BACKGROUND: Anti-programmed cell death protein 1 and programmed cell death ligand 1 (PD-L1) agents are broadly used in first-line and second-line treatment across different tumor types. While immunohistochemistry-based assays are routinely used to assess PD-L1 expression, their clinical utility remains controversial due to the partial predictive value and lack of standardized cut-offs across antibody clones. Using a high throughput immunoassay, the reverse phase protein microarray (RPPA), coupled with a fluorescence-based detection system, this study compared the performance of six anti-PD-L1 antibody clones on 666 tumor samples.

Pausing of RNA polymerase II (Pol II) close to promoters is a common regulatory step in RNA synthesis, and is coordinated by a ribonucleoprotein complex scaffolded by the noncoding RNA RN7SK. The function of RN7SK-regulated gene transcription in adult tissue homoeostasis is currently unknown. Here, we deplete RN7SK during mouse and human epidermal stem cell differentiation. Unexpectedly, loss of this small nuclear RNA specifically reduces transcription of numerous cell cycle regulators leading to cell cycle exit and differentiation. Inflammation is a hallmark of aging and accelerated aging syndromes such as Hutchinson-Gilford progeria syndrome (HGPS). In this study, we present evidence of increased expression of the components of the NLRP3 inflammasome pathway in HGPS skin fibroblasts, an outcome that was associated with morphological changes of the nuclei of the cells. Lymphoblasts from HGPS patients also showed increased basal levels of NLRP3 and caspase 1. Consistent with these results, the expression of caspase 1 and Nlrp3, but not of the other inflammasome receptors was higher in the heart and liver of Zmpste24

Insect seminal fluid, the non-sperm component of the ejaculate, comprises a variegated set of molecules, including, but not limited to, lipids, proteins, carbohydrates, salts, hormones, nucleic acids, and vitamins. The identity and functional role of seminal fluid proteins (SFPs) have been widely investigated, in multiple species. However, most of the other small molecules in insect ejaculates remain uncharacterized. Metabolomics is currently adopted to deepen our understanding of complex biological processes and in the last 15years has been applied to answer different physiological questions. An open-ended hollow coaxial cable resonator probe configured to receive an aerosol sample for analysis. A metal post shorts the resonator's inner and outer conductors. A metal plate is spaced apart from an open end of the resonator by a dielectric layer that contains the received aerosol sample. Interrogator circuitry coupled to the resonator transmits an electromagnetic wave within the resonator and generates an electric field at the open end of the resonator. The interrogator circuitry is responsive to the generated electric field for determining a resonance frequency and an impedance of the resonator when the aerosol sample is present …

Grassland bird populations are experiencing major declines due to habitat degradation, pesticide use, and fire suppression throughout North America. Large-scale grassland restoration efforts to improve and provide suitable habitat are ongoing, but there is little data on productivity of birds breeding in restored habitats, nor on the impact of specific vegetation characteristics on reproductive success. Since 2005, agriculture fields at Panola Mountain State Park in central Georgia have been undergoing restoration to warm-season grasslands; however, until now, data on nest success or productivity was lacking. This article comprehensively details the culture clash that took place between Native Americans, the North Group Science Panel, and the Science Advisory Team (SAT) involved in the Marine Life Protection Act (MLPA). In 1999, the California Legislature passed the Marine Life Protection Act to create a statewide, science-driven network of marine reserves along the 1,100 miles of the California coast. The MLPA was meant to protect marine areas from overharvesting, and an initiative was formed to create a state marine reserve system advised by regional science panels. Models that describe species distributions are valuable in guiding management decisions. We compared and combined two avian datasets during the 2010 breeding season in northern California, USA. These datasets were a large-scale avian diversity survey from McGrann and Furnas (2016; 2018) and combined data from Biological Information Serving Our Nation (BISON) and Global Biodiversity Information Facility (GBIF). Our objective was to compare the utility of these two datasets, that employ separate field protocols, to model habitat use for the Black-headed Grosbeak, Hairy Woodpecker, and Yellow-rumped Warbler, three common forest birds in our study area that occupy distinctive habitat types. Like other fields, the field of psychology can trace its roots back to the pioneering efforts of early scientists, scholars, and professionals. The current commentary fills a gap in psychology's history by providing an inside look into the accomplishments of Dr. Williams. Particular attention is paid to four notable accomplishments: the founding of the Association of Black Psychologists (ABPsi), the development of the Black Intelligence Test of Cultural Homogeneity (BITCH-100), the coining of the term Ebonics, and the development of the concept of racial scripting. This paper presents a review of the historical context and the prominent debates about rural education that occurred from 1900-1940 and connects current trends in rural education to this history. Outmigration of rural youth and the decline of rural populations spurred the development of the Country Life Commission (Danbom, 1979) which sought to address this problem through rural education reform. Outmigration of rural youth continues to be a concern for rural communities, and the continued and important role of modern education in this phenomenon is discussed. In the United States, an estimated 135,000 lesbian, gay, bisexual, transgender, queer (LGBTQ+) people will be affected by a cancer diagnosis in 2020, a significantly higher statistic than equivalent measures in non-LGBTQ+ populations (American Cancer Society, 2020). As the number of sexual and gender minority (SGM) individuals affected by this disease continues to increase and intergroup disparities in care become more empirically documented, the need for multi-level advocacy in the field of LGBTQ+ psychosocial cancer care becomes apparent. This paper aims to gauge the impact of internet blockade on the research students pursuing research in Kashmir post abrogation of article 370 of Indian constitution that granted political autonomy to erstwhile state of Jammu & Kashmir. In order to collect the data survey method was employed. A close-ended questionnaire was distributed among the select group. Animals utilise a wide range of regulatory systems to control the conditions within their bodies or homeostasis. These regulatory systems control for example, body temperature, nutritional state, water balance, social interactions and fear and these systems react to environmental and endogenous stimuli so as to correct or prevent displacements from the optimal range. The behavioural and physiological responses utilised by the animal are usually proportional to the challenge to homeostasis. The objective of this study was to determine the effect of the environmental factors on the animal behaviour of grazing dairy cows, estimated through the THI (Temperature-Humidity Index) and the relationship with the daily milk production, quality of milk, consumption and the detection of zeal. The study was carried out in Azul , in the province of Buenos Aires, Argentina and was developed from December 1999 to February 2000. Dags are the accumulation of faeces on wool in the perianal (breech) region of sheep and are a common problem in sheep fed good quality ryegrass based pastures in New Zealand. Their removal, or prevention, is costly but the consequences of doing nothing are severe. There is a high correlation between dags and flystrike (development of maggots) on sheep with dags, and dags reduce fleece value, live-weight gain and reproductive performance. Information summarised here suggests a range of factors contribute to the incidence of dags: lush (high quality) feeds, high voluntary feed intakes, presence of parasites in the gastro-intestinal tract, …

The aim of this study was to gain an understanding of how the Jordanian government has responded and continues to respond to the COVID-19 pandemic. It utilizes the interpretive policy analysis approach through document analysis. The analysis showed that Jordan created social protection policies to assist people who lost their jobs or whose work was suspended due to the coronavirus. The economic policies build solidarity and facilitate the private sector’s recovery. The health care measures firmly applied included lockdown, wearing masks, and restrictions on gatherings and public events. Fragile X syndrome (FXS) is a monogenic disorder characterized by high rates of autism spectrum disorder (ASD) and anxiety. A longstanding "hyperarousal hypothesis" in FXS has argued that ANS dysfunction underpins many symptoms of FXS. However, the developmental onset and trajectory of ANS dysfunction, as well as the consequences of ANS dysfunction on later psychiatric symptoms, remain poorly understood in FXS. Insight into the emergence, trajectory, and consequences of ANS dysfunction across early development in FXS has critical implications for prevention, intervention, and optimal outcomes in both typical and atypical development. Ground-based animal models have been used extensively to understand the effects of microgravity on various physiological systems. Among them, hindlimb suspension (HLS), developed in 1979 in rats, remains the gold-standard and allows researchers to study the consequences of total unloading of the hind limbs while inducing a cephalic fluid shift. While this model has already brought valuable insights to space biology, few studies have directly compared functional decrements in the muscles of males and females during HLS.
