cu levels in these areas. The mammalian DNA methylome is formed by two antagonizing processes, methylation by DNA methyltransferases (DNMT) and demethylation by ten-eleven translocation (TET) dioxygenases. Although the dynamics of either methylation or demethylation have been intensively studied in the past decade, the direct effects of their interaction on gene expression remain elusive. Here, we quantify the concurrence of DNA methylation and demethylation by the percentage of unmethylated CpGs within a partially methylated read from bisulfite sequencing. After verifying 'methylation concurrence' by its strong association with the co-localization of DNMT and TET enzymes, we observe that methylation concurrence is strongly correlated with gene expression. …

Platelets play a key role in thrombosis and hemostasis. Platelet count (PLT) and mean platelet volume (MPV) are highly heritable quantitative traits, with hundreds of genetic signals previously identified, mostly in European ancestry populations. We here utilize whole genome sequencing from NHLBI's Trans-Omics for Precision Medicine Initiative (TOPMed) in a large multi-ethnic sample to further explore common and rare variation contributing to PLT (n = 61 200) and MPV (n = 23 485). BACKGROUND: Selinexor is an oral, selective nuclear export inhibitor. STORM was a phase 2b, single-arm, open-label, multicenter trial of selinexor with low dose dexamethasone in patients with penta-exposed relapsed/refractory multiple myeloma (RRMM) that met its primary endpoint, with overall response of 26% (95% confidence interval [CI], 19 to 35%). Health-related quality of life (HRQoL) was a secondary endpoint measured using the Functional Assessment of Cancer Therapy - Multiple Myeloma (FACT-MM). This study examines impact of selinexor treatment on HRQoL of patients treated in STORM and reports two approaches to calculate minimal clinically important differences for the FACT-MM.

Military installations are valuable in global biodiversity conservation as they secure representative ecosystems from land conversion and protect many threatened or endangered species. Selecting suitable areas for biodiversity conservation within military installations is a challenging problem as this must not impede military training activities. The issue gets more complicated when considering multiple cohabiting species in a metacommunity with species dependency. In this paper, we present an example for the conservation of two cohabiting species, Gopher Tortoise (GT) and Gopher Frog (GF), located within the boundaries of a military installation, Fort Stewart, Georgia, United States. Cytosolic double-stranded RNA (dsRNA) initiates type I IFN responses. Endogenous retroelements, notably Alu elements, constitute a source of dsRNA. Adenosine-to-inosine (A-to-I) editing by ADAR induces mismatches in dsRNA and prevents recognition by MDA5 and autoinflammation. To identify additional endogenous dsRNA checkpoints, we conducted a candidate screen in THP-1 monocytes and found that hnRNPC and ADAR deficiency resulted in synergistic induction of MDA5-dependent IFN responses. RNA-seq analysis demonstrated dysregulation of Alu-containing introns in hnRNPC-deficient cells via utilization of unmasked cryptic splice sites, including introns containing ADAR-dependent A-to-I editing clusters. This work aimed to determine the genetic diversity of Colletotrichum gloeosporioides infecting Stylosanthes spp. in Brazil, China and India. A total of 132 isolate originating from S. seabrana, S. macrocephala, S. capitata, S. scabra, and S. guianensis were used. Four major genetic groups were identified from an analysis of genetic diversity using selection-neutral DNA markers. Group 1 contained 20 isolates and this may represent a genotype that migrated from the center of diversity in Brazil and Colombia to Australia, Thailand and India. The investigation was carried out on a densely wooded area in the Mopani savanna of South Africa. Seven plots were subjected to different intensities of tree thinning, ranging from a totally cleared plot (0 %) to plots thinned to the equivalent of 10 %, 20 %, 35 %, 50 % and 75 % of the leaf biomass of a control plot (100 %). The grasses responded positively to the tree thinning in terms of total dry matter yield. Growth and senescence are very important determinants of grassland productivity and correspond to key features to be considered for grazing management purposes. The present study was carried out at ESALQ, Piracicaba, S.P., Brazil and evaluated growth and senescence as a function of sward surface height. Mathematical models generated revealed that a similar pattern of response occurred as described in the literature for temperate grass species. However, a seasonal variation in response curves for growth and senescence was also observed and could be related to climatic events. Seven timothy (Phleum pratense L.) cultivars were evaluated over three pasture seasons under rotational grazing to 7 or 11 cm post-grazing heights and regrowth periods of four to seven weeks. There was a trend for the pasture type timothy cultivars to outyield Champ timothy (check). Dry matter yields were lower for the 7 cm than 11 cm post-grazing height. Although the overall cultivar x post-grazing height interaction was not significant (P > 0.05), there was variability in grazing tolerance among timothy cultivars. A technique for assessing the fate of seed grazed by sheep in small field plots was examined using the pasture species Trifolium michelianum cv. Paradana, Medicago truncatula cv. Mogul, Trifolium spumosum and Trigonella balansae. Trifolium spumosum lost the greatest proportion of seed from dry pasture residues (88% seed eaten), whereas Trigonella balansae, Paradana and Mogul lost 61%, 55% and 53% respectively. However, Trifolium spumosum and Paradana had high levels of seed in the faeces (56.8 kg/ha and 65.2 kg/ha respectively), whereas Mogul and Trigonella balansae had low levels (10.3 kg/ha and 11.0 kg/ha respectively). Noise-Induced Hearing Loss (NIHL) is a global health problem, often underestimated, and leads to permanent hearing loss. The prevalence of NIHL in adolescents has increased in line with the increasing use of cell phones. This study aimed to determine the prevalence of NIHL and analyze the population at risk (age, gender, and school) in adolescent students. The study design was a cross-sectional study.
