Exercise intolerance (EI) is the primary manifestation of chronic heart failure with preserved ejection fraction (HFpEF), the most common form of heart failure among older individuals. The recent recognition that HFpEF is likely a systemic, multiorgan disorder that shares characteristics with other common, difficult-to-treat, aging-related disorders suggests that novel insights may be gained from combining knowledge and concepts from aging and cardiovascular disease disciplines. This state-of-the-art review is based on the outcomes of a National Institute of Aging-sponsored working group meeting on aging and EI in HFpEF. Developing new, cheap, sensitive and selective chemiluminescence (CL) systems with multiple detection properties is still a big challenge for biological and environmental applications. Here, we report a new CL system having multiple detection applications (environmental and biological). The developed lucigenin-pyrogallol system gave an enhancement (190 times) over the conventional lucigenin-H2O2 CL system. Cobalt ion and tyrosinase can inhibit lucigenin-PG CL intensity. Based on these phenomena, we have developed new CL methods for the detection of pyrogallol (LOD = 0.94 μM), lucigenin (LOD = 0.42 μM), cobalt (LOD = 68 μM), and tyrosinase (LOD = 1.8 μg/mL). A novel magnetic adsorbent solid-phase Fe3O4@ZIF-8 extraction method coupled with UHPLC Q-Orbitrap high resolution mass spectrometry for the sensitive and high efficiency detection of fluoroquinolones, tetracyclines, dyes and amphenicols in eggs is described. Under optimum conditions, CCβ and CCα of 0.62 to 26.97 μg.kg–1 and 0.36 to 16.37 μg.kg–1 was obtained, which is much better than that obtained by conventional methods. The accuracy, expressed as recovery, were achieved falling between 75% and 104%. Herein, we describe a rapid, sensitive, and nondestructive method—extractive nanoelectrospray ionization-mass spectrometry (EnESI-MS)—for traditional Chinese medicine (TCM) authentication. The mass-spectral fingerprints of volatile compounds released from various TCMs can be rapidly acquired using EnESI-MS without sample pretreatment. EnESI-MS was applied to successfully differentiate between two commonly used medicinal herbs, Schisandra Chinensis and Schisandra sphenanthera, which are morphologically similar but exhibit different therapeutic effects. Specific volatile compounds of each herb in a ten-component Chinese herbal product, Jia Wei Xiao Yao San, were also identified, and the method was applied to discriminate between the commercial product and a substandard version.

Lactoferrin (LF) is secreted by mammals and displays extensive biological effects. We previously reported that bovine LF (BLF) can boost the cold tolerance of a well-applied probiotic strain, Lactobacillus rhamnosus GG (LGG), to grow robustly under a cold environment, but the molecular mechanism is not clear. Here, RNA-seq analysis was conducted to ascertain molecular pathways underlying cold tolerance exerted by BLF. LGG was cultured in a cold environment (22°C) in the presence or absence of BLF. Humanized bone marrow-liver-thymic (hu-BLT) mice develop a functional immune system in periphery, nevertheless, have a limited reconstitution of human myeloid cells, especially microglia, in CNS. Further, whether bone marrow derived hematopoietic stem and progenitor cells (HSPCs) can enter the brain and differentiate into microglia in adults remains controversial. To close these gaps, in this study we unambiguously demonstrated that human microglia in CNS were extensively reconstituted in adult NOG mice with human interleukin-34 transgene (hIL34 Tg) from circulating CD34+ HSPCs, nonetheless not in hu-BLT NOG mice, providing strong evidence that human CD34+ HSPCs can enter adult brain and differentiate into …

Abstract The seagrass Zostera marina (eelgrass) is typically found in shallow protected areas of portions of eastern Long Island Sound (LIS). Although the species and its associated ecosystem have been well studied in many places, work in LIS has been limited. A field study was conducted to assess the fouling communities associated with eelgrass beds around Pine Island, in eastern LIS. Samples were collected from four sites between July and October, 2011. A total of 15 species of fouling organisms were found on eelgrass leaves throughout the study period. Recruitment patterns varied spatially and temporally. Differences in the composition of …

Our research aimed to evaluate the effectiveness of the democracy and human rights course taught with a “Learning Model Based on Democratic Life.” The study was conducted with an action research design. The research study group, determined using purposeful sampling, consisted of 10 students taking a Democracy and Human Rights course at a state university in Turkey. Data were collected using “Process Evaluation Forms” and “Student Letters.” Content analysis was used in the analysis of the data. Background: We previously reported that the tomato glycoalkaloid tomatine inhibited the growth of Trichomonas vaginalis strain G3, Tritrichomonas foetus strain D1, and Tritrichomonas foetus-like strain C1 that cause disease in humans and farm and domesticated animals. The increasing prevalence of antibiotic resistance requires development of new tools to enhance or replace medicinal antibiotics. Methods: Wild tomato plants were harvested and divided into leaves, stems, and fruit of different colors: green, yellow, and red. Samples were freeze dried and ground with a handheld mill. The resulting powders were evaluated for their potential anti-microbial effects on protozoan parasites, bacteria, and fungi. A …

Background: Grewia asiatica Linn, or phalsa, is a commonly consumed fruit in Pakistan. The fruit is employed in the traditional medicine practice of Pakistan as a smooth muscle relaxant in different gastrointestinal (GI) and cardiovascular diseases. In this investigation, we show the antispasmodic and vasorelaxant actions of Grewia asiatica fruit extract.
Methods: A 70% methanolic crude extract of the plant material was prepared (Ga.Cr). Different isolated GI tissue preparations and endothelium-intact aortas from rats were utilized to observe the pharmacological actions of the extract.

Mitonuclear Mismatch Is Associated With Increased Male Frequency, Outcrossing, And Male Sperm Size In Experimentally-Evolved C. Elegans, Brent William Bever

We provide the first controlled study of how male frequencies and rates of outcrossing evolve in response to mitonuclear mismatch by allowing replicate lineages of C. elegans nematodes containing either mitochondrial or nuclear mutations of electron transport chain (ETC) genes to evolve under three sexual systems: facultatively outcrossing (wildtype), obligately selfing, and obligately outcrossing. In partial support of a tenet of the mitonuclear sex hypothesis, which predicts that outcrossing will be favored in cases of mitonuclear mismatch, we found evolution of increased male frequency in at least one replicate line of all four ETC mutant backgrounds tested--nuclear isp-1, mitochondrial …

The vitamin D hormone, 1,25dihydroxyvitamin D3 (1,25(OH)2D3), and related compounds derived from vitamin D3 or lumisterol as a result of metabolism via the enzyme CYP11A1, have been shown, when applied 24 hours before or immediately after UV irradiation, to protect human skin cells and skin from DNA damage due to UV exposure, by reducing both cyclobutane pyrimidine dimers (CPD) and oxidative damage in the form of 8‐oxo‐7,8‐dihydro‐2’‐ deoxyguanosine (8‐OHdG). We now report that knockdown of either the vitamin D receptor or the endoplasmic reticulum protein ERp57 by siRNA abolished the reductions in UV‐induced DNA damage with 20‐hydroxyvitamin D3 or 24‐hydroxylumisterol3, …

The purpose of this presentation is to show through RAM-based practice, understanding the suffering of people in various situations leads to the healing of patients, and it is possible for students to find meaning in each patient's situation. The faculty and students have learned from the RAM how to adapt to the environment, and the coping ability of patients has a great effect on the everyone's nursing practice.

Loblolly pine (Pinus taeda L.) is a commercially important timber species planted across a wide temperature gradient in the southeastern U.S. It is critical to ensure that the planting stock is suitably adapted to the growing environment to achieve high productivity and survival. Long-term field studies, although considered the most reliable method for assessing cold hardiness of loblolly pine, are extremely resource-intensive and time-consuming. The development of a high-throughput screening tool to characterize and classify freeze tolerance among different genetic entries of seedlings will facilitate accurate deployment of highly productive and well-adapted families across the landscape. Introduction: Past studies demonstrate that stress and anxiety affect emergency medicine physicians, but the causal factors identified are usually from sources outside the work shift. We attempt to show the relationship between intrinsic factors of a work shift and anxiety perceived by residents, while also examining differing gender responses.
Methods: In 2018, a cross-sectional survey of emergency medicine residents in the United States was distributed anonymously through the Emergency Medicine Residents Association. The survey consisted of demographic questions, novel questions identifying intrinsic factors, and the Generalized Anxiety Disorder 7-item (GAD-7) scale. BACKGROUND: Comprehensive testing of cognitive functioning is standard practice in studies of Alzheimer disease (AD). Short-form tests like the Montreal Cognitive Assessment (MoCA) use a "sampling" of measures, administering key items in a shortened format to efficiently assess cognition while reducing time requirements, participant burden, and administrative costs. We compared the MoCA to a commonly used long-form cognitive battery in predicting AD symptom onset and sensitivity to AD neuroimaging biomarkers.

Skeletal muscle contraction depends on activation of clustered acetylcholine receptors (AchRs) and muscle-specific Na+ channels (Nav1.4). Some Nav1.4 channels are highly enriched at the neuromuscular junction (NMJ) and their clustering is thought to be essential for effective muscle excitation. However, this has not been experimentally tested, and how NMJ Na+ channels are clustered is unknown. Here, using muscle-specific AnkyrinR, AnkyrinB, and AnkyrinG single, double, and triple-conditional knockout mice we show that Nav1.4 channels fail to cluster only after deletion of all three ankyrins. The aim of the present study was to asses the influence of the frequency of defoliation and density of sowing on yield and nutritive value of Coursetia ferruginea. Three defoliation frequencies (6, 8 and 10 weeks) and three densities of planting (1 x 1 m, 1 x 1.5 m and 1 x 2 m) were evaluated. A randomized block design, in a split plot arrangement and four repetitions, was used. Frequency of defoliation has a strong significant (P< 0.01) effect on dry matter (DM) yield (13.6, 18.1 and 27.8 g DM/plant for 6, 8 and 10-week defoliation frequencies), plant height (63.5, 76.6 and 101.2 cm) and plant survival (71, 74 and 86%). Nutritive value was significantly (P< 0.05) affected by the frequency of defoliation. Mineral content was reduced as the defoliation interval increased. Density of planting did not affect (P> 0.05) any of the variables. It is concluded that, C. ferruginea is a forage shrub with potential to improve animal feed …

The leaf elongation rate of a temperate (Festuca arundinacea Schreb.) and a Mediterranean (F. arundinacea var. letourneuxiana) tall fescue cultivar in response to water deficit was studied in a glasshouse experiment. Plants of both cultivars were grown in the same containers and received water daily with gradation in intensity of water deficit achieved by varying the daily water ration per container. Leaf elongation rate (LER) was calculated for eight successive subperiods during the application of the water treatments. The objective of this work was to estimate dry matter production of Cynodon grasses as a function of photothermal units (PU). Total PU values were calculated for the period from February 1 to April 24, 1996, from a location in southeastern Brazil. Regression analysis was run between PU and herbage production from a published data set on five Cynodon cultivars, with an r2 of 0.9983. Differences between measured and estimated values were under 5%. Model testing using an independent data set on herbage dry matter production from grazed Cynodon pastures showed an overestimation, probably due to water deficit during …
