The use of three-dimensional (3D) culture systems (hydrogels) and adipose-derived stem cells (ADSCs) in regenerative medicine to advance early-stage investigation and modeling of the mechanisms of diseases, treatments, targets, etc. has recently increased. ADSCs, specifically, are utilized due to their innate programming during embryogenesis and in adult tissues in addition to their ability to differentiate into mesodermal, endodermal, and ectodermal cell-specific lineages. The microfluidic concentration gradient generator (μCGG) is an important device to generate and maintain concentration gradients (CGs) of biomolecules for understanding and controlling biological processes. However, determining the optimal operating parameters of μCGG is still a significant challenge, especially for complex CGs in cascaded networks. To tackle such a challenge, this study presents multi-fidelity surrogate and reduced-order model-based optimization methodologies for accurate and computationally efficient design of μCGGs.

Evidence suggests attention deficit/hyperactivity disorder (ADHD) may be associated with an increased incidence of concussion; however, no study has cross-sectionally (Aim 1) and longitudinally (Aim 2) assessed the associations of ADHD with concussion. Additionally, there is reason to expect cognitive alterations following concussion in athletes with ADHD. Further, there is currently no study that has evaluated the influence of common medications often taken to mitigate some symptoms of ADHD, such as methylphenidate and amphetamines. Brands have always exuded personality and status to consumers in order to promote a competent impression and induce positive perceptions and behaviors (Griskevicius, et al., 2007). In order for individuals to identify with brands, the brands must become more personable by consciously embodying a specific personality trait. The main topics of this dissertation are: 1) the study of hydrophilic-lipophilic balance (HLB) polymer monoliths for solid-phase extraction (SPE) applications, 2) molecularly imprinted polymers (MIPs) for capturing the CO2 gas and 3) the application of symmetry-adapted perturbation theory (SAPT) calculations to examine the origins of non-covalent interactions.

Several vector-borne diseases have recently resurfaced, and many have suddenly spread to new areas in the United States (U.S.) due to climate change and tropicalization. Two vectors of public health importance are Aedes aegyptiand Aedes albopictus. The Southeastern region of the U.S. appears to be a preferential ecological niche for both Aedes species. Aedes aegyptiand Aedes albopictus are highly susceptible to environmental conditions, such as humidity, precipitation, temperature. This thesis is motivated by real world problems faced in aquatic environments. It addresses the problem of area coverage path planning with robots - the problem of moving an end-effector of a robot over all available space while avoiding existing obstacles. The problem is considered first in a 2D space with a single robot for specific environmental monitoring operations, and then with multi-robot systems — a known NP-complete problem. Next we tackle the coverage problem in 3D space - a step towards underwater mapping of shipwrecks or monitoring of coral reefs.

This study involved educational innovation and implementation of a task-based, learner-centered curriculum within a foreign language program. The concerns-based assessment model (originally proposed by Hall et al., 1973, and further developed by Hall & Hord, 1987; 2011) was used to investigate the degree to which language instructors adopted the innovation. An adapted form of the concerns-based assessment model’s levels of use focused interviews and instructional artifacts were used to measure the level of implementation of the task-based approach. Most youth form relationships with extra-familial adults, sometimes called natural mentoring relationships, and these connections appear to benefit youth in several ways. Previous research demonstrates that the presence of a mentor can positively impact student outcomes including educational expectation, educational attainment, and social success. However, little research has considered how the impact of a natural mentorship may differ based upon the role of the adult in the youth’s life. Coastal wetland mapping is often difficult because of the heterogeneous vegetation compositions and associated tidal effects. Past studies in the Gulf/Atlantic coast states have reported acute marsh dieback events in which marsh rapidly browned and thinned, leaving stubble of dead stems or mudflad with damaged ecosystem services. Reported marsh dieback in South Carolina (SC), USA, however, have been limited. Previous studies have suggested a suite of possibly abiotic and biotic attributes responsible for salt marsh dieback. Numerous studies linked endoplasmic reticulum (ER) stress and unfolded protein response (UPR) to physiological responses to stress and various human pathologies, including metabolic disorders. Although variability in stress responses among individuals has been widely recognized, how such variations in the UPR affect susceptibility to diseases remains unknown. Using outbred deer mice as a model, we explored the consequences of the differential propensity to UPR at the transcriptomic, cellular, and organismic level.

CDK8 and its homolog CDK19 are essential for transcription regulation and their dysregulation has been identified in numerous diseases, especially in cancers. CDK8 and CDK19 have been implicated in cancer development through their kinase activity and kinase independent functions. Despite the existing CDK8/19 inhibitors, there is still a need to develop novel CDK8/19 inhibitors with improved potency and PK profile. It is also necessary to develop CDK8/19 degraders that are able to eliminate the kinase-independent functions. AbstractAn examination of the applicability and accuracy of the empirical wave inversion method in the presence of swell waves is presented. The ability of the method to invert Doppler spectra to wave directional spectra and bulk wave parameters is investigated using one-month data from a 12 MHz WERA High Frequency (HF) radar system and in-situ data from a wave buoy. Three different swell inversion models are evaluated: LPM (Lipa et al. 1981), WFG (Wang et al. 2016) and EMP, an empirical approach introduced in this study. Cardiovascular diseases are the leading cause of death worldwide. A completely autologous treatment can be achieved by using elastogenic mesenchymal stem cell (MSC)-derived smooth muscle cells (SMC) at the affected tissue site of vascular diseases such as abdominal aortic aneurysms (AAA). Thus, our work focused on evaluating the efficacy of (a) the combination of various growth factors, (b) different time periods and (c) different MSC lines to determine the treatment combination that generated SMCs that exhibited the greatest elastogenicity among the tested groups using Western blotting and flow cytometry. Background: Reductions in the use of broad-spectrum antibiotics is a cornerstone of antimicrobial stewardship. We aim to demonstrate use of the Desirability of Outcome Ranking Approach for the Management of Antimicrobial Therapy (DOOR MAT) to evaluate the treatment of Escherichia coli and Klebsiella pneumoniae bloodstream infections in patients from the Veterans Health Administration (VHA) across a decade. Methods: Using electronic records, we determined empiric and definitive antibiotic treatments, clinical characteristics, and 30-day mortality of patients with monomicrobial E. coli and K. pneumoniae bloodstream infections hospitalized in VHA medical centers from 2009 to 2018. The neonatal intensive care unit (NICU) and pediatric intensive care unit (PICU) are highly operational and highly active environments. Health care workers must react quickly to care for patients and provide life-saving measures fundamental to favorable patient outcomes. Adolescents with a history of child maltreatment experience increased risk for psychopathology that sets them on a trajectory towards a range of difficulties in adulthood. Various factors influence caregivers’ decisions to seek mental health services (MHS) that could improve developmental outcomes. The present study applied a machine learning algorithm, elastic net, to a sample of 878 adolescent-caregiver dyads from the Longitudinal Studies of Child Abuse and Neglect. Analyses simultaneously examined a large number of factors to determine their ability to discriminate between caregivers who perceived a need for MHS and those who did not, as well as caregivers who utilized …
