Background and purpose: In recent years, questions regarding the genetic and environmental factors affecting variation in human craniofacial morphology have received increasing attention. Medical and clinical genetic research using family studies has proven foundational for our understanding of which genes affect craniofacial variation. On the other hand, cephalometricbased studies showed a relationship between the cranial base and midface characteristics among individuals with different skeletal pattern of malocclusion. Although data describe the presence and increase of inflammatory mediators in the local environment in periodontitis vs. health in humans, details regarding how these responses evolve in the transition from health to disease, changes during disease progression, and features of a resolved lesion remain unknown. This study used a nonhuman primate model of ligature-induced periodontitis in young, adolescent, adult, and aged animals to document features of inflammatory response affected by age. Among novel compounds under recent investigation as potential new antimalarial drugs are three independently developed inhibitors of the Plasmodium falciparum P-type ATPase (PfATP4): KAE609 (cipargamin), PA92, and SJ733. We assessed ex vivo susceptibilities to these compounds of 374 fresh P. falciparum isolates collected in Tororo and Busia districts, Uganda, from 2016 to 2019. Median IC50s were 65 nM for SJ733, 9.1 nM for PA92, and 0.5 nM for KAE609. Sequencing of pfatp4 for 218 of these isolates demonstrated many nonsynonymous single nucleotide polymorphisms; the most frequent mutations were G1128R (69% of isolates mixed or mutant), Q1081K/R (68%), G223S (25%), N1045K …

Rural Students' College Choice And The Impact Of Dual Enrollment Programs And College Cost, Elise J. Cain

This article explores the college-choice processes of two college students who graduated high school from rural areas in New York State. Hossler and Gallagher’s (1987) three-stage model of the college-choice process was used as the theoretical framework for this study. To gain in-depth knowledge about students’ predisposition, search, and choice stages, narrative inquiry was employed. This article specifically highlights how dual enrollment courses played key roles within the college-choice processes of these students. These findings suggest that there is a need for additional education about college costs and dual enrollment programs for rural students and their families.

Trace Fossils, Algae, Invertebrate Remains And New U-Pb Detrital Zircon Geochronology From The Lower Cambrian Torneträsk Formation, Northern Sweden, Stephen Mcloughlin, Vivi Vajda, Timothy P. Topper, James L. Crowley, Fan Liu, Ove Johansson, Christian B. Skovsted

Nineteen ichnotaxa, together with algal and invertebrate remains, and various pseudo-traces and sedimentary structures are described from the Torneträsk Formation exposed near Lake Torneträsk, Lapland, Sweden, representing a marked increase in the diversity of biotic traces recorded from this unit. The “lower siltstone” interval of the Torneträsk Formation contains mostly simple pascichnia, fodinichnia and domichnia burrows and trails of low-energy shoreface to intertidal settings. The assemblage has very few forms characteristic of high-energy, soft-sediment, foreshore or upper shoreface environments (representative of the Skolithos ichnofacies).

Gout is the most frequent form of inflammatory arthritis in the world. Its prevalence is particularly elevated in specific geographical areas such as in the Oceania/Pacific region and is rising in the US, Europe, and Asia. Gout is a severe and painful disease, in which co-morbidities are responsible for a significant reduction in life expectancy. Expansion of the polyglutamine tract in the N terminus of Ataxin-1 is the main cause of the neurodegenerative disease, spinocerebellar ataxia type 1 (SCA1). However, the C-terminal part of the protein - including its AXH domain and a phosphorylation on residue serine 776 - also plays a crucial role in disease development. This phosphorylation event is known to be crucial for the interaction of Ataxin-1 with the 14-3-3 adaptor proteins and has been shown to indirectly contribute to Ataxin-1 stability. In vitro MICs and in vivo pharmacodynamics of ceftazidime and cefepime human-simulated regimens (HSR) against modified carbapenem inactivation method (mCIM)-positive Pseudomonas aeruginosa isolates harboring different OXA-10-like subtypes were described. The murine thigh model assessed ceftazidime (2 g every 8 h [q8h] HSR) and cefepime (2 g and 1 g q8h HSR). Phenotypes were similar despite possessing OXA-10-like subtypes with differing spectra. Ceftazidime produced ≥1-log10 killing in all isolates. Cefepime activity was dose dependent and MIC driven. This approach may be useful in assessing the implications of β-lactamase variants.

Most breast cancer deaths are caused by estrogen receptor-α-positive (ER+) disease. Preclinical progress is hampered by a shortage of therapy-naïve ER+ tumor models that recapitulate metastatic progression and clinically relevant therapy resistance. Human prolactin (hPRL) is a risk factor for primary and metastatic ER+ breast cancer. Because mouse prolactin fails to activate hPRL receptors, we developed a prolactin-humanized Nod-SCID-IL2Rγ (NSG) mouse (NSG-Pro) with physiological hPRL levels. Here, we show that NSG-Pro mice facilitate establishment of therapy-naïve, estrogen-dependent PDX tumors that progress to lethal metastatic disease. Aim The aim was to evaluate the efficacy of interrupted course of antitumor necrosis factor alpha (TNF-α) blocking agents (etanercept and adalimumab) as 3-month-on-and-3-month-off regimen versus combined regimen of physiotherapy and NSAIDs in patients with ankylosing spondylitis (AS) within a 2-year period. Patients and methods A total of 60 patients who fulfilled the Modified New York Criteria for AS were enrolled in the current study and divided into two equal groups: one received interrupted course of anti-TNFα agents and one received NSAIDs plus physiotherapy. Introduction: Human onchocerciasis caused by the filarial nematode parasite Onchocerca volvulus remains a major cause of debilitating disease infecting millions primarily in Sub-Saharan Africa. The development of a prophylactic vaccine, along with mass drug administration, would facilitate meeting the goal of onchocerciasis elimination by 2030.

Areas covered: Models used to study immunity to Onchocerca include natural infection of cattle with Onchocerca ochengi and O. volvulus infective third-stage larvae implanted within diffusion chambers in mice. Prior researchers have observed the effect of simulated reduced-gravity exercise. However, the extent to which lower-body positive-pressure treadmill (LBPPT) walking alters kinematic gait characteristics is not well understood. The purpose of the study was to investigate the effect of LBPPT walking on selected gait parameters in simulated reduced-gravity conditions. Twenty-nine college-aged volunteers participated in this cross-sectional study. Participants wore pressure-measuring insoles (Medilogic GmBH, Schönefeld, Germany) and completed three 3.5-min walking trials on the LBPPT (AlterG, Inc., Fremont, CA, USA) at 100% (normal gravity) as well as reduced-gravity conditions of 40% and 20% body weight (BW). The current standard of practice to manage postoperative pain includes the use of opioids. The purpose of this evidence-based project was to evaluate the effectiveness of music listening as an adjuvant to traditional pain management strategies to relieve postoperative pain and to assess the dosage of opioids used to help manage postoperative pain. Kolcaba's Comfort Theory was used to guide this evidence-based project.

Chagas’ disease (also known as American trypanosomiasis) and leishmaniasis are two major classes of NTDs. Expansion of the polyglutamine tract in the N terminus of Ataxin-1 is the main cause of the neurodegenerative disease, spinocerebellar ataxia type 1 (SCA1). However, the C-terminal part of the protein - including its AXH domain and a phosphorylation on residue serine 776 - also plays a crucial role in disease development. This phosphorylation event is known to be crucial for the interaction of Ataxin-1 with the 14-3-3 adaptor proteins and has been shown to indirectly contribute to Ataxin-1 stability. Determining Effective Drug Concentrations For Selection And Counterselection Genetics In Drosophila Melanogaster, Nick Matinyan, Yezabel Gonzalez, Herman A Dierick, Koen J T Venken

We recently integrated into fly genetics a set of four selection and two counterselection markers and their corresponding drugs that can be used individually or in combination. These markers eliminate the need to visually screen progeny. Before using these markers in new genetic backgrounds, effective selection/counterselection concentrations should be established for each marker/drug combination. This protocol describes how to set up, perform, and analyze a drug titration curve to determine the effective selection/counterselection drug concentrations for their corresponding markers.
